China Pharmacy 2016;27(25):3518-3520
OBJECTIVE:To study the effects of alprostadil on the expression of vascular endothelial growth factor (VEGF) and endothelial nitric oxide synthase (eNOS) in human pulmonary microvascular endothelial cells (HPMECs) with hypoxia-in-duced injury. METHODS:HPMECs were divided into group A(normal cultural environment),group B(3%O2),group C(3%O2+15 μg/L alprostadil)and group D(3%O2+45 μg/L alprostadil)for 24 h culture. Cell morphology was observed;MTT method was conducted to detect cell activity(recorded by absorbance);flow cytometry was adopted to detect apoptosis rate;Western blot was used to detect relative expressions of VEGF and eNOS in cells. RESULTS:Cell density in group A and D was relatively high, that in group B and C was low. Compared with group A,cell activities decreased and apoptosis rates increased in group B,C and D,expressions of VEGF and eNOS enhanced,eNOS/VEGF decreased (P<0.05). Compared with group B,cell activities in-creased in group C and D,VEGF expression decreased (P<0.05);apoptosis rate in group D decreased,eNOS expression de-creased and eNOS/VEGF increased. Compared with group C,cell apoptosis rate was decreased in group D,VEGF expression de-creased and eNOS/VEGF increased(P<0.05). CONCLUSIONS:Alprostadil maybe play effect on HPMECs with hypoxia-induced injury by up-regulating the expression level of eNOS/VEGF.

Objective To explore different approaches in administrating polyethylene glycol (PEG) solution in bowel preparation. Methods From January 2015 to June 2015, 218 patients eligible for this study were randomly divided into three groups (group A, 2 L PEG solutions 4 hours before colonoscopy; group B, 2 L PEG solutions 6 hours before colonoscopy; group C, 1 L PEG the night before and 1 L at least 4 hours before colonoscopy). The quality of bowel preparation was assessed on the basis of the Boston bowel preparation scale. A questionnaire was also completed to assess the subjective feelings of the preparation, including the overall satisfaction and discomfort with drug preparation, the feeling of abdominal distension, abdominal pain, nausea and vomiting, and the character of stool. Results In terms of intestinal cleanliness, statistical difference was shown between groups. Significant difference was found between group A and group C. However, the PDR between these three groups showed no statistical difference. No signiifcant difference was found between the three groups considering the overall satisfaction and subjective feelings of preparation. Conclusions Without affecting the subjective feelings of patients, splitting dose of PEG solution improve the quality of bowel preparation and is worthy to be recommended.

Objective:To explore the value of the quantitative fecal immunochemical test (FIT) for primary colorectal cancer screening in health check-up population. Methods:A total of 468 health check-up participants who underwent quantitative FIT and colonoscopy at the First Affiliated Hospital of Soochow University from January 2018 to December 2019 were enrolled into this study. The participants were divided into two groups-the negative group(FIT<100 μg/L)and the positive group(FIT≥100 μg/L) according to the results of the quantitative FIT. We compared the detection rate of colorectal polyps and colorectal advanced cancer between the two groups, and analyzed the sensitivity and specificity of the quantitative FIT for advanced colorectal cancer and the risk factors of colorectal polyps. Results:The positive rate of quantitative FIT in the healthy population was about 4.6% (95% CI: 4.27%-4.93%). The detection rate of colorectal polyps in the positive group was significantly higher than that of the negative group (51.53%, 95% CI: 45.81%-57.25%) vs (34.28%, 95% CI: 27.25%-41.31%, P<0.001). The sensitivity and specificity of the quantitative FIT for advanced colorectal cancer was 98.55% and 56.77%, respectively. The positive predictive value of the quantitative FIT for advanced colorectal cancer was 50.37%, while the negative predictive value was 98.88%. With the increase of FIT value, the detection rate of advanced cancer was increased (χ2 =20.08, P<0.001). The multivariate logistic regression of colorectal cancer suggested that the risk of colorectal polyps in patients with a history of diabetes was significantly higher ( OR=3.28, 95% CI: 1.45-7.41, P=0.004); the risk of colorectal polyps in men was also significantly higher ( OR=3.05, 95% CI: 1.40-6.65, P=0.005). Conclusions:Quantitative FIT is an ideal non-invasive examination for primary colorectal cancer screening for a health check-up population. Patients with negative FIT are less likely to develop colorectal cancer. Diabetes history, male, and age are independent risk factors for colorectal cancer.

Objective To investigate the expression of P-glycoprotein(P-gp), multidrug resistance-associated protein (MRP), lung resistance-related protein (LRP) and predicts treatment outcome in the elderly acute myeloid leukemia. Methods Multi-parameter flow eytometric assay was used to quantify expression of P-go, muhidrug-associated resistance protein (MRP) and lung resistance-related protein (LRP) in bone marrow viable blasts from 12 eldely acute myeloid leukemia patients at diagnosis (M2a:4, one is come from MDS, M3a:2, M5a:5, M6a:1). Correlation of the MDR protein expression with treatment outcome were analysed. Results The frequency of expression of P-gp, MRP and LRP was 58.33 %, 8.33 %, 50 %, respectively and P-gp(+)/MRP(+) 0, P-gp(+)/LRP(+) 33.33 %, MRP(+)/LRP(+) 0, P-gp(+)/MRP(+)/LRP(+)8.33 %, P-go(-)/MRP (-)/LRP(-) 33.33 %;respectively. The frequency of overexpression of P-gp or LRP alone and both of them were relatively higher. The rate of complete remission (CR) and overall survival (OS) at one year of P-gp(+) group were significantly lower than those of P-go(-) group, and LRP(+) group also were lower than LRP(-) group. Conclusion The Frequency of expression of P-gp or LRP and coexpression of both of them were higher in elderly patients with AML. The overexpression of P-gp and/or LRP was a poor prognotic factor for eldely myeloid leukemia in elderly patients.
