1.Therapeutic effect of Donepezil combined nimodipine treatment in 45 cases with vascular dementia

Objective:To observe the therapeutic effect of donepezil combined nimodipine on vascular dementia. Methods:A total of 45 patients with vascular dementia aging 54~76 years were randomly divided into donepezil +nimodipine group (n= 24)and huperzine + piracetam group (n= 21).Scores of mini-mental state examination (MMSE)and activity of daily living (ADL)were examined in two groups before and after treatment,and therapeu-tic effect was compared between two groups.Results:Compared with before treatment,after treatment,MMSE score [(15.8±6.3)scores vs.(21.6±7.4)scores]significantly rose and ADL score [(50.3±6.4)scores vs.(40.2 ±7.1)scores]significantly reduced in donepezil + nimodipine group;MMSE score [(14.6±6.9)scores vs.(18.5 ±7.2)scores]significantly rose,ADL score [(49.3±7.8)scores vs.(43.5±8.2)scores]significantly reduced in huperzine + piracetam group,P <0.01 all;but there were no significant difference between two group after treat-ment (P =0.08,0.07).Conclusion:Therapeutic effect of donepezil combined nimodipine on vascular dementia is quite to huperzine + piracetam.
2.Experimental study on construction of ultrasound microbubbles co-carrying pAd-EGFP/SDF-1α and pAd-RFP/BMP2

Objective To construct the uItrasound microbubbIes co-carrying recombinant adenovirus containing stromal cell derived factor 1 (SDF-1α) and bone morphogenetic protein 2 (BMP2),and to study the maximum efficiency of carrying adenovirus and the optimum proportion of double gene combined with ultrasound contrast agents.Methods Microbubbles were combined separately with recombinant adenovirus co-expression of enhanced green fluorescent protein and SDF-1α(pAd-EGFP/SDF-1α) as well as red fluorescence protein and BMP2 (pAd-RFP/BMP2) via biotin-streptavidin method,and the maximum efficiency of carrying DNA in microbubbles was detected.Three microbubbles with binary vectors were prepared by blending the two above-mentioned pAd at different ratio (1 ∶1,1 ∶ 2,2 ∶ 1) into the microbubbles.The microbubbles with binary vectors were evaluated though physiochemical properties,fluorescence microscope and flow cytometry to test the carrying rate of DNA in microbubbles.Results There was no significant difference in PH,average diameter and concentrations between targeted microbubbles and control group (P >0.05).The carrying efficiency of DNA increased with virus loads in microbubbles,but lowered if further increasing virus amount after reaching saturation.When the proportion of binary vectors and microbubbles was 1 ∶ 1,its efficiency of carrying SDF-1α gene and BMP-2 was approximately equal,and flow cytometry demonstrated that the positive rate of microbubbles labeled by both fluorescein isothiocyanate(FITC) and rhodamine was (65.6 ± 0.5)%.However,it was (59.0 ± 2.3)% when their proportion was the 2 ∶ 1,which was significantly lower than those when other two proportions (1 ∶ 1 and 1 ∶ 2).Under the fluorescence microscope,the targeted microbubbles were equally surrounded by bright green or red fluorescence.Conclusions Ultrasound microbubbles of double genes carrying EGFP/SDF-1a and RFP/BMP2 is made successfully via biotin-streptavidin method.The optimal proportion of combining microbubbles with double gene is 1 ∶1,which can reveal the optimum load rate and stable combination.
3.Study on fingerprint for the quality control of Banlangen Granule

AIM: To establish a fingerprint analytical method for ensuring the quality of Banlangen Granule (Isatis indigotica Fort). METHODS: According to pre column derivatization HPLC method, gradient elution with sodium acetate buffer (pH6.4) acetonitrile (95∶5) in a BDS C 18 column, was applied to detect amino acid at UV 360nm; and the coefficients of cosine of include angle and distance were employed to judge the quality similarity of the samples. RESULTS: The HPLC method is accurate and reproducible and makes almost all the peaks separated; and the two coefficients can show the similarity of each sample. CONCLUSION: The developed method is simple, accurate and operable and can efficiently control the quality of Banlangen Granule.

Corona virus disease 2019 (COVID-2019) is an acute respiratory tract infectious disease caused by infection with 2019 novel coronavirus(2019-nCoV), which is mainly characterized with pulmonary inflammatory lesions, strong infectivity and long incubation period. At present, the gold standard for diagnosis of COVID-2019 is positive nucleic acid test result. However, nucleic acid test Results: of some patients are false negative, and the number of patients need to be screened is so large. For asymptomatic patients and the large number of patients needed to be screened, imaging examinations are particularly important. The main points of CT and ultrasonic examination in auxiliary diagnosis of COVID-2019, the advantages of ultrasonic examination and protective specifications were reviewed in this article.
5.Experimental study of urokinase concentration after in vivo thrombolysis with urokinase targeting microbubbles combined with ultrasound

Objective:To prepare urokinase targeted thrombus microbubbles combined with low frequency ultrasound to dissolve thrombus in rabbit femoral artery, and to explore the mechanism of thrombolysis through the change of urokinase concentration.Methods:Twenty-four rabbits with thrombosed femoral artery were randomly divided into four treatment groups: urokinase alone (UK) group, ultrasound with non-targeted microbubble and urokinase (US+ M+ UK) group, platelet-targeted microbubble with urokinase (R+ UK) group, ultrasound with platelet-targeted microbubble and urokinase (US+ R+ UK) group. Thrombus-targeted microbubbles were injected through the ear vein and irradiated by ultrasound for 30 minutes, and the pulsed Doppler blood flow meter was used to continuously monitor the blood flow at 0 min, 10 min, 20 min, 30 min, 60 min, 90 min and 120 min after injection. At the same time, the changes in urokinase concentration were monitored, and the characteristics of blood flow and urokinase concentration changes were analyzed.Results:UK and US+ M+ UK groups failed to show recanalization at 120 minutes after treatment.In contrast, the US+ R+ UK and R+ UK groups both achieved persistent recanalization( P<0.001). Compared with the basal state. Both the R+ UK group and R+ UK+ US group showed a decrease at 60 minutes, and the difference was statistically significant( P<0.05). Conclusions:In the thrombolysis process of low-frequency ultrasound combined with a targeted contrast agent carrying urokinase, the targeting can make the local urokinase concentration of thrombus reach the highest level. At the same time, ultrasound and targeted microbubbles promote urokinase to enter the thrombus, and finally achieve the strongest thrombolytic effect.
6.Improvement of instrument for preparation of medical patch material

Objective To modify the existing preparation instrument for medical patch material to realize its scale and automatic production.Methods The instrument had its shaking table improved and a cylindrical processing system added with comprehensive analysis on raw materials and kinds of process flows.A spreading and fixation mechanism for animal materials was placed in the cylindrical processing system,which had the holes for liquid inlet and outlet respectively on its top and bottom.The cylindrical processing system was fixed to the base of the shaking table.Results The instrument enhanced preparation efficiency significantly,and had the raw material utilization rate increased by 20%,product qualification rate raised by 35%,preparation cycle reduced by 33% and total cost saved by 40%.Conclusion The instrument behaves well in adaptability to multi animals,preparation process,inter-assay difference,raw material utilization rate,product qualification rate and cost reduction,which is of great significance to promote the clinical application of medical biological patch.
7.Bridging the US and China together to conquer cancer: report of the 4th annual meeting of the US Chinese Anti-Cancer Association (USCACA).

A global collaborative effort is pivotal to conquer cancer. Themed "Emerging role of China in global clinical development of novel anti-cancer drugs", the US Chinese Anti-Cancer Association (USCACA) held its 4th annual meeting in Chicago on June 2, 2012, in conjunction with the American Society of Clinical Oncology (ASCO) annual meeting to further bridge the US and China together to outsmart cancer. Although a young organization, USCACA has made significant contributions to this goal in the 3 years since its inception through extensive collaboration with academic organizations, the pharmaceutical industry, and governmental agencies. USCACA has engaged various stakeholders in developing translational and personalized medical strategies to facilitate new anti-cancer drug development and clinical trials in China. USCACA has initiated and implemented the USCACA-National Foundation for Cancer Research (NFCR) scholarship to encourage overseas returnees to continue cancer research in China. USCACA announced the Hengrui-USCACA scholarship to fund clinical trial staff from China to conduct the observation of early oncologic clinical trials in the US. During the annual meeting, distinguished panelists and the audience discussed the following critical topics:(1) oncologic translational research and early development capabilities in China;(2) novel chemical entity development and partnership with Chinese companies; and (3) Chinese participation in global anti-cancer drug development. USCACA will continue to promote collaborations among cancer researchers and clinicians in the US and China by engaging in more frequent communications and joint efforts across fields, disciplines, and countries, diligently working together toward curing and eliminating cancers.

8.The prevalence of HIV-1 drug-resistant strains in Beijing, in 2008

Objective To study the prevalence of HIV-1 drug-resistant strains in antiretroviral therapy-naive HIV-1 infectors,and provide background information for HIV-1 drug resistance survey and clin-ical antiretroviral therapy in Beijing in 2008. Methods Referring to the guidelines for HIV drug resistance threshold survey(HIVDR-TS) of WHO, collecting 60-70 plasma samples of HIV-1 infectors who were detec-ted in 6 months and not more than 25 years,we detected HIV-1 pol genotype and genetic mutations associated with drug resistance,counted the prevalence of drug-resistant strains, and evaluated the prevalent level. Re-Sults Of 61 plasma samples answering for the standards, 50 were successfully sequenced and genotyped pol sequence. The major infection route was homosex, which accounted for 62%. B, CRF01_AE, and CRF07_ BC were major genetic subtype, which accounted for 42%, 28% and 26%, respectively. One Pl-resistant strain was found, the incidence of which was 2% (1/50). One NRTI-resistant strain was found, the inci-dence of which was 2% (1/50). No NRTI-resistant strain was found, the incidence of which was 0. The in-cidence of drug-resistant strains in the protease (PR) region was 2%, and the incidence of reverse tran-scriptase (RT) region was also 2%. Both of the prevalence were classified as low level ( <5% ). Conclu-sion PR, RT-resistant HIV-1 strains were found in drug-naive infectors, and the prevalence was low in Beijing. Current antiretrovirai therapy regiments were still feasible. Most of the AIDS patients did not need to test drug resistance before antiretroviral therapy.
9.A study of the frequencies of HLA-DQA1 alleles and their clinical application values in the donor-recipient HLA-10/10 matched hematopoietic stem cell transplantation

Objective To analyze the frequencies of HLA-DQA1 alleles and their clinical values in the donor-recipient HLA-A,-B,-C,-DRB1,-DQB1 (10/10) matched hematopoietic stem cell transplantation (HSCT).Methods This study recruited 127 patients who received allogeneic HSCT and 127 unrelated donors.High-resolution (High Res) DNA typing for HLA-DQA1 alleles were performed on the 254 subjects by using sequence specific oligonucleotide probes (SSOP) and high resolution of sequence specific primer(High Res SSP).Results The DQA1 allele genotypes of 36 pairs of donor-recipient were directly identified by using SSOP.The ambiguous DQA1 allele genotypes of the rest 91 pairs were identified by using High Res SSP.Among the 127 pairs of donor-recipient,5 pairs were HLA-DQA1 alleles mismatched,while the others were all matched.No significant differences in the distribution of HLA-DQA1 alleles were observed between the donors and the recipients.Sixteen HLA-DQA1 alleles were detected in the 127 donors,which were DQA1 * 02 ∶ 01 (19.3%),DQA1* 01 ∶ 02(19.3%),DQA1 * 03 ∶ 02/03 (17.0%),DQA1 *01∶03 (9.8%),DQA1*06∶01(9.1%),DQA1*05∶ 01(7.1%),DQA1*05∶05(5.9%),DQA1*03∶01 (4.7%),DQA1*01 ∶04(2.4%),DQA1*01∶05(2.0%),DQA1*01∶01(1.2%),DQA1*05 ∶ 03(0.8%),DQA1 *05 ∶ 08(0.8%),DQA1*04 ∶ 01(0.4%),DQA1*05 ∶ 06(0.4%) from high to low frequency.Moreover,a new allele was detected in the patients.The haplotypes' frequencies and linkage disequilibrium(LD) analysis of HLA-DQA1 and HLA-DQB1 showed that the most common haplotype was DQA1 *02 ∶ 01-DQB1 *02 ∶ 02(16.1%),followed by DQA1 *03 ∶ 02/03-DQB1 *03 ∶ 03 (11.8%)and DQA1 *01 ∶ 03-DQB1 * 06 ∶ 01 (9.1%).Stronger LD were observed between DQA1 * 02 ∶ 01 and DQB1*02 ∶ 02,DQA1 *03 ∶ 02 and DQB1*03 ∶ 03,DQA1 *01 ∶ 03 and DQB1*06 ∶ 01,HLA-DQA1*06∶01 andDQB1*03 ∶ 01,DQA1*05 ∶ 01 and DQB1*02 ∶ 02(P<0.001).Conclusion There was strong linkage disequilibrium between HLA-DQA1 and HLA-DQB1 genes.The polymorphism of HLA-DQA1 gene was less than that of HLA-DQB1 gene.No more guidance was provided to donor selection in unrelated donor-recipient HLA matched HSCT by adding HLA-DQA1 genotyping,but it might have clinical application values in HSCT with HLA Ⅱ locus mismatched donor and recipient.
10.Study on visual display terminal mental fatigue status based on physiological information.

A visual display terminal (VDT) mental fatigue task was designed to search for the objective indicator to diagnose and evaluate the VDT mental fatigue status. The signals of temperature and electrocardiograph (ECG) are collected from the subjects. The temperature, heart rate (HR) and heart rate variability (HRV) are extracted as the objective parameters after analyzing the temperature signals in time domain and the ECG signals in time domain and frequency domain. Compared with mental fatigue pre-experiment, the temperature increased significantly (P<0.001), and the RR-PNN50 increased obviously (P<0.05), while the S wavelet of ECG signals decreased obviously (P<0.05) after mental fatigue experiment. Compared with the end of mental fatigue experiment, the temperature increased, the changes of RR-PNN50 were not obvious, the S wavelet increased significantly (P<0.01), and the mental fatigue was somewhat remittance after natural recovery. The results showed that the objective indicator to evaluate the VDT mental fatigue status would be hopeful to be found out by analyzing the signals of temperature and ECG.
