0.05).The cytological false negative rate was 23.58% for LSIL,17.57% for high squamous intraepithelial lesion(HSIL),0.00% for squamous cell cancer(SCC),and 50.00% for adeno-carcinoma(AC);the false positive rate was 14.29% for LSIL and 3.85% for HSIL.Conclusion Liquid-based cytology test is an effective method for the screening of cervical cancer and precancerous lesion.The high-quality cervical cell specimen is important to ensure the diagnosis accuracy.

3.Antigen screening and identification of Helicobacter pylori outer membrane proteins from murine infection model

Objective:To screen antigen of Helicobacter pylori outer membrane proteins by murine infection model.Methods:Parallel two-dimensional gel electrophoresis (2D) of outer membrane proteins extracted from Helicobacter pylori strain SS1 was performed.Western blot of a duplicate 2D gel hybridized with serum from H.pylori-infected murine was employed.Immunogenic H.pylori proteins identified in this way were digested in gel by trypsin and the mass of generated peptides were measured by matrix assisted laser desorption ionization time of flight mass spectrometry (MALDI-TOF-MS).The data obtained from peptide mass finger-printing (PMF) were searched using the internet available database.Results:32 proteins were identified and they are in good agreement with typical protective antigens which reacted with serum from H.pylori-infected patients.Conclusion:The results suggest that murine model of H.pylori may be valid to screen antigens for human vaccination and the proteins identified in this paper are valuable for the selection of H.pylori protective antigens as well.

Objective:To detection antibody against heat-stable enterotoxin by fusion protein.Methods:Mutant heat-stable enterotoxin precursor gene was ligated in vector pGEX-4T-2 to inductively express as a fusion protein GST/proST_m with glutathione S-transferase(GST).To investigate the antigenic action,serum and fecal antibodies against heat-stable enterotoxin was detected with this fusion protein.Results:The fusion protein was a about 32 kD protein.All the samples contain the antibody against ST.Conclusion:Such strategy was a promising method to detect antibody against heat-stable enterotoxin.
5.Relations between flexor hallucis longus changes and pes planus in patients with rheumatoid arthritis

Objective To assess the correlation between tenosynovitis or rupture of the flexor hallucis longus (FHL) and pes planus in patients with rheumatoid arthritis (RA).Methods The data of 47 RA patients (97 feet) with painful feet was retrospectively analyzed which was collected From February 2008 to June 2011.In accordance with the results of the FHL changes got by ultrasonography,the patients were divided into 3 groups.Group A consisted of those with normal FHL (27 feet).Group B consisted of those with FHL tenosynovitis (40 feet),and Group C eonsisted of those with FHL rupture (27 feet).Load bearing lateral radiographs were taken for all the patients.The correlation study was carried out based on the pes planus-related parameters,such as the measurement of heel pitch angle (HPA) and tarsal and 1st metatarsal angle (TM1).Factors like patients' age and course of disease were also taken into the consideration during the study.Results The mean age of those in group A was 49.9±9.2 years vs.56.2±9.2 years in group B and 54.7±8.0 years in group C.The mean duration was 4.7±2.6 years in Group A vs.16.2±7.4 years in Group B 5°±3.6° in Group C.The mean TM1 was 2.5°±2.2° in Group A vs.6.5°±3.2° in Group B and 11.2°±4.9° in Group C.Significant statistics difference were found among the three groups in these parameters.HPA< 20° was found among 6/27 feet in group A,31/40 in group B and 27/27 in group C.TM1 >4° was found among 5/27 feet in group A,30/40 in group B and 27/27 in group C.Conclusion FHL changes were common in RA patients and its severitv had positive correlation to pes planus.These changes were affected by age and duration,and could be evaluated effectively by ultrasound.
6.Study on the diagnosis of pulmonary aspergillosis in patients with non-granulocyte deficiency by GM detection of bronchial alveolar lavage

Objective To evaluate the diagnostic effect of galactomannan (GM) on bronchial alveolar lavage in patients with non-granulocyte deficiency.Methods 64 patients with suspected invasive pulmonary aspergill were enrolled in our hospital from May 2016 to May 2017.According to the relevant diagnostic criteria,they were divided into the diagnosis group (n =3),the clinical diagnosis group (n =20),the proposed group (n =14) and non-fungal infection group (n =27).All subjects underwent bronchial alveolar lavage and GM testing.Results When the GM value of bronchoalveolar lavage fluid and serum GM value increased from 0.5 to 1,the sensitivity decreased,while the specificity and positive predictive value increased.The sensitivity,positive predictive value and negative predictive value of GM detection method for bronchial alveolar lavage fluid were significantly higher than that of serum GM (P < 0.05) at the same GM boundary value,and the specificity was significantly lower than that of serum GM (P < 0.05).When the GM boundary value of bronchial alveolar lavage fluid was ≥ 0.86,the diagnostic efficiency of invasive pulmonary aspergillosis was the highest,the sensitivity and specificity reached 80.26%,75.15%,and the area under the curve was 0.945 (95% CI:0.782-0.986).Conclusions GM detection of bronchial alveolar lavage fluid is more helpful for the diagnosis of invasive pulmonary aspergillosis in patients with non-granulocyte deficiency than serum GM.The optimal threshold is 0.86.
7.Expression of snake venom thrombin-like enzyme calobin in Pichia pastoris.

Thrombin-like enzymes (TLEs) are studied widely because of their therapeutic potential in myocardial infarction and thrombotic diseases. We synthesized the DNA fragment encoding thrombin-like enzyme calobin from Agkistrodon caliginosus (Korean Viper) venom by fusion PCR and expressed it in Pichia pastoris. After induction by 0.5% methanol for 48 h, the expression level of recombinant calobin reached 3.5 g/L in medium. The recombinant calobin was purified by Q-Sepharose Fast Flow ion-exchange chromatography and Sephacryl-S-100 gel filtration chromatography. Purified sample had a molecular weight of 32 kD shown in SDS-PAGE. It hydrolyzed fibrinogen and formed a light white hydrolysis circle in fibrinogen plate. SDS-PAGE analysis showed that recombinant calobin cleaved Aalpha-chain of fibrinogen specifically, and produced an appropriately 40 kD new band. However, we failed to find its fibrin-clot formation activity.

8. Comparison of the diagnostic efficacy of arthroscopy and magnetic resonance on acute injury of the lateral ankle liga-ment complex

Objective:
To compare the diagnostic consistency and efficacy of arthroscopy and MRI on the severity of acute lateral ligament complex (LLC) injury, and to find an effective method to diagnose acute LLC injury accurately in early stage.
Methods:
Data of 37 patients (27 males, 10 females), with acute lateral ligament complex injury who were treated from February 2015 to December 2016 were retrospectively analyzed. The average age was 28.6±6.2 years (range, 18-42 years) and duration from injury to operation was 4.6±3.3 days (range, 0.3-8 days). The results of MRI and ankle arthroscopy for detecting LLC injury were compared, and the consistency between the two examination results was analyzed.
Results:
Different consistencies were found between the two examination methods according to different parts of LCC injuries. The consistency of injuries (partial tear,complete tear) at the starting point of anterior talofibular ligament and complete tear at the starting point of calcaneofibularliga-ment was ordinary (kappa=0.641, 0.624, 0.680), while the consistency of damage in other parts of LLC was poor (kappa< 0.4). Compared with arthroscopy, MRI has a diagnostic accuracy of 62% (23/37) for partial tear of anterior talofibular ligament, and 62% (23/37) for complete tear. And that of the calcaneofibularligament complete tear was 78% (29/37).
Conclusion
MRI had low capability in detecting the severity of acute LLC injury accurately and the diagnosing efficacy was insufficient, while ar-throscopic exploration had a high diagnostic value for acute ankle joint injuries.
9.Relationship between rheumatoid arthritis flatfoot and flexor pollicis longus tendon lesion

Objective To study the relationship between the rheumatoid arthritis (RA) flatfoot and flexor pollicis longus tendon (FHL) lesion.Methods A total of 120 rheumatoid arthritis flatfoot patients (120 feet) was admitted to our hospital,and were examined by uhrasonography.Foot flexor hallucis longus tendon lesions were analyzed.Theses patients were divided into intact tendon group (group A) with 48 cases,tendon inflammation group (group B) with 45 cases,and tendon rupture group (C group) with 27 cases according to the ultrasound flexor hallucis longus tendon lesions degree.Foot bearing radiographs of all patients were measured,and hallux valgus angle between the first and second metatarsal bones were measured,and the relationship between the flexor pollicis longus tendon lesion degree and hallux valgus angle were analyzed.Results There were 48 cases (40%)with normal FHL,45 cases(37.5%)with FHlL inflammation,and 21 cases(22.5%) with FHL fractures.It revealed that HVA was positively correlated with IMA in group A and B (P < 0.05).But there was no correlation between HVA and IMA in group C (P > 0.05).Conclusion FHL is a common RA flatfoot disease,but FHL is not the initial cause of RA hallux valgus,but is positively correlated with the degree of hallux valgus.
