Objective To explore the reversing mechanism of multidrug resistance of ciclosporin(CsA) on K562/A02 cell line,attenuating DNA damage repair through H2AX suppression.Methods K563 and K562/A02 respectively co-cultured with adriamycin(ADM) of different concentrations and CsA.MTT assay was employed to determine the inhibitory concentration of 50 percent(IC50),the resistance times and the reversal times.The K562/A02 cells treated with CsA,and reverse transcriptase(RT)-PCR technique was used to examine the mdr1 and H2AX mRNA level.Flow cytometry was used to measure P-glycoprotein(P-gp) and H2AX expression.Neutral comet assay was used to detect the level of DNA double strands break(DSBs) of the K562/A02 treated with ?H2AX antibody.Results 2?10-3g/L CsA could increase the sensitivity of K562/A02 to ADM,and the reversal times was 5.28.Mdr1 mRNA level and H2AX mRNA level were decreased when treated with CsA(P

Objective To observe the effect of low arsenic sub-chronic exposure on blood routine test index in rabbits to pave a way for screening early injury of the low arsenic exposure. Methods Twelve healthy male rabbits were randomly divided into four groups. They were administrated with As at the concentration of 0(control), 10, 50 and 250 μg/L in the drinking water. Blood samples were collected from the vein of the ear edge in 8 weeks, and blood test routine including leukocyte (WBC), lymphocyte (LYM), lymphocyte percentage (LYM%), neutrophil (GRA), neutrophil percentage (GRA%), monocyte (MON), monocyte percentage (MON%), red blood cell (RBC), hemoglobin (HGB), hematocrit (HCT), mean corpuscular volume (MCV), mean corpuscular hemoglobin content (MCH), mean corpuscular hemoglobin concentration(MCHC), RDW, platelets(PLT), mean platelet volume(MPV), platelet hematocrit(PCT) and platelet distribution width (PDW), were detected by the ABX-60 hemocyte analyzer. Results Compared with the control group, the WBC, GRA and GRA% increased in 0, 50 and 250 μg/L groups, but there was no significance(P > 0.05). PLT and MPV had a statistical significance in 4 groups(F = 4.07,4.20, all P < 0.05). Compared with the control group[(292.00±16.97)×109/L, (7.10±0.99)fL], PLT decreased in the 250 μg/L group [(221.33±22.50)×109/L] and MPV decreased in the 50μg/L group [(5.57±0.46)fL] significantly (P < 0.05). The other index didn't change obviously. Conclusions Sub-chronic low level arsenic exposure may lead to the change in the blood system. The blood routine test may be considered for early injury of the arsenic poisoning.

OBJECTIVETo investigate the association of Coxsackie B virus (CBV) with habitual abortion.

METHODSCBV IgM antibody, viral RNA and virions were detected in 86 women with habitual abortion and 40 with induced abortion by means of enzyme-linked immunosorbent assay (ELISA), RT-PCR and virus isolation, respectively.

RESULTSThe positivity rate of CBV IgM were 87.2% and 35% in the two groups, respectively, and the detection rate of the viral RNA was 53.5% and 17.5% in blood lymphocytes, and 59.3% and 17.5% in the placentas. The virions were found in the placentas in 41.9% and 6.9% of the women, respectively. The positivity rates of CBV IgM, viral DNA and virions showed significant difference between the two groups (P<0. 01).

CONCLUSIONCBV might be one of the causes responsible for habitual abortion.

OBJECTIVETo investigate the effect of an oral preparation of Alternathera philoxeroides Griseb (APG) against respiratory syncytical virus (RSV) in mice.

METHODSAPG preparation was administered orally in RSV-infected mice at different daily doses (2.5, 4.5 and 6.5 g/kg) to observe the therapeutic effect of the preparation.

RESULTSDistinct differences were observed between the death rate of the mice treated with APG at daily dose of 4.5 and 6.5 g/kg and that of the untreated mice with infection. After AGP treatment of the mice at 6.5 g/kg, the detection rate of the virus was 31.3% in the blood and 37.5% in the lung tissue, significantly lower than that in the untreated mice. The virus detection rate was 43.8% in the lung tissues of mice treated with APG at 4.5 g/kg, also significantly lower than that in the untreated control. APG treatment at the 3 doses resulted in different lung indices from that of the control.

CONCLUSIONAPG may be effective for treatment of RSV infection.

Objective To evaluate the prevalence and prognosis of hepatitis E virus ( HEV) su- perinfection in patients with chronic hepatitis B ( CHB) ． Methods Electronic databases of Pubmed， CNKI，and Wanfang were searched，and references of the relevant articles for literatures associated with HEV superinfection in CHB patients in China published during 2008－2019 were screened． Studies which met the inclusion and exclusion criterias were selected and related data were pooled for analysis． The Ｒ 3. 5. 2 and Ｒevman 5. 3 were used to measure heterogeneity with the I2 statistic and pooled incidence and odds ratio ( OＲ) ． Ｒesults 28 literatures were eligible for inclusion． The results showed the rate of HEV superinfection in hospitalized CHB ( CHB-HEV) patients was 13. 6% ( 95%CI: 12. 5% －14. 9%) ． The pooled incidence of liver failure in CHB-HEV patients was 34. 7% ( 95%CI: 29. 6%－40. 1%) ，which was higher than CHB patients ( OＲ= 10. 9，95%CI: 6. 8－17. 4) ，CHB patients superinfected with hepatitis A virus ( CHB-HAV) ( OＲ= 3. 8，95%CI: 2. 4－6. 2) ，and hepatitis E patients ( OＲ= 5. 1，95%CI: 3. 8－ 6. 8) ． The pooled mortality of CHB-HEV patients was 13. 8% ( 95%CI: 10. 3－17. 8) ，which was higher than CHB patients ( OＲ = 8. 5，95%CI: 4. 0－18. 3) ，CHB-HAV patients ( OＲ = 14. 4，95%CI: 4. 8－ 42. 9) ，and hepatitis E patients ( OＲ= 4. 4，95%CI: 3. 0－6. 4) ． Conclusion In China，HEV sueperinfection on chronic hepatitis B patients are common，and is an important cause of liver failure and death．

Objective To study the risk factors related to spastic type of cerebral palsy through a screening program.Methods With a 1 ∶ 2 matched case-control study design,145 cases were selected from the Cerebral Palsy Treatment and Management Center in Heilongjiang Province from Dec.,2007 through May 2009.Another 290 cases of children as control group were chosen from the First Affiliated Hospital of Jiamusi University,in the same period.Controls were excluded the central nervous system diseases,matching with cases at the age differences no more than 3 months,with same sex and ethnicity.Questionnaire survey was used to access relevant information,while logistic regression analysis was used to screen the risk factors.Results Data from Single factor logistic regression analysis notified 39 items as statistically significant factors.Through multivariate logistic regression analysis,11 factors in all the cerebral palsy cases,and incidence of cerebral palsy risk factors related to low or high birth weight were discovered as follows:mother with college degree,newborns with high muscle tone,skin color as purple,taking antipyretic analgesics drugs during pregnancy,experience significant events during pregnancy,situation of environment pollution around the house,having intracranial hemorrhage and birth asphyxia,fetal distress during pregnancy or neonatal stage etc.Conclusion Pre/Nconatal factors were closely related to cerebral palsy,suggesting that health care knowledge be popularized and related management be strengthened,to reduce the cerebral palsy incidence in the newborns.

BACKGROUND & OBJECTIVE:Glutathione is involved in cellular protection against radiation damage and drug detoxification.This study was to investigate the circadian variation of plasma cortisol and whole blood reduced glutathione(GSH)levels in nasopharyngeal carcinoma (NPC)patients to provide references for chronotherapy for NPC.METHODS:A total of 13 NPC patients and 14 healthy volunteers were involved.Peripheral venous blood was sampled every 4 h during one 24hour period starting at 12:00 am.The plasma cortisol concentration was determined by radioimmunoassay;the GSH concentration was determined by high performance liquid chromatography.RESUL-TS:Plasma cortisol levels of both groups displayed clear and similar circadian rhIythms.The cortisot level peaked in both groups in the morning and was the lowest at mid-night.In both groups,GSH concentrations showed significant differences according to sampling time(ANOVA for Repeated Measures,F=5.18,P=0.02).By cosinor analysis.the circadian variation of the GSH Ievel in NPC group was marginally statistically significant(Cosinor analysis,P=0.06)with the acrophase appeared at 05:02:the GSH Ievel in control group displayed an obvious circadian rhythm and the acrophase appeared at 07:44+01:56(P＜0.01).The rhythm-adjust mean value of glutathione was(19.60+1.11)nmol/mg protein in NPC group and(8.95,±0.46)nmol/mg protein in control group.CONCLUSIONS:The circadian rhythm of the plasma cortisol level is maintained in NPC patients,even in some patients at advanced stages.In NPC patients,GSH shows a trend of circadian variation which is similar to that in healthy controls.These results could be used to select special schedules for chrono-radiotherapy and chrono-chemotherapy for NPC patients.

OBJECTIVE: To study genetic polymorphism of 7 Y-specific short tandem repeats (STR) and assess their usefulness in forensic casework. METHODS: 7 Y-STR have been amplified in two multiplex reactions, (Multiplex I:DYS391, GATA-A4, GATA-A10 and GATA-H4. Multiplex II:DYS439,DYS437 and DYS434). PCR products were separated by polyacrylamide gels electrophoresis followed by silver stain. RESULTS: When 372 unrelated individuals from the Han population in Guangdong were detected by those system, DYS391, GATA-A4, GATA-A10, GATA-H4,DYS439,DYS437 and DYS434 showed 5,7,6,5,6,4,4 alleles, respectively. A total of 254 different haplotypes were identified, of which 201 (79.13%) were found in single individuals. The overall haplotypes diversity reached 0.9960. CONCLUSION The 7 Y-STR loci are highly genetic polymorphism and they will be very powerful for establishing Y-STR database, understanding human origin, paternity testing and personal identification.

Objective To compare the diagnostic performance of prospective electrocardiogram (ECG)-triggered computed tomography coronary angiography (CTCA) versus retrospective ECG-gated CTCA.Methods Patients with suspected coronary artery disease divided into two groups which underwent 64-slice CTCA with prospective ECG-triggered or retrospective ECG-gated scanning (n=100 each,HR≤65 bpm).Multi-planar reconstruction (MPR),curved-planar reconstruction (CPR),maximum intensity projection (MIP) and volume rendering (VR) were made to demonstrate the coronary arteries.The image quality was defined as excellent,good and poor by motion and stair-step artifacts.Individual radiation exposure dose was estimated from the dose-length product.Results The mean effective radiation dose of prospective ECG-triggered CTCA[(2.81±0.48)mSv] was significantly lower than that of retrospective ECG-gated CTCA[(10.16±1.09)mSv,P＜0.01].Segments of diagnostic image quality (95.2%,1165/1224) and non-diagnostic coronary segments (4.8%,59/1224) in prospective ECG-triggered group were similar as those of retrospective ECG-gated group (94.1%,1186/1261 and 5.9%,75/1261,all P＞0.05).Conclusion Though the radiation exposure dose required is significantly lower,the diagnostic performance of prospective ECG-triggered 64 slice CTCA is comparable with that of retrospective ECG-gated 64 slice CTCA on patients with stable heart rates up to 65 bpm.
