1.lmprovement effect and anti-oxidative stress mechanism of rosiglitazone against cisplatin-induced nephropathy in rats

OBJECTlVE To investigate the improvement effect of rosigIitazone(ROm)against cis-pIatin(DDP)-induced nephropathy in rats and its mechanim of resistance to oxidative stress. METHODS Invitro:the survivaI rate of HEK293 ceIIs was measured with mTT assay incubated with DDP 0.4 -100 μmoI·L-1 aIone or with DDP 25 μmoI·L-1 and ROm 0.01-1000 μmoI·L-1 for 48 h. Then the content of maIondiaIdehyde(mDA)and the activity of gIutathione(GSH)in HEK293 ceIIs co-incubated with DDP 25 μmoI·L-1 and ROm 100 μmoI·L-1 were assayed using chromatometry. In vivo:sixty maIe SD rats were randomIy divided into normaI controI group,modeI group(DDP 5 mg·kg-1 )and modeI+ROm 5,10 and 20 mg·kg-1 groups. The modeI of DDP-induced nephropathy was estabIished by taiI vein injection of DDP 5 mg·kg-1 ,once a week,for 3 times. After the first injection,the rats in modeI+ROm 5,10 and 20 mg·kg-1 groups were ig given ROm 5,10 and 20 mg·kg-1 ,once a week,for 8 weeks. The bIood urea nitrogen(BUN)and creatinine(Cr)were assessed with an automatic biochemicaI anaIyzer. Nitric oxide(NO)and mDA content,GSH and superoxide dismutase( SOD)activities in the kidney tissue were determined using chromatometry and histopathoIogicaI changes in renaI tissues were detected by hematoxyIin-eosin staining. RESULTS In vitro:IC50 of DDP to HEK293 ceII survivaI for 48 h was 21.0 μmoI·L-1 . Co-incubation with ROm 1-1000 μmoI·L-1 and DDP 25 μmoI·L-1 for 48 h couId signifi-cantIy increase the totaI ceII survivaI rate compared with DDP 25 μmoI·L-1 group(P﹤0.05,P﹤0.01), whiIe co-incubation with ROm 100 μmoI·L-1 and DDP 25 μmoI·L-1 for 48 h couId significantIy decrease the IeveI of mDA(P﹤0.01)and increase GSH activity(P﹤0.01),respectiveIy. In vivo:compared with normaI group,serum BUN and Cr content in modeI group were significantIy increased(P﹤0.01),renaI tissue GSH and SOD activities decreased(P﹤0.01),renaI tissue mDA content increased(P﹤0.01), and renaI tissue NO IeveI and NOS activity aIso decreased significantIy(P﹤0.05). Compared with modeI group,serum BUN content in modeI+ROm 5,10 and 20 mg·kg-1 decreased from(17.0±1.3)mmoI·L-1 to 14.0±4.1,11.2±1.8 and(6.1±1.0)mmoI·L-1(P﹤0.01),serum Cr content in modeI+ROm 10 and 20 mg·kg-1 decreased from(124.6±39.8)mmoI·L-1 to 49.0±5.2 and(47.1±2.9)mmoI·L-1(P﹤0.01), renaI tissue GSH and SOD activities in modeI+ROm 10 and 20 mg·kg-1 group increased significantIy (P﹤0.05,P ﹤ 0. 01),mDA IeveI decreased(P﹤0.05,P ﹤ 0. 01),NO IeveI and NOS activity aIso increased(P﹤0.05,P﹤0.01),and renaI pathoIogicaI tissue damage couId be improved. CONCLUSlON ROm pIays a protective roIe in rat kidney damage caused by DDP,which may be reIated to improving oxidative stress resistance of the kidney.
2.Progress in molecularly targeted anti-tumor drugs derived from natural products or their derivatives

Conventional chemotherapy drugs, molecularly targeted drugs, and immune checkpoint inhibitors are the major constituents of anti-tumor drugs in clinical settings at present. Molecularly targeted drugs specifically target the key proteins, genes, or signal transduction pathways in tumor cells which are essential for initiation and development of tumor, resulting in selective activity to induce cell death or growth inhibition. Molecularly targeted drugs have emerged as the mainstream in the research and development of anti-tumor drugs due to its high selectivity and low toxicity. Natural products refer to the chemical constituents or metabolites originated animals, plants, or microorganisms, which have been recognized as one of the important sources of drug discovery with abundant resources and diversified structures. At present, a number of molecularly targeted anti-tumor drugs derived from natural products or their derivatives have been approved for cancer therapy or in clinical trials. This review will summarize the molecularly targeted anti-tumor drugs derived from natural products or their derivatives according to their different cellular targets, and also outline the molecular mechanism, progress, and perspectives of these drugs.
3.Echocardiography Combined with Flash CT in Diagnosis of Partial Anomalous Pulmonary Venous Connection

PurposeTo investigate clinical significance of echocardiography combined with Flash CT in diagnosis of partial anomalous pulmonary venous connection (PAPVC). Materials and Methods Twenty-eight cases of PAPVC were recruited in the study. All the subjects underwent both preoperative echocardiography and coarse pitch Flash CT scanning. Imaging findings were compared with the results of surgery, including the type of PAPVC, size of atrial septal defect (ASD), and other combined malformations. Results Among 28 patients, 16 cases and 5 suspicious cases (4 cases of PAPVC and 1 case of pulmonary vein embolism) were diagnosed by echocardiography. Six cases were missed, and 1 case was misdiagnosed. CT misdiagnosed 1 case. The accurate diagnosis rate of echocardiography (71.4%, 20/28) was lower than that of Flash CT (96.4%, 27/28) (P<0.05). There were 44 combined malformations in 28 cases, 43 lesions were diagnosed by echocardiography, and 34 lesions were diagnosed by Flash CT. The accurate diagnosis rate of echocardiography (97.7%, 43/44) was higher than that of Flash CT (77.3%, 34/44) (P<0.05). The average DLP was (15.5±3.2) mGy·cm and average effective dose (ED) was (0.354±0.058) mSv.Conclusion Flash CT has a higher diagnosis accuracy of PAPVC but a lower diagnostic accuracy of combined malformations when compared with echocardiography. Moreover, echocardiography can make comprehensive evaluation in valve disease, pulmonary artery pressure and cardiac function. Flash CT combined with echocardiography may improve preoperative diagnosis.

We carried out the multiplicity teaching by remodeling experiment process, designing integral test, and resolving practical issue and setting up comprehensive trail. It was proved by fact that the multiplicity teaching increased the study interesting of students, inspired their activity, initiative and creativity.
5.Polymorphism of Escherichia coli Isolated from the Fecal

In this study, three methods for identification of E.coli were compared. The conventional method was employed to select and identify the suspicious E.coli isolates from a fecal sample. PCR based ARDRA analysis was then carried out to distinguish these E.coli isolates, E.coli MG1655 and other bacterial species. All the potential E.coli isolates and E.coli MG1655 had the identical ARDRA banding pattern while the other bacterial species showed the different patterns.The result indicated that the ARDRA analysis was consistent with the traditional method for identification of E.coli and could be the practical method for distinguishing E.coli from other intestinal bacterial species. The ERIC-PCR analysis provided abundant polymorphism between different E.coli isolates, and might be a powerful approach for elucidating the genetic diversity among isolates of the same species.
6.Amniotic membrane transplantation for corneal perforation associated with ulceration

Objective To investigate the efficacy,safety and localization of multilayer amniotic membrane transplantation(AMT) for corneal perforation associated with ulceration.Design A retrospective clinical case series.Participants Nine patients(9 eyes)with corneal perforation secondary to ulceration were enrolled into this study.These were little response to medicine,including bacterial keratitis(4 eyes ),fungal keratitis(2 eyes),fungal and bacterial mixed keratitis( 1 eye),virus keratitis( 1 eye),and Mooren's ulcer ( 1 eye).Size of perforation was 0.5~3.0 mm in diameter.Methods The AMT was applied to tamp the perforation,fill the ulcer and cover the surface of ulcer.After surgery,the medicine was continued to be used to treat the original corneal ulcer.The follow-up ranged from 6~20 months.In the suffering eyes,reformation of anterior chamber,healing of ulcer,complications and recurrence of ulceration were observed.Main Outcome Measures Of the postoperative eyes,reformation of anterior chamber,healing of ulcer,complications and recurrence of ulceration.Results The anterior chamber reformed at the first postoperative day in 9 eyes,and kept in normal depth in the follow-up time.At postoperative 2 months,the ulcer healed with sear and a smooth surface.Corneal thickness of the ulcer area recovered almost to normal.During follow up,no recurrence of ulceration or severe complications was noted.Conclusions Multilayer AMT is a safe and effective method for the management of small corneal perforation associated with ulceration,but the ulcer healed with scar.(Ophthalmol CHN,2008,17:101-103)
7.Biological characteristics and hematopoietic support of bone marrow-derived mesenchymal stem cells in patients with systemic lupus erythematosus

Objective To explore the biological characteristics and karyotype of bone marrow-derived mesenchymal stem cells(MSCs)in patients with systemic lupus erythematosus(SLE)and hematopoietic sup- port of MSCs.Methods MSCs were isolated from bone marrow of 11 SLE patients and 6 healthy controls by density centrifugation and adhesive culture in vitro.The surface markers were detected by flow cytometry (FCM).The morphological changes of MSCs were observed in primary and passage cultures.The growth curves were assayed.The karyotype of MSCs was detected by blocking cellular mitosis with colchicines.The MSCs from SLE patients and healthy controls were infused to ICR mice after high-dose chemotherapy.The changes of peripheral blood counts of the mice were recorded.Results Approximately(6~9)?10~9 MSCs from SLE were obtained after 5 passages and their growth was slower than normal controls(P<0.01).Both groups were positive for CD29,CD44 and CD105,and negative for CD14,CD34,CD45 and HLA-DR.MSCs from SLE had a normal karyotype.MSCs infusions of the two groups were accompanied by no adverse event and the recovery of white blood cell,hemoglobin and platelet count was quicker when compared with the controls(P<0.05).Conclusion MSCs from SLE have demonstrated abnormalities in expansion in vitro.MSCs from SLE have a normal karyotype.Ex vivo MSCs infusion from SLE patients can support hematopoiesis as normal MSCs.
8.Choroidal Neovascularization Model of Rats Induced by Diode Laser

Objective To evaluate the feasibility of inducing choroidal neovascularization(CNV) in Brown Norway(BN) rats by diode laser with wavelength of 810 nm. Methods Thirty-five BN rats were divided into(7 groups.)In each groups,one rat served as control,while the other four recieved retinal photocoagulation by diode(laser) in two eyes,at a spot size of 75 ?m with a duration of 100 ms and power of 140 mW.On 1,3,7,14,21,28 and 56 days after photocoagulation,the formation and nature process of CNV were observed by fundus fluorescein(angiography)(FFA),indocyaninegreen angiography(ICGA) and light microscopy(LM). Results FFA,ICGA and LM demonstrated that CNV began to form on the seventh day after photocoagulation,increased on the fourteenth day,reached the peak on the twenty-first day and kept stable until the fifty-sixth day. Conclusion Diode Laser can successfully induced CNV in BN rats,and it is an ideal animal model for further study.
9.Multilayer amniotic membrane transplantation for the management of corneal perforation associated with ulceration

AIM: To investigate the efficacy, safety and localization of multilayer amniotic membrane transplantation (AMT) for the management of corneal perforation associated with ulceration.METHODS: Six eyes (6 patients) were treated with multilayer AMT for corneal perforation associated with ulceration. The perforation size was 0.5-2mm in diameter. The amniotic membrane (AM) was applied to tamp the perforation, fill the ulcer and cover the surface of ulcer. The follow-up ranged from 5-19 months.RESULTS: The anterior chamber reformed at the first postoperative day in all patients, and kept in normal depth in the follow- up time. The epithelialization of the AM grafts was observed in about 3 weeks after surgery. During the first postoperative month, the corneal thickness at the perforation site gradually increased, and the stromal inflammatory cell invasions surrounding the corneal ulcer were gradually subsided.Two months postoperatively, the ulcer healed with scar with part neovascutarization; the corneal thickness of the ulcer area was stable and recovered almost normal. The vision was not improved in all patients after operation. During-follow up,no severe complications or recurrence of ulceration was noted.CONCLUSION: Multilayer AMT is an effective and safe method for the management of small corneal perforation associated with ulceration, but the ulcer healed with scar.
10.Amniotic membrane transplantation for limbal pathological changes of vernal keratoconjunctivitis

AIM: To evaluate the efficacy and safety of annular bubble conjunctival excision combining with amniotic membrane transplantation(AMT) for vernal keratoconjunctivitis(VKC) with gelatinous swellings(papillae) at the limbus.METHODS: Twenty eyes(10 patients) with VKC, were characterized by gelatinous swellings(papillae) at limbus and were underwent annular bubble conjunctival excision associated with AMT. The follow up ranged 5-21months.RESULTS: After operation, the symptoms were subsided remarkably: itching,discharge and foreign body sensation were almost disappear; photophobia and tearing were obvious in the first week after surgery, but were gradually alleviated, and almost disappear after one month of postoperative.Yellow-brown or filthy red gelatinous protuberances at limbus were cleaned away by operation and did not recur during the follow up. Corneal superficial punctuate keratitis(11 eyes) and corneal ulcers(3 eyes) were healed during the first week after surgery and did not recur. Conjunctival congestion was gradually lessened after surgery, which could not be noted 1months after surgery in 19 eyes, and the conjunctiva around the limbus, where AMT was performed, was whiter than the normal. But conjunctival congestion in 1 eye lasted for about 5mon accompanying with a little of itch, and this eye was treated with eyedrops with improvement but recurrence. No severe complications had been seen during the follow up.CONCLUSION: Annular bubble conjunctival excision combining with amniotic membrane transplantation is an effective and safe method for the treatment of vernal keratoconjunctivitis with gelatinous swellings (papillae) at limbus. But the candidates must be chosen with discretion.
