1.mazEF gene searching and molecular evolution in genome sequenced strains of Escherichia coli and Shigella

Objective To perform molecular evolution analysis of mazEF gene in genome sequenced strains of Escherichia coli and Shigella. Methods Pathway Tools (version 13.5) provided by BioCyc was used to search encoding gene mazF of toxin MazF ( chpA) and encoding gene mazE of antitoxin MazE (chpR) in genome sequenced 10 strains of Escherichia coli, 6 strains of Shigella and 1 strain of unkown Enterobacteria. Then Minimum Evolution method in MEGA4. 1 software was used to analyze molecular evolution of MazE and MazF. Results Encoding gene mazF of toxin MazF was found in 12 strains, and encoding gene mazE of antitoxin MazE was found in 11 strains, while neither mazE nor mazF was found in rest 5 strains. Both mazE and mazF had good conservation in molecular evolution analysis. Conclusions MazEF is the first toxin-antitoxin system found in prokaryotic chromosomes, but not in all strains of Escherichia coli and Shigella. MazEF deletion is associated with antibiotic resistance and it also mediates programmed cell death in bacteria, so MazEF might be a new target for antimicrobial agents.
2.Molecular evolution and binding free energy analysis on substrates of KPC carbapenemases

Objective To analyze molecular evolution and binding free energies in substrates of KPC-2,KPC-5 and KPC-10 carbapenemases.Methods Minimum Evolution method in MEGA 4.1 was used to analyze molecular evolution of KPC-2,KPC-5 and KPC-10 carbapenemases,Dock module in ArgusLab 4.1 was used to perform molecular docking of these 3 carbapenemases to 10 kinds of β-lactams substrates,and calculate binding free energies(△G).Results Ambler Class A β-lactamases with carbapenemase activities were grouped in the same cluster and had good conservation,while ordinary Ambler Class A β-lactamases without carbapenemase activities were groupod in the other cluster.Binding free energies of KPC-2,KPC-5 and KPC-10 carbapenemases were lower to carbapenem antibiotics than the thirdgeneration cephalosporins,while binding free energies to aztreonam and clavulanic acid were of comparatively higher levels.Conclusion Catalytic activities of KPC to carbapenem antibiotics are higher than those to the third-generation cephalosporins,but the activities to aztreonam and clavulanic acid are low.
3.Association of sleep to osteoporosis

In the view of physiology,sleep can be described as an important modulatory factor for hormone release,glucose regulation,and cardiovascular function.The circadian rhythms of hormones is mainly associated with sleep,which is largely mediated by neuroendocrine system.Sleep deprivation will disrupt this rhythm,leading to a series of metabolic diseases.We published an article in 《J Clin Endocrinol Metab》 titled Association between sleep duration,daytime nap duration,and osteoporosis varies by sex,menopause,and sleep quality,which suggested that sleep durations of 7-8 h/day,9-10 h/day,and ≥ 10 h/day,as well as longer daytime napping times,tend to present higher risks of having osteoporosis,and this tendency is especially obvious in postmenopausal women reporting good quality sleep.

The trace elements (Mn, Cu, Zn, Fe) andCalcium, Magnesium in aged person's serum oflong-life area and control area were determined. Itwas found that the contents of Manganese, cop-per, and Calcium of aged person's serum in long-life area were significantly higher than those incontrol area (Manganese and Calcium are essentialelements for health and antiaging, Copper is an es-sential trace element to health). In the long-lifearea the contents of six kinds of elements (Mn,Cu, Zn, Fe, Ca, Mg)in the serum have no signifi-cant distinction between the male aged and thefamle aged. In the long-life area the contents of sixkinds of elements (Mn, Cu, Zn, Fe, Ca, Mg )inaged (61~90) person's serum have no significantincrease or decrease with the increase of age.Judged from serum compositions, the agedperson's serum of the long-life area have a betterspectrum of elements. The relationship betweenthe elements in this experiment and antiaging effect was discussed.

6.Effect of Modified Constraint-induced Movement Therapy for Motor Function of Upper Extremity after Stroke

ObjectiveTo investigate the therapeutic effects of modified constraint-induced movememt therapy(mCIMT) on upper extremity motor function of stroke patients with hemiparasis, and to compare the effects of mCIMT with those of conventional rehabilitation.MethodsThirty stroke patients with hemiparesis were divided into mCIMT group and control group (conventional rehabilitalion), 15 cases in each group. The mCIMT group received mCIMT (by shaping) in the affected extremity without restriction of movement of intact upper extremity, 6 h a day, five times a week, for two weeks while the control group was treated with traditional rehabilitation in upper-limb and hands functional exercises. Motor activity logs(MAL) score and simple test for evaluating hand function(STEF ) score were measured in two groups before treatment and 2 weeks, 1 month and 3 months after treatment.ResultsThere were improvement in MAL and STEF scores in both mCIMT group and control group 2 weeks after treatment compared with pre-treatment(P<0-01), and the scores of MAL and STEF on post-treatment 1 month and 3 months were higher than those of pre-treatment in mCIMT group (P<0-05, P<0-01), but there was no significant difference in MAL and STEF score between pre-treatment and post-treatment 1 month and 3 months in control group(P>0-05).ConclusionSignificant improvement in the affected hand function could be achieved with modified constraint-induced movement therapy in stroke patients, better than conventional rehabilitation therapy.
7.Iodine nutritional status in adolescents after a decade of salt iodization in three cities of Guangdong

A cross-sectional survey of 3589 adolescents was conducted in three cities from different typical geographical zones of Guangdong province. The median urinary iodine concentrations (MUI) of adolescents in Nanxiong, Guangzhou and Maoming were 286.6,204.1 and 166.0μ/L, respectively. The MUI of all these adolescents Was 231.7μg/L, which was slightly higher than the current World Health Organization recommendation.
8.Effects of Yishen Jiangzhuo Granule on the bone metabolism of patients with stage 3-4 chronic kidney disease and its correlation with the immune indices.

UNLABELLEDOBJECTIVE To explore the changes of bone gla protein (BGP) and tartrate resistant acid phosphatase (TRACP) in patients with stage 3 -4 chronic kidney disease (CKD) before and after treatment, to study their correlation with interleukin-17 (IL-17) and regulatory T cells (Treg), and the effects of Yishen Jiangzhuo Granule (YJG) on the bone metabolism.
METHODSFifty-three patients with stage 3-4 CKD were randomly assigned to the treatment group and the control group using random digit table. The following parameters in blood were detected: Treg (CD4+ CD25+ CD127lo) using tri-chrism fluorescent labeling by flow cytometry; levels of TRACP, BGP, and IL-17 by double antibody sandwich ELISA. The hemoglobin (HGB) content was detected using Beckman-Coulter heme/analysis. The urinary contents of creatinine (UCr) were determined using reversed HPLC. The blood contents of calcium (Ca), phosphate (P), blood urea nitrogen (BUN), serum creatinine (SCr), and plasma albumin (ALB) were determined using automatic biochemical analyzer. Then the calcium-phosphate (Ca x P) product was calculated on the basis of blood contents of Ca and P. The clearance rate of endogenous creatinine (CCr) was calculated on the basis of blood BUN and SCr contents.
RESULTS(1) There was no obvious change in CD4+ CD25+ CD127lo in the two groups before and after treatment (P > 0.05). Compared with before treatment in the same group, there were statistical difference in the levels of CD4+ and TRACP in the two groups, as well as the IL-17 level in the control group (P < 0.01, P < 0.05). But compared with the healthy group, statistical difference was shown in each index (except CD4+) (P < 0.01). Compared with the control group after treatment, there was no statistical difference in each index of the treatment group after treatment (P > 0.05). Compared with before treatment in the same group, the levels of Hb, ALB, and CCr increased (P < 0.05, P < 0.01), and the SCr level decreased in the two groups after treatment (P < 0.05). Compared with the control group after treatment, the SCr level decreased and the CCr level increased more obviously in the treatment group (P < 0.05). There was no correlation among the levels of IL-17, TRACP, BGP, and Treg between before and after treatment in the two groups.
CONCLUSIONSYJG could improve the kidney function and delay the progression of micro-inflammation of stage 3 -4 CKD patients. It could not improve the level of CD4+ CD25+ CD127lo. It also showed no effects on bone metabolism. The CD4+ T cells were differentiated to Th17 cells in stage 3-4 CKD patients. Their immunity was in a state of anergy but continually activated. The inflammatory factors in patients with stage 3-4 CKD play important roles in inducing the activation of osteoclasts.

9.FAK gene silencing induces apoptosis of leukemic cells in vitro

AIM:Targeting of focal adhesion kinase (FAK) gene,we aim to construct FAK shRNA lentiviral vector and to identify its function on the growth of leukemic cells.METHODS:FAK shRNA was chemically synthesized,and inserted into a GFP-lentiviral plasmid through molecular biology methods.After packaged and concentrated,the lentiviral-FAK-shRNA-vector was transduced into a human leukemic cell line.FAK gene expression was detected by reverse transcriptional PCR and Western blotting.Cell apoptosis was measured by Annexin V labeling.RESULTS:The results showed that FAK shRNA was successfully inserted into the lentival vector,and the infection efficiency varied from 10% to 25%.Compared to the control vector (lentival vector without FAK shRNA),FAK shRNA inhibited the expression of FAK mRNA and protein by 40% and 70%,respectively.Moreover,the results of apoptosis experiment showed that the percentages of Annexin V+ cells in control vector group and FAK shRNA group were (4.19 ? 0.36) % and (8.48 ? 0.58) % respectively,the difference was statistically significant (P
