1.THE EFFECTS OF hTERT AND C-MYC ANTISENSE OLIGODEOXYNUCLEOTIDE ON THE TELOMERASE ACTIVITY IN HL-60 CELLS

Objective To investigate the effects of human telomerase reverse transcriptase(hTERT) and c-myc antisense oligodeoxynucleotide(ASODN) on the telomerase activity in HL-60 cells,and to explore the relationship between the telomerase activity and the expression of hTERT and c-myc in HL-60 cells. Methods The expression of hTERT and c-myc was sealed by ASODN in HL-60 cells,and was detected by RT-PCR.The telomerase activity was determined with TRAP-ELISA and PAGE-silver staining. Results After treated with ASODN for 72 hours the expression of hTERT and c-myc mRNA was inhibited in different degree,and was the lowest in group of 30??mol/L ASODN.As for the TRAP-ELISA detection,the difference was significant when compared with groups of 10,20,30??mol/L hTERT ASODN and 20、30??mol/L c-myc ASODN with group of untreated(P0.05).PAGE-silver staining detection showed that comparing ASODN groups with SODN groups the telomerase image strips were decreased and was the least in group of 30??mol/L,and the telomerase image strips were also decreased when compared the same density group of hTERT ASODN with c-myc ASODN.Conclusion hTERT and c-myc ASODN may inhibit the expression of the genes respectively and down-regulate the telomerase activity in HL-60 cells.
2.Neuroprotection of Copolymer 1 in glaucoma rats with elevated intraocular pressure

Objective To observe the neuroprotective effect of Copolymer 1 ( Cop 1) in rats with elevated intraocular pressure (IOP), and explore the clinical application of Cop 1 in the prevention of optic nerve atrophy induced by glaucoma. Methods Thirty adult Wistar rats were rendered for elevation of IOP in both eyes by Shareef-Sharma method. At the third week of IOP elevation, a mixture (1:1) of Cop 1 (100 μg) emulsified in complete Freund's adjuvant (CFA) was injected intracutaneously at two hind footpads in 15 rats (Cop 1 model group), and the other 15 rats were given the same dose of PBS emulsified in CFA (model control group) . Retinal ganglial cell (RGC) retrograde labeling was performed with Fluorogold 6 d later, and retina was taken after 24 h to compared the density of RGC between two groups. Another normal rats were served as blank controls. Results It was found that lymphocytes were accumulated on the histological slices of retina and optic nerve in Cop 1 model group. The rates of RGC loss of Cop 1 model group and model control group were 10. 24% ± 3.75% and 29.00% ±6.92%, respectively. The RGC density in Cop 1 model group was significantly lower than that in model control group (P <0. 05), while there was no significant difference in that between Cop 1 model group and blank control group (P >0.05). Conclusion Intracutaneous injection of Cop 1 can reduce the damage of RGC induced by elevated IOP.
4.Explore the TCM pathogenesis based on the analyses of TCM pattern elements among 84 ;patients with middle-late pancreatic cancer

Objective To explore the TCM pathogenesis based on the analyses of TCM pattern elements. Methods TCM pattern elements of 84 patients with middle-late pancreatic cancer to analyze the TCM disease location and characteristics by their frequency and the contribution scores. Results A total of 84 patients were included, TCM disease location with the top 10 rank of the contribution scores were spleen, stomach, liver, gallbladder, kidney, intestine, lung, heart, spleen, stomach, while they were significantly higher than others (P<0.05). And the TCM pattern elements with the top 5 rank of the contribution scores were qi deficiency, dampness, heat, blood stasis, toxin, while they were significantly higher than others (P<0.01). Conclusions TCM disease locations of middle-late pancreatic cancer were mainly in spleen and stomach, and TCM pattern element were excessive patterns with qi deficiency, deficiency patterns with dampness, heat, blood stasis and toxin.
5.Clinical observation of acute promyelocytic leukemia remission induction and sequential treatment programs

Objective To observe efficacy of acute promyelocytic leukemia (APL) by chemotherapy induction and treatment programs after remission.Methods 28 cases of newly diagnosed APL were treated with combined arsenic trioxide and retinoic acid which was called induced program,the patients whose leukocytes >10×109/L would be given anthracycline chemotherapy (DNR 40 mg × 3 d).Remission induction anthracycline chemotherapy was given after three courses of intensive therapy,the standard-dose cytarabine was used to the patients with high white blood.After the intensive treatment and with the recorery of the patient' s blood,bone marrow sequential therapy was used [arsenic trioxide (As2O3),retinoic acid (ATRA),6-MP + MTX and chemotherapy],during which recurrence rate and adverse reactions in the patient' s course of treatment were observed.Results Except 1 was died of intracranial hemorrhage at the beginning of treatment,the remaining 27 patients had received hematologic remission with PML/RARα fusion gene negative.Fusion gene of 5 patients remained negative even after maintenance treatment 2 years later.Conclusion As2O3,ATRA combined with chemotherapy in the treatment of APL is effective,the hematologic response time may be shortened,and sequential therapy can significantly reduce the relapse rate and hospitalization time,patient compliance is increased and the quality of life in patients is improved.

7.Expression of TNF-? and TNF-R1 in retinal glial cells of a rat chronic elevated intraocular pressure model

AIM:In order to study the effects of retinal glial cells on glaucomatous retinal ganglion cells damage,expression of TNF-? and TNF-R1 in retinal glial cells was observed in rat model with chronic elevated intraocular pressure glaucoma.METHODS:(1)Rats were rendered elevated intraocular pressure by ligating 2 episcleral veins with subconjunctival injection of 5-Fu.(2)Four weeks after operation,immuno-histological assays were carried out on cryostat sections.The co-expressions of TNF-?-GFAP,TNF-?-OX42,TNFR-1-GFAP,and TNFR-1-NeuN were observed via a confocal laser scanning microscope,respectively.RESULTS:(1)Elevated intraocular pressure was consistently maintained for up to 4 weeks in model group.(2)The co-expressions of TNF-? and GFAP,TNF-? and OX42 were detected in retina in treatment group,respectively.(3)The co-expressions of TNFR-1 and GFAP were also detected in retina,but the co-expressions of TNFR-1 and NeuN were not detected in retina in treatment group.CONCLUSION:TNF-? that activated retinal glial cells may play an important role in glaucomatous retinal ganglion cell damage.

9.Automatic Oral Drug Dispensing System Conducive to Hospital Pharmaceutical Care

OBJECTIVE:To realize automatic central drug dispensing model in hospital pharmacy.METHODS:The construction,running condition,major function and flow-sheet of automatic oral drug dispensing system of our hospital was introduced,and the automatic drug dispensing system was compared with traditional dispensing system.RESULTS:The automatic oral drug dispensing system could enhance the speed and accuracy of drug dispensing,shorten waiting time of nurse,improve health condition and decrease medicine pollution.CONCLUSION:The automatic oral drug dispensing system is suitable for drug dispensing in hospitals and conducive to the enhancing of pharmaceutical care level.
10.Effect of Buyang Huanwu decoction on rehabilitation of stroke patients

ObjectiveTo evaluate the clinical effect of Buyang Huanwu decoction on motion functions and complications of stroke patients.Methods86 acute stroke patients were randomly divided into the therapy group (n=46) and control group (n=40). Despite of routing neurological medicine, rehabilitation therapy including decubitus position, neurodevelopmental therapy and activities of daily living (ADL) and low frequency electronic stimulation on both two groups, Buyang Huanwu decoction was used on the therapy group. Patients of two groups were evaluated with Fugl-Meyere motion index and modified Barthel index before and after therapy, and complications of all cases were observed, including shoulder pain, joints motion limitation, sensation disorder and stembrocker syndrome.ResultsThe motion functions of patients in two groups were improved significantly after therapy (P<0.01). While effect of the therapy group was better than that of the control group (P<0.01), and the accidence of complications in the therapy group was lower than that in the control group (P<0.01).ConclusionBuyang Huanwu decoction can improve the motion function and ADL, and decrease the complications of acute stroke patients.
