4. The peptide containing the conserved sequence of HPV L1 was used in the detection of polyvalent HPV-positive clinical specimens

Objective: To confirm that the antiserum induced by short peptide in the region of AA 448-477 at HPV16 L1 C-terminal can respond with many types of HPV L1. Methods: This sutdy synthesized a short peptide containing the conserved sequence of HPV L1 and immunized animal with the synthetic peptide to obtain the polyclonal antiserum. The type of HPV in clinical specimens, such as cervical cancer and condyloma acuminatum tissues, was identified by PCR. ELISA, Western blotting, and inummohistochemistry were used to determine whether the anti-serum of the short peptide could react with different types of HPV-positive specimens. Results: ELISA assay showed that the reaction between antigen and antibody was positive in HPV6, 11, 16, and 18 and other HPV-positive clinical specimens. Western blotting revealed that there was a strip at 56 kDa in all types of HPV-positive clinical specimens, but there was no strip in HPV-negative clinical specimens. Immunohistochemistry analysis demonstrated that the histological reaction had varying features in different clinical specimens. The positive-reaction appeared only in the epithelium tissues, which was regional and had apparent borders. Conclusion: This study confirms that the common conserved sequence really has HPV L1 common epitope. Its anti-serum can respond with many types of HPV L1. These findings provide an important basis for further researching a broad-spectrum HPV L1 vaccine or publicizing broad-spectrum HPV detection kits.
5.Analysis of Allergens Detection in Asthmatic Infants and Young Children

Objective To analyze the allergens detection in asthmatic infants and young children. Methods The specific IgE test and allergens screening test were conducted in 243 infants and young children with whooping diseases including asthmatic bronchitis, asthmatic pneumonia and asthma by skin prick test and Pharmacia UniCAP test. Results 1. Food allergen was the allergen detection most frequently in infants(P = 0.03). The proportion of aeroallergens increased significantly with age (positive rate:67. 2%). Milk and egg white was the main food allergen detection in infants and young children,and the main aeroallergen detection was house dust mite. 2. The allergen - specific IgE test using UniCAP test system was more accurate than skin prick test. Conclusions The infants are more susceptible to food hypersensitivity; the increasing opportunity of aeroallergen exposure may lead to a higher prevalence of aeroallergens in young children. UniCAP system is more accurate than other methods.

7.Role of T?SPOT.TB test in diagnosis of active tuberculosis

Objective To assess the value of T?SPOT.TB test in the diagnosis of active tuberculosis. Methods The clinical data of 975 hospitalized patients receiving T?SPOT.TB test were collected in our hospital. The clinical information and testing results were analyzed. The receiver operating curve (ROC) was used to determine the optimal threshold of T?SPOT.TB test for differentiating active tuberculosis. Results T?SPOT.TB test results showed that the positive rate was 29.26%for the non?active tuberculosis group(n=793),but was 91.21%for active tuberculosis patients group (n = 182),which indicated that the test had a significant value in active tuberculosis detection(P<0.001). The sensitivity of T?SPOT.TB test was 0.912 and the specificity was 0.707. The detection threshold of T?SPOT.TB was optimized. As the spot?forming count(sfc)of ESAT?6 antigen threshold was 11.5 and that of the CEP?10 threshold was 9.5,the efficiency of T?SPOT.TB test for detection of active pulmonary tuberculosis was the highest. Conclusions T?SPOT.TB test has a good diagnostic performance for active tuberculosis, and it can be further optimized to better serve the clinical practice.
8.Genomic level analysis of the molecular resistance mechanism for Pseudomonas aeruginosa PA902

Objective To investigate the resistance mechanism of Pseudomonas aeruginosa PA902 by high -throughput sequencing.Method PA902 was sequenced by the Illumina Miseq platform,and sequencing data were analyzed by bioinformatic techniques.Results PA902 was resistant to all common clinical antibiotics except amikacin.Analysis of data reveal resistance genes to β-1actams (blaOXA-10,blaPER-1 和 blaVIM-2),aminoglycosides (aph (3 ‘)-Ⅱd,aac (6')-Ⅰb-cr,aacA4 和 aadA2),sulfonamides (sul1),tetracyclines (tetG),chloramphenicol (floR).Several putative chromosome-located resistance genes were also identified.The results were fully in accordancc with thc susceptibility results.Sequence analysis of the contigs containing resistance genes revealed that they were always clustered on the same contig and correlated mobilization sequences.MLST identified PA902 as ST389,which was first reported in China.Conclusion High throughput sequencing demonstrated the molecular resistance mechanism of PA902,and the findings were in accordance with the susceptibility results.The technique will provide solid support for the traditional clinical microbiology methods.
9.Investigation on the Performance of Chlorine Dioxide Generators for Drinking Water Disinfection

Objective To preliminarily investigate the conditions of the performance of chlorine dioxide generator for drinking water disinfection and know the overall situation of the industry.Methods From 2007 to 2008,6 companies'products were chosen for this investigation according to certain rules,and their performance indicators,such as chlorine dioxide output and conversion rate were tested by using the Five-step iodimetry method.Results On-site generating technology was commonly used,and the average conversion rate of the compound type generator was 57.06%,and the pure type generator was 84.78%.Conclusion The conversion rate of the pure type generator is better than the compound type,and the newly invented pure type generator has much better performance and lower disinfection cost.
10.Analysis on the Utilization of Cardiovascular and Cerebrovascular Drugs in a Military Sanatorium During 2007~2009

OBJECTIVE: To analyze the use situation and trend of cardiovascular and cerebrovascular drugs during 2007~2009 in a military sanatorium, and provide reference for the rational use of cardiovascular and cerebrovascular drugs and its essential medicines. METHODS: Cardiovascular and cerebrovascular drugs in the rehabilitation center during 2007~2009 were analyzed statistically in the respect of category, consumption sum and DDDs. RESULTS: The consumption sum and DDDs of cardiovascular and cerebrovascular drugs were increasing in the past three years. And its essential drugs account for 94.8%.The most widely used drugs were cerebrovascular drug, brain metabolism drug, nootropics and antihypertensive drugs. Oxiracetam and Xueshuantong were widely used cerebrovascular drug, brain metabolism drug and nootropics. Amlodipine were widely used antihypertensive drugs in the clinic. CONCLUSION: Cardiovascular and cerebrovascular drugs and its essential drugs were characterized with complex classification, numerous categories, extensive application and potential market demand, which require rational use of drug should be strengthened.
