1.Curative Effect of Lacidophilin Vaginal Capsules Combined with Loop Electrosurgical Excisional Procedure in the Treatment of Cervical Erosion with Chronic Cervicitis

Objective:To discuss the curative effect of lacidophilin vaginal capsules combined with loop electrosurgical excisional procedure ( LEEP) in the treatment of cervical erosion with chronic cervicitis. Methods:Totally 72 cases of patients with cervical ero-sion with chronic cervicitis were selected and divided into the observation group and the control group at random. The patients in the two groups were given routine LEEP with the application of antibiotics to prevent infection for 1 week. The patients in the observation group were additionally given lacidophilin vaginal capsules, placed in the vagina after the operation, one capsule per time at every night for 14 days. The vaginal fluid time and vaginal bleeding time in the two groups were observed. The clinical curative effect and adverse drug reactions ( ADRs) after 8 weeks of the operation were studied and evaluated again. Results: The vaginal bleeding amount and bleeding time in the observation group after the operation was less or shorter than that in the control group (P<0. 05). According to the further consultation after 8 weeks of the operation, the total clinical efficiency in the observation group was 94. 44%, which was ob-viously higher than that in the control group(75. 00%,P<0. 05). There was no obvious ADR appeared in the two groups during the treatment. Conclusion:Lacidophilin vaginal capsules combined with LEEP has reliable curative effect on cervical erosion with chronic cervicitis, which can obviously shorten the wound healing time and decrease the vaginal bleeding amount and time with high security.
2.Proteomics analysis for the signal transduction under stimulation of prolactin on Jurkat cells

Objective:The experiment was designed to apply proteomics to analyze the signal transduction molecules in T lymphocyte strain JurkatD1.1 cells with the stimulation of prolactin.Methods:JurkatD1.1 cells were cultured in the presence of recombinant human prolactin(rhPRL).Phosphated metal affinity chromography(PMAC) and immunoprecipitation(IP) assay were applied to enrich phosphoproteins from the total cell lystates which were resolved by one-dimensional electrophoresis(1 DE) or 2 DE.The different bands and different spots in the gels between control group and rhPRL-treated group were excised,digested,and analyzed by MALDI-TOF mass spectrometry.Results:Three major protein bands were observed specifically in the rhPRL-treated group and other two major protein bands were observed specifically in the control group.The bands were excised and analyzed by MALDI-TOF mass spectrometry.Protein band a in rhPRL-treated group was successfully identified as hot shock protein 90(hsp90).Nine different protein spots which were observed in the 2 DE gels between the two groups were excised and analyzed by MALDI-TOF mass spectrometry.In the rhPRL-treated group the protein spot 4 was identified as nuclear receptor co-repressor 2 variant(NcoR 2 variant);The protein spot 5 was identified as galactose-1-phosphate uridyl transferase;And the protein spot 6 was identified as zinc finger ZIM3.Conclusion:Hot shock protein 90 participates the signal transduction pathways of prolactin.Nuclear receptor co-repressor 2 variant and zinc finger ZIM3 may play a role in the process of prolactin modulating target genes' expression.
3.HIF-1αprotein expression in oral submucous fibrosis tissue

Objective:To evaluate the effects of HIF-1αprotein in the pathogenesis of oral submucous fibrosis(OSF)tissue.Meth-ods:HIF-1αprotein expression in 30 cases of OSF,5 cases of normale control oral mucosa tissue was studied by SP immunohisto-chemistry with HIF-1αrabbit anti-human polyclonal antibody.Results:The negative expression of HIF-1αwas found in normal oral mucoa tissue and possitive in early,intermediate and advanced stage of OSF(P<0.05 ).Conclusion:HIF-1αprotein may play an importent role in the pathogenesis of OSF.
4.Expressions of HIF-1αand CTGF proteins in oral submucous fibrosis

Objective To evaluate the effects of hypoxia inducible factor-1 alpha ( HIF-1α) and connective tissue growth factor (CTGF) proteins in the pathogenesis of oral submucous fibrosis (OSF). Methods Expressions of HIF-1αand CTGF proteins in 30 cases of OSF, and 5 cases of normale control o-ral mucosa tissue were studied with streptomyces peroxidase ( SP) immunohistochemistry with the HIF-1αand CTGF rabbit anti-human polyclonal antibodies.Results The negative expressions of HIF-1αand CT-GF proteins were found in normal control and significant increase was found in OSF ( P <0.05).Conclu-sions HIF-1αand CTGF proteins have positive expression in OSF, both of them may act as an important role in pathogenesis of OSF.
5.Observation of the effect of house dust mite vaccine in the treatment of allergic rhinitis

ObjectiveTo observe the efficacy and safety of house dust mite vaccine in the treatment of house dust mite-allergic rhinitis.Methods47 patients with allergic rhinitis were recruited into the study.The course of SIT was treated by house dust mite vaccine for 51 weeks.Self-contrast method was used to compare the symptom score,nasal airway resistance before and after treatment.Then we observed the security in the process of injection.ResultsThe symptom score was (7.40 ± 1.64) vs ( 3.40 ± 1.50 ) pre and post therapy.Nasal airway resistance were (2.02 ±0.72) vs ( 1.06 ± 0.81 ) kPa · s -1 · L-1.Both the values of the data had significant differences( t =15.49,4.44,all P < 0.01 ).No serious adverse reaction was observed.ConclusionSIT was able to improve the clinical symptoms and decrease nasal airway resistance.House dust mite vaccine was an effect and safe drug for the dust mite allergic asthma patients.

7.Effect of PRL on the expression of CD154 in Jurkat cells

Expression of PRL receptor mRNA in Jurkat D1.1 cell line was confirmed by RT PCR. CD154 expression of Jurkat cells was significantly increased when these cells were incubated with PHA and recombinant human PRL, suggesting that binding of PRL to its receptor promotes CD154 expression which plays a role in immune regulation.
8.Expression of PRLr mRNA in human immune system

Objective:To investigate the expression of prolactin receptor(PRLr) in human immune system.Methods:Specimens of human immune system were obtained from both central and peripheral immune organs, including thymus(thymoma), bone marrow, lymph node and peripheral blood mononuclear cells. Specific fragments of PRLr mRNA were obtained through RNA isolation and RT-PCR amplification and confirmed by DNA sequencing.Results:PRLr mRNA was detected from the thymoma, bone marrow, lymph node and peripheral blood mononuclear cells. All the PRLr cDNA fragments generated by PCR were as long as the expected length, which was 276 bp. The sequencing result of the cDNA was identical to the sequence of PRLr cDNA in GeneBank.Conclusion:The study confirmed that prolactin receptor mRNA was expressed in human central immune organs such as thymus and bone marrow and peripheral immune organs such as lymph node and peripheral blood mononuclear cells. This phenomenon provided a biologic structural evidence that neuro-endocrine hormone PRL might play a modulating role on immune cells from receptor's point of view.

[Summary] The controversy remains on whether the Oddi sphincter function should be preserved or deprived in the treatment of hepatolithiasis .The surgical approaches achieving both hepatobiliary lesions clearance and Oddi sphincter preservation have been widely accepted .The biliary-enterostomy is suspected to increase the risk of reflux cholangitis and cholangiocarcinoma due to iatrogenic Oddi sphincter deprivation.Endoscopic sphincterotomy (EST) as a minimally invasive method still needs further research to determine its role in hepatolithiasis management .

Objective To explore the influence of recombinant human erythropoietin(rhEPO) injection about nitric oxide(NO) and nitric oxide synthase(NOS) level in the brain and endothelin(ET) level in the blood after hypoxia-ischemia brain damage(HIBD) in neonatal rats,and elucidate the possible mechanism of the neuroprotective effect of rhEPO administered to the rat pup prior to cerebral hypoxia-ischemia.Methods Seven days-old SD rats were divided into control group,hypoxia-ischemia group(HI) and EPO group.NO activity,NOS level in the brain and ET in the blood were determined at 6 h after hypoxia-ischemia.Results NO and NOS levels in the brain and ET level in the blood were increased after HIBD in neonatal rats(P
