1.Influence of Circ_HECTD1 on OGD/R-induced hippocampal neuronal damage by regulating the miR-135a-5p/TP53INP1 axis

Objective To observe the influences of circular RNA HECT domain E3 ubiquitin ligase 1 (Circ_HECTD1) on the proliferation and apoptosis of HT22 cells and its regulatory mechanism on the microRNA-135a-5p (miR-135a-5p)/tumor protein 53-induced nuclear protein 1 (TP53INP1) axis by in vitro oxygen-glucose deprivation/reoxygenation (OGD/R)-induced hippocampal neuron damage.Methods HT22 cells were routinely cultured,and the cells were separated into Con group,OGD/R group,si-NC group,and si-Circ_HECTD1 group,miR-NC group,miR-135a-5p group,si-Circ_HECTD1+anti-miR-NC group,and si-Circ_HECTD1+anti-miR-135a-5p group.qRT-PCR method was used to detect the expression of Circ_HECTD1,miR-135a-5p and TP53INP1 mRNA;MTT was used to detect cell viability;flow cytometry was used to detect apoptosis;dual-luciferase reporter gene experiment was applied to verify the targeting relationship between Circ_HECTD1 and miR-135a-5p,miR-135a-5p and TP53INP1;Western blot was applied to measure the protein expressions of Bax,Bcl-2 and TP53INP1.Results After OGD/R induction,the expressions of Circ_HECTD1 and TP53INP1 in HT22 cells were up-regulated,the expression of miR-135a-5p was down-regulated,the cell survival rate and the expression of Bcl-2 protein were remarkably decreased,the apoptosis rate and the expression of Bax protein were remarkably increased (all P<0.05).Silencing the expression of Circ_HECTD1 could remarkably up-regulate the expression of miR-135a-5p in OGD/R-induced HT22 cells,down-regulate the expression of TP53INP1,increase cell survival rate and Bax protein expression,decrease cell apoptosis rate and Bcl-2 protein expression (all P<0.05).There was a targeting relationship between Circ_HECTD1 and miR-135a-5p,between miR-135a-5p and TP53INP1.Overexpression of miR-135a-5p could remarkably down-regulate the expression of TP53INP1,increase cell survival rate and Bcl-2 protein expression,decrease cell apoptosis rate and Bax protein expression (all P<0.05).Inhibition of miR-135a-5p expression could partially reverse the protective effect of silencing Circ_HECTD1 on HT22 cell damage.Conclusion Silencing Circ_HECTD1 can regulate the miR-135a-5p/TP53INP1 axis and promote cell survival,inhibit cell apoptosis,and protect against OGD/R-induced hippocampal neuron damage.
2. Reviews on Chemical Compositions and Pharmacological Effect of Gelsemium elegans

Gelsemium elegans,the evergreen woody vine of the genus Gelsemium of the Loganiaceae family in China,is a traditional Chinese medicinal plant. It is spicy,bitter,warm,highly toxic and commonly used for dispelling wind-evil,attacking poison,reducing swelling and relieving pain. In this article, the researches on the chemical constituents and pharmacological effects of G. elegans in recent years were retrieved,reviewed and summarized. So far,alkaloids,iridoids,triterpenes,phenolic acids,steroids,coumarins,lignans,megastigmane glycosides and other ingredients have been separated from G. elegans. Alkaloids, mostly of indole alkaloids,are the main active ingredients there of which can significantly inhibit central nervous activity. Modern pharmacological studies have shown that alkaloids have a variety of pharmacological activities,which can achieve the purpose of anti-tumor by regulating cell cycle; enhance macrophage phagocytosis,protect white blood cells,and promote immune regulation;relieve cancer pain and long-term pain;reduce the myocardial contractility,get vasodilation to achieve antihypertensive effect;and also play an important role in the treatment of anxiety disorder and dermatosis. In the future research on G. elegans,it is still necessary to further study the chemical constituents,develop promising lead compounds,conduct in-depth research on its toxicology and clinical pharmacology,and clarify its mechanism of action,make it used more fully and reasonably,and lay the foundation for the application and safe use of G. elegans.

5.Treatment of type C3 distal femoral fractures with double-plating fixation via anteriormiddle approach.

OBJECTIVETo investigate clinical efficacy and feasibility of double-plating fixation via anteriormiddle approach in treating type C3 distal femoral fractures.
METHODSFrom August 2008 to August 2011, 12 cases with type C3 distal femoral fractures were treated, including 5 open fractures and 7 closed fractures. Among them, there were 8 males, 4 females with an average of 40 years (ranged, 25 to 55 years). There were 7 in left side, 5 in right side. Nine cases were caused by car accident, 3 cases by falling down. The duration from injury to hospital was form 20 minutes to 5 days (mean 135 min). After tibia bone traction for 5 to 8 days, the operation were performed by double-plating fixation via anteriormiddle approach, and autograft of iliac bone or allograft bone grafting were given to bone defect. Knee joint function was evaluated according to Merchanetal criteria.
RESULTSThe operation time was from 110 to 160 min, with an average of 135 min, the blood loss was from 300 ml to 500 ml,with an average of 400 ml. Post-operative wound were stage I healing. All patients were followed up from 16 to 36 months (mean 24 months). No infection, reduction loss, nonunion, deep vein thrombosis occurred. Bone healing time was for 18 to 24 weeks with an average of 21 weeks. According to the Merchanetal criteria, 4 cases got excellent results, 6 good, 1 fair and 1 poor.
CONCLUSIONDouble-plating fixation via anteriormiddle approach for type C3 distal femoral fractures is an effective way, which has advantages of obvious exposure, simple manipulation, anatomical reduction, stable fixation. However,operation indications and operating instructions should be strictly followed.

6.Chemical Constituents of Fibraurea recisa Pierre

Objective The purpose of this study was to investigate the chemical constituents of Pierre. Methods The chemical compositions were isolated by silica gel column chromatography, semi-preparative HPLC,Sephadex LH-20, and MPLC packed with MCI gel, and their structures were identified on the basis of chemical and spectroscopic evidence (NMR and MS) . Results Eight compounds were obtained and determined as fibraurin (1),makisterone A (2),cycloartenol (3),feruloylphenethylamine (4),trans- -feruloyltyramine(5),β-sitosterol(6),1-monopalmitin(7),1-sinapoyl-β-D-glucopyranoside(8). Conclusion Compounds 2-8 were isolated from this plant for the first time.
7.Qualitative study on the influencing factors of medical insurance-related doctor-patient disputes based on grounded theory

The medical insurance policy has entered into a deep reform and can effectively alleviate the tensions between doctors and patients. However, medical insurance-related doctor-patient disputes still occur during the actual implementation process. Based on the grounded theory research method, semi-structured interviews were conducted to intuitively and realistically describe the experiences of both doctors and patients in medical insurance-related doctor-patient disputes. Nvivo 11 software was used to conduct three-level coding on interview data and extract the reasons that trigger medical insurance-related doctor-patient disputes. in response to the influencing factors of medical insurance-related doctor-patient disputes, four core concepts of “medical staff, patient groups, medical environment, and social environment” were extracted. Additionally, countermeasures to alleviate medical insurance-related doctor-patient disputes were proposed, including the government strengthening overall planning and diversifying policy publicity; hospitals refining services and optimizing the evaluation system for medical staff; improving the professional quality of medical staff and guiding the standardized practice; and transforming patients’ medical thinking and improving their policy awareness.
8.Research advances on the application of carbon dots in wound treatment.

Chronic and infectious wound healing has always been an issue of concern in clinical and scientific research, in which bacterial infection and oxidative damage are the key factors hindering wound healing. Carbon dots, as a new material, has attracted much attention because of its unique physical and chemical properties and good biological safety. In recent years, the researches on the antibacterial property, antioxidant, and photoluminescence properties of carbon dots are more and more extensive and carbon dots have great potential in the treatment of chronic and infectious wounds. This paper reviews the research progress of carbon dots in three aspects: antibacterial, anti-oxidation and monitoring of wound infection are reviewed, and further discusses its specific mechanism, potential research direction, and application prospect.

9.Isolation of a methane-utilizing Klebsiella sp. strain and its application for detecting methane.

We have isolated a strain C611 that used methane as the sole carbon sources for growth from paddy soil in Taiyuan of Shanxi province. Based on the physiological characteristics and 16S rDNA sequence analysis, we identified the strain as Klebsiella sp.. We used statistic-based experimental design (RSM) to optimize the culture conditions for C611 strain. The optimum conditions were as follows: temperature of 24.4 degrees C, inoculum volume of 6.7% and methane content of 25%. We studied the response time and the relationship between consumption of dissolved oxygen and methane gas contents with PVA-H3BO3 immobilized cell of C611 using electrochemical method. The response time was no more than 100 s of this reaction system, and the linear range of detection of methane content was from 0 to 10%. The standard gas sample 3% methane was measured by this method with the mean content value of 3.09%, RSD of 3.48%, and the relative error of 3%. Hence, it has the potential in developing biosensor for methane.

10. Clinicopathological features of neuroendocrine carcinoma of the cervix: an analysis of 35 cases

Objective:
To analyze the clinical and pathological features and prognostic factors of neuroendocrine carcinoma of the cervix (NECC).
Methods:
The clinical data of 35 cases of NECC treated in Shanxi Provincial Cancer Hospital from January 2006 to May 2014 were retrospectively analyzed.
Results:
The median age of 35 cases of NECC was 43 years old. The infection rate of human papillomavirus (HPV) 18 type was 66.7% (10/15) in 15 NECC patients who were tested with HPV. The accuracy rate of diagnosis was 14.3% (5/35) before procedure. The positive detection rate of Syn, CgA, AE1/AE3, P63 and NSE were respectively 100.0% (35/35), 80.0% (28/35), 100.0% (35/35), 34.3% (12/35) and 57.1% (20/35). The 3-year overall survival rate of 35 NECC cases was 34.29% (12/35). The age, volume of local tumor and degrees of interstitial infiltration were the main prognostic factors of patients with NECC(all
