1.Evaluation on expression of recombinant S gene of human hepatitis B virus in vitro and in vivo

Objective To construct a DNA vaccine capable of expressing S gene of hepatitis B virus(HBV) and evaluate the expression of recombinant S gene in vitro and in vivo. Methods A cloned S X gene fragment was inserted into a eukaryote expression vector to construct a recombinant plasmid. The S gene was transcribed in vitro and expressed in a transfected cell line, and the efficiency of HBsAg in eliciting anti HBs was evaluated in mice. Results The expression of S gene was confirmed by Northern blotting, Western blotting, and ELISA(for both antigen and antibody detection). Conclusions The recombination and expression of S gene is achieved successfully in vitro.
2.The relationship between estrogen levels and carotid atherosclerosis in postmenopausal women with type 2 diabetes

Objective To observe the correlation of estrogen,fasting insulin(FINS),fasting plasma glucose (FPG),glycosylated hemoglobin(HbA1c),blood lipids and carotid arteries -membrane thickening(IMT)in the postmenopausal women with type 2 diabetes.Methods 63 postmenopausal women with type 2 diabetes were divided into normal IMT group(group A),IMT thickening group(group B),plaque group(group C)according to the vascular ultrasound carotid artery intima thickness,at the same time 15 cases of healthy physical examination as control group (group D).All of the groups were detected the plasma total cholesterol(TC),high-density lipoprotein cholesterol (HDL-C),low density lipoprotein cholesterol(LDL-C),fasting insulin(FINS),fasting plasma glucose(FPG),gly-cosylated hemoglobin(HbA1c),follicle-stimulating hormone(FSH),estradiol(E2).Results Compared with group A,the levels of FINS,IR,HbA1c,TC,LDL-C in group C was higher(t=16.83,14.44,2.50,2.38,2.42,all P<0.01 or P<0.05),however,the results of E2,HDL-C was decreased(t=7.32,5.51,all P<0.01).The levels of FINS,HbA1c,TC,LDL-C,FSH,IR in group D was lower than those of group C(t=19.74,36.44,8.64,7.95, 27.13,6.71,all P<0.01),On the contrast,the levels of HDL-C,E2 were elevated(t=11.34,8.72,all P<0.01). The Pearson's correlation analysis showed that E2 had negative correlation with FINS,IR,TC,LDL-C (P<0.01 or P<0.05).E2 had positive correlation with HDL-C(P<0.05).Logistic regression analysis showed that IMT and FINS,IR,HbA1c,LDL-C were positively correlated(OR=2.94,72.63,27.30,98.18,all P<0.01),while nega-tively correlated with E2 levels(OR=2.94,P<0.01).Conclusion The increased level of estrogen was a protective factor of carotid artery atherosclerosis in postmenopausal women with type 2 diabetes.
3.The Analysis of Craniocerebral Trauma with Negtive Results on Emergency CT (A Report of 51 Cases)

Objective To explore the causes of negtive results on emergency CT and prevent method in craniocerebral trauma.Methods 51 cases of craniocerebral trauma with negtive results on emergency CT were analyzed respectively during 2002 to 2004.Results The causes of negtive results on emergency CT in craniocerebral trauma were:the insufficient experience for technicians and doctors (12.8%);the atypical signs in early phase(48.6%);the delayed lesions after injury(16.2%) and the limitation of CT (24.2%).Conclusion CT technicians and doctors should be well trained primarily. Furthermore, CT images, clinical manifestations and other examinations should be combined with to obtain precise diagnosis. Finally, uncertaint cases should be examined again with CT or MRI.

Object To explore the feasibility of breeding genetic modified (GM) medicine by expressing human cytokine in transgenic Chinese materia medica Methods Human interferon ? gene and RANTES gene available from the amplification in vitro were enzymatically excised, recoveried, and inserted into intermediate vectors The recombinants were identified by double enzyme digestion of EcoRⅠand HindⅢ The plasmids were extracted from Escherichia coli and introduced into A tumefaciens, and the transformants harboring binary vectors were screened by addition of antibiotics of kanamycin (Km) and rifampicin (Rif), and the explants of M charantia and P vulgaris were transformed by co cultivation of leaf disks with A tumefaciens strain Results RT PCR was applied to detect the transient expression of human interferon ? gene and RANTES gene in transformed medicinal herbal calli Conclusion The expression of recombinant human interferon ? gene and RANTES gene in transgenic M charantia and P vulgaris cells was firstly reported, which opens an alternative road to antivirus, especially anti AIDS virus, by using transgenic Chinese materia medica
5.CT and MRI Findings of Esthesioneuroblastoma

Objective To study CT and MRI findings of esthesioneuroblastoma.Methods Nine cases of esthesioneuroblastoma proved pathologically were presented.There were four male and five female,with mean age of 34.6 years.CT scan in four cases and MRI in seven cases were done and 2 cases had enhanced MRI.Results The location of lesions were in nasal cavity in 2 cases and in nasoethmoid region in 7 cases.Among nine cases,six cases were seen in the right side,and three cases in the left side.On CT and MRI,the lesion demonstrated a destructive solid mass,CT value ranged 29~36 HU.The lesions appeared as hamogeneous signal intensity which was higher than tongue muscle on T_1WI,and higher than grey matter on MRI T_2WI . Turbinal and ethmoid bony destruction were showed on all nine cases. The lesions invaded into opposite nose and ethmoid sinus in four cases,intruded into intracrainial cavity in eight cases, frontal sinus in six cases, orbit in eight cases in which two cases had eye protrusion, maxillary sinas in five cases and sphenoid sinus in five cases.Conclusion A esthesioneuroblastoma can be considered if a destructive solid mass occurs in nasal cavity and ethmoid sinus,with a middle signal intensity on T_2WI.
6.Resource Investigation for Endangered Wild Ferula sinkiangensis Based on Low Altitude Remote Sensing

As the specific endangered medicinal plant in Xinjiang, resources and distribution of Ferula sinkiangen-sis are important for biodiversity conservation and sustainable development of Chinese medicine resources. The spa-tial distribution and resources of F. sinkiangensis were researched based on low altitude remote sensing and sample investigation. The results showed that the optimum working time for F. sinkiangensis monitoring by low altitude remote sensing was the nearby 5 hills, which covered about 0.88 km2. It was suggested that the fence area should be expanded for protection. According to the results of low altitude remote sensing, the amount of F. sinkiangensis in yellow (diameter exceeding 0.3 m) was about 3 191. However, the sample investigation on amount of F. sinkiangensis in yellow (diameter exceeding 0.3 m) was about 2 752. The error between them was 14%. The monitoring time and range for F. sinkiangensis by low altitude remote sensing were also ensured. It was concluded that low altitude re-mote sensing had the advantage of quickly receiving distribution situation of F. sinkiangensis, which can effectively evaluate dynamic changes of F. sinkiangensis in Xinjiang.
7.Biomechanical study of swan-like memory compressive connector for treating fractures and nonunions of humerus

Objective:To study the biomechanical basis of swan like memory compressive connector (SMC) for treating fractures and nonunions of humerus. Methods:The study was performed by electrometric, two and three dimensional (3D) photoelasticity techniques. By gauging and multiple time standardization, load strain curve was got, then the dynamic memory force of SMC was calculated. The dynamically compressive force of swan neck compressive part was calculated by 2D photoelasticity. By precision moulding, the epoxy resin 3D photoelasticity humeral model was established, then it was fixated by SMC, meanwhile, an epoxy resin cirque was used to share the continuously memorial compress. The 3D stress distribution of humeral model was got, and 6 space component stress of the inspected node was divided by Leize polarizing microscope. Results:The continuously memory force of SMC was 110.35 N. The longitudinal dynamical compressive memory force was 163.88 N. 3D photoelasticity method showed that the swan neck compressive part of SMC, swan body connecting part of SMC and swan wing holding part of SMC were in consistent with each other, memorably holding bone, 3D fixation style was formed, and overall stress distribution was produced.. Conclusion:The stress field produced by 3D memory fixation of SMC is good for the treatment of fractures and nonunions. Early function exercises are allowed, and the fracture healing is prompted.
8.A dosimetric study of volumetric modulated arc therapy with a simultaneous integrated boost for preoperative chemoradiotherapy in patients with locally advanced rectal cancer

Objective To investigate the dosimetric feasibility of volumetric modulated arc therapy (VMAT)with a simultaneous integrated boost(SIB-VMAT58.75 Gy)for preoperative chemoradiotherapy in patients with locally advanced rectal cancer(LARC),and to provide a basis for clinical practice.Methods Nine patients with stage Ⅱ-Ⅲ rectal cancer who underwent preoperative concurrent chemoradiotherapy were involved in the study,and two plans were performed for each patient:SIB-VMAT58.75 Gy and VMAT50.00 Gy. For the SIB-VMAT58.75 Gy plan,the prescribed dose was 58.75 Gy(2.35 Gy/fraction)for the local rectal tumor and positive lymph nodes(GTV 58.75 Gy),and 50 Gy(2 Gy/fraction)for the regions at high risk of harboring microscopic disease(pelvic lymphatic drainage area)(PTV 50Gy).For the VMAT50.00 Gy plan,the prescribed dose was 50 Gy(2 Gy/fraction)for the regions at high risk of harboring microscopic disease(pelvic lymphatic drainage area)without a boost. The conformity index(CI),homogeneity index (HI),and dose for target areas and organs at risk(OAR)were assessed according to the dose-volume histogram. The paired t-test or nonparametric rank test was used to compare the differences between the two plans. Results Both plans met the prescription goal for PTV dose coverage. There was no significant difference in CI for the PTV between the two plans(1.0±0.0 vs. 1.0±0.0,P>0.05).The SIB-VMAT58.75 Gy plan had a worse HI than the VMAT50.00 Gy plan(0.2± 0.2 vs. 0.1± 0.0,P<0.05).There was no significant difference in V10-V50of the small intestine,bladder,femoral heads,and pelvis between the two plans(P>0.05),but D 2 cm3of the small intestine was significantly higher in the SIB-VMAT58.75 Gy plan than in the VMAT50.00 Gy plan(P=0.038). Conclusions The SIB-VMAT58.75 Gy plan for LARC achieves required target volume dose coverage and OAR dose constraints,which is safe and feasible in terms of dosimetry,and its clinical efficacy and adverse effects need further evaluation.
