1.The antagonistic effect of notoginsenoside-Rg_1 injected in striatum on immunoinflammatory injury in rat model of Parkinson's disease

Objective:To study the effect of notoginsenoside-Rg 1 on immunoinflammatory injury in rat model of Parkinson′s disease.Methods:The rat model of Parkinson′s disease was established and treated with notoginsenoside-Rg 1 by injecting in the striatum.The change of rotational behavior,levels cytokines in striatum and splenocyte proliferation were measured.Results:After treatment with notoginsenoside-Rg 1 the rotational behavior and levels of TNF-?,IL-1?,IL-6 were decreased.The splenocyte proliferation was increased.Conclusion:Notoginsenoside-Rg 1 can regulate the immunological function and reduce the pathological injury to dopaminergic neuron caused by immunoinflammatory reaction.
2.Experimental study on removal melanin granules from acellular dermal matrix of giant nevus.

OBJECTIVETo study the possibility of removal melanin granules from autogenic acellular dermal matrix of giant nevus tissue by H2O2 bleaching technique.
METHODSA total of 32 skin specimens (0.5 cm x 0.5 cm) from giant nevus tissue and 1 piece (0.5 cm x 0.5 cm) of normal skin were obtained from the surgical removal. One giant nevus tissue was chosen as control. The others and the normal skin tissue were treated with solution of 0.25% Dispase II for digestion for 24 hours under normal temperature to remove epidermis. Then each piece was immerged into solution of 0.5% Triton X-100 for digestion for 48 hours in normal temperature. One giant nevus tissue and the normal skin tissue were chosen as control. The others were immerged into solution of different concentrations of H2O2, treated under different temperature and lasting for different period. Lastly, all specimens were treated with HE staining, immunohistochemical staining, light microscopy and so on.
RESULTSAfter giant nevus tissues were treated with solution of 0.25% Dispase II and immerged into solution of 0.5% Triton X-100 in normal temperature, nevus cells and all other cellular components of pigmented nevus tissues can be effectively removed, there were the cavities left by removal of cells without any residual cell debris, but still remaining part of pigment. Then each specimen were immerged into solution of different concentrations of H2O2, under different temperature and lasting for different period which can remove residual melanin granules. In solution of 3% H2O2 for 36 h under 37 degrees C, can remove all the melanin particles, the content of collagen type I in the obtained specimen was not changed. Collagen fibers were uniform in thickness, regular in arrangement with no obvious degeneration.
CONCLUSIONSWith solution of 0.25% Dispase II and solution of 0.5% Triton X-100 in normal temperature, all cells in nevus tissue can be removed effectively. Further treatment with 3% H2O2 at 37 degrees C for 36 h can remove all the melanin particles, while collagen type I has no obvious change. The preparation of acellular dermal matrix of the giant nevus may possibly be applied as autologous tissue implant to repair tissue defects.

3.Clinical application of dynamic contrast-enhanced MRI of the breast

Objective To evaluate the clinical application of dynamic contrast-enhanced MRI of the breast.Methods In this study, 56 cases received dynamic contrast-enhanced MRI of the breast, among them 37 cases with a total 45 solid masses were confirmed by pathology.The clinical value of breast MRI was analyzed retrospectively with the combination of surgery and pathology.Results Except for multiple bilateral cysts in one case among the 45 solid masses, 17 masses were benign and 28 were malignant.MRI detected multiple carcinoma in 2 cases, residual carcinoma in one case and intraductal carcinoma in 2 cases.The smallest mass detected was 4 mm in diameter.Tumor detection rate in this series was 100%, with the diagnostic sensitivity, speciality and accuracy being 92%, 92% and 90% respectively.Conclusion Dynamic contrast-enhanced MRI of the breast has high sensitivity and helps to make a accurate diagnosis.
4.Influence of Shenmai injection for the postoperative infection and related serum indexes of patients with breast cancer

Objective To observe the influence of Shenmai injection for the postoperative infection and related serum indexes of patients with breast cancer,in order to provide evidence for the prevention of patients'postoperative infection.Methods 90 patients with breast cancerwere selected as the study object,and all the patients were randomly divided into control group(conventional postoperative treatment group)45 cases and observation group (conventional treatment and Shenmai injection group)45 cases,then the postoperative infection rates,inflammatory indexes and immune indexes before the treatment and after the treatment at different time were compared .Results The total postoperative infection rate of observation group was one case (2.22%),and it had no obviously difference compared with four cases (8.89%) of control group,the inflammatory indexes including PCT, TNF-α, IL-6 and hs-CRP at first,third and seventh day after the treatment were all lower than those of control group ,the immune indexes including CD3 +、CD4 +、CD8 +及 CD3 +CD4 +were all better than those of control group ( all P<0.05 ) .Conclusion The Shenmai injection can effectively control the postoperative infection rate of patients with breast cancer,and it also plays active improvement role for the infection related indexes.
5.Measurement of hip and thigh profile in young females from the central plain area of China and its clinical significance

Objective To investigate the relevant data and aesthetic standards of hip and thigh profile in young females from the central plain area of China, aiming to provide a credible reference for the clinical application. Methods Measurements were made in 551 young healthy females aged 16 to 21 years, including waist circumference, hip circumference, maximum thigh circumference, thigh length and the ratio between thigh length and height. Based on these data, the features of the hip and thigh profile were analyzed. Results Waist circumference, hip circumference, maximum thigh circumference, thigh length and the ratio between thigh length and height were ( 70. 74 ± 5.86 ), ( 87.16 ± 4. 19 ), ( 51.87 ± 3.27 ),(42.35 ±2.38) cm and (26. 1 ± 1.23) %, respectively. The hip and maximum thigh circumference showed significant difference between different height and weight groups. The thigh length and the ratio between thigh length and height were positively correlated with height. Conclusion The parameters of hip and thigh profile in young females from the central plain area are of significant values in providing quantitative references for the plastic surgery and evaluation of the operation,especially in liposuction.

8.Effect of collateral circulation on prognoses of patients with acute cerebral ischemia after thrombolysis

Objective The goal of this study is to compare the prognosis of recombinant tissue plasminogen activator (rt-PA) thrombolysis for middle cerebral artery (MCA) occlusion with patients with good and poor cerebral collateral circulation.Methods This retrospective study included 49 patients diagnosed with acute MCA occlusion and treated with rt-PA in the First Affiliated Hospital of Nanjing Medical University between October 1,2014 and February 1,2016.Patients were divided into good collaterals group (n =31) and poor collaterals group (n =18) according to their distribution of leptomeningeal arteries with CTA.Thirty day mortality rate,the incidence of symptomatic intracranial hemorrhage,24h and 30 day Stroke scores with National Institute of Health Stroke Scale (NIHSS) were compared between the two groups.Corrected chi-squared test,Fisher's exact test,or t test was used to statistical analysis as appropriate.Results The 30 day mortality rate of good collaterals group was significantly lower than that of poor collaterals group (0% vs.16.7%,P < 0.05).There were no significant differences in the incidence of symptomatic intracranial hemorrhage and 24h NIHSS score between the two groups (P > 0.05),however,30 day NIHSS score of good collaterals group was significantly lower than that of poor collaterals group (7.2 ± 3.1 vs.9.6 ± 2.7,P < O.05).Conclusion For patients with MCA occlusion and receiving intravenous thrombolysis,good cerebral collateral circulation may reduce their mortality and improve their clinical outcome after thrombolysis.However,good cerebral collateral circulation does not reduce the risk of symptomatic intracranial hemorrhage in those patients.

Objective To establish an easily reproducible rabbit model of acute pulmonary embolism (APE) with right ventricular dysfunction (RVD).Methods Two gelfoam strips (5 rnm×5 mm× 10 mm) were squeezed and were introduced into the pulmonary arteries of each healthy rabbit (n=12).Pulmonary and systemic hemodynamic function were recorded.All rabbits underwent CT pulmonary angiography (CTPA) and pathological examination after the introduction of APE.Results All gelfoam strips located in the bilateral lower lobe arteries.Compared with baseline mean pulmonary artery pressure (mPAP) ([9.75±1.75] mmHg),mPAP increased to (20.58 ± 5.86) mmHg immediately after embolism (P < 0.001),and then decreased to (18.78 ±4.80) mmHg 1 h after embolism (P<0.001).Right ventricle/left ventricle diameter ratio (RV/LV) increased from baseline (0.67±0.09) to (1.90±0.28) 45 min after embolism (P<0.001).Conclusion An easily reproducible rabbit model of APE with RVD are established and may be suitable for study of APE pathophysiology.
10.Expression of recombinant human tumor suppressor NDPK-A in E. coli

AIM: To construct E. coli expression plasmid of recombinant human NDPK-A with a 6?His tag, optimize the expression condition and identify the activity of the product. METHODS: nm23-H1 was subcloned from plasmid pBVNMH1 to pQE40 which contain 6?His purification tag. The expression condition was modulated in grades to get the optimal expression. We purified protein with the Ni+-NTA affinity chromatography column, identified the immunogenicity of the product with Western blot, and measured the kinases activity with HPLC. In addition, angiogenesis inhibition activity of rhNDPK was identified by CAM. RESULTS: The sequence of nm23-H1 subclone in pQE40 was exactly correct. The expression rate of rhNDPK-A was 49 6%. Purified rhNDPK-A specially recognized the antiserum of NDPK-A. It also inhibited angiogenesis. CONCLUSION: PQE-nm23H1 containing 6?His can express target protein at high level. This purification method is simple than other methods, and the product has the same activity as natural human NDPK-A.
