Objective To investigate the retinoic acid incubation effect on proliferation of U251 cell line and effect on MAPK signal pathway. Methods ATRA solution of different concentration on the U25 1 glioma cells were incubated, the influence of ATRA on the proliferation of U25 1 cells were detected, and the proteins of MKPs and MAPK signaling pathways were detected by qRT-PCR and Western blot.Using Graph Prism 5 software for quantitative analysis of experimental results. Results Compared with control group,ATRA could effectively inhibit the proliferation of U25 1 glioma cells, in a concentration dependent manner.QRT-PCR results showed that, different concentrations of ATRA after incubation for 48 hours, the expression of MKPs mRNA changed, but the changes of MKP-5 and expression of 67LR was different, explained the main differences between the two methods of the MAPK signaling pathway was the regulation of MKP-5.Western blot results showed that the ATRA,after 48 hours of incubation, the protein MAPK pathway had changed in phosphorylation, which showed that ATRA protein in the MAPK signaling pathway through control of the degree of phosphorylation on U25 1 cell line regulation. Conclusion Retinoic acid and retinoic acid receptor play its physiological effects and regulate human glioma cell line U25 1 proliferation through different combination.Retinoic acid could not only reduce the expression of phosphorylated ERK1/2 to inhibit tumor proliferation, but also regulate three kinds of protein phosphorylation, therefore its mechanism will be more complex,at the same time that the MAPK signaling pathway plays a crucial role in tumor proliferation process.

7.Effects of Candesartan on the Expression of Cerebral NAD(P) H Oxidase mRNA in Salt-loaded Stroke-Prone Spontaneously Hypertensive Rats
Chinese Journal of Hypertension 2007;0(03):-
Background Oxidative stress has been implicated in the pathogenesis of angiotensin Ⅱ(Ang Ⅱ)related hypertension.Superoxide anion, produced by NAD(P)H oxidase, plays important roles in hypertension and its complications.Ang Ⅱ infusion increases cerebral NAD(P)H oxidase activity, which is inhibited by angiotensin Ⅱ receptor blocker(ARB)candesartan and NAD(P)H oxidase inhibitor. However, the underlying mechanism of ARB in preventing hypertensive stroke is not elucidated clearly. Objective To investigate the effects of candesartan on the expression of cerebral NAD(P)H oxidase mRNA in salt-loaded stroke-prone spontaneously hypertensive rats(SHRsp). Methods Twelve-week-old salt-loaded SHRsp were treated with candesartan [1.0 mg/(kg?d)],trichlormethiazide [TCM,1.6 mg/(kg?d)] or vehicle(n=12 in each)for 2 weeks.Age-matched salt-loaded WKY rats were served as control(n=12).Blood pressure was measured every week. After two weeks, cerebrums were harvested and 24 h urine was collected.Urinary albumin was examined by ELISA.Cerebral cortex NAD(P)H oxidase subunits(p22phox,p47phox and gp91phox)mRNA expression were assayed by real-time PCR.Results The systolic blood pressure was increased significantly in salt-load SHRsp.Candesartan and TCM reduced SBP to the similar level.Urinary albumin excretion and cerebral cortex NAD(P)H oxidase subunits were markedly higher in salt-loaded SHRsp than those in salt-loaded WKY rats.Incidence of stroke was significantly reduced in candesartan treated group as compared with TCM treated SHRsp(P

Journal of Biomedical Engineering 2013;30(6):1239-1243
In the present study, the accuracy of the 4-dimensional model single photon emission computed tomography (4D-MSPECT) and quantitative gated single photon emission computed tomography (QGS) was investigated for assessing left ventricular end-diastolic (EDV), end-systolic volume (ESV) and ejection fraction (EF) from gated myocardial perfusion imaging (G-MPI) using left ventriculography (LVG) as reference. From December 2008 to June 2011, 85 patients, who underwent rest G-MPI and LVG (within 30 days) in West China Hospital, Sichuan University, were retrospectively recruited. EDV, ESV, and EF were calculated from G-MPI using 4D-MSPECT and QGS. Eighty-five patients (47 men, 38 women; age 57 +/- 13 years) were finally analyzed. Correlation between results of G-MPI and LVG was high for EDV, r = 0.89 (4D-MSPECT), r = 0.81 (QGS); ESV, r = 0.97 (4D-MSPECT), R = 0.95(QGS); EF, r = 0.95 (4D-MSPECT), r = 0.93 (QGS). 4D-MSPECT and QGS underestimated EDV significantly compared with LVG [(125 +/- 20) mL (4D-MSPECT), (118 +/- 39) mL (QGS), (131 +/- 33)mL (LVG)]. The ESV, 4D-MSPECT and QGS values did not differ significantly from LVG [(47 +/- 32) mL (4D-MSPECT), (53 +/- 29) mL (QGS), (49 +/- 37) mI (LVG)]. For LVEF, only QGS yielded values were significantly lower than LVG [61% +/- 21% (4D-MSPECT), 55% +/- 17% (QGS), and 63% +/- 19% (LVG)]. EDV, ESV, and EF as determined by 4D-MSPECT and QGS from G-MPI agree well with relevant values with LVG. However, Algorithm-inherent also showed slightly over- or under-estimation of volumes. Therefore, separated normal databases should be set up for each algorithm.

Chinese Journal of Geriatrics 2015;34(4):431-433
Objective To investigate the expression changes of fractalkine (FKN)in focal cerebral ischemia and reperfusion penumbra, and to explore its variation law and role in the inflammation of cerebral ischemia-reperfusion injury. Methods The cerebral ischemia reperfusionmodel was established by intraluminal thread occlusion in the middle cerebral arteries occlusion (MCAO).FKN protein expression in focal cerebral ischemia and reperfusion penumbra was detected by immunohistochemistry and Western blot. Results The results of immunohistochemistry stain showed that the chemokine FKN was expressed in a low level in the normal group and the sham operation group, and there were no significant differences among the two groups (P> 0.05).Compared with the humbers of FKN in normal group (37.03± 6.28) in focal cerebral ischemia and reperfusion penumbra, the expression of FKN in model group was increased after 3 h of reperfusion (48.58±7.29) (P<0.05),peaked at 24 h (112.08±8.26) (P<0.05],and then decreased gradually at day 7 after reperfusion, but had no significant difference (40.73 ± 4.02) (P> 0.05).FKN was expressed in a low level in the sham operation group (0.527±0.002),then up-regulated after 3 h of reperfusion [(0.598±0.004),P<0.05],peaked at 24 h [(0.833±0.005),P<0.05],maintained a high level till 48 h after reperfusion [(0.735±0.002),P<0.05],and return to baseline level at day 7 after reperfusion [(0.533±0.004),P>0.05].Conclusions Fractalkine is upregulated after focal cerebral ischemia/reperfusion,and has a dynamical change, which indicates that fractalkine might involve in the inflammatory process after cerebral ischemia-reperfusion injury.
