1.The shRNA-mediated downregulation of protein kinase D-2 enhanced chemosensitivity of Tca8113.

OBJECTIVETo explore the effect of silencing protein kinase D (PKD)-2 on Tca8113 cell proliferation, programmed cell death, and chemosensitivity.
METHODSThe stable cell lines of pkd-2 gene silencing and empty vector plasmid group were established. The proliferation and 50% inhibitory concentration (IC50) of shRNA-mediated Tca8113 chemotherapy drugs were detected through methyl thiazolyl tetrazolium assay (MTT). The programmed cell death rate and sensitivity to Tca8113 chemotherapy drugs before or after pkd-2 gene silencing were measured through flow cytometry. P-glycoprotein (P-gp) expression of pkd-2 silencing cells was identified by immunohistochemical methods.
RESULTSStable cell lines of pkd-2 gene silencing were established. Compared with parental cells, the proliferation of shRNA-mediated Tca8113 was not significantly different, but its IC50 was lower. Meanwhile, cell programmed death rate and sensitivity to chemotherapeutic drugs of shRNA-mediated Tca8113 significantly increased. Compared with wild group Tca8113, a significant decrease in P-gp expression was induced by chemotherapy drugs with shRNA-pkd-2 gene silencing.
CONCLUSIONThe pkd-2 gene of shRNA interference silencing Tca8113 promotes programmed cell death of Tca8113, reduces the IC50 of the chemotherapy drugs, and significantly improves the sensitivity of Tca8113 to chemotherapeutic drugs while reducing the expression of P-gp.

2.Effects of Melatonin on Oxidative Stress and Neuronal Apoptosis in Hippocampus of Rats with Epilepsy

Objective To investigate the effects of melatonin on oxidative stress and neuronal apoptosis in hippocampus of epileptic rats and the mechanism. Methods Seventy-two adult male Sprague-Dawley rats were equally divided into control group, model group, low dose group and high dose group. The model group was injected coriamyrtin 50μg/kg in the lateral ventricle, while the low dose group and high dose group were injected melatonin 20 mg/kg and 60 mg/kg, respectively, 30 minutes before injection of coriamyrtin. The contents of malo-ndialdehyde (MDA) and superoxide dismutase (SOD) were detected with ultraviolet spectrophotometer, the apoptosis was detected with TU-NEL, and the ultrastructural changes of neurons and mitochondria in hippocampal CA3 region were observed with electron microscopy, af-ter 60 minutes of epilepsy. Results The neurons and mitochondria in hippocampus were damaged, the number of apoptotic cells significant-ly increased (P<0.001), the content of SOD decreased (P<0.001), and the content of MDA increased (P<0.001) in the model group, com-pared with the control group. In the low dose group, the ultrastructural damage relieved, the number of apoptotic cells decreased (P<0.01), the content of SOD increased (P<0.05), and the content of MDA decreased (P<0.05);and for the high dose group, the ultrastructural damage relieved very much, the number of apoptotic cells decreased (P<0.001) and was not significantly different from the control group (P>0.05), SOD increased (P<0.001), and MDA decreased (P<0.001), compared with the model group. Conclusion Exogenous melatonin may signifi-cantly reduce neuronal apoptosis in rat hippocampal after epilepsy, and high dose is more effective, which may relate with resistance of oxi-dative stress, alleviate neuronal mitochondrial damage.
3.Influence of evidence based nursing strategies on negative emotion and daily behavior in aged patients with coronary heart disease

Objective: To explore influence of nursing strategies based on evidence based medical theory on negative emotion and daily behavior in aged patients with coronary heart disease (CHD).Methods: A total of 88 CHD patients, who were treated in our department from Oct 2012 to Mar 2015, were selected.According to random number table, they were randomly and equally divided into routine nursing group and evidence based nursing group (received evidence based nursing based on routine nursing group).Unhealthy lifestyles, scores of self-rating anxiety scale (SAS) and self-rating depression scale (SDS), satisfaction for nursing and incidence rate of major adverse cardiovascular events (MACE) were measured and compared between two groups before and four months after intervention.Results: Compared with routine nursing group after four-week intervention, there were significant reductions in percentages of smoking (40.9% vs.22.7%), drinking (54.5% vs.38.6%), high-fat diet (43.2% vs.29.5%), physical inactivity (45.5% vs.25.0%) and overweight (52.3% vs.29.5%), scores of SAS [(50.4±3.3) scores vs.(45.8±3.2) scores] and SDS [(53.4±4.7) scores vs.(48.0±4.2) scores], and incidence rate of MACE (25.0% vs.11.4%), and significant rise in satisfaction for nursing (86.4% vs.97.7%) in evidence based nursing group (except for the P of overweight was <0.01, others were P<0.05 all).Conclusion: Evidence based nursing mode can improve unhealthy lifestyle and relieve negative emotional status in aged patients with coronary heart disease, which is worth extending in clinic.
4.Meta- analysis of the effect of single hand pipe method on venous indwelling needle puncture

Objective To evaluate the effect of single hand pipe method on venous indwelling needle puncture. Methods Literature about the impact of single hand pipe method on venous indwelling needle puncture was retrieved from digital databases of PubMed, Medline, EMbase, The Cochrane Library, SinoMed, VIP database, China National Knowledge Infrastructure(CNKI), and WanFang Data. The quality of literature was evaluated by the Cochrane Handbook (5.1.0). The Meta data was analyzed by RevMan 5.3. Results Ten random control trials were included . Ten studies indicated that single hand pipe method increased success rate of puncture[related to the risk(RR)=1.16, 95%confidence interval(CI) 1.11-1.22]. And, it decreased the complication rate (3 studies) (RR=0.32, 95%CI 0.13-0.79) and pain rate(2 studies) (RR=0.25, 95%CI 0.13-0.47) as well as shorten the time of tube(2 studies) (WMD:-1.68, 95%CI-2.44--0.93). Conclusions Single hand pipe method can improve a puncture success rate and reduce complications and pain rate and shorten the time of tube, so that get more time to rescue the patient, and improve the level of the working efficiency and nursing for the nursing staff. In addition, to some extent, save manpower. It is worth being popularized.
5.Research on mental status and coping style in aged cardre ward patients with long- term hospitalization

Objective To explore the prevalence of anxiety, depression, loneliness in elderly patients with long- term hospitalization, its coping style and the correlation between them. Methods 48 hospitalized elderly patients with long- term hospitalization were investigated by Standard Scale and self- administrated questionnaire, the Pearson's correlation was used to find the correlation. Results It was concluded that anxiety, depression and other negative emotions were commonly seen in elderly patients with long term hospitalization, these patients usually adopt the avoidance and yield coping style as well,and there was a positive correlation between them, the r value was 0.438, 1.473 amd 0.501 respectively. Conclusions This research confirmed the possibility of negative emotion in elderly patients with large long- term hospitalization, but also closely related to the choice of coping styles. The nursing staff should closely observe and assess the psychological status of elderly patients, planned, targeted to help elderly patients use more positive coping style.
6.Th17 cells and psoriasis

Psoriasis is a common autoimmune disease and mainly affects skin,joints,or both.Psoriasis is also a chronic relapsing disorder that can cause physical and psychological burdens to patients.Currently it is widely accepted that the immune system is involved in the development of psoriasis.Th17 cells,a subtype of CD4 +T lymphocytes,are characterized by its ability to secrete proinflammatory cytokine IL-17.Recent studies indicate that Th17 cells play a predominant role in the pathogenesis of psoriasis and other immune-mediated inflammatory diseases.Moreover,targeted therapies have been developed and approved for the treatment of moderate-to-severe plaque psoriasis or psoriatic arthritis.This review summarizes the role of Th17 cells in the pathogenesis of psoriasis and several therapeutic biologics targeting this pathway in psoriasis.
7.Comparative study on the relationship between loneliness and the quality of life of urban and rural absent parents in the Inner Mongolia Autonomous Region

Objective To know the relationship between loneliness and the quality of life of urban and rural absent parents in the Inner Mongolia Autonomous Region,clear the influence factors of loneliness in the absent parents.Methods 441 cases of urban and rural absent parents were selected from the Chifeng district of the Inner Mongolia Autonomous Region were valuated with the general information questionnaire,UCLA Loneliness Scale and MOS 36 item short form health survey (SF-36).Results Rural cases had a higher level of loneliness when compared with the urban cases,t=4.60,P<0.01.There were significant differences in the general score and the scores of each dimension between the two groups,P<0.05.Loneliness score was significantly negative correlation with the quality of life in all dimensions,P<0.01.Regression analysis showed that health,marital status,income and the education level were the main influence factor of loneliness in the urban and rural absent parents.Conclusions Compared with the urban absent parents,rural cases had more obvious loneliness and lower quality of life.Loneliness levels influence the quality of life of absent parents.Loneliness condition of urban and rural absent parents were influenced by various factors,the cases with characteristic of frail,single,low education level and poor economic condition should be the focus group to intervene.
8.Relationships among psychological capital, social support and mental health of medical staffs

ObjectiveTo explore the relationships among psychological capital,social support and mental health of medical staffs.MethodsAccording to convenience sampling principle,101 medical staffs from one three-grade class A hospital in Hebei participated in the study.They were investigated with questionnaires such as Psychological Capital Questionnaire ( PCQ-24),Social Support Rating Scale (SSRS) and Symptom Checklist 90(SCL-90).Results①The scores of self-efficacy,hope,resilience and optimism were(3.73 ±0.58 ),(3.59 ±0.60),(3.81±0.49),(3.70 ± 0.32),all of which are higher than middle points.There were statistic difference in four subscales of psychological capital(P < 0.05 ).Compared with nurses,doctors scored higher in self-efficacy ( (3.92 ±0.66),(3.59 ±0.48) ).The difference was significant(P<0.05).Among different educational background there were also statistic differences(P < 0.05 ).②Psychological capital was negatively related to the whole factors of SCL-90(P<0.0l,or P < 0.05 ),however,social support was negatively related to only part of it(P <0.05 ).③Psychological capital could negatively predict mental health(P < 0.01 or P < 0.05 ).ConclusionThis research outlines a strong relationship between psychological capital and mental health of medical staffs.But social support has no a significant impact on mental health.
9.Topical aminolevulinic acid-photodynamic therapy in acne

Objective To investigate the effect and safety of photodynamic therapy (PDT) with topical aminolevulinic acid (ALA) in moderate to severe acne. Methods Seventy patients with moderate to severe acne were randomly divided into two groups, one group was treated with topical ALA-PDT for 1-3 sessions with an interval of 2 weeks, the other group received oral isotretinoin 10 mg twice a day for 6 weeks. Patients were evaluated for efficacy on week 2, 4 and 6 after the initial treatment. A 3-month follow up was performed to monitor adverse events following the end treatment. Results The total effective rate was 97.1% in ALA-PDT group after 1-3 sessions of treatment, 80.0% in the control group on week 6, and the difference was significant between the two groups (P<0.05). Acne lesions tended to recur more slightly with a longer remission period in ALA-PDT group than in the control group. A temporary and local pigmentation was observed in some patients treated with ALA-PDT, but no scar was noticed. Conclusion Topical ALA-PDT is a simple and effective treatment option for moderate to severe acne with mild side effects.
10.Clinical significance of S-100B protein in children with different degrees of amniotic fluid meconium contamination

Objective Detection of S-100B protein in different degree meconium pollution level in serum of children with value.MethodsIn 2012 June to 2013 December in our department were simple meconium stained amniotic fluid in term newborns in 73 cases, and set up for the observation group, and according to the amniotic fluid pollution degree is divided into 47 cases of amniotic fluid of Ⅰ-Ⅱdegree pollution group and 26 cases in grade Ⅲmeconium group during the same period, selected 20 cases without amniotic fluid contamination in term healthy newborns for the control group, the groups were compared in S-100B protein content difference.ResultsⅢmeconium stained amniotic fluid were within 6h serum S-100B was significantly higher than that in control group(P<0.01), and the degree of contamination of amniotic fluidⅠ-Ⅱgroup compared with the control group, no significant difference(P>0.05).Ⅲ meconium stained amniotic fluid in children with 72h also increased.ConclusionThird degree meconium stained amniotic fluid but normal Apgar score of newborns may still exist in clinical brain injury, so pay close attention to.
