1.Relationship beween cerebrospinal fluid changes and prognosis in patents with neurosyphilis

Objective To investigate the relationship between cerebrospinal fluid (CSF) changes and prognosis in patients with neurosyphilis.Methods The CSF and the clinical data of 55 syphilis patients were analysed retrospectively,including 30 neurosyphilis cases and 25 syphilis cases during incubation period.The CSF leukocyte, protein and pressure of the two groups were measured,meanwhile, rapid plasma regain circle card(RPR) and treponema pallidum particle assay (TPPA) were simultaneous determination. All cases were treated with penicillin G normally during the whole range.Two groups of patients were followed-up by telephones or outpatient department in two year and the prognosis were evaluated. 2.Acupoint Sticking at Shenque (CV 8) with Ginger-preparedBan Xia (Rhizoma Pinelliae) for Nausea and Vomiting Induced by Amifostine

Objective: To observe the treatment effect of acupoint sticking at Shenque (CV 8) with ginger-preparedBan Xia (Rhizoma Pinelliae) on nausea and vomiting induced by Amifostine for myelodysplastic syndromes (MDS).
Methods: Totally 124 eligible subjects intervened by Amifostine were randomized into 2 groups by the visiting order,an observation group and a control group,62 in each group. The control group was intervened by conventional treatment, while the observation group was by acupoint sticking at Shenque (CV 8) with ginger-preparedBan Xia (Rhizoma Pinelliae) in addition to the same conventional treatment. The occurrence rate of nausea and vomiting in the two groups were observed.
Results: After intervention, the occurrence rate of nausea and vomiting in the observation group was significantly lower than that in the control group (P<0.01).
Conclusion: Acupoint sticking at Shenque (CV 8) with ginger-prepared Ban Xia (Rhizoma Pinelliae)can produce a content effect on nausea and vomiting induced by Amifostine for MDS.
3.Detection of the tumor markers in patients with gastric precancerous lesions

It is confirmed that chronic atrophic gastritis (CAG) caused by Helicobacter pylori is the main cause of gastric precancerous lesions.CAG is also the key determinant in gastric cancer risk assessment,which affects pepsinogen and gastrin-17 secretion.Most of the gastric cancer patients have poor prognosis,and non-invasive tools for gastric cancer screening and diagnosis are lacking.Therefore,the early detection of gastric cancer in order to reduce the disease mortality is necessary.Pepsinogen and gastrin-17 are biomarkers of gastric mucosa and gastric antra.The serological testing for the stomach-specific biomarkers offers the possibility to know preneoplastic gastric mucosal conditions.
4.Effect of long-term glucocorticoid administration on nondepolarizing muscle relaxants-induced neuromuscular blockade in rat diaphragma

Objective To evaluate the effect of long-term glucocorticoid administration on nondepolarizing muscle relaxants-induced neuromuscular blockade in rat diaphragma in vitro.Methods Forty adult male SpragueDawley rats,aged 8 weeks,weighing 180-220 g,were randomized into 2 groups (n =20 each):control group (group C) and dexamethasone group (group D).In group D,dexamethasone 0.6 mg/kg was injected intraperitoneally once a day for 14 consecutive days,while the equal volume of normal saline was injected instead of dexamethasone in group C.The left phrenic nerve-hemidiaphragm was removed at 24 h after the last injection to evaluate the effect of d-tubocurarine.Different concentrations (0-10 μmol/L) of d-tubocurarine were added.The d-tubocurarine concentration-effect curve was drawn.The half inhibitory concentration and 95 % confidence interval of d-tubocurarine were calculated.Right diaphragma was removed for determination of the expression of embryonic nicotinic acetylcholine receptor (nAChRγ) mRNA and adult nicotinic acetylcholine receptor (nAChRε) mRNA by RTPCR.Results Compared with group C,the concentration-effect curve was shifted to the right,the half inhibitory concentration was significantly increased,and the expression of nAChRγ mRNA and nAChRε mRNA was up-regulated in group D (P < 0.05).Conclusion Long-term glucocorticoid administration can weaken nondepolarizing muscle relaxants-induced neuromuscular blockade in rat diaphragma in vitro,and up-regulation of nAChR mRNA expression may be involved in the mechanism.
5.Analysis of necessity and tolerance of operation for hip fracture in the elderly

Objective To investigate the necessity and tolerance of operation for hip fracture in patients aged 75 years and over. Methods The general tolerance including the self-care ability, comorbidities and postoperative complications was retrospectively analyzed in 70 elderly patients with hip fracture. Then the relationship between operation and survival status was estimated. Results The average tolerance score was significantly higher in 46 patients who accepted the operation than in 24 patients who didn't C(76. 1±12. 2) vs. (65. 5±13. 9), P<0. 01]. The median survival period was significantly higher in operation group than in non-operation group ( 97 mouths vs. 48 mouths, P< 0. 05). Kaplan Meier analysis showed that the survival status was better in operation group than in non-operation group. Conclusions For elderly patients over 75 years old with hip fracture, comprehensive evaluations of surgical tolerance before operation are very important for assessing the perioperative risk and feasibility of surgery. The operative treatment is a reasonable choice, as long as the operation is well tolerated by the patients.

8.Research of Human-mouse Chimeric Antibodies Against Ebola Virus Nucleoprotein.

The Ebola virus is highly infectious and can result in death in ≤ 90% of infected subjects. Detection of the Ebola virus and diagnosis of infection are extremely important for epidemic control. Presently, Chinese laboratories detect the nucleic acids of the Ebola virus by real-time reverse transcription-polymerase chain reaction (RT-PCR). However, such detection takes a relatively long time and necessitates skilled personnel and expensive equipment. Enzyme-linked immunosorbent assay (ELISA) of serum is simple, easy to operate, and can be used to ascertain if a patient is infected with the Ebola virus as well as the degree of infection. Hence, ELISA can be used in epidemiological investigations and is a strong complement to detection of nucleic acids. Cases of Ebola hemorrhagic fever have not been documented in China, so quality-control material for positive serology is needed. Construction and expression of human-mouse chimeric antibodies against the nucleoprotein of the Ebola virus was carried out. Genes encoding variable heavy (VH) and variable light (VL) chains were extracted and amplified from murine hybridoma cells. Genes encoding the VH and VL chains of monoclonal antibodies were amplified by RT-PCR. According to sequence analyses, a primer was designed to amplify functional sequences relative to VH and VL chain. The eukaryotic expression vector HL51-14 carrying some human antibody heavy chain- and light chain-constant regions was used. IgG antibodies were obtained by transient transfection of 293T cells. Subsequently, immunological detection and immunological identification were identified by ELISA, immunofluorescence assay, and western blotting. These results showed that we constructed and purified two human- mouse chimeric antibodies.

9.Correlation of aspirin resistance, gene polymorphism of platelet membrane glycoprotein Ⅱb HPA-3 and recurrent ischemic stroke

ObjectiveToinvestigatethecorrelationofaspirinresistance(AR),genepolymorphismof platelet membrane glycoprotein (GP)Ⅱb HPA-3 and recurrent ischemic stroke. Methods The consecutive patients w ith acute ischemic stroke and gender, age-matched healthy subjects w ere enrol ed. Adenosine diphosphate (ADP) and arachidonic acid (AA) w ere used as inducing agents. The platelet aggregation rate (PAgT) was detected by flow cytometry. AR was defined as PAgTADP≥39.27% and PAgTAA≥34.27%. Polymerase chain reaction-restriction fragment length polymorphism w as used to detect the GPⅡbHPA-3 genotype. Results A total of 224 patients w ith acute ischemic stroke (case group) and 98 healthy subjects (control group) were enroled. In the case group, 162 patients had the first-ever stroke (first-ever stroke group) and 62 had recurrent stroke (recurrent stroke group). The incidence of AR in the case group w as 15.18%, in w hich the incidence of AR in the recurrent group w as significantly higher than that in the first-ever stroke group (27.42%vs.10.49%; χ2 =9.977, P=0.002). The frequencies of bb genotype ( P=0.004) and b alele (P=0.001) in the recurrent group were significantly higher than those in the first-ever stroke group. In the case group, the frequencies of bb genotype ( P=0.028) and b alele (P=0.004) in the AR group w ere significantly higher than those in the non-AR group. Multivariable logistic regression analysis show ed that AR (odds ratio [OR] 2.933, 95%confidence interval [CI] 1.326-6.486;P=0.008) and bb genotype (OR 2.198, 95%CI1.164-4.149, P=0.015) w ere the independent risk factors for recurrent ischemic stroke. Conclusions AR and GP Ⅱb HPA-3 bb genotype are associated w ith recurrent ischemic stroke.
10.Effects of butorphanol pretreatment on myocardial damage induced by limb ischemia/reperfusion in rats

Objective To evaluate the effects of butorphanol pretreatment on myocardial damage induced by limb ischemia/reperfusion (I/R) in rats.Methods One hundred and eight male Sprague-Dawley rats,weighing 180-220 g,were randomly divided into 3 groups (n =36 each) using a random number table:sham operation group (group S),I/R group and butorphanol pretreatment group (group B).Limb ischemia was induced by occlusion of bilateral hind limbs for 2 h followed by reperfusion in I/R and B groups.At 10 min before ischemia,butorphanol 0.2 mg/kg was injected via the right jugular vein in group B,while the equal volume of normal saline was injected in group I/R.Six rats were chosen randomly before ischemia (baseline,T0) and at 2,6,12,24 and 48 h after reperfusion (T1-5) and blood samples were taken from the abdominal aorta for determination of the serum creatine kinase isoenzyme-MB (CK-MB) activity and cardiac troponin Ⅰ (cTnI) concentration.The animals were then sacrificed and myocardial specimens were obtained for determination of myocardial apoptosis (using TUNEL) and the expression of c-Jun N-terminal kinase (JNK) and extracellular signal-regulated protein kinase (ERK) (using Western blot analysis).Apoptosis index was calculated.Results Compared with group S,the serum CK-MB and cTnI levels were significantly increased at T1-5,and the apoptosis index and expression of JNK and ERK were increased at T2-5 in group I/R (P < 0.05).Compared with group I/R,the pathological changes of myocardium were significantly reduced at T4,the serum CK-MB and cTnI levels were decreased at T2-5,the apoptosis index was decreased at T4,5,the expression of ERK was up-regulated at T2-5,and the expression of JNK was down-regulated at T2-4 (P < 0.05).Conclusion Butorphanol pretreatment can reduce apoptosis in myocardial cells through activating MAPK signal transduction pathway,up-regulating ERK expression and down-regulating JNK expression,thus attenuating myocardial damage induced by limb I/R in rats.
