1.Research progress on use of polypeptide in tumor therapy
Journal of International Oncology 2010;37(9):643-646
Besides being able to specially target tumor tissues, anticancer peptides have other advantages such as easy synthesis, low immunogenicity, high potency, low toxicity, and easy access. These peptides exert their anti-tumor effects through mechanisms such as modulating immune system, killing tumor cells directly, and/or inhibiting tumor metastasis.
2.Promotion of TRIM59 on neuroglioma genesis
Journal of Shanghai Jiaotong University(Medical Science) 2017;37(6):719-725
Objective · To demonstrate the effect of TRIM59 on gliomagenesis and the molecular mechanism. Methods · TRIM59 protein expression in glioma specimens was analyzed by immunohistochemical staining, and in several glioma cell lines by Western blotting and quantative PCR.After TRIM59 was knocked down by shRNAs,cell proliferation, migration, colony formation, and orthotopic xenograft brain tumor development were detected. The signaling pathway of TRIM59 in gliomas was also explored by RNA-Seq and KEGG PATHWAY analyses. Results· The levels of TRIM59 protein expression in clinical glioma specimens were positively correlated with glioma malignancy.TRIM59 was highly expressed in LN229 and U87 glioma cells compared with normal human astrocytes.Knockdown of TRIM59 in these two cell lines with lentivirus-mediated shRNAs inhibited their proliferation, migration, and colony formation.Compared with the control xenograft models, knockdown of TRIM59 significantly inhibited glioma tumor growth.RNASeq and KEGG PATHWAY analyses identified that TRIM59 knockdown down-regulated 306 genes, among which PI3K/AKT signal pathway-related genes were the most.Moreover,TRIM59 knockdown suppressed AKT phosphorylation, whereas overexpression of a constitutively actived AKT (Myr-AKT)rescued TRIM59 knockdown-inhibited cell proliferation. Conclusion· TRIM59 is a new glioma oncogene, which may take effect through activating PI3K/AKT signaling pathway.

Objective To investigate the retinoic acid incubation effect on proliferation of U251 cell line and effect on MAPK signal pathway. Methods ATRA solution of different concentration on the U25 1 glioma cells were incubated, the influence of ATRA on the proliferation of U25 1 cells were detected, and the proteins of MKPs and MAPK signaling pathways were detected by qRT-PCR and Western blot.Using Graph Prism 5 software for quantitative analysis of experimental results. Results Compared with control group,ATRA could effectively inhibit the proliferation of U25 1 glioma cells, in a concentration dependent manner.QRT-PCR results showed that, different concentrations of ATRA after incubation for 48 hours, the expression of MKPs mRNA changed, but the changes of MKP-5 and expression of 67LR was different, explained the main differences between the two methods of the MAPK signaling pathway was the regulation of MKP-5.Western blot results showed that the ATRA,after 48 hours of incubation, the protein MAPK pathway had changed in phosphorylation, which showed that ATRA protein in the MAPK signaling pathway through control of the degree of phosphorylation on U25 1 cell line regulation. Conclusion Retinoic acid and retinoic acid receptor play its physiological effects and regulate human glioma cell line U25 1 proliferation through different combination.Retinoic acid could not only reduce the expression of phosphorylated ERK1/2 to inhibit tumor proliferation, but also regulate three kinds of protein phosphorylation, therefore its mechanism will be more complex,at the same time that the MAPK signaling pathway plays a crucial role in tumor proliferation process.
7.Clinical Characteristics and Speech Training of Bilabial Consonant Articulation Disorders
Chinese Journal of Rehabilitation Theory and Practice 2014;(8):763-766
Objective To discuss the clinical features and speech training of bilabial consonant articulation disorders. Methods 92 patients with functional articulation disorders (FAD) and 85 patients repaired cleft palate were studied. Then 20 patients with bilabial consonant articulation disorders were selected respectively from two types of patients for speech training. Results FAD patients who replaced /p/with /b/ accounted for 78.8% in patients with bilabial consonant articulation disorders, and the patients repaired cleft palate dropped or replaced /b/ accounted for 30.2% and 60.4% respectively. Both types of the patients with bilabial consonant articulation disorders made less mistakes after speech training (P<0.001). Conclusion Bilabial consonant articulation disorders mainly show unaspirated about /p/ in FAD patients, and show dropping and replacement about /b/ in the patients repaired cleft palate. The speech training is significantly effective.

Objective To investigate the effect of swimming exercise on insulin sensitivity and interleukin-1 beta(IL-1β)mRNA expression in muscle tissue of insulin-resistant (IR) rats induced by high-fat diet. Methods The IR rat model was induced by high-fat diet feeding for 10 weeks and assessed by hyperinsulinemic-euglycemic clamp technique (HEC). The rats were divided into the fllowing three groups: normal group without exercise, IR groups with and without swimming exercise. After 4 weeks swimming excersie, insulin sensitivity and serum IL-1β levels were measured. Results Glucose infusion rate (GIR) of the IR rats was significantly elevated than that of non-exercise IR rats (P<0.01). Moreover, serum IL-1β levels were reduced markedly in IR rats (P<0.01). Conclusion High-fat diet feeding can partly reverse IR of rats to normal value. Swimming exercise improves insulin sensitivity and reduces the levels of IL-1β in IR rats induced by high-fat diet.
