1.Bioactive glass modified polyethylene terephthalate promotes bone healing

BACKGROUND:Polyethylene terephthalate (PEA) holds good mechanical properties, but its biocompatibility needs to be improved. OBJECTIVE:To investigate the effect of bioactive glass modified PEA on bone healing. METHODS:In vitro experiment:PEA (control group) and bioactive glass modified PEA (experimental group) were respectively co-cultured with MC3T3-E1 cells. The cell morphology was observed at 3 days of culture, and the cell proliferation and alkaline phosphatase activity were detected at 1, 3 and 5 days of culture. In vivo experiment:24 New Zealand white rabbits were enrolled, modeled into unilateral knee ligament rupture, and then randomly allotted to two groups. Bioactive glass modified PEA and PEA were implanted into experimental and control groups, respectively. Biomechanics and histological changes were compared between groups at 6 and 12 weeks after transplantation. RESULTS AND CONCLUSION:In vitro experiment:the cell density in the experimental group was significantly higher than that in the control group after 3-day culture (P<0.05);the cell proliferation and alkaline phosphatase activity were significantly higher than those in the control group at 3 and 5 days of culture (P<0.05). In vivo experiment:the maximum axial pull-out strength in the experimental group was significantly greater than that in the control group at 6 and 12 weeks after implantation (P<0.05). In the control group, the defect was obvious and there were a large number of inflammatory cells at 6 weeks after implantation;the defect region became fuzzy, abundant inflammatory cells existed and new bone formed on the surface at 12 weeks after implantation. In the experimental group, there were newly formed bones and trabeculae formed on the interface, the scar tissue became less, and the interface width was narrow at 6 weeks after implantation;numerous new tissues were observed, and only a few inflammatory cells observed in the defect region at 12 weeks after implantation. These results indicate that the modified PEA with bioactive glass possesses good biocompatibility and biomechanics, which can promote bone healing.
2.Investigation of glucose metabolism in rats with polycystic ovary syndrome by isotope tracer

Objective To establish a rat model of polycystic ovary syndrome (PCOS),and to study the changes in glucose metabolism by isotope tracer.Methods Female SD rats aged 21 days were divided into control group and PCOS group (made by means of testosterone propionate and high-fat diet).After 8 weeks of molding,glucose tolerance test and rat tail vein perfusion experiment was conducted,by application of the principle of stable isotope tracer technique,rate of glucose appearance was compared in two groups and thus glucose metabolism was measured.Results Compared with control group,PCOS group showed polycystic ovarian changes and significantly elevated serum testosterone levels.After 8 weeks of molding,fasting blood glucose showed no significant difference,while glucose tolerance was impaired and rate of appearance of 6,6-d2-glucose increased significantly in PCOS rats.Conclusions The testosterone propionate combined with high-fat diet could induce PCOS rat model,and the PCOS rats demonstrated a significant increase of glucose rate of appearance despite of normal blood glucose level.Therefore it is suggested that during the early stage of PCOS,change in the glucose metabolism has appeared,and early intervention may be beneficial.
3.Reconstruction of the anterior cruciate ligament using Ligament Advanced Reinforcement System: its biocompatibility and bone healing

BACKGROUND: Ligament Advanced Reinforcement Sytstm (LARS) artificial ligament can avoid the interfibrillar friction compared wiht traditional ligaments, which is conductive for autogenous tissue growth. OBJECTIVE: To investigate the biocompatibility of LARS artificial ligament used for cruciate ligament reconstruction and bone healing. METHODS: Twenty goats were randomly divided into two groups: the anterior cruciate ligament insertion at the femur was cut as stump group; the anterior cruciate ligament insertions at the femur and tibia, and all the ligaments and synovial tissues between them were cut as stump resection group. LARS artificial ligament was then implanted into both two groups. At 20 months after implantation, the ligament healing was observed using CT and MRI examinations, the fibrous tissues were observed under scanning electron microscope, and the growth of newborn ligaments was observed by histological staining. RESULTS AND CONCLUSION: CT findings showed that in the stump group, the number of femoral tunnel enlargement was significantly more than that in the stump resection group (P < 0.05), while the number of tibial tunnel enlargement in the stump group was significantly less than that in the stump resection group (P < 0.05). LARS artificial ligament in the femoral and tibial tunnel and free part of the joint presented with hypointensity on T1- and T2-weighted images, which showed no significant difference between two groups. Under the scanning electron microscope there were obvious dense fibrous tissues between LARS artificial ligaments in both two groups; and the autologous fibrous tissues arranged closely and evenly under high magnification. Hematoxylin-eosin staining results showed that in the two groups, the LARS artificial ligaments in the free part of the articular cavity and bone tunnel were surrounded by a large number of autologous tissues in a fusiform arrangement; autologous fibrous tissues and artificial ligament were wrapped well but arranged irregularly; mature fibrous tissues and few capillary infiltration were found in the bone tunnel and free part of the joint cavity, but without necrotic tissues. Masson staining results showed that the inner and outer parts of the LARS in the bone tunnel were wrapped by blue collagen, the red-stained bones located outside the collagen, and no obvious transitional zone appeared. These results showed that the LARS artificial ligament holds good biocompatibility and fibrous tissue induction, which can promote bone healing.
4.Application of Wood's lamp in the diagnosis of superficial cutaneous fungal infections

Objective To estimate the performance of Wood's lamp examination in the diagnosis of superficial cutaneous fungal infections. Methods Totally, 129 patients, who were diagnosed with superficial cutaneous fungal infections according to clinical medical history and signs, were enrolled in this study. Wood's lamp examination of lesions was carried out. Cutaneous samples were obtained from the patients and subjected to microscopic examination and fungal culture. Results Wood's lamp examination was positive in 84% and 85.7% of patients with tinea versicolor and malassezia folliculitis, respectively; among these patients positive for Wood's lamp examination, 92.9% were positive for fungal culture, and 87.5% for microscopic examination. In patients clinically diagnosed with tinea manus and pedis, tinea corporis or tinea cruris, 8.3% were positive for Wood's lamp examination, while 85.4% were positive for fungal examination. There was a high consistency between Wood's lamp examination and fungal examination in patients with tinea versicolor and malassezia folliculitis, but not in those with tinea manus and pedis, tinea corporis or tinea cruris. Conclusions Wood's lamp examination shows a high specificity and sensitivity and is useful in the diagnosis of tinea versicolor and malassezia folliculitis, but seems unapplicable for the diagnosis of tinea manus and pedis, tinea corporis or tinea cruris.
5.Countermeasures for invasive catheter infections in neurosurgical intensive care unit

Objective To study the characteristics of invasive catheter infection in neurosurgery intensive care unit (NICU) to provide theoretical basis for the prevention of the infections.Method A retrospective analysis was done into the clinical data of 563 patients with indwelling catheter from January 2011 to December 2012 in our department.Results Among 563 cases undergoing invasive catheteration,there were 106 cases of catheter-associated infections with an incidence of 18.8%. The urinary catheter-associated infection was 24.5% and epidural drainage catheter-associated infection was 16.9%.Conclusion The enhanced consciousness of critical catheter infections,abiding by principles for aseptic operation and strengthened nursing to catheters or tubes are critical for reducing the incidence of invasive catheter-associated infections and ensuring the safety patients.
6.The effects of Tangzhiping granules on blood glucose and IGF-Ⅰ in T2DM rats

Objective To explore the effects of Tangzhiping granules on the blood glucose and IGF-Ⅰ in T2DM rats.Methods 40 SPF grade Wistar rats whose weight were about (200±20)g were selected as the subjects for this study,one half were male and another half were female.After be adapted to the normal food for 1 week,8 rats were selected randomly from all of rats following their weight as the blank group (ordinary feed).The remaining the others were fed with the high-fat and high-sugar food.The 6th week,the other rats would be undergo intraperitoneal injection once in left lower belly with 40 mg/kg streptozotocin (STZ),and enable them to become T2DM rats.The 8th week,we would select successful animal models 32 which were divided randomly into a modle group,a Tangmaikang (TMK) granules group,a Tangzhiping granules (TZP)group.The 12th week,we would execute all of rats and collect the blood serums which were tested for the blood glucose and IGF-Ⅰ.Results () Compared model group with blank group,the levels of the blood glucose of the model group increased to (28.33±0.50)mmol/L,while the IGF-Ⅰ decreased to (3.40 ± 0.83) pg/ml,which showed the model of type 2 diabetic rats was successfully set up.② The levels of the blood glucose of the Tangzhiping granules group and Tangmaikang granules group were lower than that of the model group to (15.06±0.39)mm ol/L,(20.12±0.58)mmol/L,but the levels of the IGF-Ⅰ were higher to (9.39±2.91)pg/ml,(5.58± 1.09)pg/ml,which implied that each group of pharmacological treatment had a good effect.③ The level of the blood glucose and IGF-Ⅰ of the Tangzhiping granules group was also significantly different from that of the Tangmaikang granule group,the therapeutic effect of Tangzhiping granules group was better than that of Tangmaikang granules group.Conclusion Tangzhiping granules can reduce the blood glucose of T2DM rats.And also can increase IGF-Ⅰ of T2DM rats.Tangzhiping granules can improve the various symptoms of diabetes mellitus at the same time.
7.Clinical observation of benazepril plus metoprolol in the treatment of chronic congestive heart failure

8.A study on SNP polymorphisms in the hypermethylated region upstream of the human H19 gene

Objective To establish a simple and high-performance analytical technique for detecting DNA methylation markers and SNPs simultaneously,and obtain the population genetics data of some SNPs in the hypermethylated region upstream of the human H19 gene.Methods The haplotypes of H19FR1 and H19FR2 which located in the promoter region upstream of the human H19 gene were investigated from 232 unrelated Chinese individuals living in Wuhan by means of PCR and subsequent denaturing gradient gel electrophoresis(DGGE).Based on the methylation status of the genomic DNA,selective detection of the parental alleles for H19FRs was examined by using the methylation-sensitive restriction enzyme(msRE) Hpa II or Hha I.Results Five haplotypes and nine phenotypes were observed for H19FR1 in Chinese Han population in Wuhan,and the power of discrimination(DP),polymorphism information content(PIC) and probability of paternity exclusion(PE) were 0.803,0.58 and 0.322 respectively.For the H19FR2,two haplotypes and three phenotyes were detected,and the DP,PIC and PE were 0.626,0.37 and 0.162 respectively.Sequencing results showed that there were 3 SNPs,a7342g,a7357g and g7547a,and one g7351c point mutation in H19FR1.In the H19FR2,there was only one SNP,a8097g.The msRE,HpaⅡ or HhaⅠ,could digest the maternal allele,and only a single band derived from the paternal allele was detected by post-digestion PCR-DGGE(PDP-DGGE) technique.Conclusion PDP-DGGE is a simple,sensitive and effective technique for analyzing DNA methylation status and SNPs simultaneously,and can be used for discriminating the parental origin of alleles.
9.Effect of repeated debridement on the efficacy of endoscopic sinus surgery

OBJECTIVE To investigate the effect of repeated debridement on the efficacy of endoscopic sinus surgery(ESS). METHODS A total of 67 patients underwent ESS were divided into intervention group and control group. Debridement was performed at the 1st, 4th, 8th week after operation for the patients in intervention group, while once per week for the patients in control group. The primary outcome measure was visual analogue scale(VAS)and Lund-Kennedy endoscopic score(LKES). RESULTS Comparing the two groups on all domains of digital VAS and LKES at the 4th, 8th week after operation, there were no significant difference between the two groups. CONCLUSION Benefit of frequent debridement during the early postoperative period was no positive correlation with patients recovering after ESS. Therefore, in terms of subjective recovery and health care costs,appropriate extending postoperative management time and decreasing intervention frequency do not decrease the efficacy of endoscopic sinus surgery.
