1.Advance in chimeric antigen receptor-T cell immunotherapy for multiple myeloma

In recent years,considerable progress has been made in the treatment of multiple myeloma(MM).However,despite the current improved prognosis of this malignancy,it always ends in relapse and therefore new therapeutic approaches are urgently needed to overcome it.The chimeric antigen receptor(CAR)-T cells targeting B cell maturation antigen(BCMA),cluster of differentiation 19(CD19),cluster of differentiation 38(CD38) and kappa light chains have been evaluated,and have achieved remarkable results in clinical trials.However,even when MM is treated with CAR-T cell therapy,most patients eventually relapse,which is the greatest limitation of this therapy.This paperreviewedthe research progress,limitations and optimization of CAR-T cell immunotherapy in the treatment of MM.
2.Determination of the antibacterial agent content of compound dexamethasone acetate cream and its antibacterial effect evaluation

Objective To establish a HPLC method to determine the antiseptics in compound dexamethasone acetate cream and evaluate the usage rationality of antimicrobial preservatives. Methods The content of ethyl hydroxybenzoate was calculated by external standard with HPLC.The effectiveness of antimicrobial preservatives was measured according to the preservative effectiveness test in ChP(2015edition) by inoculating 5 kinds of test microorganisms including Pseudomonas aeruginosa,Escherichia coli,Staphylococcusaureus,Candida albicans and Aspergillusniger.Results The linear ranges of ethyl hydroxybenzoate was 6.63-132.6 μg/mL ( r =0.999 7 ) .The average recoveries was 99.2%.The samples from different batche had different antimicrobial effectiveness due to the content of ethyl hydroxybenzoate.Conclusion The described method can be used in the determination of the preservatives of ethyl hydroxybenzoate in compound dexamethasone acetate cream.At present the use of antibacterial agent is arbitrary, it is necessary to strengthen research and supervision.
3.Mechanisms of drug resistance to gefifinib and erlotinib

Gefitinib and erlotinib are the first generation of targeted therapeutic agents approved for the treatment of advanced non-small cell lung cancer. Despite a dramatic initial response, most patients subsequently and ultimately become resistant to the drugs. Recent studies have proposed various mechanisms of such resistance, some of which related to a second mutation in epidermal growth factor receptor, aberrant activation of other tyrosine kinase receptors, bypassing signal pathways and constitutive activation of downstream signal molecules. Other researches also associated the drug resistance to internalization of epidermal growth factor receptor and changes in tumor microenvironment. Furture studies need to focus on overcoming or reversing resistance to the drugs and developing new targeted therapeutic agents.
4.Qualitative research on the job experience of the middle youth patients with peritoneal dialysis

Objective To interview the work feelings of the Middle youth patients with peritoneal dialysis. Methods Using the purposive sampling method, 11 patients were interviewed, and analyzed the content of the interview. After the interview, data were transcribed and extracted the keywords. Results Eight keywords were extracted, the patients had four positive feelings:self-perceived burden relived;more social support; more glad, negative feelings relived; peritoneal dialysis could be sure. 4 negative themes were extracted:tedious;had economic pressure;lack of self-realization;could not take the family burden. Conclusions Work can improve the living conditions of middle youth patients with peritoneal dialysis. More patients should be encouraged to return to work.

6.Clinical analysis on cognitive dysfunction early after non-cardiac surgery in elderly

Objective To analyse the cognitive dysfunction early after non-cardiac surgery in elderly. MethodsEighteen elderly patients undergoing selective non-cardiac surgery were investigated. General anesthesia was used in 11 patients and combined epidural-spinal anesthesia in 7 cases. The operations involved orthopedic, thoracic, abdominal and urologic surgeries. The patients were premedicated with phynoharbital 0. 1g and scopolamine 0. 3mg. Results All patients suffered from typical POCD that occurred during 1 to 3 days after operation. The symptoms were more severe at night. The patients with POCD were treated effectively with intravenous injection of midozolam and muscle injection of haloperidol in 15 cases. The symptoms of POCD were attenuated by injection of pethidine in 2 cases. One case had to be given intravenous infusion of propofol continually for 16 hours. All patients were cured without psychological complications before discharge Conclusion POCD is a common complication in the elderly and should be treated actively.
7.Molecular mechanism of G2 phase cell cycle arrest in tumor cells

The formation and progression of tumor is inseparable from the changes of cell cycle regulatory proteins.The G2-M period block effect of tumor cells is especially important in slowing the growth of tumor and improving the curative effect.DNA damage receptor,signal transduction factor and effectors are the classical pathways to mediate G2 phase arrest of tumor cells,and have the potential to be new targets for tumor therapy.
8.Analysis of Teaching Work for Clinical Pharmacy Undergraduate Interns in Our Hospital

OBJECTIVE:To explore the teaching method for clinical pharmacy undergraduate interns,and provide reference for improving graduation practice effect. METHODS:Starting from the actual teaching work of clinical pharmacy undergraduate in-terns,the teaching work situation was emphatically introduced,development of teaching plan,determinationof teaching duties and development of management practices were described in detail,the working experience and deficiency were summarized,and ap-propriate improvement measures were proposed. RESULTS & CONCLUSIONS:According to the contents and characteristics of clinical pharmacy,teaching plans based on clinical practice was developed for the basic premise of standardization and systematiza-tion by combining with the teaching syllabus;setting up teaching working group,developing management practices for interns and knowing responsibilities of teachers are the fundamental guarantees for successful completion of the internship. Practice has proved that the improvement of the teaching method of teaching was beneficial to improve the effect of clinical pharmacy undergraduates.
9.Proteomics analysis for the signal transduction under stimulation of prolactin on Jurkat cells

Objective:The experiment was designed to apply proteomics to analyze the signal transduction molecules in T lymphocyte strain JurkatD1.1 cells with the stimulation of prolactin.Methods:JurkatD1.1 cells were cultured in the presence of recombinant human prolactin(rhPRL).Phosphated metal affinity chromography(PMAC) and immunoprecipitation(IP) assay were applied to enrich phosphoproteins from the total cell lystates which were resolved by one-dimensional electrophoresis(1 DE) or 2 DE.The different bands and different spots in the gels between control group and rhPRL-treated group were excised,digested,and analyzed by MALDI-TOF mass spectrometry.Results:Three major protein bands were observed specifically in the rhPRL-treated group and other two major protein bands were observed specifically in the control group.The bands were excised and analyzed by MALDI-TOF mass spectrometry.Protein band a in rhPRL-treated group was successfully identified as hot shock protein 90(hsp90).Nine different protein spots which were observed in the 2 DE gels between the two groups were excised and analyzed by MALDI-TOF mass spectrometry.In the rhPRL-treated group the protein spot 4 was identified as nuclear receptor co-repressor 2 variant(NcoR 2 variant);The protein spot 5 was identified as galactose-1-phosphate uridyl transferase;And the protein spot 6 was identified as zinc finger ZIM3.Conclusion:Hot shock protein 90 participates the signal transduction pathways of prolactin.Nuclear receptor co-repressor 2 variant and zinc finger ZIM3 may play a role in the process of prolactin modulating target genes' expression.
10.Preparation and Clinical Use of Pefloxacin Mesylate Eye Drops

OBJECTIVE: To prepare pefloxacin mesylate eye drops.METHODS: The content of pefloxacin was determined with UV- spectrophotometry.The therapeutic effect of the eye drops was observed in 170 out- patients.RESULTS: The linearity range was 2~ 14μ g/ml with a regression equation of A=0.1 018C and r=0.9 999(n=7).The average recovery was 99.64% with a RSD of 0.43% .After treatment,150 out- patients recovered,16 out- patients improved and 4 out- patients had no efficiency with a total effective rate of 97.6% .CONCLUSION: The eye drops has definite therapeutic effect.The UV- spectrophotometry method is simple,rapid and accurate in determination of the content and no special reagent is needed.
