Finite element analysis (FEA) is broadly used in biomechanics.Being widely used in clinical studies on cervical spine injury,cervical degeneration and stimulating a variety of cervical spine surgeries,cervical finite element model is becoming more and more accurate in recent years.This paper aims to review the development of cervical finite element model,to introduce the application progress of the modeling and analysis in cervical spine injury,cervical disc arthroplasty,cervical interbody fusion,cervical degeneration and cervical instability,and to prospect the foreground of cervical finite element model in future.
2.A clinical comparative study on Naodekang capsule combined with western medicine for treatment of patients with mental disorders after bilateral cerebral frontal lobes contusion and laceration

Objective To study the clinical therapeutic effect of combination of Naodekang capsule and western medicine for treatment of patients with mental disorders after bilateral cerebral frontal lobes contusion and laceration.Methods A prospective study was conducted. Sixty-two patients with mental disorders after bilateral frontal lobes contusion and laceration hospitalized from June 2012 to January 2014 in the Department of Neurosurgery in Liangzhou Hospital, Wuwei City, Gansu Province were enrolled. According to random principle, they were divided into combined traditional Chinese and western medicine treatment group (32 cases) and western medicine treatment control group (30 cases). Conventional symptomatic therapy was given to both groups. In the western medicine control group, based on the above conventional treatment, antipsychotic drug risperdal (risperidone) was given, the beginning dosage was 4 mg daily and gradually reduced to 1 mg daily. In combined traditional Chinese and western medicine group, beside the conventional therapy, oral Naodekang capsules and oral risperdal were given. The ingredients of the capsule included: Astragalus 60 g, Radix Paeoniae Rubra 15 g, Rhizoma Chuanxiong 25 g, Angelica 15 g, Earthworm 10 g, Semen Persicae 10 g, Safflower 15 g, Salvia miltiorrhiza 15 g, Rhizoma Gastrodiae 15 g, Uncaria 10 g, Bupleurum 20 g, Pueraria 15 g, Rhizoma Acori Tatarinowii 15 g; the patients took once 3 capsules, three times a day, and risperdal 2 mg per day at the beginning and gradually reduced to 1 mg daily. Before treatment and after treatment for 2, 6 and 12 weeks, the brief psychiatric rating scale (BPRS) score was applied to evaluate the clinical therapeutic effect on the situation of patients' improvement of symptoms; in the two groups, the clinical curative effects were observed and when the psychiatric symptoms were controlled, the mean dosage of antipsychotic western medicine used daily in each patient was calculated.Results After treatment, the BPRS scores in the two groups were lower than those before treatment, from 6 weeks after treatment, the degree of descent in the BPRS score in the combined treatment group was more significant than that in the western medicine group (38.68±7.81 vs. 54.05±6.75,P < 0.05), and the descent was persistent until 12 weeks after treatment (15.46±8.51 vs. 23.98±8.03,P < 0.05). The cure rate of the combined group was markedly higher than that in western medicine group [84.4% (27/32) vs. 76.6% (23/30),P < 0.05]. When the psychiatric symptoms were controlled, the dosage of antipsychotic western drug used in each patient in the combined group was less than that in western medicine group (mg/d: 1.5±0.1 vs. 2.8±0.2,P < 0.05).Conclusion The westernconventional therapy combined with traditional Chinese medicine Naodekang capsule for treatment of mental disorders after bilateral cerebral frontal lobes contusion and laceration has relatively good therapeutic effect.
3.Construction of Newcastle disease virus hemagglutinin-neuraminidase (HN) gene and its antitumor immune response effects

Objective: To construct an eukaryotic expression plasmid containing gene coding for the hemagglutinin neuraminidase(HN) of Newcastle disease virus (NDV), and to study its mechanism and value in antitumor therapy. Methods: The HN cDNA was abtained from NDV with RT PCR and an eukaryotic expression vector of HN gene ( pcDNA3 HN ) was constructed. The antitumor effect was evaluated after injecting pcDNA3 HN into mice bearing B16 melanoma. Results: The HN cDNA of NDV was successfully cloned and pcDNA3 HN had a good expression in COS 7 cells. Animal experiments suggested that the pcDNA3 HN could significantly increase CTL and NK activity of tumor bearing mice. Conclusion:The eukaryotic expression plasmid containing the gene coding for the (HN) has the function of increasing CTL and NK activity of tumor bearing mice.
4.The relationship between pillow height and cervical intervertebral disc stress in lateral decubitus position by three-dimensional finite element analysis

Objective To analyze the relationship between pillow height and cervical intervertebral disc stress in lateral decubitus position of the human body using three-dimensional finite element method,and to provide the basis for appropriate pillow selection.Methods A cervical spine CT scan was performed on a normal male volunteer.The medical 3D image reconstruction software (Mimics),reverse engineering software (Geomagic Studio) and finite element software (MSC.Patran) were collectively used to establish three-dimensional finite element models of the whole cervical spine under three lateral decubitus conditions,includes pillow height of 10 cm (cervical spine leftward flexion),17 cm (cervical spine neutral),and 25 cm (cervical spine rightward flexion).Moreover,the finite element analysis and calculation of the three models were conducted.Results The Von-Mises stress,maximal principal stress and maximum shear stress of the model in cervical spine neutral (pillow height=17 cm) were all significantly lower than those of the cervical spine lateral model.Conclusions When the cervical coronal plane is in neutral position,the cervical discs can achieve the best stress distribution state.In lateral decubitus position of the human body,a appropriate pillow height should make the cervical vertebrae in neutral position.
5.Effects of PM2.5 on the differentiation of splenic CD4 + T lymphocyte of acute asthma mice

Objective To evaluate the effects of PM2.5 on the differentiation of splenic CD4 + T lymphocytes in acute asthma mice.Methods (1) Mouse models of acute asthma were established by ovalbumin (OVA) sensitization and challenge.(2) PM2.5 was collected in the urban area of Zhanjiang city under heavy traffic and serious air pollution from total suspended particulate(TSP) mid-flow sampler and multistage particles cutters and the dry powder of PM2.5 was prepared.(3) Specific-pathogen free Balb/c mice,female,at 6 to 8 weeks of age were randomly divided into 8 groups (8 mice each group):a negative control group (NC group),asthma control group (AC group),sensitized mice treated with different doses of PM2.5 groups (SP groups) and asthmatic mice treated with different doses of PM2.5 groups (AP groups).SP groups and AP groups were respectively divided into 3 subgroups according to the dose of PM2.5.The AC group,SP groups and AP groups were sensitized on D0,D7 and D14,and the NC group was treated with NS as controls.The SP1/AP1 group,SP2/AP2 group and SP3/AP3 group were respectively given 50 μL PM2.5 suspension.NC group and AC group were instilled with NS as controls.AC group and AP groups were challenged by aerosol of OVA,and NC group and SP group were treated with NS as controls.Twenty-four hours after last challenge,all the mice were sacrificed,and the percentage of regulatory T cells (Treg),T helper cell type 1 (Th1),Th2 and Th17 of splenic CD4 + T lymphocytes was detected by flow cytometry (FCM).Results (1) An OVA-induced mouse models with acute asthma were successfully established.(2) Comparison of the percentage of Treg of splenic CD4 + T lymphocytes:SP group [(12.28 ± 0.73) %,(11.93 ± 0.81) % and (11.70-± 1.14) %] and AC group [(12.18 ± 1.00) %] were lower than that in the NC group[(13.50 ± 0.39) %] (P < 0.05),AP3 group [(10.58 ± 0.65) %] was lower than that in the AC group and AP1 group [(11.91 ± 0.79) %] (P < 0.05).(3) Comparison of the ratio of Th1/Th2 of splenic CD4+ T lymphocyte:SP1 group [(7.74 ± 1.21)%] was higher than that in the NC group [(5.52 ± 1.06) %] (P <0.05),SP2 group[(6.30 ±0.58) %] was lower than that in the SP1 group(P <0.05),SP3 group [(4.87 ± 0.82) %] was lower than that in the SP2 group (P < 0.05);AC group [(3.69-± 0.47) %] was lower than that in the NC group and SP3 group (P < 0.05);AP3 group [(2.92 ± 0.57) %] was lower than that in the AC group(P < 0.05).(4) Comparison of the percentage of Th17 of splenic CD4+ T lymphocyte:AP3 group [(1.46 ± 0.39) %] was higher than that in the NC group [(0.89 ± 0.24) %] and the AP2 group [(0.83 ± 0.15) %] (P < 0.001).Conclusions PM2.5 can inhibit splenic CD4 + T lymphocyte of acute asthma mice differentiation into Treg and Th1,and promote their differentiation into Th2 and Th17,through which aggravates inflammation reactions in the airway.
6.Pharmaceutical Practice for a Patient with Hiatal Hernia Complicating with Venous Catheter-related Infection

OBJECTIVE:To provide reference for clinical desicion,drug use and ADR avoidance. METHODS:Clinical pharmacists participated in the consultations and therapy plan about a patient with hiatal hernia complicating with venous catheter- related infections. Clinical pharmacists suggested pulling out peripherally inserted central catheter(PICC)and placing a catheter again. Anti-infective therapy plan had been adjusted:vancomycin 0.5 g/time,ivgtt,q12 h;fluconazole 0.2 g,ivgtt,qd(0.4 g,ivgtt,qd on the first day),cefoperazone/sulbactam 2 g,ivgtt,q8h. Vancomycin and fluconazole were additionally used for anti- infective therapy. RESULTS:Doctors adopted the suggestions of clinical pharmacist,and pulled out PICC;and then the infection of the patient was controlled effectively. Finally,the infection had been cured. CONCLUSIONS:The participation of clinical pharmacists in the consultation contributes to rational drug use,and assists doctors to make the right clinical decisions and avoid adverse drug events.
7.Effect of Hyperbaric Oxygen Therapy on Abilities of Learning and Memory and Hippocampal Pyramidal Cell in Vascular Dementia Rats

Objective To study the effect of hyperbaric oxygen(HBO)therapy on abilities of learning and memory and hippocampal pyramidal cell in vascular dementia rats.MethodsThe vascular dementia model was reproduced and divided into control,sham and treatment groups.After 30 d of HBO therapy,the rats were assessed with the Morris water maze,and their hippocampal pyramidal cells were observed after HE staining.ResultsCompared with the control group,the ability of learning and memory of rats in treatment group increased,as well as the hippocampal pyramidal cells did.ConclusionHyperbaric oxygen therapy can be used to treat vascular dementia.
8.PTEN inhibits the proliferation of neural stem cells via the antagonism of PI3K/Akt signaling pathway

ABSTRACT:Objective To explore the role of PTEN in the suppression of neural stem cells so as to clarify whether neural stem cell proliferation can be promoted by regulating the PI3K-Akt/PTEN expression level. Methods We removed the hippocampus from neonatal 24 h Kunming mice,isolated and cultured the generation of neural stem cells,which were then identified by immunofluorescence test.We randomly grouped the cultured neural stem cells into normal group,ischemia model group,hypoxia group (hypoxia + ischemia model group),Lip2000 group (hypoxia+ ischemia model group + Lip2000 null transfection group,PTEN transfection group (hypoxia +ischemia model group+Ad5-PTEN transfection group),and PTEN interference group (hypoxia+ ischemia model group+Lip2000+PTENsiRNA interference group).We detected the proliferation of neural stem cells in the groups at different time points,and PTEN protein expression of neural stem cells in each group after PTEN transfection,
and the effect on iconic protein on Akt-PI3K signaling pathway.Results (1 )Nestin identification of the neural stem cells was tested by immunofluorescence.We observed green bright and typical cell spheroids under fluorescence microscope;the clear structure could be seen within spheroids.(2)We determined the proliferation of neural stem cells at different time points by MTT.After 36 h of culture,the neural stem cells had obvious proliferation in the hypoxia group and Lip2000 group compared with the model group, the PTEN transfection group and PTEN interference group (P <0.05),including the cell proliferation of PTEN interference group compared to that of the model group,and PTEN transfection group obviously,the differences were also significant (P < 0.05 );(3 ) Compared with the cells in the normal group,PTEN expression increased in the model group and PTEN gene transfection group (P <0.05);it was all lower in other groups than in the normal group (P <0.05).The trend of decrease was similar in hypoxia group,Lip2000 group,and PTEN interference group (P >0.05);(4)After PTEN plasmid transfection,there were no apparent changes in the total Akt and bad in the groups,but compared with the normal group,PI3K,p-Akt,and p-bad expressions in model group and PTEN gene transfection group reduced (P <0.05),with the more obvious changes in the PTEN transfection group.PI3K,p-Akt,and p-bad expressions in the hypoxia group, Lip2000 group and PTEN interference group increased (P < 0.05 ). Conclusion Hypoxia contributes to promoting the proliferation of neural stem cells,and the inhibition of PTEN on PI3K/Akt might be the key factor for blocked proliferation of adult neural stem cells.
9.Effect of Basic Fibroblast Growth Factor on Magnetic Resonance Spectroscopy of Vascular Dementia Rats

Objective To observe the effect of basic fibroblast growth factor (bFGF) on magnetic resonance spectroscopy of vascular dementia (VD) rats.Methods After the VD model was reproduced, the 12 rats were randomly divided into the therapy group and dementia group with 6 animals in each group. Another 6 rats were selected as sham operation group. The VD rats in therapy group were treated with bFGF by hypodermic injection. After 5 weeks, abilities of learning and memory of three groups' rats were tested by Morris water maze. The changes of magnetic resonance spectroscopy and N-acetyl aspartate (NAA), NAA/(Cr+Cho) in lobus temporalis of VD rats were also observed.Results Abilities of learning and memory of rats significantly improved in the therapy group than that in the dementia group (P<0.01). The values of NAA, NAA/(Cr+Cho) in lobus temporalis of VD rats significantly increased in the therapy group than that in the dementia group (P<0.01).Conclusion bFGF by hypodermic injection can obviously elevate abilities of learning and memory and the values of NAA, NAA/(Cr+Cho) in lobus temporalis of VD rats.
10.Prediction of Infantile cytomegalovirus infection with viral load in urinary epithelial cell

Objective To investigate the application value for predicting human cytomegalovirus(HCMV) infection with viral load in urinary epithehal cell (EC).Methods Peripheral blood and urine specimens from 82 infants with HCMV latent infection and 84 infants with HCMV active infection were collected respectively.Plasma HCMV DNA load and the levels of HCMV lgM/IgG antibody were detected by real-time fluorescence quantitative polymernse chain reaction (FQ-PCR) and chemiluminescence immunsassay.HCMV pp65 antigen in peripheral blood polymorphonuclear leukocytes (PMNLs) was detected by indirect immunofluorescence assay.The urinary EC count and HCMV DNA load were detected by UF-100 automated urine sediment analyzer and FQ-PCR,respectively.HCMV DNA load in urinary EC was calculated accordingly.At the same time,the sensitivity and specificity for diagnosis of active HCMV infection were evaluated by receiver operating characteristic curve (ROC).Results The positivity of HCMV DNA in urinary EC was 94.58% (157/166),which was the highest among the urinary EC from 166 cases of HCMV infection.HCMV DNA load ranged from 5.67×102to 1.31×107 copies/103 EC There was no significantly statistical difference among urine specimens from different periods of time(P>0.05).HCMV DNA load in active infection group [5.13±0.99(copies/103EC,lg)]is significantly higher than that in latent infection group [3.92±0.82 (copies/103 EC,lg),t = 8.52,P < 0.01];According to ROC curve analysis,its sensitivity and specificity were 71.4% and 75.2% respectively when cut-off value was 4.55.The virus load was significantly decreased in urinary EC in post-treatment infants as compared with pre-trestment(t=5.44,P<0.01).Conclusion Detection of HCMV DNA load in the urinary EC is a cost-effective method and can be used to predict HCMV active infection in infants and monitor treatment of HCMV infection.
