1.Effect of lanthanum chloride on the proliferation and expressions of cyclin D1 and cyclin dependent kinase 4 in rat hepatocellular carcinoma CBRH-7919 cells

BACKGROUND: It has been proved that the rare earth compounds can obviously restrain the growth of tumor in vitro and in vivo.OBJECTIVE: To check the effect of chlorinated lanthanum (LaCl3) on the expressions of cyclin D1 and cyclin dependent kinase 4 (CDK4) of hepatocellular carcinoma cells.DESIGN: Controlled observation.SETTING: Department of Nephropathy, First Clinical Hospital, Jilin University. MATERIALS: The experiment was carried out in the cell culture laboratory (province-level) of Department of Histology and Embryology, Jilin University from July 2002 to June 2003. Wells of the microwell plate seeded with CBRH-7919 cells were randomly divided into 4 groups according to the dose of LaCl3: control, 0.01, 0.1, 1.0 mmol/L LaCl3. And each group was further divided into three subgroups according to the culture time: 1, 3, 5 days, with 6 wells for each.METHODS: The growth of CBRH-7919 cells was monitored following treated with 0.01, 0.1, 1.0 mmol/L LaCl3 for 1, 3, 5 days in vitro, respectively. ① Absorbance(A) of each well was measured by MTT assay.② The changes of distribution of cells into different phases of cell cycle were detected by flow cytometry. ③ Cyclin D1 and CDK4 expression was detected by immunocytochemical analysis.MAIN OUTCOME MEASURES: The expression of cyclin D1 and CDK4 in CBRH-7919 cells of different doses of LaCl3 at different time.RESULTS: Effect of LaCl3 on the growth of CBRH-7919: On days 3 and 5, the A value of each well in the 0.1 and 1.0 mmol/L LaCl3 groups was respectively significantly lower than that in the control group (day 3: 1.140±0:070, 0.706±0.092,1.461±0.087; day 5: 1.888±0.020,0.625±0.037,2.544±0.032, respectively) (P < 0.05,0.001 ). ②Effect of LaCl3 on the percentage of CBRH-7919 cells at G0/G1 stage: On days 1, 3, and 5, the percentage of CBRH-7919 cells at G0/G1 stage in the 1.0 mol/L LaCl3 group was respectively significantly higher than that in the blank control group [(60.70±0.20)%, (39.49±0.67)%; (61.66±0.97)%, (45.56±1.00)%; (69.92±0.18)%, (49.24±0.27)%, (P < 0.01,0.001 )]; On day 5, the percentage of CBRH-7919 cells at G0/G1 stage in the 0.1 mol/L LaCl3 group was respectively significantly higher than that in the blank control group[(58.88±0.73)%, (49.24±0.27)%, P < 0.05]. ③ Effect of LaCl3 on the expressions of cyclin D1 and CDK4 of CBRH-7919 cells: On day 1, the expressions of cyclin D1 and CDK4 of CBRH-7919 cells in the 0.1 and 1.0 mmol/L LaCl3 groups were respectively lower than those in the blank control group (expression of cyclin D1: 562±35,453±22,860±82;expression of CDK4: 705±84,680±28,762±16, P < 0.05, 0.01 and 0.001).CONCLUSION: LaCl3 inhibited the growth of CBRH-7919 cells correlating down-regulating cyclin D1 and CDK4 as reflected on the cell cycle arrest of CBRH-7919 cells from G1 stage to S stage.
2.Protection of superior parathyroid glands by improving dissection procedure of thyroid superior pole in total thyroidetomy surgery

Objective To investigate the protection of superior parathyroid glands by improving dissection procedure of thyroid superior pole in total thyroidectomy surgery.Methods 136 consecutive cases undergoing two-step dissociating procedure of thyroid superior pole(the improved group) in total thyroidectomy plus central compartment dissections from Sep.2012 to Oct.2015 were retrospectively analyzed.Intraoperative observation of superior parathyroid and incidences of postoperative hypoparathyroidism were compared with conventional surgical methods (the conventional group) containing 113 cases from Jun.2009 to Sep.2012.All operations were performed by the same treatment group.Results Intraoperatively,249 (249/272,91.5%) superior parathyroid glands were identified in the improved group and 215 (215/226,95.1%) superior parathyroid glands were identified in the conventional group.The difference had no statistical significance (P=0.114).Whereas,among identified superior parathyroid glands,242 (242/249,97.1%)of them were conserved in situ in the improved group and 198 (198/215,92.0%) were conserved in situ in the conventional group.The difference had no statistical significance (P=0.013).Among superior parathyroid glands conserved in situ,225 (225/242,93.0%) had good blood supply in the improved group and 170(170/198,85.9%) in the conventional group.The difference had statistically significance (P=0.001).28 (28/136,20.5%) patients displayed temporary hypoparathyroidism and 2 (2/136,1.5%) cases suffered permanent hypoparathyroidism in the improved group after surgery.Whereas,in the conventional method group,a higher incidence was observed in 37 (37/113,32.7%) cases of temporary hypoparathyroidism and 7 (7/113,6.2%) cases of permanent hypoparathyroidism respectively.The difference had statisticall significance (P=0.003).Conclusions Two-step dissociating procedure technique offers protective effect on superior parathyroid glands function.It is worthy of popularization.
3.Distribution of pathogenic bacteria of nosocomial infections in intensive care unit and its drug-resistances

Objective To investigate the distribution of pathogenic of nosocomial infections in intensive care unit (ICU) and the tendencies of its drug-resistances.Methods All of 130 patients of nosocomial infection from March 2009 to March 2013.Results Of all the nosocomial infection pathogens,gram-negative(G-) bacteria was the main(53.76%).The first five pathogens were E.coli(13.9%),E.faecalis (11.3%),A.baumannii (10.3%),S.aureus (9.8%) and K.pneumoniae (8.3%).The lower resistant to pseudomonas aeruginosa were amikacin (5.3%).A.baumannii increases rapidly and its resistances was severe.Two trains of E.faecalis were resistant to vancomycin.Conclusion G-bacteria is the main pathogens in ICU,fungal has upward tendency.The multidrug resistances of A.baumannii and E.faecalis were severe; Target monitoring is an effective surveillance method to reduce the prevalence of the nosocomial infection in ICU.
4.Determination of the Related Substances in Nortriptyline Hydrochloride by HPLC

Objective: To establish a determination method for the related substances in nortriptyline hydrochloride by HPLC. Methods:An Agilent C18 (250 mm × 4. 6 mm,5 μm) column was adopted with a mobile phase consisting of acetonitrile-0. 2% trieth-ylamine(pH 3. 0)(34 ∶66). The flow rate was 1. 0 ml·min-1, the detection wavelength was set at 210 nm, the column temperature was 40℃ and the injection volume was 20 μl. Results:The resolutions of the related substances were acceptable. The detection limit and the quantitation limit of the samples was 0. 40 ng and 1. 35 ng, respectively. Conclusion: The method is sensitive, accurate and specific, and can be used to determine the related substances in nortriptyline hydrochloride.

Objective To investigate the protective role of two-step decapping technique for superior parathyroid glands in total thyroidectomy surgery.Methods The clinical data of 136 consecutive cases underwenttwo-step decapping technique in total thyroidectomy plus central compartment dissection from April 2013 to January 2016 were retrospectively analyzed.And the incidences of postoperative hypocalcemia were was compared with conventional consecutive decapping surgical methods containing 113 cases from January 2010 to March 2013.All operations were performed by the same treatment group.Results In the two-step decapping group,28 cases(28/136,20.6%) displayed postoperative temporary hypoparathyroidism and a higher incidence were was observed in 37 cases (37/113,32.7%) in the conventional method group,the difference was statistically significant(x2 =4.737,P =0.03).Whereas,2 cases(2/136,1.5%) suffered permanent hypoparathyroidism in the two-step decapping group and 7 cases (7/113,6.2%) of permanent hypoparathyroidism in the conventional method group,the difference was statistically significant (x2 =3.954,P =0.047).Conclusion Two-step decapping technique can more effectively protect the parathyroid gland function.It is worthy of popularization.
6.Effects of LaCl3 on P16 and P21 expressions of hepatocellular carcinoma cells

BACKGROUND:There is often space between implant and bone during immediate implantation.Whether biological membrane is needed to guide bone regeneration remains poorly understood.OBJECTIVE:To createdifferent sizes of space between femurand implantsindogs and to observe the effects of biological membrane on bone regeneration capacity of bone defects surrounding implants.DESIGN,TIME AND SETTING:A self-control animal experiment was performed at the Laboratory Animal Center,Norman Bethune College of Medicine,Jilin University and School of Stomatology,Jilin University between March and December 2005.MATERIALS:BLB hydroxyapatite-coated implant was provided by Beijing Leiden Biomaterial Co.,Ltd.,China;BME-10X collagen membrane was purchased from Fujian Better Biotechnology Co..Ltd.,China.METHODS:BLB implants were installed in the bilateral proximal femoral bone to create standard gradient bone defects with horizontal width 3 mm.vertical depth 5 mm,and horizontal lengths of 0,1,2,3,and 4 mm Bone defects on the left femur were sutured directly and those on the right femur were covered with biological membrane prior to suture.All animals were sacrificed at 3 months after surgery.Specimens containing implants were harvested to prepare tissue blocks for radiological observation.MAIN OUTCOME MEASURES:The quantity,color,and texture of newly formed bone surrounding implants were observed from the surface and profile levels.The implant-bone integration and new bone formation were also examined by soft X-ray photography.RESULTS:Grossobservation results revealed that when the horizontal length of bone defect was 3 mm or less,there was no significant differenee in bone density between the newly formed bone and the host bone no matter whether biological membrane existed or not;when the horizontal length of bone defect was 4 mm the bone density was better when biological membranes were used than not.Soft X-ray photography results revealed that when the horizontal length ofbone defect was 3 mm or less.no significant difference in bone density and bone trabecular morphology and orientating was found between newly formed bone and host bone no matter whether biological membrane was used or not;in the 4-mm-length bone defect areas.implants contacted with newly formed bone directly,but the calcified degree ofnewly formed bone was poor,bone trabecula was thin,and bone trabecular course was irregular,nevertheless,the calcified degree of newly formed bone was better under the condition of being with biological membrane than without biological membrane.CONCLUSION:Biological membrane exhibits strong capacity to promote the regeneration and repair of bone defect tissue with a horizontal length of 3 mm or less,and plays an important role in repatr of large sizes of bone detect
9.Analysis of surgical treatment of hashimoto's disease(HD) complicated with thyroid multiple nodules

Objective To investigate the diagnosis、 treatment and prognosis of hashimoto's disease(HD) complicated with thyroid multiple nodules.Methods Clinical data of 129 cases of HD complicated with thyroid multiple nodules under want surgery in three years were analyzed retrospectively and followed up.Results All patients were received different scope of thyroidectomy according to the situation of thyroid nodules.The histologic examination after operation indicated there are 36 cases(27.9%) complicated with nodular goiter;23 cases(17.8%) complicated with thyroid cancer;3 cases(2.3%) complicated with thyroid adenomas;1 cases(0.78%) complicated with toxic goiter.Conclusion HD complicated with thyroid multiple nodules patients had difficulty in preoperative diagnosis of nodular nature and should be operate.
10.Adsorption and desorption of lycopene and ?-carotene using three macoroporous adsorption resins

Object To study the separation effect of three macoroporous resins for lycopene and ? carotene, in order to optimize the suitable one. Methods The performances of adsorption and desorption for lycopene and ? carotene with three macroporous resins D4020, X 5 and AB 8 were compared, and adsorption dynamics process of resins was studied. Results The adsorptive capacities of X 5 resin for lycopene and ? carotene are the most, they are 7 44 and 11 2 mg/g, respectively. Desorption ratio of X 5 resin is the highest. Conclusion X 5 resin is an ideal adsorbent for lycopene and ? carotene extraction.
