Objective To study the relationship between blood uric acid(BUA)level and oxidative stress in the elderly. Methods One hundred and eighteen healthy subjects who aged 60years and over were recruited.They were divided into 3 groups according to the serum uric acid level:(1)Control group:BUA 142-416 μmol/L;(2)Hyperuricemia group Ⅰ:BUA 417-470 μmol/L;(3)Hyperuricemia group Ⅱ:BUA＞470 μmol/L.Serum xanthine oxidase(XO)activity,superoxide dismutase(SOD)activity,total antioxidation capacity(TAOC)and peroxide concentration were examined. Results (1)No significant differences in TAOC[control group:(8.40±2.09),Hyperuricemia group Ⅰ:(8.13±1.92)and Hyperuricemia group Ⅱ:(7.95±1.54)U/ml,all P＞O.05)and in SOD activity[control group:(80.57±15.44),Hyperuricemia group Ⅰ:(81.37±11.92)and Hyperuricemia group Ⅱ:(87.45±11.38)U/ml,all P＞0.05)among the 3 groups were found.The serum peroxide level and XO activity were significantly higher in hyperuricemia group Ⅰ and Ⅱ[XO activity:(9.17±1.68) vs.(9.32±1.87)U/ml,all P＜0.05;serum peroxide:(149.5±25.7)vs.(150.7±23.1)μmol/L,all P＜0.05]than in group control[XO activity:(7.81±1.84),serum peroxide:(122.5±33.8)].(2)Multifactorial regression analysis showed TAOC was related to age,triglyceride and uric acid(all P＜0.05),XO activity was related to uric acid(P＜0.05)and peroxide concentration was related to age and uric acid(all P＜0.05). Conclusions This study supports the presence of the xanthine oxidase-related oxidative stress in the elderly with hyperuricemia.

Objective To evaluate effects of human papilloma virus(HPV)16E7 on expressions of six tumor suppressor genes(including MT1G, NMES1, RRAD, SFRP1, SPARC and TFPI2)in a cell line SiHa, as well as on its proliferation and apoptosis. Methods SiHa cells were divided into two groups to be transfected with a small interfering RNA targeting HPV16E7(E7SiRNA, experimental group)and an empty vehicle(negative control group) respectively, with SiHa cells receiving no treatment serving as the blank control group. After 48 hours, qPCR was performed to measure the mRNA expressions of E7 and six tumor suppressor genes, CCK?8 assay to evaluate cellular proliferative activity, and flow cytometry to assess apoptosis of SiHa cells. Results At 48 hours after the transfection, the experimental group showed significantly decreased E7 mRNA expression(0.25 ± 0.036, P<0.05), but increased mRNA expressions of the six genes(MT1G 1.403 ± 0.190, NMES1 1.720 ± 0.060, RRAD 1.390 ± 0.160, SFRP1 1.493 ± 0.120, SPARC 2.157 ± 0.144, TFPI2 2.060 ± 0.122, all P < 0.05). The proliferative activity of SiHa cells was significantly decreased(0.554 ± 0.130 vs. 1.028 ± 0.236 and 1.220 ± 0.126, both P<0.05), but the apoptosis rate was significantly increased(9.222%vs. 0.246%and 0.123%, both P<0.05)in the experimental group compared with the negative control group and blank control group. No significant differences were observed between the negative control group and blank control group in proliferative activity or apoptosis rate of SiHa cells(both P>0.05). Conclusion E7 may participate in HPV16?induced cellular malignant transformation by suppressing the mRNA expressions of 6 tumor suppressor genes, including MT1G, NMES1, RRAD, SFRP1, SPAR and TFPI2.

Objective To evaluate the safety and effectiveness of 2μm Thulium laser vaporization enucleation (Thu-VEP) in the treatment of benign prostate hyperplasia (BPH). Methods A total of 145 patients with BPH were randomized into two groups including patients underwent ThuVEP and patients underwent standard transurethral resection of the pros-tate (TURP). The intraoperative blood loss,flushing fluid quantity,operation time,bladder irrigating time,catheterization time,the international prostate symptom score (IPSS),quality of life score (QOL),the maximum flow rate (Qmax),the post-void residual (PVR) and complications were observed in two groups. Results There were no significant differences in pa-tient age, preoperative duration, prostate weight, IPSS score,QOL score,Qmax and RUV between two groups. Patients in two groups were performed surgery successfully. The values of blood loss, bladder irrigating time and catheterization time were significantly less in ThuVEP group than those of TUEP group (P＜0.05). There were no postoperative complications in pa-tients of ThuVEP group. There were 5 cases of complications in TUEP group after operation. The values of IPSS score,QOL score,Qmax and RUV were significantly different after 3-12-month follow up than those before operation(P＜0.01),but no significant difference between two groups. Conclusion ThuVEP is a safe and more effective treatment than that of TURP for patients with BPH. ThuVEP can significantly improve the quality of life, and reduce complications in patients with BPH.

Objective To explore the effect of postural reduction and balloon dilation on the correction of vertebral height following balloon percutaneous kyphoplasty (PKP) for osteoporotic vertebral coinpressive fracture (OVCF).Methods From June 2012 to December 2013,47 OVCF patients involving 57 fractured thoracolumbar vertebrae with intact posterior wall underwent PKP.In all patients postural reduction was performed under general anesthesia.The percentages of vertebral height loss before and after balloon dilation were compared.The curative effects were evaluated by comparing preoperative and postoperative visual analogue scale(VAS) scores and cobb angles.Results Compared with preoperation,the vertebral heights of anterior and middle columns were significantly improved after postural reduction,balloon dilation and PKP (P ＜ 0.05),but there was no significant improvement before and after balloon dilation (P ＞0.05).The postoperative average cobb angle (22.8°± 8.1°) was significantly decreased compared with the preoperative one (22.8° ± 8.1°) (P ＜ 0.05).The VAS scores at preoperation,one day postoperation and the final follow-up were respectively 8.5 ± 0.4,3.4 ± 0.2 and 3.1 ± 0.3.The postoperative ones were significantly lower than the preoperative one (F =7.518,P =0.006).Conclusions In PKP,postural reduction under general anesthesia plays an important role,but balloon dilation may exert an insignificant effect on restoration of vertebral height.

Objective:To investigate the effects of Bailing capsules on the expression of TGF-β1 in the peritoneal dialysis solution for peritoneal dialysis patients. Methods: Totally 40 patients treated with peritoneal dialysis ( PD) were randomly divided into two groups (20 cases in the control group and 20 cases in the experimental group). All the patients were treated with PD by 1. 5% perito-neal dialysis effluent (6 000 ml everyday), and the patients in the experimental group were additionally treated with Bailing capsules (5 capsules each time, three times a day after meals) for 6 months. The adequacy of PD (including Kt/V and Ccr) of the two groups was examined after the one-month treatment. The renal function and the level of TGF-β1 in the effluent in 1, 3 and 6 month were com-pared between the two groups. Results:The adequacy of PD ( Kt/V and Ccr ) had no significant difference between the two groups (P<0. 05). The level of serum creatine was decreased significantly in both groups after the one-month treatment (P<0. 05), and no significant change was shown in 3 and 6 month (P>0. 05). In the control group, the expression of TGF-β1 in the effluent in 1, 3 and 6 month was increased gradually with significant difference (P<0. 05). In the experimental group, the expression of TGF-β1 in the effluent had no significant difference in 1, 3 and 6 month (P>0. 05), which was lower than that in the control group (P<0. 05). Conclusion:Bailing capsules can decrease the expression of TGF -β1 in the effluent for the patients treated with PD and inhibit the peritoneal fibrosis.

Objective:To investigate the effect of 3 HLA matching measures on preventing hyperacute rejection(HAR). Methods:Among 1 152 cases,550 cases received anti donor lymphocytotoxic crossmatch(LXM),335 cases received LXM and HLA serological test,267 cases of the latter again received panel reaction antibody(PRA)test, the change of the HAR rate with different HLA matching measures were analyzed. Results:If LXM value ≤10% or HLA matched loci ≥3,the rate of HAR was unrelated to LXM value or HLA matched loci. HAR rate decreased significantly in patients with LXM and HLA serological test, and decreased more markedly in those with additional PRA test compared with those only with LXM test. Conclusion:HLA test and PRA are necessary and effective to avoid HAR.

Objective To evaluate the effects of lentivirus-delivered short hairpin RNA (shRNA) targeting human papillomavirus 16 (HPV16) E7 gene on the expression of 4 kinds of DNA methyltransferases (DNMTs),including DNMT1,DNMT3A,DNMT3B and DNMT3L,in HPV16-positive cervical cancer cell line SiHa.Methods The recombinant plasmid containing HPV16 E7 gene-targeting shRNA was constructed firstly.Then,the BLOCK-iTTM lentiviral RNAi expression system kit was used to package the lentiviral vector,which was transfected into 293T cells.The lentivirus-containing supernatants were collected at 48 and 72 hours after transfection.The SiHa cells were divided into 3 groups to be cultured with lentiviral supernatant containing HPV16 E7 gene-targeting shRNA recombinant plasmids mixed with complete medium at a ratio of 1:1 (shRNA group),lentiviral supernatant containing empty plasmids mixed with complete medium at a ratio of 1:1 (negative control group),and complete medium alone (blank control group),respectively.Real-time fluorescence-based quantitative PCR (qRT-PCR) was performed to measure mRNA expression of HPV16 E7 and 4 kinds of DNMTs in the above 3 groups at 0,48,96 hours after infection,and Western blot analysis to determine protein expression of the 4 DNMTs at 48,96 hours after infection.Results There were no significant differences in the mRNA expression of HPV16 E7 and the 4 DNMTs among the shRNA group,negative control group and blank control group at 0 hour after infection (all P ＞ 0.05).At 48,96 hours after infection,the mRNA expression of HPV16 E7 and the 4 DNMTs decreased significantly in the shRNA group compared with the negative control group and blank control group (all P ＜ 0.05),but did not differ between the negative control group and blank control group (all P ＞ 0.05).Additionally,E7,DNMT1,DNMT3A,DNMT3B and DNMT3L gene-silencing efficiencies in the shRNA group were 71.13％,50.53％,13.72％,46.27％ and 17.92％ at 48 hours,and 83.50％,74.2％,47.8％,64.7％ and 48.9％ at 96 hours after infection,respectively.Western blot analysis showed that the protein expression of the 4 DNMTs significantly decreased in the shRNA group compared with the negative control group and blank control group at 48,96 hours after infection (all P ＜ 0.01).Moreover,the protein expression of DNMT1,DNMT3A,DNMT3B and DNMT3L in the shRNA group gradually decreased over time,and was inhibited by 84％,37.2％,59.8％ and 49.3％ at 48 hours respectively,and by 73.1％,68.7％,55.5％ and 65.5％ at 96 hours after infection respectively.Conclusion Targeted silencing of E7 gene in HPV16-positive SiHa cells can interfere with the mRNA and protein expression of DNMT1,DNMT3A,DNMT3B and DNMT3L.

Objective To find out about the temperature ,relative humidity, concentration and species of air microbes in the warship , and to detect the possible pathogenic agents responsible for the high incidence of respiratory diseases among the crew.Methods A digital hygrometer was used to measure the indoor tempreture and relative humidity .Open agar set-tling plates were used to sample the air bacteria in the ship .Pure bacterial colonies were isolated from those on the original plates by repeated streak inoculation .The species of bacteria were determined by gram staining ,light microscope observation and 16S rDNA sequencing .Results Spots that met the standard requirement of temperature accounted for 20%but relative humidity was up to the stevrdard ineach place investigated .Although the concentration of airborne microbes was under the standard limit,bacteria were various.There were 18 different bacteria isolated from the air sample , including Chryseobacteri-um jejuense,Acinetobacter baumannii, Bacillus sporothermodurans, Staphylococcus aureus, Staphylococcus capitis, Staphylo-coccus hominis subsp.hominis strain and subsp.novobiosepticus, Staphylococcus cohnii subsp.cohnii and subsp.urealytic-us,Enhydrobacter aerosaccus,Corynebacterium casei,Macrococcus caseolyticus,Corynebacterium ammoniagenes,Brevibacillus panacihumi,Comamonas koreensis,Bacillus subtilis,Sphingobium yanoikuyae, Chryseobacterium hominis.Conclusion The high incidence of respiratory diseases among officers and soliders might be related to the lower tempreture and some airborne bacteria in the warship .
