[Objective]To investigate the endogenous erythropoietin expression of neural stem cells(NSCs) of rats after hypoxia.[Method]We used a rat NSCs culture model.EPO expression after 4 hours of hypoxia was studied. It was examined again following 12 hours of hypoxic lethal insult which was applied various times after hypoxic preconditioning.EPO expression was studied by western blotting.[Result]Following the preconditioning, expression of EPO increased immediately and reached a peak 4 hours later. It lasted till 8 hours later. While applied the lethal hypoxia,EPO expression of cells in preconditioning 24 hours group were was significantly increased than that in preconditioning 4 hours group.[Conclusion]The hypoxic preconditioning can lead an increased endogenous EPO expression and decrease the threshold of EPO expression against subsequent lethal stimulation during a certain period.

Objective To analysis the flash electroretinogram (FERG) of diabeic retionpatby patients. Method 90 cases of DR patients were selected. According to the partial signs and the overall symptoms, the patients can be divided into three syndromes - dry heat by yin deficiency, deficiency of both qi and yin, deficiency of yin and yang. FERG were detected. Result There was close relation between the occurrence and development of DR and the evolution of the syndrome from yin deficiency to yang involved by deficient yin. Retina actional potential will change along with the change of syndrome. Conclusion The retina actional potential which recorded by FERG can be used as an objective criterion of syndrome differentiation, thus we can establish a diagnosis and treatment system of DR.

Objective To study the association between the CTLA-4 gene polymorphism and Graves disease. Methods A total of 120 unrelated patients with Graves disease southern Han nationality of China were recruited from clinics in the Department of Endocrinology and Metabolism, the First Affiliated Hospital of Sun Yat-sen University from 2001 to 2003.Ethnically matched 123 control subjects with no history of autoimmune disease were from Guangdong. We investigated the polymorphism of the cytotoxic T lymphocyte antigen 4 gene on chromosome 2q33.1 microsatellite markers of CTLA4 were chosen, this was done by PCR amplification of marker sequences using fluorescently labeled primers and subsequent analysis of the PCR products on ABI prism 377.Results Twenty alleles were observed in the population, with sizes ranging from 84 to 144 bp.There was no significant difference when compared with controls in the distribution of the genotypes(?2=21.428,P=0.208).This investigation indicated that the gene of CTLA4 was not susceptible gene for Graves disease. Conclusion This study suggests that CTLA4 gene may not be susceptible gene to Graves disease for the Chinese southern Han population.

Chinese Journal of Tissue Engineering Research 2014;(1):75-80
BACKGROUND:Compared with mesenchymal stem cells from bone marrow and fat, human umbilical cord-derived mesenchymal stem cells, as one of the most potential cellsources to repair the central nervous system, are easy-based, more primitive, and not limited by ethical and legal.
OBJECTIVE:To explore differentiation of human umbilical cord-derived mesenchymal stem cells into neural stem cells induced by the recombinant human basic fibroblast growth factor and recombinant human epidermal growth factor.
METHODS:The human umbilical cord-derived mesenchymal stem cells were induced by basic fibroblast growth factor and epidermal growth factor in vitro. The cellmorphology was observed by invert microscope. The differentiation status was detected by immunofluorescence. The Nestin expression in mRNA level before and after induction was detected by real-time PCR.
RESULTS AND CONCLUSION:The neural stem cellbal s were observed after induction. And the Nestin was detected by immunofluorescence and real-time PCR. Nestin could further differentiate to the neuronal markproteins neuron-specific enolase, microtubule-associated protein 2 protein and glial fibril ary acidic protein. Results from this study show that basic fibroblast growth factor and epidermal growth factor can induce human umbilical cord-derived mesenchymal stem cells to differentiate to neural stem cells, neurons and glial cells.

Objective To compare the effects of remifentanil infused at different rates on median effective target plasma concentration (EC50) of propofol inhibiting responses to laryngeal mask airway (LMA) insertion and determine the optimum infusion rate of remifentanil when used for fiberoptic bronchoscopy in pediatric patients. Methods Eighty-four ASA Ⅰ or Ⅱ pediatric patients, aged 7 months-3 years, scheduled for elective fiberoptic bronchoscopy, were randomly assigned into 3 groups (n =28 each):normal saline group (group C),remifentanil infused at 3 ng· kg-1 ·min-1 group (group R1) and remifentanil infused at 5 ng· kg-1 · min-1 group (group R2).Responses to LMA insertion were defined as body movement and/or bucking during insertion. The initial target plasma concentrations of propofol were 5.2,4.8 and 4.4 μg/ml in groups C,R1 and R2,respectively.The target plasma concentration of propofol was determined by up-and-down sequential allocation. Each time the target plasma concentration increased/decreased by 0.2μg/ml.EC50 and 95 % confidence interval of propofol blunting responses to LMA insertion were determined by probit method. Results EC50 (95 % confidence interval) of propofol was 5.03 (4.92-5.12) μg/ml,4.71 (4.58-4.84) μg/rnl and 4.46 (4.20-4.94) μg/ml in groups C,R1 and R2,respectively.There was no significant difference in EC50 of propofol between groups R1 and C (P > 0.05).EC50 of propofol was significantly lower in group R2 than in groups C and R1 (P < 0.05).Conclusion The infusion rate of remifentanil should not be lower than 5 ng· kg-1· min-1 when combined with propofol in pediatric patients undergoing fiberoptic bronchoscopy.
