2.High mobility group box 1 protein and intracerebral hemorrhage
International Journal of Cerebrovascular Diseases 2016;24(6):565-568
High mobility group box l protein (HMGB1) is a highly conserved non-histone DNA binding protein, which is widely distributed in eukaryotic cells.HMGB1 is an important proinflammatory cytokine, involving in various pathological and physiological processes, such as inflammation, neurogenesis and angiogenesis. In the early stage of intracerebral hemorrhage,HMGB1 can induce secondary brain injury, such as inflammatory reaction, neuronal necrosis, and brain edema. However,in the late stage of intracerebral hemorrhage,HMGB1 can promote angiogenesis and neurogenesis, and thus improving neurological function.HMGB1 will become a novel therapeutic target for intracerebral hemorrhage in the future.

ObjectiveTo summarize the short-term effects o f Symmetry aortic co nnector in off-pump coronary artery bypass grafting (OPCAB) operation. MethodsTwo hundreds and four patients suffering from coronary artery d isease underwent OPCAB from September 2002 to June 2003. Among the 204 patients, saphenous vein grafts were used in 167 patients and Symmetry aortic connectors were adopted in 100 of them.ResultsOf 142 connectors used, 2 connectors fai led and manual r epairs were required in 3 cases because of anastomotic leakage, the success rate being 96 5% (137 of 142). The number of proximal anastomosis was (1 4?0 5) per case and the number of distal anastomosis (3 5?0 8) per case. The proximal a na stomotic blood flow was (46 5?22 3) ml/min. The fitting time for connectors w as (4 0?1 1) min, and anastomoses were all accomplished within 10 seconds. The op eration time was (3 2?0 4) hours. The operative death rate was 1% (1 of 100). R e-operation of thoracotomy was required in 1 case because of bleeding but not an astomotic bleeding. No myocardial infarction or stroke occurred in the peri-oper ative period. The postoperative drainage amount was (547?247) ml. The blood tra nsfusion rate was 36 0% (36 of 100). The postoperative hospital stay was (7 8 ?1 6) days.ConclusionsSymmetry aortic connector can be saf ely and effectively used in coronary artery bypass grafting operations, with satisfactory short-term clinical effects. The procedure may shorten operation time, avoid aortic clampi ng, and lower the incidence of stroke.

Objective To summarize the mid-term outcomes of minimally invasive direct coronary artery bypass (MIDCAB). Methods A series of 33 patients underwent MIDCAB from November 2000 to April 2003: 29 of them received MIDCAB only and 4 received the hybrid approach operation (combining MIDCAB of the left anterior descending artery with percutaneous coronary intervention of the remaining diseased coronary arteries). Out of the 33 cases, conventional MIDCAB was carried out in 25 cases, 4 underwent thoracoscopy-assisted operation and 4 received the surgery with the assistance of the AESOP robot system. Results No operation-related deaths or complications were seen in the study. The extubation time was (7.5?1.9) hours, the chest drainage volume was (274?197) ml, the blood transfusion rate 9.1% (3/33) and the postoperative hospital stay (6.9?1.7) days. Follow-up in all the 33 cases for (24.3?5.7) months revealed no long-term deaths and the recurrence rate of angina was 9.1% (3/33). Postoperative coronary angiography in 4 cases found 1 case of 50% stricture of anastomosis and 3 cases of anastomotic patency between the anterior descending artery and left internal mammary artery. Postoperative catheter revascularization was required in 2 cases (6%). Conclusions The mid-term outcomes of MIDCAB is satisfactory.

Objective To Construct fusion gene DNA vaccine pcDNA3/STxB-VP1 and fusion gene DNA vaccine pcDNA3/MDC-VP1,then the two vaccines were inoculated to mice and stuy their immunological effects. Methods B subunit of Shiga Toxin(STxB)gene fragment were amplified by PCR from the DNA of Shigella dysenteriae and Macrophage-derived chemokine(MDC)gene fragment was amplified by RT-PCR from the total RNA of a mouse spleen cells. The DNA fragments of STxB and MDC waere respectively linked to coxsackievirus B3(CVB3)VP1 by a DNA sequence which encode a flexible polypeptide(15 amino acids)to construct eukaryotic expression plasmids pcDNA3/STxB-VP1 and pcDNA3/MDC-VP1.BALB/c mice were randomized to 6 groups which were respectively immunized pcDNA3,pcDNA3/STxB,pcDNA3/MDC,pcDNA3/VP1,pcDNA3/STxB-VP1 and pcDNA3/MDC-VP1 for 3 times at 3-week intervals,intramuscularly(i.m.)in tibialis anterior muscle. The levels of the serum neutralizing antibodies were detected 20d after each inoculation. The mice were challenged with 7 LD50 CVB3 3 weeks after the last immunization. Results The fusion gene vaccines pcDNA3/STxB-VP1 and pcDNA3/MDC-VP1 were constructed successfully. The survival rates of each group were 10%,10%,15%,40%,20% and 75%,respectively,and the levels of neutralizing antibody titers, virused titers, were all consistent with those survive rates in each group. Conclusion Comparing with pcDNA3/VP1,pcDNA3/MDC-VP1 vaccine can induce a high level of neutralizing antibody titers and result in a higher survival rate in mice, while pcDNA3/STxB-VP1 induce a low level of neutralizing antibody titers and result in a lower survival rate in mice.

Acta Nutrimenta Sinica 1956;0(01):-
Zinc, copper, manganese, cadmium, vitamin A, E and folic acid status of 36 pregnant and 40 nonpregnant women was studied. The results showed that dietary nutrient intakes of the pregnant women were more than that of the nonpregnant controls, but both of which were less than the corresponding values of the RDA. Serum zinc, folic acid and hair manganese concentrations were lower, while serum copper, cadmium, vitamin A and E were higher in pregnant women than that in nonpregnant women. The simultaneous occurrence of serum zinc decrease and cadmium increase during pregnancy may be a potential risk for the growth and development of foetus. So the interfering role of cadmium should not be neglected whenever we estimate zinc or copper status.

Objective To develop autosomal SNP fluorescent-multiplex PCR system, and investigate the allele frequencies of these 13 SNP loci in Southern Liaoning Han population. Methods After selected 13 autosomal SNP loci,SNP fluorescent-multiplex system was developed based on the principle of fragment length discrepant allele specific PCR,and the allele frequencies of these SNP loci in Southern Liaoning Han population were investigated using the established system. Results For the same SNP locus, the homozygote showed a single product peak, and the heterozygote showed two product peaks with different length. For the different SNP loci, the lengths of their PCR products were different. Therefore,we could genotype the 13 SNP loci simultaneously according to the length of products and the amount of product peaks, and the results were in accordance with that of direct sequencing. In the meanwhile, the allelic frequencies of these 13 SNP loci in Southern Liaoning Han population were obtained. Conclusion The SNP fluorescent-multiplex system based on the fragment length discrepant allele specific PCR strategy is simple and economical, and is of a high application value in forensic medicine.
