Objective To analyze the clinicopathological characteristics, immunophenotypes and immu-noglobulin heavy chain gene rearrangement of gastric mucosa-associated lymphoid tissue (MALT) lymphoma. Methods The clinical data of 35 patients with gastric MALT lymphoma who had been admitted to Southwest Hospital from January 1984 to June 2003 were retrospectively analyzed. The specimens of the gastric MALT lymphoma were obtained and their pathomorphological classifications were studied. The immunophenotypes and immunoglobulin heavy chain gene rearrangement were detected by immunohistochemistry and polymerase chain reaction, respectively. All the data were analyzed by chi-square test and Mann-Whitney U test. Results The main manifestations of patients with gastric MALT lymphoma were epigastric discomfort and abdominalgia. The percentage of patients with age≥40 accounted for 74% (26/35). The gastric MALT lyrnphoma has low differentia-tion in 32 patients, and high differentiation in 3 patients. There were 31 patients in Ⅰ E stage, 2 in Ⅱ E1 stage and 2 in Ⅳ stage. Pathological examination showed that gastric MALT lymphoma was composed of centrocyte-like cells and mononuclear-like B cells or lymphoplasma cells. Twenty-nine specimens were with lymphoepithelial lesions (83%), 2 with follicular colonization (6%), and 11 with plasma cells segregated beneath the surface epithelium (33%). All the 35 specimens were B cell immunotype. Restrictive expression of κ and λ light chains of the immunoglobulin was found in 19 cases, and Bcl-2 positive expression was found in 16 cases. High expres-sion of proliferating cell nuclear antigen was found in gastric MALT lymphoma with transformed blasts, which indicated that gastric MALT lymphoma with transformed blasts was more active in proliferation than those of low grade. A monoclonal pattern of immunoglobulin heavy chain gene rearrangement was detected in 33 patients (94%). Conclusions Digestive symptoms are the main manifestations of patients with gastric MALT lymphoma. The incidence of gastric MALT lymphoma increases with age. The final diagnosis of gastric MALT lymphoma depends on the pathological, immunohistochemical examination and laboratory test.

Objective To study the effect and mechanism of Hedyotic diffusa injection in reversing multi-drug resistance of BEL-7402/5-FU cells. Methods The IC50 of BEL-7402/5-FU cells was examined by MTT,apoptosis was detected by flow cytometry, and the content of tyrosine kinase in total protein of tumor cells was detected by enzyme-linked immunosorbent assay (ELISA).Results The IC50 of Hedyotic diffusa injection (6 μL/mL ) combined with 5-FU (636.5 μmol/L)decreased significantly compared with that of 5-FU applied alone (1 071.6 6μmol/L). The reversal on BEL-7402/5-FU was 1.68-fold.After injection of 5-FU plus Hedyotic diffusa ,the apoptosis rate of BEL-7402/5-FU cells was increased significantly and the tyrosine kinases of BEL-7402/5-FU cells decreased significantly. Conclusion Hedyotic diffusa injection plays a certain role in reversing multi-drug resistance of BEL-7402/5-FU cells, which may be related to decreasing their tyrosine kinases.

Chinese Journal of Tissue Engineering Research 2014;(32):5178-5183
BACKGROUND:Tol-like receptor 4 (TLR4) and its ligand, lipopolysaccharid, are closely associated with the occurrence and development of periodontitis. Meanwhile, the immunological properties of periodontal ligament stem cells (PDLSCs) play an important role in the reconstruction of periodontal tissue and cel-based therapy of periodontitis. However, the effect of TLR4 and lipopolysaccharid on the immunological properties of PDLSCs remains unclear. OBJECTIVE:To investigate the effect of TLR4 on the immunological characteristics of human PDLSCs. METHODS:PDLSCs were isolated by enzyme digestion method as previously reported, and were cultured in the medium containing 10 mg/L lipopolysaccharid, the ligand of TLR4 for 3 days. Using un-treated PDLSCs as controls, we then investigated whether lipopolysaccharid-treated PDLSCs could cause the proliferation of al ogeneic T lymphocytes as wel as the effect of lipopolysaccharid-treated PDLSCs on the mixed lymphocytes reaction and proliferation of lymphocytes induced by phytohemagglutinin. PDLSCs, peripheral blood mononuclear cells and phytohemagglutinin were co-cultured, and the concentration of prostaglandin E2 in the culture supernatant was examined. Then we added indomethacin, which is the inhibitor of prostaglandin E2, into the co-culture system of PDLSCs, peripheral blood mononuclear cells and phytohemagglutinin, and tested the proliferation of lymphocytes. RESULTS AND CONCLUSION:Lipopolysaccharid-treated PDLSCs did not lead to the proliferation of al ogeneic T lymphocytes just as un-treated PDLSCs, and could suppress the mixed lymphocytes reaction and proliferation of phytohemagglutinin-induced lymphocytes. However, the inhibitory ability of lipopolysaccharid-treated PDLSCs was significantly lower than that of un-treated PDLSCs. The levels of prostaglandin E2 were significantly elevated in the co-culture of PDLSCs, peripheral blood mononuclear cells and phytohemagglutinin. After adding of indomethacin, the PDLSCs-suppressed proliferation of lymphocytes restored to normal levels. Lipopolysaccharid weakened the immunosuppressive capacity of PDLSCs, which may be due to the decreasing secretion of prostaglandin E2.

Aim To investigate the effects of total gin-senosides ( TG) on microvascular regeneration and car-diac function in rat after acute myocardial infarction ( AMI ) . Methods The acute myocardial infarction model was created with left coronary artery ligated in male Sprague Dawley rats. The model rats were ran-domly divided into sham, model, TG low and high dose groups. TG groups were injected into abdominal cavity with TG(20 mg·kg-1·d-1, 40 mg·kg-1· d-1 ) . Sham and model groups were injected with e-qual-volume normal saline. On the 35th day post-opera-tion, heart function was examined by color doppler ul-trasoundination. HE, Masson and immunohistochemis-trical staining were used to observe the histopathologi-cal changes of myocardium and micro vessel density. The level of vascular endothelial growth factor( VEGF) and basic fibroblast growth factor( bFGF) mRNA were detected by real-time PCR. Results Compared with the model group, high and low dose TG obviously de-creased the left ventricular end diastolic dimension, the left ventricular end-systolic dimension, the left ventric-ular end-diastolic volume and the left ventricular end-systolic volume(P<0. 05 and P<0. 01), and signifi-cantly increased the ejection fraction and the fractional shortening ( P <0. 01 ) . The histopathological changes of myocardium on myocardial infarction and fibrosis were dramatically reduced by TG. But ventricular wall was thicker. Two dose TG remarkably increased the expressions of VEGF and bFGF mRNA and micro ves-sel density of compositive CD31 + cells in ischemic myocardial tissue and around of infarct area(P<0. 01, P <0 . 0 5 ) . Conclusions TG could improve the car-diac function of acute myocardial infarction rat. The mechanism may be related to the upregulation of VEGF and bFGF gene expression, the promotion angiogene-sis, then the improvement of blood supply in myocardi-al infarction area.
