Ten pharmacophore models of beta-tubulin inhibitors were established from the training set of seventeen beta-tubulin inhibitors (two categories) with comformer analysis by using the Catalyst software. The optimal pharmacophore model with two hydrophobic units and two hydrogen bond acceptor units were confirmed (RMS = 0.43, Correl = 0.98, Weight = 2.06, Config = 15.97). This pharmacophore model is able to predict the activity of known beta-tubulin inhibitors and can be further used to identify structurally diverse compounds with higher activity.

Objective To investigate the significance of microsatellite instability(MSI) and expressions of cyclin D1 and P53 in 7,12 dimethylbenz(a)anthracene (DMBA) induced breast carcinoma in rats. Methods Animal model of rat breast carcinoma was duplicated in 30 female Wistar rats by gastric irrigation of DMBA at the dose of 200 mg/kg. Eight microsatellite loci(D1Mitl4, D1Mgh2, D1Mgh6, D3Mit3, D3Mgh9, D5Mitll, D5Mgh5 and D7Mgh1) were selected for the examination of MSI of breast carcinoma by PCR analysis. S P immunohistochemical staining was used to study the expressions of cyclin D1 and P53 in DMBA induced breast carcinoma in rats. Results Of the 27 cases of breast carcinoma, there were 2 cases of MSI at 8 loci, 9 cases with P53 overexpression and 12 cases with cyclin D1 overexpression. All these were associated with histological grading. Conclusion MSI is infrequent in breast carcinogenesis in rats. Cyclin D1 overexpression may be the early event but P53 protein overexpression may be the late event in breast carcinoma.

A method was developed to assay α-Hydroxyltriazolam and α-Hydroxyalprazolam, which are the major metabolites of triazolam and alprazolam respectively,in human urine. After addition of 2-hydroxyflurazepam (interal standard) and hydrolysis with β-glucuronidase, the hydroxy-metabolites were extracted with hexane-dichloromethane (1∶1) at pH 10.8, then were derivated with (BSTFA). The analysis was performed on a HP-5 capillary column with electron-capture detector. The detection limits of analysis in urine were about 1μg/L.The method was successfully applied to urine specimens collected from healthy human volunteers who ingested 0.5 mg of triazolam or 0.8 mg alprazolam. The method was enough sensitive to assay urine specimen excreted at 24 h by volunteers after taking the medicine.

Objective To study the correlation between the expressions of Bcl-2 and Caspase-3 in intestinal epithe-lial and inflammatory cells of lamina propria and the staging and pathogenesis of ulcerative colitis (UC). Methods The pathomorphological changes of 25 samples of UC in the active phase and remission phase and 25 samples of control were ob-served under microscope. The expressions of Bcl-2 and Caspase-3 were detected by immunohistochemistry SP method. The correlations between expressions of Bcl-2, Caspase-3 and UC staging were analyzed. Results There were no significant dif-ferences in expressions of Bcl-2 and Caspase-3 in intestinal epithelial cells between three groups (P>0.05). The expression of Bcl-2 in inflammatory cells of lamina propria was significantly higher in active UC group than that of normal control group (P<0.05), no significant difference between UC remission group and normal control group (P>0.05). There was a higher positive expression of Caspase-3 in inflammatory cells of lamina propria in UC remission group. There was correlation be-tween Bcl-2, Caspase-3 and UC staging (r=0.371 and 0.4453 respectively). Conclusion The expression levels of Bcl-2 and Caspase-3 were related with pathogenesis of UC. The detection of expression levels of Bcl-2 and Caspase-3 has a cer-tain application value in UC staging.

Objective To investigate the changes of intracellular GSH content in HepG2 cells infected with dengue virus(DV)or stably expressing E or NS3 protein. Methods HepG2 cells were cocultured with DV or inactivitated DV,l h later the viral supernatant was removed. The infected HepG2 cells were collected 10,20,30,40,60 min,and 2,6,12,24,48 h after the beginning coculture and intracellular GSH content was detected by spectrophotometry.Intracellular GSH content was also detected in HepG2 cells stably expressing E protein/NS3(pReceiver-E/HepG2,pReceiver-NS3/HepG2)and those containing empty plasmid(pReceiver/HepG2).Results GSH content showed a decreasing tendency after DV-2 infection. The lowest values were seen 30 min and 2,24 h after infection, which were of significant difference in comparison with those in inactivated DV infected HepG2 cells as well as at other time points.GSH levels in pReceiver-E/HepG2,pReceiver-NS3/HepG2 were significantly lower than those in pReceiver/HepG2.Conclusion DV-2 infection might lead to the GSH depletion in HepG2 cells, and GSH lost from HepG2 cells might undergo a three-step process:virus adsorption/penetration,protein synthesis and budding.E or NS3 protein stably expressed in HepG2 cells can also decrease the GSH levels.
