Chinese Journal of Tissue Engineering Research 2005;9(13):196-198
BACKGROUND: It is well known that central noradrenergic system takes part in learning and memory, but there are a few researches focused on the effects of impairing the central noradrenergic system on rat' s P3-like cognitive potential.OBJECTIVE: To study the effects of impairing the central noradrenergic system on rat's P3-like cognitive potential.DESIGN: Randomized controlled study based on the experimental animals.SETTING: Department of physiology in a medical college of a university.MATERIALS: Totally 30 male SD rats, clean degree, weighing from 500 to 600 g, were selected from the Experimental Animal Center of Sun Yat-sen Medical University(certificate: 26-99A018), drank freely and fed timely under the normal condition. After Y-maze test of all the rats, 24 rats with normal learning and memory function, which met the criteria specified in the description of Y-maze test, were randomly divided into three groups(8 rats in each group): control group, 6-oxidopamine (6-OHDA) injection group and saline control group.INTERVENTIONS: The models of central norndrenergic system impairment were made by injecting 6-hydroxysopamine(6-OHDA) into the bilateral dorsal noradrenergic bundle in dentate granule(DG). Then Y-type maze test and elicitation of P3-likewere carried out respectively before and after the models were built.MAIN OUTCOME MEASURES: Main outcome: P3-like latency, total reaction time(TRT) and error number(EN). Secondary outcome: Results of P3-like test recorded in the 12 days after the models were built.RESULTS: As compared with the controls, the model rats' P3-like latency and total reaction time(TRT) prolonged and error number(EN) increased statistically ( P < 0.05 ).CONCLUSION: Central NE takes part in the activities of learning and memory and plays a certain role in the production and integration of P3 -like.

Objective In order to study the biological significance of HBV/C gene variant in vitro. Methods Retroviral vector pXT1 was used to construct the HBV/C gene expression vector and the recombinant plasmid pXT1-HBV/C was used to transfect immortalized human peripheral blood B cell lines to express HBcAg steadily in the host cells. Results plasmid pXT1-HBV/C was detected positive by PCR as well as enzyme digestion with Bgl Ⅱ and Xho Ⅰ.Meanwhile.transfected cells was detected to contain HBV/C gene by PCR and HBcAg was expressed in 47.4%of the ceils by means of flow cytometry. Conclusion Retroviral expression vector with HBV/C gene can transfeet into eucaryotic cells effectively and express the goal gene steadily. The recombinant cells may be used in the systematic studies on the biological significance of HBV/C gene variant.

Objective To study cervical human papillomavirus (HPV) infection and genotype distribution characteristics in patients with vaginal disease. Methods The PCR dot blot was used to carry out HPV detection of cervical exfoliated cells in 816 women with cervical lesions in the hospital, and 23 kinds of HPV genotypes were identified. Results Among the 816 cases receiving HPV type detection, HPV of 532 cases (65.20%) was positive, including 371 cases (69.74%) with low-risk HPV (HR-HPV) infection, 161 cases (30.26%) with low-risk HPV infection (LR-HPV) (P<0.05).The HPV infection rate below 45 years old was relatively higher. The HPV infection rate in patients with cervical cancer (97.10%) was higher than that in patients with cervical intraepithelial neoplasia (CIN) II -III (80.54%), patients with CIN Ⅰ(41.55%) and patients with cervicitis (17.20%) (P<0.05).The top five types of HPV infection were HPV16, 58, 18, 11 and 56 type, respectively. The single infection rate (79.70%) was higher than multiple infection rate (20.30%) (P<0.05); HR-HPV infection was the main infection in single infection, and double infection was the main infection in multiple infection. Conclusion Below 45 years old women are the high-risk groups of HPV infection.16, 58, 18, 11 and 56 are the main types of HPV infection. The infection rates of patients with cervical cancer and precancerous lesions are relatively higher.Paying attention to the screening of HPV infection has positive significance in the prevention and treatment of cervical cancer.

Objective To study the biological significance of the common mutant preC/C gene in clinical HBV in China. Methods Site directed mutagenesis based on the unique enzyme site elimination was used to construct eukaryocyte expression vector with mutant HBV/C gene(V60、G87、L97). Expression vectors with wild and mutant preC/C gene were transferred into HepG2 cell. Culture supernatant was detected by ELISA for HBeAg. Results Result of DNA sequencing showed that the constructed mutant HBV preC/C gene had only one specific site variation compared with the wild type sequence. Goal DNA fragment was detected positive in the HepG2 cells transferred with wild and mutant preC/C gene. A value of HBeAg in the supernatant of the cells harboring L97 variant was higher than that of the wild and other variant strains( P

Medical Journal of Chinese People's Liberation Army 2001;0(11):-
To study the characteristics of EBV transformed human peripheral blood B cell lines from hepatitis B patients and to provide a basis for further studies on the cellular immune function of hepatitis B patients.High titer of EBV was produced by B95 8 cell by induction with sodium n butyrate, and peripheral mononuclear cells from two different groups of hepatitis B patients were harvested and infected with EBV. The results showed that 4~8 weeks after infection 19 EBV transformed B cell lines from hepatitis B patients were established. It was found that colony formation of cells from A group appeared 2~3 weeks later than B group. Cells grew healthily and had good activity after 4 weeks of freezing.CD19 and CD20 were detected in 88 34% and 37 48% of the immortalized B cell membrane respectively. HBV DNA could not be detected in a11 19 immortalized B cell lines. It suggested that the time needed for the establishment of B cell lines was related with the immune state of the patients.HBV DNA could not exist persistently, and it would disappear finally in the immortalized B cell lines.

Chinese Journal of Tissue Engineering Research 2017;21(17):2759-2765
BACKGROUND: Bone marrow mesenchymal stem cells (BMSCs) with multiple differentiation potential can be induced into osteogenic or adipogenic differentiation under certain conditions. OBJECTIVE: To review the related factors regulating the adipo-osteogenic differentiation of BMSCs. METHODS: A computer-based search of CNKI and PubMed databases was performed for literature concerning the related factors regulating the adipo-osteogenic differentiation of BMSCs published from January 2006 to August 2016. The search terms were bone marrow mesenchymal stem cells, osteogenic differentiation, adipocyte differentiation in Chinese and English, respectively. RESULTS AND CONCLUSION: Signaling pathways, transcription factors, and smal molecule compounds that are interacted are key factors in the regulation of BMSCs differentiation, so the techniques to intervene BMSCs differentiation based on these key molecules may correct bone or fat abnormality and can be applied to tissue engineering and regenerative medicine in the future. Additionally, the biological clock is also one of the most important factors for adipo-osteogenic differentiation of BMSCs by regulating signaling pathways or transcription factors.

Objective Effects of exercise training and Chinese Medicine Compound for Invigorating Qi and Tonifying the Kidney on testicular StAR and P450 SCC mRNA expression in rats were observed. Methods 50 Wistar rats were randomly divided into 4 groups: sedentary control group(SC, n=10), sedentary with medicine group (SM,n=10), training group(T, n=15) and training with medicine group(TM, n=15). After 6 weeks swimming training , serum testosterone level of rats were tested through radioimmunological method; using RT-PCR, we measured testicular StAR and P450 SCC mRNA levels of rats. Results (1) In T group, serum testosterone level was lower than that in SM and TM groups. Whereas, compared with SC group, the serum testosterone level in TM group kept steadiness. (2) In T group, the gene expression of StAR was markedly lower than that in SC control (P
