2.Efficacy of propofol compared with sevoflurane on inflammatory factor and liver function after hepatectomy

Objective To evaluate efficacy of propofol compared with sevoflurane on inflammatory factor and liver function after hepatectomy. Methods 40 cases with hepatectomy were divided into propofol group ( P group) and sevoflurane group ( S group) , 20 cases in each group.The P group and S group respectively were given propofol or sevoflurane maintained anesthesia.The alanine aminotransferase( ALT) , total bilirubin( TBil) , aspartate amino transferase(AST), albumin(ALB), plasma tumor necrosis factor(TNF-α), interleukin-6(IL-6), interleukin-10(IL-10)and complication rate were compared between two groups.Results Postoperation 1d, 3d and 7d, the AST, ALT, and TBiL levels of S group were significantly lower than those of P group (P<0.05).There was no significant difference of ALB level between two groups.The plasma TNF-α, IL-6 and IL-10 levels of S group were significantly lower than those of P group (P<0.05).There was no significant differece of the rate of postoperative complications between S group and P group [6(30.0%) vs.10 (20.0%); χ2 =1.67, P >0.05].Conclusion Sevoflurane inhalation could effectively inhibit the plasma inflammatory cytokines secretion after hepatectomy, and promote liver function recovery, its effect is better than that of propofol.
3.Management of Antimicrobial Drugs During Perioperative Period:Application of Information Technology

OBJECTIVE In order to improve the traditional management mode,we try to explore the application of information technology for management of antimicrobial drugs during perioperative period. METHODS Through analysis of our research program case,the differences between the traditional management mode and the information-based management mode were compared by the method of reviewing literature data and analyzing hierarchy process of administration. RESULTS There were obvious limitations in the traditional management mode,but the information-based management mode exhibitsed evident advantages,because it focused on node control and process management. CONCLUSIONS The information-based management mode can establish a long-term management mechanism of antimicrobial drugs during perioperative period,and also can individualizedly implement precise management goal.It is an innovation of the traditional management mode.
4.Content Determination of Hesperidin in Qiansheng Baizhusan Jiawei Granule by HPLC

Objective To establish a method for the determination of hesperidin content in Qiansheng Baizhusan Jiawei Granule (QBJG). Methods The content of hesperidin was determined by HPLC. Results Hesperidin showed a good linearity in the range of 0. 22-1.1ug(r = 0. 9998). The average recovery was 103. 0 % and RSD was 3. 12 %. Conclusion The methods are accurate and can be used for the quality control of QBJG.
5.Interleukin-12 blocking GATA-3 expression in treatment of bronchial asthma:an experimental study

Objective:To observe the expression of GATA-3in asthmatic mice lungs and to explore the feasibility of in-terleukin(IL-12)blockading GATA-3expression in treatment of bronchial asthma.Methods:C57BL/6mice were randomly divided into3groups:control group(group A),asthmatic model group(group B)and IL-12injection group(group C).Asth-matic models were established in group B and C.Normal saline(0.1ml)and IL-12(1?g)were injected in group B and C re-spectively on the1,3,7,9,25,26,27and28d.Six mice from each group were obtained for analyses of bronchoalveolar lavage fluid(BALF)on d31.The airway pathology changes and the expression of GATA-3were observed by hematoxylin/eosin(H-E)and immunohistochemistry respectively.Results:There was no symptom in group A,and the symptoms of group B were more severe than those of group C.Eosinophil(EOS)was not seen in the BALF of group A,while EOS in group B and group C were(20.0?4.0)%and(0.2?0.1)%respectively.In the same microscopic visual field,there was no inflam-mation cell in group A and a large number of inflammation cells in group B,but inflammation cells in group C were signifi-cantly decreased.Immunohistochemistry showed that the expression of GATA-3in group B was strong and no GATA expres-sion was found in group A and group C.Conclusion:The expression of GATA-3in asthmatic mice is high;IL-12can inhibit asthmatic airway and lungs inflammtion,whose mechanism may be the blockade of GATA-3expression in asthmatic models.[
6.The effects of a high-intensity pulsed electromagnetic field on the expression of β-catenin in the neural stem cells of neonatal rats

Objective To study the mechanism by which a high-intensity pulsed electromagnetic field (HIPEMF) (0.1 Hz,4 T,8 pulses) facilitates the proliferation of neural stem cells by detecting the expression of β-catenin genes and protein.Methods Neural stem cells (NSCs) were isolated from the sub-ventricular zone (SVZ) of neonatal rats and cultured in supplemented,serum-free medium for two weeks.The NSCs were then divided into an experimental group exposed to a HIPEMF for 8 pulses and a control group given sham stimulation.The gene and prorein expression of β-catenin in the NSCs were assayed by RT-PCR and Western blotting on the 1st,3rd,5th and 7th day after the stimulation.Results The NSCs' cloned spheres were round and translucent,and showed red fluorescence after staining with anti-nestin (cy3).The RT-PCR results showed β-catenin genes were highly expressed in the exposed group (significantly more than in the controls).The Western blotting showed that expression of β-catenin protein was also higher in the experimental group,especially at the 7th day after stimulation,a difference which was also statistically significant Conclusion HIPEMF at 0.1 Hz,4 T,in 8 pulse trains can promote NSC proliferation,perhaps through the Wnt/β-catenin signaling pathways.
7.Study on in vitro Anticancer Activity of Conjugated Linoleic Acid-Gemcitabine Conjugate

OBJECTIVE:To study in vitro anticancer activity of conjugated linoleic acid-gemcitabine conjugate (CLA-GEM). METHODS:IC50 of different tumor cells (breast cancer MCF-7 cell,breast cancer MDA-MB-231 cell,lung cancer A549 cell, small cell lung cancer NCI-H446 cell,glioma C6 cell)were investigated after treated with different concentrations(0.001-100μmol/L)of CLA-GEM and gemcitabine(GEM)for 72 h;survival rates of MCF-7 cell were investigated after treated with above so-lution for 24,48 and 72 h. The dependence of 0.001-100 μmol/L CLA-GEM and GEM to nucleoside transporter was investigated (by IC50)through MCF-7 cells and MDA-MB-231 cells treated with nucleoside transporter inhibitors(NBMPR,100 μmol/L)and di-pyridamole(4 μg/ml). The change of MCF-7 cell cycle was investigated after treated with 1 μmol/L CLA-GEM and GEM for 24 h. RESULTS:Compared with GEM,IC50 of MCF-7,MDA-MB-231 and NCI-H446 cells became lower after treated with CLA- GEM (P<0.01),there were no statistical significances in IC50 between A549 and C6 cells(P>0.05). Survival rate of MCF-7 cells de-creased significantly after treated with GEM for 48 h and CLA-GEM for 24 h. Survival rate of MCF-7 cells was the lowest,being 21% after treated with GEM for 72 h,while tumor cells were sacrificed by CLA-GEM completely. Compared with GEM or CLA-GEM,IC50 of MCF-7 and MDA-MB-231 cells increased significantly after treated with NBMPR,dipyridamole combined with GEM (P<0.01);there were no statistical significance in IC50 after treated with NBMPR,dipyridamole combined with CLA-GEM (P>0.05). Compared with GEM,CLA-GEM could prolong 6% of S stage of MCF-7 cells (P<0.01). CONCLU-SIONS:Compared with GEM,CLA-GEM exhibits significant antitumor activity and rapid action,and it isn’t influenced by nucle-ic acid transportation.
8.Advances of Notch signaling regulating fetal liver stem/progenitor cells: implications for the therapy of end-stage liver diseases

Cell therapy has a very promising potential for end-stage liver diseases (ESLD).Fetal liver stem/progenitor cells (FLSPCs) have advantages of safety,high survival and proliferation rates,and a small volume,all which make them ideal for liver disease stem cell therapy.During the early phase of our study,we applied a three-step separation method to enrich FLSPCs and obtained a separation efficiency similar to that of the flow-cell sorting method.Additionally,using a fulminant hepatic failure model in rats,we have demonstrated that FLSPCs can contribute to morphological and functional recovery of the liver.This manuscript will discuss how FLSPCs can be induced to accurately differentiate into hepatocytes and cholangiocytes and how FLSPCs maintain self-renewal.The Notch signaling plays a critical role in regulating the differentiation and self-renewal of many types of stem cells.Our previous findings have shown that the Notch signaling plays an important role in FLSPCs differentiation into hepatocytes.Therefore,the Notch signaling might be involved in the differentiation and self-renewal of FLSPCs.We conducted a study on the regulatory effects and relative molecular mechanisms of the Notch signaling on FLSPCs and found the corresponding interfering target,which might become an index for the clinical application of FLSPCs.

Objective To explore the value of cardiac troponin-I (cTnI), B-type natriuretic peptide (BNP) and blood lactic acid (Lac) on evaluation of severity and prognosis in patients with septic myocardial dysfunction (SMD). Methods According to retrospective analysis of clinical data,161 cases with sepsis were divided in to SMD group and non-SMD group. And the SMD group was further divided in to death group and survival group. Blood cTnI, BNP and Lac value in each group were detected respectively. The ROC curve was used to evaluate the forecast value of cTnI, BNP and Lac on prognosis for patients. Results The value of cTnI, BNP and Lac in SMD group were significantly higher than those in non-SMD group(P<0.05);The value of cTnI, BNP and Lac in death group among the SMD patients were significantly higher than those in survival group(P<0.05);cTnI, BNP and Lac contribute to predict the 28 day mortality rate of SMD. Conclusions Blood cTnI, BNP and Lac contributes to the assessment of the severity and the prognosis of septic patients with myocardial dysfunction.
10.Clinical Observation of Oral Use and Acupoint Application of Chinese Herbal Medicine for Strengthening Spleen and Regulating Stomach, Lowering Rebellious Qi to Stop Vomiting in Treating Spleen-stomach Deficiency Pregnant Pernicious Vomiting

Objective To observe the clinical effect of oral use and acupoint application of Chinese herbal medicine with the actions of strengthening spleen and regulating stomach, lowering rebellious Qi to stop vomiting in treating pregnant pernicious vomiting. Methods One hundred and twenty-two spleen-stomach deficiency pregnant pernicious vomiting cases were randomly divided into observation group ( N=63) and control group (N=59). At the end of the trial, 2 cases of the control group dropped out, and the total case number was 57 in the control group. Both groups were treated with intravenous infusion, dietary care, daily life and emotional nursing, and oral use of Chinese herbal medicine with the actions of strengthening spleen and regulating stomach, lowering rebellious Qi to stop vomiting. Additionally, the observation group was given acupoint application of herbs for strengthening spleen and regulating stomach, lowering rebellious Qi to stop vomiting. The treatment was performed once a day, 4 days constituted one treatment course, and the treatment lasted two continuous courses. The improvement of vomiting, therapeutic effect and the safety were compared between the two groups after treatment. Results ( 1) The total effective rate was 96.8%in the observation group and was 71.9%in the control group, the difference being significant (P<0.01). (2) The time for stopping vomiting in the observation group was significantly less than that in control group (P<0.01). (3) During the treatment, no obvious adverse reaction was found in the two groups. Conclusion Acupoint application of herbs for strengthening spleen and regulating stomach, lowering rebellious Qi to stop vomiting has obvious synergistic effect on the clinical efficiency of spleen-stomach deficiency pregnant pernicious vomiting treated by oral use of herbal medicine, intravenous infusion, dietary care, and daily life and emotional nursing.
