1.Observing the influences of liver function and virus outcome with telbivudine combined with adefovir dipivoxil

Objective To observe the clinical effects of anti-hepatitis B virus with telbivudine combined with adefovir dipivoxil.Methods 82 cases of patients diagnosed with chronic hepatitis B from January 2014 to February 2016, were randomly divided into two groups, the control group were treated with adefovir ester treatment, the observation group were treated with telbivudine combined with adefovir dipivoxil treatment, patients were followed up and recorded Child-Pugh score, aspartate aminotransferase ALT, total bilirubin TBIL, aspartate aminotransferase AST, albumin ALB, HBV-DNA negative rate, HbeAg seroconversion rate case, the use of statistical methods for data analysis.Results After treatment, the observation group Child-Pugh score, ALT values were(4.01±0.79)points,(37.19±4.82)U/L, were better than the control group(5.46±1.16)points,(61.49±9.78)U/L, and the differences were statistically significant (P<0.05).Observation group after treatment TBIL, AST, ALB, respectively(34.91±5.49)μmol/L,(56.49±5.28)U/L,(45.51±4.24)g/L, were better than the control group(61.62±10.06)μmol/L,(78.64±8.16)U/L,(38.76±3.21)g/L, and the differences were statistically significant (P<0.05).The observation group after treatment for HBV-DNA negative rate and HbeAg seroconversion rate(82.93%, 58.54%)respectively were higher(58.54%, 29.27%), and the difference was significant (P<0.05).Conclusion Telbivudine combined with adefovir dipivoxil anti-HBV effect is good, better than medication alone is worthy of further research and application.
2.Anticipatory nausea and vomiting in cancer patients with chemotherapy: A review

Anticipatory nausea and vomiting (ANV) is one of the common adverse effects in cancer patients undergoing chemotherapy.ANV is related to classical conditioning,psychosocial and demographic factors.Psychological intervention such as systematic desensitization,hypnosis and biofeedback therapy could be used to treat ANV,while some psychotropic medication including benzodiazepines and olanzapine is also helpful to prevent ANV.Now the recognition about ANV in clinical oncology is not adequate and large sample epidemiological survey of ANV is lacked.So far there are no special tools to predict and assess ANV,and the effects of pharmacological interventions also need solid evidence from large sample double-blind randomized controlled trials.
3.Effects of 1 0-week group psychotherapy on mood and quality of life in patients with early and mid-term colorectal cancer

Objective:To determine if a group psychotherapy programme offers significant benefits to psycho-logical wellbeing for early and mid-term colorectal cancer patients compared to control group.Methods:Totally 67 patients previously diagnosed with colorectal cancer who participated in this study,34 patients accepted an invitation to join the intervention group,received 10 consecutive,weekly sessions group psychotherapy,and 33 patients served as control subject with usual care only.All participants were assessed at baseline and post-therapy (week 10 follow-ing enrollment or equivalent ).Assessment comprised self-completed Cancer Quality-of-life Core Questionnaire (QLQ-C30),Self-Rating Depression Scale (SDS)and Self-Rating Anxiety Scale (SAS).Results:At baseline, 67participants completed questionnaires,and 60 questionnaires returned at post-therapy,from 30 intervention group participants and 30 control group participants.Baseline scores for SAS and SDS were higher in intervention than control group[(42.0 ±7.7)vs.(36.0 ±6.8),(44.4 ±11.5)vs. (38.7 ±9.0),Ps <0.05],while other itemswere not differ(Ps>0.05 ).Post-therapy,intervention group participants reported significantly scores for anxiety, depression and fatigue compared with controlled group.In the comparison of before-post difference between inter-vention and controlled group,the difference of SAS score,SDS score,role function score and fatigue score showed significant discrepancy,intervention group improve significantly (Ps <0.05 ).Conclusion:Group psychotherapy could improve anxiety and depression,for colorectal cancer patients,and may help to improve quality of life,possi-bly through improving role function and decreasing fatigue symptoms.
4.Study on the Expression of HER2 and TOPOⅡ? in Human Breast Cancer and Relationship between them

Objective To detect the expression of HER2 and TOPOⅡ? in human breast cancer to explore the relationship between them and the potential value in the chemotherapy of breast cancer. Method 24 breast cancer tissues and paired normal tissues were investigated. RT-PCR and imumohistochemical staining were used to detect the expressions of HER2 and TOPOⅡ? at the transcription and protein level respectively. Results HER2 mRNA expressed positively in 29% (7/24) breast cancer tissues vs 4% (1/24) in paired normal tissues. The positive expression of TOPOⅡ? mRNA in breast cancer tissues and paired normal tissues were 92% (22/24) and 21%(5/24) respectively. The result of the immunohistochemical staining showed that the positive rates of HER2 and TOPOⅡ? in breast cancer tissues were 30%(6/20) and 94%(16/17) respectively, which was consistent with RT-PCR. 5.Extraction Technology of Volatile Oil from Qingjie Granules and Optimization of Inclusion Technology by Box-Behnken

Objective To optimize the extraction of volatile oil from Qingjie Granules and process of inclusion compound of beta-cyclodextrin. Methods Water Distillation was used for extraction. Extraction time, grinding degree, and the amount of water were set as inspection factors, and volatile oil volume was set as the evaluation index to inspect extraction process of volatile oil from Qingjie Granules. With inclusion rate as the evaluation index, the single factor test and the Box-Behnken combined with the response surface method were used to choose the optimum inclusion process. Results The optimum extraction process for Schizonepetae Herba, Saposhnikoviae Radix and Forsythiae Fructus coarse powder should be with 10 times amount of water, extracting 3 h. Inclusion method should be saturated water solution method, and the inclusion process of volatile oil as feed and beta-cyclodextrin inclusion ratio was 1:12; the temperature was 40 ℃; inclusion time was 3.5 h. By means of TLC, UV and IR spectra, the formation of the inclusion compound of volatile oil in clear solution particles was preliminarily proved. Conclusion The optimum extraction and inclusion process of volatile oil from Qingjie Granules are stable and feasible, which can be used in industrial production.

BACKGROUND:The differentiation of neural stem cells (NSCs) is a hot research. Whether brain-derived neurotrophic factors (BDNF) can induce the differentiation of NSCs into neurons has not been detailed studied. OBJECTIVE:To investigate the effect of different concentrations of BDNF on the differentiation of NSCs from adult rat hippocampus into neurons in the medium with epidermal growth factor. DESIGN:A controlled study. SETTING:Department of Human Anatomy, China Medical University. MATERIALS:The experiment was performed at the Neurotomia Laboratory of China Medical University in August 2007. NSCs isolated from hippocampus of adult SD rats were inoculated in a serum-free medium. Three clean adult SD rats (200-250 g each) were provided by Laboratory Animal Department of China Medical University. Dispositions to the rats were consistent with ethical standards of animals. Epidermal growth factor (EGF) and BDNF were bought from R&D Company. METHODS:①NSCs obtained from rat hippocampus were cultured in the serum-free medium containing basic fibroblast growth factor (bFGF), EGF and B27. ②Single cell clone was obtained via limited dilution for the fourth passage of cells. Identification of NSCs for the passage of cells from monoclonal spheres was performed by Nestin immunocytochemistry. Neurons and astroglial cells were identified by immunocytochemistry for neuron specific enolase (NSE) and glial fibrillary acidic protein (GFAP) a week after differentiation. ③Differentiation for the monoclonal spheres was done to appraisal the proportion of neurons. NSCs were divided for several groups containing 0 μg/L, 50 μg/L, 100 μg/L, 150 μg/L, and 200 μg/L groups according to the doses of BDNF. EGF was added in the media of each group. Immunocytochemistry for NSE was done a week later to count the positive cells. MAIN OUTCOME MEASURES:Proportion of neurons differentiated from NSCs.RESULTS:①NSCs immunocytochemical staining showed that Nestin was positive in monoclonal spheres, and NSE and GFAP were positive in differentiated cells. ②The proportion of differentiation from NSCs into neurons was significantly higher in groups treated with 50 μg/L and 100 μg/L BDNF compared to other groups(t=2.502-5.025, P < 0.05), but there was no significant difference between 50 μg/L and 100 μg/L BDNF groups(P > 0.05). Moreover, no significant difference was detected among 0 μg/L, 150 μg/L and 200 μg/L BDNF groups (P > 0.05). CONCLUSION:The optimal concentration of BDNF is 50 μg/L for differentiation from NSCs into neurons while the concentration of EGF is 20 μg/L.

ObjectiveTo summarize the nursing measures for patients with nasal facial soft tissue defect treated with transplantation of free foot skin flap.MethodThe clinical data of 13 patients with transplantation were reviewed to summarize the nursing measures.ResultAll skin flaps of the 13 patients survived.ConclusionEnough preoperative preparation,active psychological nursing and careful postoperative observation are critical for the success of the operation.

10.Protective effect of pituitary adenylate cyclase activating polypeptide on focal cerebral ischemia reperfusion injury in rats

Aim To investigate the protective effect of pituitary adenylate cyclase activating polypeptide(PACAP)on focal cerebral ischemia reperfusion injury in rats.Methods The models of focal cerebral ischemia reperfusion in rats were established with suture-occluded method to simulate the middle cerebral artery ischemia reperfusion injury in clinic.The treatment groups were given three different dose PACAP(PACAP 10-11 mol,PACAP 10-10 mol and PACAP 10-9 mol) in 15 minutes before cerebral ischemia.PACAPs were infused into lateral cerebral ventricle by microinjector.The water content of brain,the activity of superoxide dismutase(SOD),the content of malondialdehyde(MDA) and the content of nitrogen monoxidum(NO) of the brain tissue were determined.Results Compared with NS group,the water content of brain and the concentration of MDA and NO were obviously lower in each PACAP treatment group,while the activity of SOD was elevated to different degrees.Conclusion PACAP has an obvious protective effect on ischemia-reperfusion injury of focal brain tissue in rats,in which edema-alleviating,free radicals-scavenging and lipid peroxidation-attenuating could be involved.The protective effect seems to be more clear in middle-and high-dose group than in low-dose group.
