1.Pharmacokinetics of probucol self-microemulsifying drug delivery system in rats

Objective:To investigate the pharmacokinetics of probucol(PB)self-microemulsifying drug delivery system(PB-SMEDDS) in rats.Methods:Rats were used as the models.With PB suspension as control,double cycle crossover with single oral dose was performed to investigate the pharmacokinetics of PB-SMEDDS.The HPLC method was established to detect drug concentration in the rat plasma,and the obtained data were processed with 3P97 software.Results:The plasma concentration-time profiles of PB-SMEDDS conformed to a two-compartment model(the coefficient weight is 1/C2).Cmax,tmax and AUC0~96h were 6.72?g/ml,8.58 h and 253.98(?g.h)/ml,respectively.The AUC0~96h of PB-SMEDDS was 19.08 times of PB suspension.Conclusion:PB-SMEDDS could improve the oral bioavailability of PB significantly.
2.Design, synthesis, and PPARgamma agonistic activity of novel indenone derivatives.

Agonists of peroxisome proliferator-activated receptor gamma (PPARgamma) are of interest as a treatment of type II diabetes, and indenone derivatives are a new class of non-TZD PPARgamma agonists. Based on existing indenone derivatives, a series of novel ones have been designed and synthesized. Meanwhile the structures have been comfirmed with 1H NMR and MS. Among them, 17b and 19 showed higher agonistic activities than rosiglitazone.
3.Effect of Ga-Al-As laser irradiation on CD4 + CD25 + regulatory T cells and local inflammatory media in rheumatoid arthritis

Objective To explore the mechanism of combined treatment with methotrexate (MTX) and Ga-Al-As laser irradiation for rheumatoid arthritis (RA) and to assess the effectiveness of Ga-Al-As laser therapy for RA. Methods Twenty-two patients with RA were randomly and evenly divided into two groups: the treatment group treated with Ga-Al-As laser irradiation combined with MTX and the control group treated with MTX only. Ten age-matched normal subjects were observed as normal controls. The amount of CD4 + CD25 + regulatory T cells in peripheral blood (PB) of the normal controls and that in the PB and synovial fluid (SF) of the 22 patients before and after therapy were counted by flow cytometry. Meanwhile, the amount of prostaglandin E2 (PGE2) in synovial fluid of the patients was measured before and after treatment by enzyme-linked immunosorbent assay(ELISA). Results After combined treatment the clinical symptoms of the patients were improved significantly, and the amount of PGE2in SF decreased significantly. The count of CD4 + CD25 + regulatory T cells in PB of RA patients was ( 3.84 ±3.20) % , compared to ( 10.05 ± 7.04) % in healthy individuals. The count of CD4 + CD25 + regulatory T cells in SF of RA patients was ( 14.89 ± 12.30) % , much higher than that in PB. The count of CD4 + CD25 + regulatory T cells in SF decreased significantly in treatment group compared to control group (P <0.05). Conclusion Ga-Al-As laser irradiation eombined with MTX can effectively improve the clinical symptoms of RA patients. It may be related to the decrease of amount of PGE2 and count of CD4 + CD25 + regulatory T cell in PB and SF.
5.Effect of moxibustion on the Governor Vessel in combination with acupuncture on ankylosing spondylitis

ObjectiveTo investigate the effect of moxibustion on the Governor Vessel in combination with acupuncture on ankylosing spondylitis.MethodsThirtypatetints with ankylosing spondylitis(kidneyyang deficiency pattern) were recruited and treated with moxibustion on the Governor Vessel in combination with acupuncture. The scores of Scale of Quality of Life for Ankylosing Spondylitis (SQOL-AS), Bath Ankylosing Spondylitis Disease Activity Index(BASDAI), Bath Ankylosing Spondylitis Functional Index(BASFI), Fatigue Assessment Instrument(FAI)and erythrocyte sedimentation rate(ESR) before and after treatment were evaluated.ResultsThe scores of BASDAI(0.427±0.381vs. 6.107±1.375;t=28.986,P<0.01), BASFI(0.753±0.504 vs. 7.480±1.152;t=36.580,P<0.01), FAI(40.067±14.746vs. 146.967±26.283;t=24.520,P<0.01)after treatment were significantly lower than those before treatment; ESR after treatment were significantly lower than that before treatment(19.833±13.929 mm/hvs. 31.567± 20.065 mm/h;t=3.708,P<0.01). The subscores of Physical Functioning(33.000±1.930vs. 12.033±1.790;t=62.165,P<0.01), Mental Health(25.367±2.059vs. 14.067±2.303;t=30.139,P<0.01), Social Functioning(21.267±4.025vs. 8.767±1.977;t=26.643,P<0.01) and General Health Perception (22.733± 2.690vs. 9.767±2.738;t=25.729,P<0.01) of SQOL-AS after treatment were significantly lower than that before treatment.ConclusionsMoxibustion on the Governor Vessel in combination with acupuncture can improve physical functioning and reduce disease activity in patients with ankylosing spondylitis.
6.Qualitative research on the impact of administrative penalties on medical staff in medical disputes

Objective To describe the impact of administrative penalties for medical disputes on the medical staff, including their perception on the penalty and its impact on their mind and body, professional behavior, career development, etc, and to provide a theoretical basis for health policy making and hospital management. Methods The phenomenological research method was adopted to conduct in-depth interview to 10 hospital staff in Taian who were involved in medical disputes and got punished, and the Giorgi qualitative data analysis was used in the research. Results The interviewees′perception and impacts of the punishment were concluded into five aspects:physiologically, psychologically, cognitively, behaviorally and professional development. Conclusions It is concluded that the current punishment to individuals has caused quite negative effects, and a new mechanism should be established to systematically reconstruct the procedure and reduce the medical disputes to the minimum.
7.Establishment of a three-dimensional digital model based on multi-source craniofacial profile

Objective To establish a three-dimensional digitized visible model to meet the clinical and research needs.Methods The head of the volunteer was scanned by a cone beam CT (CBCT).The bone tissue,the facial soft tissue and the teeth were extracted from the cone beam CT raw data according to the different Houns field values.The 3D digital craniofacial model was reconstructed from the three tissues extracted from the CBCT original data.The 3D dental model with high resolution was acquired by scanning the dental cast with the laser scanner.The 3D facial model with texture was acquired by scanning the volunteer's face directly by other laser scanner.With the regis tration algorithm in 3D graphics,the 3D facial model with texture and the 3D dental model with high resolution were transformed to the same coordinate which was of the 3D digital craniofacial model.Results We acquired a complicated 3D craniofacial digital model which combined the teeth model with high resolution,the bone model and the face model with texture.Conclusions The method is able to accurately establish the craniofacial digital model.
