1.Progress in research on fluorescence resonance energy transfer technology based on upconversion nanomaterials for biological detection
Chinese Journal of Biologicals 2023;36(8):996-1001
Upconversion nanoparticles(UCNPs)doped with rare earth elements have advantages in biose-nsing because of their good fluorescence stability, biocompatibility and avoidance of background fluorescence. Therefore, fluorescence resonance energy transfer(FRET)system based on upconversion particles(UCNPs based FRET)has been widely used in biological detection. This paper reviews the application and prospect of UCNPs based FRET in biological detection of biotoxins, hormones, proteins, nucleic acids, bacteria, and so on.

3.Static cold storageversushypothermic machine perfusion:changes in inflammatory cytokines and apoptotic factors in isolated liver before and after liver transplantation
Chinese Journal of Tissue Engineering Research 2015;(33):5322-5326
BACKGROUND:Static cold storage is the main storage method of liver transplantation in the past 40 years. There are success cases in foreign countries about the use of mechanical reperfusion in liver transplantation, but there is no large-scale development in China. OBJECTIVE:To investigate and compare the inflammatory cytokines and apoptotic factors in isolated liver between static cold storage and hypothermic machine perfusion before and after liver transplantation. METHODS:There were donor group, static cold storage group and hypothermic machine perfusion group in the study, and 30 dogs were in each group. Liver specimens from donor dogs were subjected to static cold storage and then transplanted into dogs in the static cold storage group. Liver specimens from dogs in the static cold storage group were subjected to hypothermic machine perfusion and then transplanted into dogs in the hypothermic machine perfusion group. Liver specimens from dogs in the hypothermic machine perfusion were used in other experiments. Dogs in the donor group were euthanatized. After liver transplantation, 20 dogs from static cold storage group and hypothermic machine perfusion group were selected respectively for further study. RESULTS AND CONCLUSION: When the liver was separated from the dogs, inflammatory cytokines and apoptosis factors had no significant difference in the static cold storage group and hypothermic machine perfusion group (P > 0.05). P53 expression in these two groups had no significant difference before and after transplantation (P > 0.05). Interleukin-1βand interleukin-8 in these two groups had no significant difference before transplantation (P > 0.05). Compared with the static cold storage group, expressions of tumor necrosis factor-α, macrophage inflammatory protein-1α, macrophage inflammatory protein-1β, CC chemokine ligand 20 and FASR were significantly lower in the hypothermic machine perfusion group before transplantation (P< 0.01). Compared with the static cold storage, interleukin-1β, tumor necrosis factor-α, interleukin-8, macrophage inflammatory protein-1α, macrophage inflammatory protein-1β, CC chemokine ligand 20, FASR and nuclear factor-κB were reduced significantly in the hypothermic machine perfusion group at 60 minutes after transplantation (P < 0.01). Taken together, hypothermic machine perfusion can significantly reduce inflammatory cytokines and apoptosis factor, which is conducive for liver transplantation.
4.Effect of atorvastatin on myocardial fibrosis induced by aldosterone
Chinese Journal of Pathophysiology 2010;26(2):227-232
AIM: To investigate the effects and mechanisms of atorvastatin on myocardial fibrosis induced by aldosterone in SD rats. METHODS: Forty male uninephrectomized SD rats were limited to drink 1% NaCl water for 4 weeks and assigned to the follow groups: vehicle control rats (CON group);aldosterone treated rats (ALD group);spironolactone + aldosterone treated rats(SPI+ALD group);atorvastatin + aldosterone treated rats (ATO+ALD group). Blood pressure was measured by catheterization. Expression of platelet-derived growth factor (PDGF-A, PDGF-B), platelet-derived growth factor receptor (PDGFR-α, PDGFR-β) and ectodermal dysplasia-1 (ED-1) were analyzed by immunohistochemistry. Collagen volume fraction (CVF) and perivascular collagen area (PVCA) were analyzed by Sirius-Red staining. Myocardium osteopontin protein was detected by Western blotting analysis. RESULTS: Mean arterial blood pressure in ALD group, SPI+ALD group and ATO+ALD group was elevated, and significant difference was observed between the three groups and vehicle control group (P<0.01 or P<0.05). Myocardial fibrosis was observed in ALD group. Compared to other three groups, the index of CVF and PVCA was increased significantly (P<0.01 or P<0.05). No significant difference of the index of CVF and PVCA between ATO+ALD group and SPI+ALD group was observed (P>0.05). Compared to other groups, the levels of PDGF-A, PDGF-B, PDGFR-α, ED-1 and OPN in ALD group were significantly increased (P<0.01 or P<0.05). The levels of PDGF-A, PDGF-B, PDGFR-α and OPN were no significant difference between ATO+ALD group and SPI+ALD group (P<0.01 or P<0.05). However, the level of ED-1 in ATO+ALD group was significantly decreased compared to SPI+ALD group (P<0.05). No significant difference of PDGFR-β level among four groups was found (P>0.05). CONCLUSION: These results suggest that atorvastatin may attenuate myocardial fibrosis induced by aldosterone. The mechanisms concern with reduction of macrophage infiltration, expression of inflammatory cytokines OPN, partially inhibition of platelet-derived growth factor and its receptor expression.

7.Judging professional management from the differences between Chinese and Australian hospital managers
Chinese Journal of Hospital Administration 2001;0(08):-
Through a comparison of the differences between China and Australia in the models of hospital management and the roles of hospital managers, the paper expounds the specific features of the model of “decentralized powers” and “professional presidents” and the model of “centralized powers” and “expert-turned presidents”. It argues that under the pressures of both internal and external changes, including rapid socioeconomic development, the deepening of the health care reform, the effect of China's accession to the WTO on the medical and health industry, the development of a contingent of professional health care managers in China has become an inevitable trend.

9.Application of epirubicin in combination with anti-CD47 antibody for therapy of breast cancer in vitro
Chinese Journal of Immunology 2014;(10):1360-1363
Objective:To explore the effects of combination of epirubicin (EPI) and anti-CD47 antibody on therapy of breast cancer.Calreticulin ( CRT) and CD47 on DC-mediated phagocytosis. Methods: Breast cancer cells MCF-7 and MDA-MB-231 were treated with EPI , and expression of calreticulin ( CRT ) and CD47 on cell surface were detected by flow cytometry or immunofluorescence.DC were isolated from human peripheral blood , and cocultured with epirubicin-treated breast cancer cells in the present of anti-CD47 or anti-CRT antibody ,and the phagocytosis of DC was assessed. Results:Untreated breast cancer cells MCF-7 and MDA-MB-231 did not express CRT on the surface of cells ,but high level of CD47 was detected. After EPI treatment,CRT exposed on the cells surface, and the expression of CD47 was reduced. The results showed that EPI or anti-47 antibody alone increased DC-mediated phagocytosis ,and combination treatment marked enhanced this effects.Further more, in addition of anti-CRT antibody reduced the phagocytosis of DC.Conclusion:EPI could induce CRT exposure on the tumor cell surface ,and enhanced DC-mediated phagocytosis when combination with anti-CD47 antibody. The phagocytosis of DC on tumor cells were modulated by both ecto-CRT and CD47.This study provide a new effective strategy for the use of anti-CD47 antibody among the chemotherapy of breast cancer.
10.Regulatory T cells and hepatocellular carcinoma
International Journal of Surgery 2011;38(9):629-632
Regulatory T cells (Tregs) are a subpopulation of CD4 +T cells highly expressing CD25 and Foxp3. Treg not only involves in autoimmune disease, infection and transplantation tolerance, but also plays a pivotal role in the suppression of anti-tumor immunity during tumor development. Current researches suggest the frequency of Treg is increased in tumor tissues and peripheral blood of patients with HCC, which is associated with HCC development, and affect survival rate and prognosis of HCC patients. Depletion of Treg together with surgical resection of the tumor could be a new approach for HCC, which can enhance tumorspecific T cell memory to remove latent metastasis and protect against recurrence for improvement of HCC therapeutic effect. This review presents the role of Treg in HCC development, the relationship between Treg and prognosis and its clinical practice.
