BACKGROUND: There are pressure sensors in carotid-sinus, which are very sensitive to blood pressure regulated by ions and play an important role in the regulation of blood pressure. But it is yet not very clear how different ions regulate the blood pressure through pressure sensors in carotidsinus.OBJECTIVE: To observe the effect of different ions at various concentrations outside the carotid-sinus.DESIGN: Self-control experiment.SETTING: Preclinical Experiment Center, Fourth Military Medical University of Chinese PLA.MATERIALS: The experiment was accomplished in the Preclinical Experiment Center, Fourth Military Medical University of Chinese PLA from December 2000 to June 2001. Totally 18 New Zealand pure strain rabbits were provided by the Aninal Experimenting Center of Fourth Military Medical University of Chinese PLA. They were standard grade Ⅱ, of either gender and body mass was (2.0±0.2) kg.METHODS: The rabbits were divided into Na+, K+ and Ca2+ groups according to random numbers, and each group consisted of 6 rabbits. After anaesthesia, tracheal intubatton was performed on the rabbit, and bilateral carotid arteries were separated with carotid-sinus separated on one side and vessel intubatton performed in the other side for blood pressure measurement. Then various concentrations of Na+, K+ and Ca2+ ions were added outside the carotid-sinus with the pipette to make the carotid-sinus completed immersed in the ion solutions. The basal blood pressure and the peak value after ions addition were recorded respectively.MAIN OUTCOME MEASURES: The basal blood pressure and the peak value after ions addition.RESULTS: After Na+ (0.15, 1.5 mol/L) was added the blood pressure was(97±12), (83±17) mm Hg. It was decreased significantly compared with the basal value (106±14), (105±12) mm Hg (t=2.946, P < 0.05). K+ (0.4 mol/L)decreased the blood pressure significantly [(106±12), (64±13) mm Hg, (t=13.496, P < 0.01)], but other concentrations of K+ were not effective. Ca2+(0.07 mol/L) increased the blood pressure to (113±16) mm Hg compared with the basal value (103±12) mm Hg (t=-3.627, P < 0.01).CONCLUSION: Na+, K+ and Ca2+ regulate the blood pressure by acting on the carotid-sinus directly. High concentrations of Na+ and K+ possess the effect of decreasing the blood pressure, while high concentrations of Ca2+increases it, which may be an important mechanism of blood pressure regulation.

Objective: To explore the influence factors of the degenerate oligonucleotide primered PCR(DOP-PCR). Methods: Genome DNA template from the mouse single oocyte or liver tissue were used to perform DOP-PCR. DOP-PCR was carried out with templates of different origin, different gradient dilution, with or without low melting point gel purified to wipe off the small fragment that might interfere with the following analysis, and then PCR of gene FTCD and CBS were carried out to evaluate the influence of these factors on the amplification efficiency and specificity. Results: Compared with genome DNA template from mouse liver, the template from single oocyte had the same efficiency and specificity but a minor yield and different gradient dilution of DNA template had no effect on the efficiency and specificity. Furthermore, there was a higher specificity in the low melting point gel-purified DOP-PCR product than in untreated ones. Conclusion: We have got a satisfactory result and increased specificity from DOP-PCR product purified with the low melting point gel. Single oocyte of mice could be used for further investigation of special genes detection by DOP-PCR and of an optimization in the yield of the products.

The mathematical models for simulation of cardiac sodium, potassium and calcium channel kinetics courses and currents were developed to simulate the properties of ionic currents and channel dynamic courses. With modifications of these models, it is possible to make them integrated for simulating the whole process of action potential, thus additional discussion on ionic mechanism could provide a theoretical foundation for further animal experiments and clinical applications.

Purpose The mean pulmonary artery (MPAP) has been widely used as an important parameter to diagnose and evaluate pulmonary hypertension (PH).The purpose of this paper is to compare the efficacy of two methods in evaluating PH,including estimating pulmonary artery systolic pressure (PASP) using Doppler ultrasonography to measure tricuspid regurgitation (TR) velocity, and directly using the peak velocity of TR.Materiasl and Methods From January 2012 to June 2013,eighty patients with left-to-right shunt congenital heart diseases (CHD) planned for closure procedure in Tangdu Hospital of the Fourth Military Medical University and the General Hospital of Shenyang Military region were included in this prospective study, who underwent right heart catheterization to measure pulmonary artery pressure, and underwent Doppler ultrasonography to measure the peak velocity ofTR.Results Using catheter-measured MPAP of≥ 25 mmHg as diagnostic reference, the false positive rate was 62.96%,and the false negative rate 0% when the estimated PASP of >30 mmHg determined by TR method was used to diagnose PH.There was high diagnostic agreement when peak velocity of TR was used to diagnose PH.When 320 cm/s and 340 crn/s were used as diagnostic cutoff values, false positive rates were 14.81% and 7.41%,and false negative rates were 15.91% and 20.45%,respectively.Conclusion In patients with left-to-right shunt CHD,peak velocity of TR measured on echocardiography can be used to diagnose PH which overcomes the high false positive rate in estimation method. It is more suitable to diagnose PH when the MPAP is used as the diagnostic criterion.

Objective To study the genotypic and biological characteristics of catheter-related MRSE isolates and to further provide information for the diagnosis and prevention of catheter-related bloodstream infection. Methods Thirty strains of catheter-related MRSE isolates were collected from venosus blood and whole blood of 30 inpatients including 20 males and 10 females from Beijing Tongren Hospital, Capital Medical University from January 2006 to December 2009. The genetic features of these strains were determined by MLST. PCR was used to detect the icoA gene (encoding the polysaccharide intercellular adhesion associated with pathogenicity), and the antimicrobial susceptibility test was detected by disc diffusion test. Results A total of 15 STs were obtained from 30 strains ST259, ST20, ST2 and ST235 were common clones obtained from 17 strains. Four novel STs were found and uploaded to the MLST database (http://www. mlst. net), including ST259 (6 strains), ST260 (1 strain), ST261 (1 strain) and ST262 (1 strain). The ST259 was the dominant clone of catheter-related MRSE isolates in this hospital, and 3 strains carrying icaA gene were detected in this study. Conclusion Some ST259 isolates express high pathogenesis among the four novel STs, which may make them as the pandemic strains in nosocomial infection, and this would increase the difficulty of the prevention and control of nosocomial infection.

Objective: To clone a novel gene and explore its expression patterns in tissues and cells,so as to find its role in the process of encephalopathy in DS.Methods: On the base of our previous microarray's result together with the tissue type,we chose EST AI480014 to carry out RACE,then analyzed its expression profiles in liver, spleen, kidney, heart, brain by multi-tissues Northern blot, after that semi-quantitive RT-PCR was used to reexamine the expression profiles.Furthermore,we used ISH to find whether aim gene expressed in neuroglial cells cultured in vitro. Finally we performed semi-quantitive RT-PCR to explore whether it expressed differently between DS and normal. Results: We gained a 682 bp new cDNA fragment(DQ275636)which expressed in all the tissues examined and had no alternative splices in them.It expressed highly in brain especially in frontal lobe and hippocampus.According to the ISH result we convinced that it expressed in neuroglial cells.Using bioinformatics we mapped DQ275636 to chromosome 5q14.Conclusion: We have obtained a new gene fragment based on the(above) results.According to its expression character and tissue type,it can be suggested that this gene has a probable role in the process of encephalopathy in DS.
