Objective To observe the changes of serumγ?glutamyl transpeptidase(GGT)levels before and after the patho?gen treatment in patients with subclinical schistosomiasis,and explore its clinical value in the diagnosis and treatment of sub?clinical schistosomiasis. Methods Totally 109 patients with subclinical schistosomiasis,who were found in the endemic inves?tigation of schistosomiasis in Ezhou City,were selected as the investigation subjects,and then they were treated with praziquan?tel. The serum GGT levels of the subjects before and after the treatment were detected and compared. Results Before the treat?ment,the average value of the GGT levels of the 109 patients was(48.1 ± 45.9)IU/L,among which,the GGT levels of 69 cases (63.3%)were normal,and the levels of 40 cases(36.7%)were increased. After the treatment,the average GGT level of the pa?tients was(32.1 ± 23.4)IU/L,which decreased by 33.3%comparing with that before the treatment,and the difference had a statistical significance(U=2.17,P=0.01). The GGT levels of 65 patients decreased in different degrees. Among the 40 pa?tients whose GGT levels had increased before the treatment,the GGT levels of 31 ones returned to the normal. Conclusion The GGT level detection can accurately reflect the liver function in the patients with subclinical schistosomiasis ,and also it has certain clinical application value to judge the liver function damage and recovery of the patients before and after the pathogen treatment.

Objective To investigate the requirements of hospitalized patients on humanized nursing in the ward of whole-process humanistic care mode and provide scientific references for continuous improvement of nursing quality.Methods A total of 408 patients from 2 wards of whole-process humanistic care mode.Patients were applied the self-designed questionnaire in combination with interview to understand the patients’ requirements of humanized nursing.Survey was carried out before the inpatients were discharged from the hospital adopting the “Evaluation Table of Patients’ Satisfaction Degree about the Nursing Service” produced and researched by our hospital independently.Results After carrying out the whole-process humanistic care service mode,the inpatients’ overall satisfaction degree and satisfaction rate with humanistic care were respectively 98.2％,97.5％.Conclusions Carrying out the quality care demonstration project and enhancing the concept of humanistic care can promote each other.Carrying out the inpatients’ care needs survey in the ward of whole-process humanistic care mode,integrating humanistic into every aspect of clinical nursing,are the crux of deepening high-quality nursing care as well as a new subject that clinical nurses should confront conscientiously.

Objective To establish an early,simple and rapid colloidal gold strip method for the detection of IgM against hantavirus nucleocapsid protein in patients with hemorrhagic fever with renal syndrome(HFRS).Methods Purified recombinant Hantavirus nucleoprotein(rNP)was labeled by colloidal gold particles and then sprayed and fixed on fiberglass membrane as the combination pad.Anti-Human IgM(μ-chain specific)antibody produced in goat was fixed in the detection area,and mouse antihantavirus antibody was fixed in the quality control area.Both of them were on nitrocellous membrane strip in tandem.Together with a specimen pad ahead.The conbination pad and the nitrocellous membrane were assembled into a test strip.The colloidal gold strip assay was compared with ELISA for evaluation of specificity and sensitivity.Results The colloidal gold strip tests showed positive in the serum samples from 50 cases of HFRS which was clinically diagnosed and then verified by ELISA within 10-15 minutes.Whereas 30 serum samples of healthy donors have tested negative.Conclusions Our new colloidal gold immunochromatographic test strip method was well concordant with the ELISA assay,but the former was more raoid and simole.It could be used primary medical services.

Objective:To explore the regulatory role of exosomes in calcification of rat vascular smooth muscle cells (VSMC) induced by high phosphorus.Methods:VSMC (A7r5 cells) were cultured in vitro and randomly divided into three groups: normal phosphorus group (0.9 mmol/L), high phosphorus group (2.6 mmol/L) and high phosphorus exosomes induction group (i.e. the exosomes extracted from high phosphorus group were added to VSMC in normal culture). Until the 7th day of culture, the culture medium of normal phosphorus group and high phosphorus group obtained during the change of cell culture medium was collected, and the precipitate was obtained by ultracentrifugation and suspended by phosphate buffered saline. The protein content of the precipitate was determined by BCA protein quantitative method. The precipitates were identified. The structure and size of exosomes were observed by transmission electron microscope. The exosomes marker proteins tumor susceptibility gene 101 protein (TSG101) and CD9 were detected by Western blotting. The miRNA in exosomes was extracted, and the expression of related miRNA (miR-30b, miR-204, miR-211) were observed by real-time quantitative PCR. After 7 days of cultivation, the exosomes uptake process of VSMC in high phosphorus exosomes induction group was observed. The calcium deposition was detected by Alizarin stain, and the calcium content was detected by O-cresol complex copper. The content of alkaline phosphatase was detected by colorimetry. The protein expression of Runx2 was quantified by Western blotting. Results:(1) The precipitate obtained by ultracentrifugation of the cell culture fluid was identified as exosomes by electron microscopy morphology. Western blotting confirmed that the expression of the exosomal marker proteins TSG101 and CD9 were positive. (2) The exosomes were rich in miRNAs. The expression of miR-30b, miR-204, miR-211, which negatively regulated the transcription of Runx2, was significantly down-regulated in the high-phosphorus group compared with the normal group ( P<0.05). (3) After culturing rat VSMC with high phosphorus for 7 days, calcium salt deposition was obvious. Compared with the normal phosphorus group, calcium content and alkaline phosphatase activity were significantly increased (both P<0.05), and Runx2 expression was also significantly increased ( P<0.05). (4) Added the obtained high-phosphorus exosomes to the normal cultured VSMC, the exosomes could be taken up by VSMC and successfully induced VSMC calcification. The levels of cell calcification indicators and Runx2 expression were significantly increased. Conclusions:High phosphorus induces calcification of VSMC and promotes the increase of Runx2 expression. The mechanism may be realized by releasing exosomes from VSMC to transmit cell signals.

Objective To investigate the preventive and therapeutic effects of Tiaozhi Baogan Capsule (TBC) on fatty liver in rats and to explore its therapeutic mechanism. Methods SD rat models of fatty liver were set up by feeding high fat diet. After the treatment of TBC, the general physical state, liver index (the ratio of liver weight with body weight) , hepatic pathological changes , blood biochemical indices were observed in model rats. AIM: To study the effect and mechanism of Baogan Xiaoshi Capsule(BXC) (Rhizoma Coptidis,Fructus Evodiae,Rhizoma Alpiniae Officinarum,Rhizoma Cyperi ,Rhizowa Atractylodis Macro cephalae,Fructus Aurantii Immaturus,Fel Bovis) preventing lithogensis of pigment gallstones in guinea pigs. METHODS: Feeding guinea pigs on lithogenous diet,BXC was taken for 3 months at the same time.In the end,the gallstone formation rate,biochemical indicators and liver morphology were observed. RESULTS: The gallstone formation rate was 76.92% in model group,and in BXC group,at middle or upper dose,it was 25%,20% respectively.BXC could increase the concentration of conjugated dilirubin and glycocholic acid,and decrease that of unconjugated dilirubin,Ca~(2+) and H~+.It could weaken the activity of endogenous ?-Glucuronidase and Ca~(2+)Mg~(2+)-ATPase,and also could strengthen that of Na~(+)K~(+)-ATPase.It could improve the fatty degeneration of liver. CONCLUSION: The results show that BXC could prevent pigment gallstone formation and the fatty degeneration of liver.The mechanism may be that BXC can weaken the activity of ?-Glucuronidase and decrease [Ca~(2+)].

OBJECTIVE:To study the litholytic,bacteriostatic and spasmolytic effects of Baoganxiaoshi capsule(BXC) in vitro.METHODS:Observed the effects of BXC on human bile pigmental and cholesterol gallstone, bacteria, duodenum smooth muscle of rabbits in vitro.RESULTS:The litholytic concentration of 7.568,15.135 and 30.270g(crude drug)/ml solutions of BXC on human bile pigmental gallstone were 5.4mg,8.3 mg and 14.7 mg respectively,and on human bile cholesterol gallstone were 2.9mg,5.1 mg and 7.1 mg respectively.So BXC have the litholytic function of bile pigmental and cholesterol gallstone.It can strongly inhibit or sterilize staphylococcus aureus, enterococcus, diphtherial bacillus, hemo1ytic streptococcus, and the MICs were 23.65mg(crude drug)/ml ,23.65mg(crude drug)/ml,23.65mg(crude drug)/ml and 47.297mg(crude drug)/ml respectively, and all of the MBCs were 47.297mg(crude drug)/ml.It can inhibit or sterilize coliform,salmonella and pneumococcus, and all of the MICs were 378.375mg(crude drug)/ml, and all of the MBCs were 756.75mg(crude drug)/ml.5.3 414mg(crude drug)/ml solutions of BXC could obviously inhibit the average tension amplified,the frequency accelerated,and the activity increaced,on the duodenum smooth muscle induced by ach(P
