Objective To investigate the protective effects of Xuebijing(XBJ)on the injury of vascular endothelial cells(VEC)induced by lipopolysaccharide (LPS),and to study the mechanisms of the production of nitric oxide (NO)and the expressions of inducible nitric oxide sytnhase (iNOS)and signal transduction under XBJ intervention condition.Methods The cultured VEC were divided into control group, LPS (1 mg · L-1 )group, LPS (1 mg·L-1)+XBJ(25 g·L-1)group,LPS(1 mg·L-1)+pyrrolidine dithiocarbamate(PDTC,20μmol·L-1) group;XBJ and PDTC were administrated 1 h before incubation of with LPS.Western blotting method was used to detect the expressions of iNOS and NF-κB p65 protein.The level of NO in the supernatant was measured by Griess reagent.Results Comparaed with control group,the NO level and the expression levels of iNOS protein and NF-κB p65 protein in VEC in LPS group were significantly increased(P<0.01).Compared with LPS group,the NO level and the expression levels of iNOS protein and NF-κB p65 protein in VEC in LPS+XBJ group were significantly decreased (P<0.05 or P<0.01);the NO level and the expression levels of NF-κB p65 protein and iNOS protein in VEC in LPS+PDTC group were significantly decreased(P<0.05 or P<0.01).There were no significant differences of the NO levels and the expression levels of iNOS protein between LPS+XBJ group and LPS +PDTC group (P>0.05),but the expression level of NF-κB p65 protein in LPS+PDTC group was lower than that in LPS+XBJ group(P<0.05).Conclusion XBJ can inhibit the production of NO and the expression of iNOS protein in VEC;its mechanism may be related to inhibiting the activation of NF-κB to control inflammation.

Objective To explore the correlated factors of bilirubin neurotoxicity. Methods From July 2007 to October 2007, 53 hospitalized patients with jaundice in neonatal ward of Peking University Third Hospital were enrolled in the study. The relationship between serum free bilirubin and parameters of brainstem auditory evoked potential(BAEP) was analyzed to screen the sensitive index of bilirubin neurotoxicity. Multivariate analysis was conducted between the sensitive parameter and various factors. Results Partial correlation analysis showed that the wave Ⅴ latency was positively correlated to serum free bilirubin(r=0. 228, P= 0. 020) after the corrected gestational age was controlled. There were no correlation between other latencies or interwave intervals and serum free bilirubin. Linear regression showed the equation was wave Ⅴ latency= 9.721-0. 082 × corrected gestational age+0. 064 × bilirubin/albumin+ 0. 504 × acidemia(F= 7. 534, P＜0.01). Conclusions Wave Ⅴ latency is the sensitive indicator of bilirubin neurotoxicity, which is correlated with the value of bilirubin/albumin and acidemia.

Objective To investigate the changes in acetylation of histone in the spinal dorsal horn in a rat model of persistent postoperative pain.Methods Ninety-six malc Sprague-Dawley rats,weighing 200-250 g,aged 6-8 weeks,were randomly divided into 2 groups (n=48 each) using a random number table:sham operation group (group S) and persistent postoperative pain group (group PPP).The rat model of persistent postoperative pain evoked by skin/muscle incision and retraction was established according to the method described by Flatters.After the rats were anesthetized with intraperitoneal chloral hydrate,the skin and superficial muscle of the medial thigh were incised and retractors inserted.This tissue was retracted for 1 h.The mechanical paw withdrawal threshold (MWT) was measured at 1 day before operation and 1,3,7,14,and 21 days after operation.Four animals were sacrificed in each group after measurement of MWT at each time point for detection of acetylated histone H3 (Ac-H3) and acetylated histone H4 (Ac-H4) expression (by Western blot analysis) and the number of Ac-H3 and Ac-H4 positive cells in the spinal cord horn (by immunofluorescence histochemistry).Results Compared with group S,the MWT was significantly decreased at 3,7,14 and 21 days after operation,the expression of Ac-H3 and Ac-H4b was significantly down-regulated at 3,7 and 14 days after operation,and the number of Ac-H3 and Ac-H4 positive cells was significantly decreased at 7,14 and 21 days after operation in group PPP (P＜0.05 or 0.01).The MWT,expression of Ac-H3 and Ac-H4b,and the number of Ac-H3 and Ac-H4 positive cells were significantly higher at 21 days after operation than at 14 days after operation in group PPP (P＜0.05).Conclusion Acetylation of histone in the spinal dorsal horn is decreased after operation,which may be involved in the development and maintenance of persistent postoperative pain in rats.

Objective Study on the spread and the development history of traditional Chinese medicine in Africa, as well as the present situation and trend, provide reference for better promoting China-Africa cooperation of traditional Chinese medicine. Methods Literature review and historical research methods are used to retrospect the process of traditional Chinese medicine into Africa, and analyze the typical event. Results traditional Chinese medicine was introduced into Africa's history dates back to the period of Zheng He's voyage. In 1963, China sent medical teams to Africa, the TCM comprehensive spread in Africa started. In 2005, China sent the first group of traditional Chinese medicine volunteers to Africa, which a new pattern of TCM spread. Conclusions By sending the medical teams, volunteers, and education training, academic conferences, traditional Chinese medicine was gradually known by the African people. TCM is developing rapidly by the supporting of the government. The opportunity should be seized to promote the comprehensive development of traditional Chinese medicine in Africa.

BACKGROUND:Bone marrow mesenchymal stem cells can differentiate into osteoblasts under inducing condition that Zouguiwan and Youguiwan coordinate inducers, but the mechanism remains to be discussed. OBJECTIVE:To observe the effects of serum containing Zuoguiwan and Youguiwan on transforming growth factorβ1 and its signal transduction protein Smad2/3 message expression during the osteogenic differentiation of bone marrow mesenchymal stem cells. METHODS:A whole bone marrow adherence method was adopted to isolate and cultivate bone marrow mesenchymal stem cells from rats. The cellcultivation was processed in five groups:bone marrow mesenchymal stem cells were respectively cultured with blank serum, serum containing Zouguiwan, serum containing Youguiwan, positive serum containing progynova+inducer (dexamethasone, vitamin C, andβ-glycerophosphate), and inducer. Western blot was applied to detect the expression of type I col agen. The immunohistochemical assay was utilized to test transforming growth factorβ1 and Smad2/3 expression in the osteoblasts. RESULTS AND CONCLUSION:It was apparently more significant for serum containing Zuoguiwan and Youguiwan on type I col agen, transforming growth factorβ1 and Smad2/3 expression, compared with blank serum group and inducer group (P<0.05);moreover, serum containing Zuoguiwan was better than serum containing Youguiwan (P<0.05). Both of serum containing Zuoguiwan and Youguiwan are able to promote osteogenic differentiation of bone marrow mesenchymal stem cells. Moreover, Zuoguiwan is much more effective indicating that this method of traditional Chinese medicine about nourishing kidneys can be better to promote osteogenic induction of bone marrow mesenchymal stem cells.

Objective To evaluate the value of endoscopic ultrasound (EUS) in differentiatied diagnosis for stromal tumour and lipoma in duodenal tract.MethodsThe EUS images of 44 cases of stromal tumour(30 cases) and lipoma( 14 cases) which were confirmed by pathological results were analyzed retrospectively.The location,size,layer of origin,margin,internal echo pattern and homogeniety of the lesion were recorded and compared.Results Compared with lipoma,stromal tumour showed a significant difference in the layer of origin,margin,internal echo and homogeniety ( P ＜ 0.05 ),but there was no statistical difference in the lesion location and size( P ＞0.05).ConclusionsEUS is greatly helpful to the differential diagnosis of stromal tumour and lipoma in duodenal tract.
