1.Effect of 99Tc-MDP on the receptor activator of nuclear factor-κB ligand/osteoprotegerin system of type Ⅱ collagen-induced arthritis in rats

Objective To study the effect of 99Tc-MDP on receptor activator of nuclear factor-κB ligand/osteoprotegerin (RANKL/OPG) system of collagen-induced arthritis (CIA) rat. Methods The CIA was established by subcutaneous injection with type Ⅱ collagen and complete Freud's adjuvant to rats, except the group (n=8, treated with 99Tc-MDP). The histopathological changes of joints were scored based on the extent of infiltration of inflammatory cells, cartilage destruction and bone erosion. The serum levels of RANKL/OPG were tested with ELISA, and the synovial membrane levels of RANKL/OPG were tested with Western blot.Repeated ANOVA and one way ANOVA were used for statistical analysis. Results The MTX group relative value of RANKL/OPG in serum [35 d (1.076±0.016), 42 d (1.140±0.005), 49 d (1.155±0.023)], and in synovial membrane (1.156±0.014), and the histopathological scores [synovial membrane (1.8±0.5), cartilage (1.9±0.6), bone (1.8±0.5) ] were lower than model group ( 1.269±0.025, 1.296±0.015, 1.340±0.011 ), (2.9±0.4, 2.6±0.5, 2.6±0.5), ( 1.340±0.013 )(P<0.01 ). And all above index of 99Tc-MDP group ( 1.035±0.034,0.986±0.019, 0.991±0.020), (1.5±0.5, 1.4±0.5, 1.2±0.5), (0.098±0.026) were notably lower than the MTX treat ment group (P<0.01). Conclusion 99Tc-MDP may retard the progression of the disease by decreasing the relative RANKL/OPG expression and it can initiate its effect earlier than MTX.
2.Effects of murine vascular endothelial growth factor (mVEGF) and anti- vascular endothelial growth factor (anti-VEGF) antibady on human umbilicus vein endothelial cell (HUVEC) proliferation

Objective To study on effects of mVEGF and anti-VEGF antibody during cultured HUVEC proliferation in vitro.Methods Endothelial cell proliferation was assayed using human umbilical vein endothelial cells stimulated with mVEGF and with CIA joint extracts and was used 3 H-TaR incorporation.Results The anti-VEGF neutralizing antibody can inhibit the proliferation of HUVEC stimulated with mVEGF and with CIA joint extracts,whose suppression percents were 72.2% and 69.9%,respectively.Conclusion mVEGF specifically promotes the growth of vascular endothelial cells.During early stage of CIA development,expression of VEGF in the joint increases and VEGF is expressed biologically active and can be inhibited by anti-VEGF neutralizing antibody.
3.MEASUREMENT AND CLINICAL APPLICATION OF GLYCOSYLATED LDL AND THE GLYCOSYLATED RATE OF LDL IN DIABETES MELLITUS

A fructosamine (DMF) method for the estimation of Serum glycosylated LDL (GLDL) is reported. The glycosylated rate of LDL (K value) is equal to GLDL/LDL. It is concluded that fructosamine method for the estimation of GLDL and K value is more convenient, fast, and has a high specificity, and is not interfered by glutathione, other reducing substances with low melecular weight, and the variation of serum proteins. GLDL and K value are thus suggested to be a better indicator for the diagnosis and evaluation of treatment in diabetes mellitus than the glycosylated serum total proteins.
4.Effects of dieting and exercise intervention on lipid level in patients with hyperlipidemia

Objective To investigate the effects of dieting and exercise intervention on lipid level in patients with hyperlipidemia.Methods Health education including dieting and exercise interventions was given to 1,800 patients with hyperlipidemia.Serum total cholesterol(TC), triglyceride(TG), high density lipoprotein(HDL-C)and low density lipoprotein (LDL-C)levels were assessed before and after interventions.Result TC, TG and LDL-C levels were decreased and HDL-C increased after interventions with statistical significance(P<0.01).Conclusions Dieting and exercise intervention can be effective in controlling lipid levels effectively.Health education should vary among individuals to achieve good effects.
5.Investigation for the expressions of Skp2 and P27 in normal and tumor salivary tissues by tissue chip technique

Objective:To study the expressions of Skp2 and P27 in the normal and tumor tissues of salivary glands,and then to evaluate their significance in the development and progression of salivary tumor.Methods:SP immunohistochemical method was used to evaluate the expression of Skp2 and P27 in the normal and tumor tissues of salivary glands. Results:The rates of Skp2 and P27 expression were neither significantly different between polymorphic adenoma and basal cell adenomas, nor significantly different between adenoid cystic carcinoma and mucoepidemoid carcinoma. The rates of Skp2 positive were significantly different between normal salivary glands and benign salivary tumor, as well as between normal salivary glands and malignant salivary tumor. The rates of Skp2 and P27 positive were significantly different between normal salivary glands and malignant salivary tumor.Conclusion:Skp2 overexpression and decreased expression of P27 might play an important role in the development and progression of salivary tumor, overexpression of Skp2 protein may lead to increase of P27 degradation.
6.Treatment of intrasacrospinal muscular approach and midline approach in lumbar interbody fixation with fusion for lumbar spinal stenosis

Objective To explore the clinical effect about the treatment of intrasacrospinal muscular approach and midline approach in lumbar interbody fixation with fusion for lumbar spinal stenosis.Methods Forty-two cases lumbar spinal stenosis were reviewed retrospectively in Meitan General Hospital from February 2009 to February 2014.All cases were divided into intrasacrospinal muscular approach and midline approach group(Group A,n =19) and midline approach group(Group B,n =23) according to the different surgical approach.Operation time,the amount of blood loss,post-operative drainage were compared.The VAS score of low back pain and leg pain preoperative and 6 months after operation were recorded.All cases were followed up for more than one year to evaluate fusion status.Results All cases were followed up for 13.0-20.0 months,with an average of (16.0± 1.2) months,all patients had bony fusion.Operation time,the amount of blood loss,postoperative drainage of Group A were significantly less than of Group B,the differences were significant ((95.1±4.5) min vs.(110.4±6.6) min,(75.2±5.1) ml vs.(159.8±7.2) ml,(53.6±4.3) ml vs.(86.2±14.1) ml;t =1.565,6.087,2.371;P<0.05).VAS of back pain and leg pain at 6 months after operation of Group A showed better results than of Group B,the differences were significant((1.43±0.76) points vs.(1.39±1.00) points,(2.78± 1.23) points vs.(2.52±0.88) points;t =1.982,1.774;P<0.05).Interbody fusion was obtained in two groups at the end of the last follow-up.Conclusion The intrasacrospinal muscular approach and midline approach for lumbar interbody fixation with fusion for lumbar spinal stenosis is better in less injury to paraspinal muscle,less blood loss and less post-operative drainage,which contributes a rapid recovery.
7.Prognostic analysis of 43 cases of secretory carcinoma of the breast

Objective To investigate the clinicopathologic features and prognosis of secretory carcinoma of the breast (SCB).Methods We enrolled 43 cases of secretory carcinoma of the breast out of invasive breast cancer patients according to the 2012 WHO Classification of Tumors of the Breast.Results The incidence of breast SCB was low,accounting for about 0.17% of all breast cancer patients.The median age of onset was 49 years old.The diameter of tumor was 1.0-10.0 cm(mean size 3.2 cm).Pathology revealed large number of eosinophilic secretory material inside or outside the cancer cells positive for periodic acid-Schiff stains (PAS) and Alcian blue (AB).The positive rate of estrogen receptor (ER) was 34.9% (15/43),progesterone receptor (PR) was 23.3% (10/43),human epithelial growth factor receptor 2 (HER-2) was 25.6% (11/43),S-100 was 97.7% (42/43),and special PAS and AB diastase were tested and positive in 8 cases.The postoperative average follow-up time was 78 months,7 cases were found with distant metastasis.2 cases dying of distant metastasis.Conclusions SCB has a good prognosis despite of occasionally axillary lymph node metastasis,local recurrence or distant metastasis.
8.A retrospective cohort study of 320 thousand subjects of colorectal cancer screening in Haining City.

OBJECTIVETo evaluate the colorectal cancer incidence among compliers and non-compliers in the population of colorectal cancer screening area, and to provide scientific basis for health economic evaluation of cancer screening.
METHODSBy screening different years build queue, to retrospectively compare the data of colorectal cancer screening from January 1, 2008 to December 31, 2013 and the data of cancer registration, and to analyze the colorectal cancer incidence rates among screening compliers and non-compliers, and to compare the average intervals between the end of screening and clinical cancer diagnosis using SPSS 19 statistical software. Mantel-Haenszel test was performed with a statistical significance level of α = 0.05.
RESULTSThe non-compliance rate was 38.24% among males and 28.49% among females (P < 0.001). The non-compliance rate was highest in the 40-44 and 70-74 years age groups and lowest in the 50-59 years age group. The compliers of the screening were followed up for 476,049 person-years, and there were 51 cases of colorectal cancer, with an incidence rate of 10.71/100,000. The non-compliers of the screening were followed up for 259 183 person-years, and there were 66 cases of colorectal cancer, with an incidence rate of 25.46/100,000, which was 1.38 times higher than that of the compliance group (χ2 = 21.699, P < 0.001). The incidence rate of colorectal cancer among subjects who were positive in initial screening but refused to receive an electronic colonoscopy was as high as 164.40/100,000, and the average delay time was 20.8 ± 16.8 months.
CONCLUSIONSThe colorectal cancer screening adherence among women is better than among men, and that of 50-54 years and 55-59 years age groups is better than in other age groups. The subsequent incidence rate of colorectal cancer in the non-compliance group is significantly higher than that of the compliance population.

9.Clinical Study on Headache due to Nitroglycerin Remitted with Ice Covering Head

Objective To study whether covering ice on head can abate headache produced by using nitroglycerin.Method 64 cases were collected from the patients who used nitroglycerin and were randomly divided into one contrasting group (group A) and one experimental group (group B).For the patients of group A,the state of headache produced by using nitroglycerin should be observed.For the patients of group B,covering ice on their head was adopted when they used nitroglycerin,and the state of headache should also be observed.Results There were obvious difference between the two groups (P<0.001).Conclusion Covering ice on head is a good method which can abate headache produced by using nitroglycerin.
10.Effect of therapeutic hypercapnia on hepatic ischemia-reperfusion injury in rat liver transplantation

ObjectiveTo investigate the effect of therapeutic hypercapnia on hepatic ischemia-reperfusion (I/R) injury in rat liver transplantation.MethodsMale specific pathogen-free adult Wistar rats aged 6 weeks weighing 220-280 g were used in this study.Sixteen rats in which liver transplantation was successfully performed were randomly divided into 2 groups ( n =8 each): liver transplantation group (group LT) and therapeutic hypercapnia group (group TH).In group TH,PaCO2 was maintained at 80-100 mm Hg by inhalation of CO2 for 1 h at the begining of reperfusion.MAP,PaO2 and PaCO2 was recorded during reperfusion.Blood samples were obtained at 2 h of reperfusion for determination of serum ALT,AST,TNF-α,IL-1 and IL-6 levels,and then the rats were sacrificed and transplanted liver was immediately removed for determination of NF-κB activity and apoptosis and microscopic examination.The apoptotic index was calculated.ResultsMAP,PaO2 and PaCO2 were higher,and serum ALT,AST,TNF-α,IL-1 and IL-6 levels,NF-κB activity and apoptotic index lower in group TH than in group LT ( P < 0.05).The histopathologic damage was ameliorated in group TH as compared with group LT.Conclusion Therapeutic hypercapnia can attenuate hepatic I/R injury in rat liver transplantation by inhibiting inflammatory response and apoptosis.
