1.Study on Formaldehyde-Induced DNA Damage in Mice Testicular Cells

Objective To exlplore the adverse effect of formaldehyde on DNA in isolated mice testicular cells. Methods The testicular cells were exposed to formaldehyde of different concentrations(0, 25, 50, 75, 100 ?mol/L), comet assay was applied to detect DNA breaks. Results Formaldehyde induced DNA breaks at 10 ?mol/L, 25 ?mol/L and 50 ?mol/L (P0.05). The critical concentration at which formaldehyde could induce DNA strand breaks of testicular cells was about 10 ?mol/L, peak point was near 50 ?mol/L. Conclusion Formaldehyde may cause DNA damage in isolated mice testicular cells in various degree.
2.Operations and Application Experience of Perpendicular Acupuncture at the Back-Shu Points of the Five Zang-organs

The Back-Shu points of the five Zang-organs are a group of points where qi and blood pour into the back and lumbar regions. Acupuncture at these points has a wide indication scope. However, it’s reported that internal organs were punctured in some cases because of improper control of needling depth, angle, and intensity during acupuncture. We used perpendicular needling instead of conventional oblique needling during the acupuncture of the Back-Shu points of the five Zang-organs. We found that when patients were completely relaxed, acupoints were correctly selected, the needling depth, the way of holding the needle, and inserting methods were well controlled, perpendicular needling of the Back-Shu points of the five Zang-organs can produce stronger needling sensation, and it’s easier to operate, safer, and more effective compared to oblique needling.
3.Modified radical mastectomy-stage Ⅰ expansion of latissimus dorsi muscle flap breast reconstruction

Background and purpose:Physical and psychological trauma has been a challenge for both clinician and patients with breast cancer after modified radical mastectomy,and now more and more patients have begun to accept breast reconstruction,especially Ⅰ stage reconstruction after surgery.The aim of our study was to investigate a breast reconstruction procedure that can not only achieve radical tumor excision but also retain the aesthetic appearance of breast.Methods:15 patients from January 2006 to April 2007 were enrolled with early stage breast cancer have received surgical treatment with conservation of breast skin,breast tissue excision and axillary lymph nodes;latissimus dorsi muscle flap has been used for breast reconstruction.Results:The results of the 15 cases:excellent in 11 cases(83.3%),good in four cases(16.7%).Conclusions:Immediate breast reconstruction after radical mastectomy with conservation of skin has shown good results in terms of aesthetic appearance of breast,this procedure has been proved to be a safe and feasible method of treatment for the patients with early stage breast cancer.
4.A COMPARITIVE STUDY ON IODINE NUTRITIONAL STATUS OF STUDENTS IN ZHOUSHAN ISLANDS

Objective: To measure the iodine nutritional status of students in Zhoushan Islands and evaluate the advantages and disadvantages of iodized salt prophylaxis. Methods: A comparative study was carried out in 12 classes which were selected from Dinghai(iodized salt district) and Daishan(non-iodized salt district) of Zhoushan Islands by random sampling. Mann-Whitney test was used to compare the urinary iodine concentration and dietary iodine intake of the two groups. Spearman correlation was used to look for the correlation between urinary iodine concentration and dietary iodine intake in the two groups. Ordinal regression was used to analyze the factors that affect the urinary iodine concentration. Poisson regression was used to analyze the risk factors of goiter.Results: The urinary iodine concentration in non-iodized salt district was 129 ?g/L,which was lower than 169 ?g/L in iodized salt district(u=6.302,P=0.000) , whereas the daily iodine intake in the two groups was 105 ?g/d and 184 ?g/d respectively, the difference was also significant(u=4.710,P=0.000). The influence of sex and iodized-salt intake was significant in the final regression model (P0.05). AIM: To observe the change of boiling point in the concentration of medical herb's extract liquor. METHODS: Extract liquors of typical herb. such as Radix salviae Miltiorrhizae, Flos chrysanthemi indici, Folium isatidis and Radix Astragli, were concentrated in the normal pressure, its boiling-points and mol concentrations were recorded and its curve was depicted. RESULTS: At the same concentration, the boiling points of different kinds of Chinese medicinal material is different. CONCLUSION: In the concentration process of distillation the change of solvent and solute should be taken into account.
6.Effects of Mirror Visual Feedback and Electromyographic Biofeedback on Upper Extremity Function in Hemiplegics after Stroke

Objective To investigate the effects of mirror visual feedback (MVF) and electromyographic biofeedback (EMGBF) on upper extremity function in hemiplegic patients after stroke based on task-oriented training. Methods 90 patients with hempiplegia after stroke were randomly divided into control group (n=30), EMGBF group (n=30) and MVF group (n=30). All patients accepted routine rehabilitation and task-oriented training once a day for 8 weeks. The EMGBF group also accepted EMGBF, and the MVF group accepted MVF in addition. They were assessed with Fugl-Meyer Assessment (FMA) and the Upper Extremity Function Test (UEFT), and their integrated electromyogram (iEMG) of affected upper extremities were recorded before and after treatment. Results All the groups improved in scores of FMA and UEFT, as well as the iEMG after treatment (P<0.05), and ranked as the MVF group, the EMGBF group and the control group from improving more to less (P<0.05). Conclusion Mirror visual feedback combined with electromyographic biofeedback may further promote the recovery of upper limb function in patients with hemiplegia after stroke based on task-oriented training.
7.Study on the event-related potentials P300 of schizophrenic patients with aggressive or violent behaviors

ObjectiveTo explore the electrophysiological mechanisms of the schizophrenic patients with aggressive or violent behaviors.MethodsAccess the aggressive behaviors of schizophrenic patients being treated in hospitals or clinics with the revised MOAS in accordance with the ICD-10 diagnostic criteria,and sort the qualified patients into two groups:the group of aggressive or violent schizophrenia (Aggressive Group,n=70) and the group of non-aggressive or non-violent schizophrenia ( Non-Aggressive Group,n =65 ) ; 60 age- and gendermatched healthy people were collected as Healthy Group.P300 tests were carried out on patients in these three groups with the MEB-9200 Nicolet Bravo Instrument by the Nihon Kohden Corporation.Results ( 1 ) latency P3a of the Aggressive Group on Cz point exceeded that of the Non-Aggressive Group (P =0.01 ),and that of the Non-Aggressive Group exceeded that of the Healthy Group.All these disparities were of statistical significance (P <0.01 ).Latency P3a of the Aggressive Group on Fz point exceeds that of the Non-Aggressive Group,and that of the Non-Aggressive Group exceeded that of Healthy Group.All these disparities are also of stafistical significance (P<0.01).(2)N2' amplitude of the Aggressive Group on Cz point was higher than those of the Non-Aggressive Group and the Healthy Group.This disparity was of statistical significance(P < 0.05 ) and the disparity between the Non-Aggressive Group and the Healthy Group did not have statistical significance (P =0.985 ).ConclusionCharacteristic electrophysiological changes exist in the event-related potentials P300 of schizophrenic patients with aggressive or violent behaviors.
8.Screening, identification and significance of the phage-display random 7 amino acid peptide specific to the sera of patients with systemic lupus erythematosus

Objective To screen and identify the phage-display random 7 amino acid peptide specific to the systemic lupus erythematosus(SLE) and analyze its practical significance. Methods Using the phage random 7 peptide library screening, the SLE specific phage clones are obtained after binding with the mixture of sera from 30 SLE patients and 30 normal controls as ligand respectively. Then the Dot-ELISA is used to identify the SLE specific phage clones reactive to sera of the SLE patients and normal controls individually. Finally the identified phage-display random 7 amino acid peptides are sequenced and it's homology with the antigenic epitope of human being and other are also analyzed. Results Total 12 of the phage-display random 7 amino acid peptide are obtained by phage peptide library screening and the Dot-ELISA identification. Sequence analysis shows that the identified phage-display random 7 amino acid peptide epitope have homology with E. coli, Salmonella and human immunodeficiency virus, but not with that of human being. Conclusion SLE-specific peptides screened by phage random peptide library maybe used to diagnosis the SLE. Meanwhile, the antibodies in SLE patients which are combined with the Pathogen epitope, suggest that SLE maybe relate to pathogen infection.
9.Analysis of peripheral blood lymphocyte subsets in patients with antinuclear antibody positive

Objective To investigate the clinical application value of peripheral blood lymphocyte subsets expression levels in autoimmune disease(AID) among the patients with antinuclear antibody(ANA) positive .Methods 200 patients with ANA positive and 196 patients with ANA negative were selected as the experimental group and control group respectively .The experimental group adopted indirect immunofluorescence assay (IFA) and immunoblotting assay(LIA) for detecting ANA ,moreover divided into the IFA group and LIA group according to the detection results .Meanwhile the flow cytometry was adopted to detect peripheral blood T lymphocytes ,helper T lymphocytes ,cytotoxic T lymphocyte ,NK lymphocytes and B lymphocytes absolute values of each group .The detection results were statistically analyzed .Results Helper T lymphocytes ,NK lymphocytes and B lymphocytes absolute values in the experimental group were significantly lower than those in the control group ,the differences were statistically significant(P<0 .01);in the experimental group ,helper T lymphocytes and NK lymphocytes absolute values in the IIF group were significantly lower than those in the LIA group ,the differences were statistically significant (P<0 .01) .Conclusion Peripheral blood lymphocytes subsets can serve as the important detection indicators during the diagnosis and treatment process of AID .
10.Relationship between chemosensitivity in vitro and the expression of GRP78 in non-small cell lung cancer.

BACKGROUNDThe expressive level of glucose-regulated protein 78 (GRP78) is elevated and correlated with resistance of chemotherapy drugs in breast cancer cell. However, little is known about the relationship between its expression and drug resistance in non-small cell lung cancer (NSCLC). The aim of this study was to explore the relationship between drug resistance and the expression of GRP78 in NSCLC.
METHODSDrug sensitivity test was used to detect the resistance to 8 chemotherapy drugs in 52 NSCLC fresh surgical samples by methylthiazoletrazolium (MTT), and expression of GRP78 was detected by immunohistochemistry method. Spearman correlation assay was used to investigate the correlation between the GRP78 expression and drug resistance.
RESULTSThe resistance rates to paclitaxel (PTX), adriamycin (ADM), carboplatin (CBP), topotecan (TPT), navelbine (NVB), vincristine (VCR), cisplatin (DDP) and etoposide (VP-16) of the 52 samples were 42.31%, 57.69%, 63.46%, 65.38%, 67.31%, 73.08%, 78.85%, 90.38%, respectively. Fourteen cases showed the complete resistance to the total 8 chemotherapy drugs. Furthermore, the expression of GRP78 was stronger in poorly differentiated cancer as compared with the well and moderately differentiated cancer (P < 0.05), so as in stage II and III cancer than in stage I cancer (P < 0.05). Spearman correlation assay showed that there was a correlation between the chemotherapeutics resistance to ADM, VP-16, VCR, TPT and the expression of GRP78 in NSCLC (P < 0.05).
CONCLUSIONSIt is feasible to detect the drug sensitivity to chemotherapy for tumor cells by MTT method. The results of chemosensitivity assay in vitro are indicative of clinical drug administration in NSCLC. The detection of GRP78 isalso indicative of the resistance to chemotherapy drugs and the differentiation and the clinical stage in NSCLC.
