1.Immunomodulatory effects of polysaccharopeptide on EAC tumor-bearing mice through MyD88-dependent signaling pathway

Objective:To detect the effects of polysaccharopeptide(PSP) on MyD88-dependent signaling pathway in EAC tumor-bearing mice,and explore the immunomodulatory mechanism of PSP.Methods: Ehrlich′s ascites carcinoma(EAC) C57BL/6 mice were used to establish the animal model for solid tumor.Mice were divided into two groups:WT group and MyD88-/-group.After 22 days of treatment,quantitative real-time PCR( Q-PCR) ,Western blot and were used to detect the related gene and protein expression of TLR4 pathway in spleens,ELISA were used to detect the terminal effect factors secretion eyeball blood from each group.Results:Related genes and proteins of TLR4 pathway in spleen were up-regulated significantly from two groups.Compared with WT group, related genes and proteins in MyD88-dependent pathway(MyD88,TRAF6,NF-κB,AP-1)was down-regulated in MyD88-/-group(P<0.05).Meanwhile,There was no significant change of the related genes and proteins of MyD88-independent pathway(TRAM,TRIF)in MyD88-/-group( P>0.05 ) .The terminal effect factors secretion of IL-2, IL-6, IL-10, TNF-αand IFN-γin MyD88-/-group were decreased significantly compared with WT group(P<0.05).Conclusion:The immunoregulatory effect of PSP on EAC tumor-bearing mice may be implement through the regulation of MyD88-dependent signaling pathway.
2.Ultrastructural study of sclerotic dentin in non-carious cervical lesions disposed by a total-etching dentin adhesive

Objective: To examine the ultrastructure of resin-infiltrated sclerotic dentine following the application of a three-step total-etching dentin adhesive. Methods: Naturally-occurring, non-carious cervical lesions on extracted premolars were bonded using a total-etching dentin adhesive-Scotchbond Multi-purpose. Artificially prepared wedge-shaped lesions were also made in sound premolars and restored using the same adhesive as control. The morphological change of the dentin surface conditioned with the etchant, and the hybrid zone and resin tags in the dentin-resin interface were studied by SEM . Results: Most dentinal tubules were obliterated by rod-like sclerotic casts in the non-carious cervical lesion, and could not be dissolved by Scotchbond Multi-purpose etchant.Both the hybrid zone and resin tags were observed in sclerotic dentin after restoration, although the resin tags were fewer and in lack of communications, the length of resin tags and the thickness of the hybrid zone were almost similar to those of the sound dentin. Conclusion: Bonding to sclerotic dentin is different from sound dentin, and may be compromised by the fewer resin tags and communications.
3.The anti-tumor activity and molecular mechanisms of an Aurora kinase inhibitor ZLJ213 in suppressing colon cancer growth.

The aim of this study is to evaluate anti-tumor activities and mechanism of a novel kinase inhibitor ZLJ213 which targeted Aurora A and vascular endothelial growth factor receptor (VEGFR) in vitro and in vivo against human colon cancer. Results showed that ZLJ213 inhibited cell proliferation and induced cell cycle arrest and apoptosis of HCT1 16 and SW48 cell lines. In HCT116-derived xenograft, ZLJ213 dosed at 100 mg · kg(-1) inhibited tumor growth by 73.24%. The IC50 of ZLJ213 on the expression of p-Aurora A was 0.258 µmol · L(-1) analyzed by ELISA. Under the concentration of 0.08 µmol · L(-1), ZLJ213 could inhibit the activities of Aurora A, Histone H3 and VEGFR of HCT116 and SW48 cell lines. Simultaneously, ZLJ213 induced activation of Caspase 3 and PARP cleavage. Above data suggested that ZLJ213 had the ability to inhibit cell proliferation and induce cell apoptosis both in vitro and in vivo in colon cancer, and down-regulate the expression of p-Aurora A and p-VEGFR. ZLJ213 might be a potential therapeutic agent against colon cancer.
4.Ultrastructural study of non-carious cervical sclerotic dentin bonded by a all-in-one self-etch adhesive

The morphological change of dentin surface conditioned by the adhesive and the dentin-resin interface were studied by SEM.Results:Most dentinal tubules were obliterated by rod-like sclerotic casts in the non-carious cervical lesion, the sclerotic casts could not be totally dissolved by the self-etch adhesive. After restoration the resin tags were fewer and shorter in sclerotic dentin than those in sound dentin.Conclusions:Bonding to sclerotic dentin is different from that to sound dentin, and may be compromised by the fewer resin tags.
5.Injection stauntoniae inhibits chronic inflammatory pain responses by changing COX-2 expression in carrageenan-inflamed mice

AIM: To investigate the effect of injection stauntoniae ( IS) on inflammatory pain responses in mice.METHODS:The carrageenan test was used to determine the anti-inflammatory and analgesic effects of IS .Except for control group, the mice in other groups received an injection of λ-carrageenan solution (1%, 50 μL) into the plantar region of the left hind paws , followed by a subcutaneous injection of IS at doses of 12.5%, 50%and 100%or equal vol-ume of 0.9%NaCl.Both paw edema and hyperalgesia to thermal stimulation were measured 4 h, 12 h, 24 h and 48 h after the injection of λ-carrageenan solution.The lumbar-5 (L5) dorsal root ganglions (DRGs) of the mice were taken to inves-tigate the cyclooxygenase 2 ( COX-2) expression by immunohistochemical staining .RESULTS:Subcutaneous injection of IS potently inhibited paw edema and hyperalgesia induced by λ-carrageenan in the mice accompanied with the inhibition of COX-2 protein expression in L 5 DRGs.CONCLUSION:IS exerts the anti-inflammatory and analgesic effects on the in-flammatory responses by inhibiting the protein expression of COX-2 in DRGs.
6.Synthesis and in vitro cytotoxic activities of sorafenib derivatives.

A series of novel sorafenib analogues were designed and synthesized. The cytotoxic activities of these compounds were tested in four tumor cell lines. Some of the compounds showed potent antiproliferative activity against the tested cell lines with IC50 = 4-20 micromol x L(-1). Some compounds demonstrated competitive antiproliferative activities to sorafenib against tested cancer cell lines. Among them, compound 7c demonstrated significant inhibitory activities on ACHN, HCT116 and MDA-MB-231 cell lines with IC50 values of 9.01, 4.97, 6.61 micromol x L(-1), respectively.
7.Design, synthesis and antitumor activity of sorafenib analogues containing 2-picolinylhydrazide moiety.

A novel series of sorafenib analogs containing 2-picolinyl hydrazide moiety were designed and synthesized. In vitro, most of synthesized compounds have antiproliferation activity on MDA-MB-231, ACHN, HepG2, Mia-PaCa-2 and SW1990 cell lines tested by MTT assay. It is worth noting that the antitumor activities of compounds 2c, 2d and 2f are more potent than that of sorafenib on pancreatic cancer cells Mia-PaCa-2 and SW1990, and the activities of compounds 3f and 3g are 2-3 times than that of sorafenib on human hepatocellular carcinoma HepG2 cell line.
8.Conditions for extraction of ergosterol from fruits of Ganoderma lucidum using supercritical fluid extration.

OBJECTIVETo study the feasibility of supercritical fluid extraction (SFE) for extraction of ergosterols from the fruits of Ganoderma lucidum.
METHODOptimum extraction conditions were studied by orthogonal tests, the ergosterol were analyzed by RP-HPLC.
RESULTThe optimal extraction conditions were pressure as 20 MPa, temperature at 45 degrees C, using ethanol as modifier carrier at the volume of 1 mL x g(-1), and 1.5 hour of extraction time.
CONCLUSIONSFE has the superiority of adjustable polarity, and has the ability of extracting ergosterol from fruits of Ganoderma lucidum.

9.Discussion of the implementation of MIMIC database in emergency medical study

To introduce Medical Information Mart for Intensive Care (MIMIC) database and elaborate the approach of critically emergent research with big data based on the feature of MIMIC and updated studies both domestic and overseas, we put forward the feasibility and necessity of introducing medical big data to research in emergency. Then we discuss the role of MIMIC database in emergency clinical study, as well as the principles and key notes of experimental design and implementation under the medical big data circumstance. The implementation of MIMIC database in emergency medical research provides a brand new field for the early diagnosis, risk warning and prognosis of critical illness, however there are also limitations. To meet the era of big data, emergency medical database which is in accordance with our national condition is needed, which will provide new energy to the development of emergency medicine.
10.Study on quality and safety management standard of lifesaving access to emergency care

A review of management problems found in lifesaving access to emergency care in healthcare organizations at all levels in China, and a reference international critical and emergency care systems and their quality and safety management standards were made by the authors. Based on such and special needs of such access, the paper studied the domestic lifesaving access to emergency care service workflow, and abstracted common management issues on the lifesaving access to emergency care. The study identified key elements of lifesaving access to emergency care management through case analysis, and formulated standard terms for such key elements. Hence a set of standards is developed for quality management fitting healthcare organizations at all levels in China′s lifesaving access to emergency care, serving as the guidance for their standardized management and evaluation.
