1.Protection of Semen Cuscuta extracts from apoptosis PC12 cell of induced by 1-methyl-4-phenylpyridinium

AIM: To establish an apoptotic model induced by 1-methyl-4-phenylpyridinium ion(MPP~+) in PC12 cell and assess the effect of Semen Cuscuta extracts on protecting PC12 cell from apoptosis induced by MPP~+. METHODS: The cell viability was analyzed by MTT method,the morphological changes were observed by Hoechst 33258 staining assay,and the apoptosis rate was measured by flow cytometry analysis(FCM).(RESULTS:) Treatment of 0.15 mM MPP~+ for 48 h induced apoptosis in PC12 cells;concentration of 50 mg/L Semen Cuscuta extracts could protect cells from apoptosis induced by MPP~+. CONCLUSION: MPP~+-induced apoptosis in PC12 cell is greatly inhibited by Semen Cuscuta extracts.

3.Application of endoscopic miniprobe ultrasonography in diagnosis and therapy of gastrointestinal submucosal tumors

Objective To evaluate the diagnostic value of endoscopic miniprobe ultrasonography (MPS) and endoscopic therapies under the guidance of MPS in gastrointestinal submucosal tumors. Methods Patients with submucosal tumors of lower digestive tract were given MPS under colonoscope. Endoscopic therapy or surgery had been taken according to different deriving layers of submucosal tumors. The ultimate diagnosis was confirmed by histology. Results Twenty patients with submucosal tumors were diagnosed with MPS under colonoscope, including 9 patients with stromal tumors, 5 patients with lipoma, 3 patients with car-cinoid, 2 patients with abscess,1 patient with intestines pneumatosis. Four patients with benign stromal tumors derived from muscularis mueosa and 2 patients with carcinoid were resected by endoscopy. Two patients with abscess and 1 patient with intestines pneumatosis were puncture under endoscopy. Three patients with benign stromal tumors from muscularis propria, 2 patients with malignant stromal tumors, 2 patients with lipama and 1 patient with careinoid were resected surgically. 88.24% submucosal tumors diagnosed by MPS were identified by pathology. Three cases were followed-up by endoscopy. Conclusions Different layers of gastrointestinal tract with submucosal tumors can be distinguished clearly by MPS leading to definite diagnosis of submucesal tumors. MPS can play an important role in selecting treatment procedures.
4.A case report of isolated marginal zone B cell lymphoma originating from the parapharyngeal space.

This is a case report of isolated marginal zone B cell lymphoma originating from the parapharyngeal space and reaching up to the side of skull base. There are no obvious clinical symptoms of the marginal zone B cell lymphoma originating in the parapharyngeal space, the imaging examination of which showed slightly blurred edge mass shadow with heterogeneous textures and no specificity. The pathological examination revealed a large number of lymphocyte proliferation, which was not specific yet. Immunohistochemistry may show the tumor was monoclonal and positive to CD20 and CD79a positive. Marginal zone B cell lymphoma originating in the parapharyngeal space is a rare tumor with no specific clinical symptoms or early signs, and is easily misdiagnosed. The diagnosis should be confirmed based on pathological and immunohistochemical examination, and the tumor can be treated by surgery combined with chemotherapy.

5.Mutation analysis of keratin 17 gene in a pedigree with steatocystoma multiplex

Objective To analyse the mutation of K17 gene in a pedigree with steatocystoma multi-plex. Methods Blood samples were obtained from 3 affected and 3 normal individuals in a family with steatocystoma multiplex, as well as from 50 unrelated healthy individuals. Mutation scanning was carried out by PCR and direct sequencing. Results A heterozygous nucleotide transition (C→T) at position 428 in exon 1 of KI7 gene, which leads to the substitution of CGC (arginine) by TGC (histidine) at codon 94, was detected in the affected individuals, but not in normal family members or the 50 unrelated individuals. Conclusion A missense mutation (428C→T) in KI7 gene has been detected in affected individuals of this family, which seems to be a molecular basis of pathogenesis of steatocystoma multiplex.
6.Infection of respiratory syncytial virus in BALB/c mice and nude mice

Objective To compare respiratory syncytial virus(RSV)infection and inflammatory responses between immunocompetent BALB/c mice and immanodeficient nude mice.Methods At various time points after BSV infection of BALB/c mice and nude mice,pulmonary viral titers were assayed.Leukocvtes in bronchoalveolar lavage fluid(BALF)and pulmonary histology were identified.F4/80+cells and CD49b+cells in lung tissue were examined by immunohistochemistry,and the cytokines of TNF-α,IL-12,IFN-r and IL-10 in BALF were assayed by ELISA.Results RSV titers in infected BALB/c mice and nude mice peaked on the 3rd day postinoculation,and nude mice had higher-level and more durative viral replication than BALB/c mice.RSV infection induced more severe pulmonary histopathology and larger number of leukocytes in airway in nude mice than in BALB/c mice.RSV infection enhanced more pulmonary F4/80+macrophages,CD49b+ NK cells in both mice.Furthermore infected nude mice had larger amount of pulmonary macrophages and NK cells than infected BALB/c mice.RSV infected BALB/c mice secreted more TNF-α,IL-12,IFN-r and IL-10 as compared with control BALB/c mice,and infected nude mice had hisher level of TNF-α.IL-12 and IL-10 than infected BALB/c mice.Conclusion Nude mice are good model for severe and pemistent RSV infection in immunocomprised hosts.The inflammation induced by RSV infection is not parallel with the immune response of T cells,and macrophages and NK cells are potent immunocytes and inflammatory cells in RSV infection especially when T lymphocytes are absent.
7.Association of ulcerative colitis with fork head/winged helix transcription factor-3 gene polymorphisms in Chinese patients

Objective To investigate the association of ulcerative colitis (UC) with fork head/ winged helix transcription factor-3 (Foxp3) polymorphisms in Han population in Zhejiang province,China.Methods A total of 381 UC patients and 490 healthy controls were enrolled in this study.The four single nucleotide polymorphisms (SNPs) of Foxp3 (rs3761547,rs2232365,rs2294021,rs3761548) were examined by SNaPshot.The analyses of linkage disequilibrium (LD) and haplotype were also performed in all study subjects.Results When male and female UC patients were compared with their corresponding controls respectively,the alleles and genotypes of the four SNPs were not statistically different (all P >0.05).According to severity and location of the disease,the UC patients were divided into different subgroups.The alleles (C,G,A) of (rs2232365,rs2294021,rs3761548) were more frequent in male patients with severe UC than in the male controls (69.6% vs 34.3%,P =0.001;69.6% vs 34.3%,P =0.001;39.1% vs 14.4%,P =0.002,respectively).As compared with the female controls,the alleles (C,G,A) and genotypes (TC + CC,AG + GG,CA + AA) of (rs2232365,rs2294021,rs3761548) were significantly increased in the female patients with severe UC (51.9% vs 38.0%,63.5% vs 39.2%,53.8% vs21.4%,80.8% vs57.7%,84.6% vs58.4%,76.9% vs34.7%,all P<0.05).The four SNPs above were shown to be in a strong LD both in male and in female subjects.When male and female UC patients were compared with their corresponding controls respectively,nevertheless,each haplotype frequency was not statistically different (all P > 0.05).Conclusions Foxp3 (rs2232365,rs2294021,rs3761548) variations might engender the increased risk of severe UC in Chinese Han patients.
8.Expression of mic2/CD99 protein and their correlation with Eber-1/LMP-1 in Hodgkin and Reed-Sternberg cells of classical Hodgkin's lymphoma

AIM: To study the expression of mic2/CD99 protein and their correlation with Eber-1/LMP-1 in Hodgkin and Reed-Sternberg cells of classical Hodgkin's lymphoma. METHODS: Immunohistochemical staining, in situ hybrization and tissue microarry technique were used to detect the expressions of mic2/CD99 and Eber-1/LMP-1 of H/RS cells in 43 cases of cHL and 16 cases of NHL. RESULTS: The positive rate of CD99 protein expression in 43 cases of cHL was 2.3% (1/43) , mic2 was 55.8% (24/43), LMP1 was 58.1% (25/43) and Eber-1 was 53.5% (23/43). The expressions of CD99 and mic2 in the NHL group were higher than those in cHL group (P0.05). There was a negative correlation between the expression of CD99 protein and LMP1 in H/RS cells (P0.05). There was a significant correlation between the high expression of LMP1 and a low expression of CD99 in the young patients (P0.05). CONCLUSION: There is a negative correlation between the expression of LMP1 and CD99 in Hodgkin and Reed-Sternberg cells of classical Hodgkin's lymphoma.
9.Expression of matrix metalloproteinase-2,cysteine rich protein61 and CD25 In esophageal carcinoma and their significance

ObjectiveTo determine the expression of matrix metalloproteinase-2 (MMP-2),cysteine rich protein 61 (Cyr61) and CD25 in esophageal carcinoma and investigate their clinical significance.MethodThe expression of MMP-2, Cyr61 and CD25 in 68 cases of esophageal tumor tissues (esophageal carcinoma group ) and part-cancer( para-cancer group ) and 68 cases of reflux esophagitis (control group ) was detected by immunohistochemical method and the results were statistically analysed. ResultsThe positive expression rates of MMP-2,Cyr61 and CD25 were 82.35%(56/68), 73.53%(50/68) and 89.71%(61/68)respectively in esophageal carcinoma group,there were significant differences compared with those in paracancer group and control group(P < 0.05) ; while there was no significantdifference between para-cancer group and control group (P > 0.05 ). The positive expression rates of MMP-2, Cyr61 and CD25 in esophageal carcinoma group in different T stage, N stage and M stage all had statisticall differences (P < 0.05),while there was no significant difference in different degree of differentiation (P > 0.05 ). ConclusionMMP-2,Cyr61 and CD25 is highly expressed in esophageal carcinoma,there is some significance for understanding the occurrence, development and prognosis of esophageal carcinoma.
10.Down-regulation of monoamine neurotransmitter synthetase expression involved in CUS-induced depression of rats

Aim To investigate whether chronically un-predictable stress (CUS)-induced depression-like be-haviors of rats is associated with the variant expression of tryptophan hydroxylase (TPH)and tyrosine hydrox-ylase (TH).Methods 30 male SD rats were ran-domly divided into depression model group(MG)and control group (CG),the former was established using CUS plus solitary condition for 28 d,whereas the latter was fed normally as five rats per cage without CUS. The open field test(OFT)and the sucrose preference test were used to evaluate depressive behaviors.Both mRNA and protein expressions of TPH and TH in hip-pocampus and forebrain cortex were determined by re-al-time fluorescent quantitative PCR and western blot (WB),respectively.Results MG rats showed obvi-ous depressive behaviors with much lower locomotive activity and sucrose preference than CG.Meanwhile, the mRNA and protein expressions of TPH and TH also significantly decreased in MG rats,compared with CG rats.Conclusion The depression behaviors of rats in-duced by CUS may be associated with down-regulation of TPH and TH expression.
