2.Morphology of rat testis preserved in three different fixatives.

Histopathological examination of testes is important in assessing spermatogenesis and testicular function. Modified Davidson's fluid (mDF) has been proposed as a superior substitute for Bouin's fluid (BF) for fixation of adult animal testes. Besides, 4% paraformaldehyde (PFA) has been commonly used to fix testes with convenience. We compared the morphology of the rat testis fixed in 4% PFA, mDF, or BF using hematoxylin and eosin (HE)-stained sections. Fixation in 4% PFA resulted in obvious tissue shrinkage artifacts, especially between seminiferous epithelium cells. Shrinkage artifacts were also observed in the central area of the testes fixed in BF. Use of mDF did not cause shrinkage artifacts between seminiferous tubules, though a small amount can be observed in seminiferous tubules between germ cells. Clarity of nuclear detail in testes fixed in mDF and BF is better compared to 4% PFA. Our study demonstrated that fixation in mDF provided better morphologic details in the rat testis as compared with 4% PFA and BF.
3.Investigation of the demands of chemometrics guidelines in China’s pharmaceutical industry

Objective: To understand and grasp the current situation and actual demands of China’s pharmaceutical industry, in order to provide a basis for the establishment of the chemometrics guideline in the Chinese Pharmacopoeia.
Methods: Online questionnaire was adopted to investigate the chemometrics backgrounds and demands of pharmaceutical industry practitioners.
Results: The practitioners of China’s pharmaceutical industry had certain expectations and demands for the chemometrics guideline, but the situation of talent reserves was not that optimistic.
Conclusions: It is extremely urgent to develop the chemometrics guideline, as general chapters of the Chinese Pharmacopoeia, which serve as the legal basis to guide the data quality control, the establishment of analytical methods and the verification of analytical methods in analytical practice, so as to ensure the scientific of multivariate analysis methods and reliability of analytical results, and will be conducive to promoting the improvement of China’s pharmaceutical level.
4.Analysis of lung function of patients with leukemia before and after transplantation of stem cells

Objective To explore the changes in pulmonary function of patients with leukemia before and after stem cells transplantation(SCT).Methods Twenty-three patients with leukemia were assigned to group A,while 35 healthy controls to group C.All spirometric values were recorded including percentage predicted forced vital capacity(FVC),residual volume(RV),total lung capacity(TLC),forced expiratory volume in one second(FEV1),maximal expiratory flow in 50% and 75% vital capacity(MEF50%,MEF75%),diffusing capacity of the lung for carbon monoxide(DLCO),and ratio of carbon monoxide diffusion capacity to alveolar ventilation(DLCO/VA).Twenty patients with leukemia undergone stem cell transplantation were assigned to group B.Changes in lung function 7 days before and 3-12 months after transplantation were recorded.Group B was sub-grouped according to the source of stem cells and extent of anemia.Changes in lung function and infection were observed subsequently.Results Patients in group A experienced a significant decrease in predicted percentage of DLCO,DLCO/VA and MEF75%(76.25%?22.48%,83.77%?21.32% and 61.13%?19.08%,respectively),which were significantly different compared with that in group C(118.21%?19.34%,112.4%?20.96% and 81.02%?12.10%,P
5.Cognitive function in women with hypertensive disorders complicating pregnancy:A follow-up study

Objectives: Hypertensive disorders complicating pregnancy(HDCP) has a long-term deleterious effect.This study aimed to investigate the changes of cognitive function in women with HDCP and explore their possible mechanism.Methods: Fifty-three patients with HDCP and 22 normal pregnant women were included in this study,their cognitive functions assessed 60-80 days after delivery by word learning and delayed recall test,symbol digit substitution test,animal category fluency test,block design,trail making test(Part A) and digit span test.The results of the cognitive tests were compared between the HDCP and normal control groups.Results: Cognitive performances of the severe preeclampsia patients differed significantly from those of the normal individuals on the tests of word learning(P
6.Fluorescence PCR for detecting ureA gene of Helicobacter pylori in feces

Objective To evaluate the application of fluorescence PCR in detecting ureA gene of Helicobacter pylori(HP)in feces.Methods Fluorescence PCR was used to detect ureA gene of HP in feces from 50 patients,including 23 confirmed by gastric biopsy urease test and histological staining.Bacterium culture and serum antibody detection were also performed, and chi-square test was used to compare the sensitivity,specificity,positive predictive value and negative predictive value among three methods.Results The sensitivity,specificity,positive predictive value and negative predictive value of fluorescence PCR were 1.00,0.96,96.O%and 100.0%,while those for HP culture were 0.78,1.00,100.0%and 84.0%,and thee for serum antibody detection Was 0.96,0.74,76.O%and 95.0%.There were significant differences in sensitivity and negative predictive value between PCR and bacterium culture (X2=5.60 and 4.44,P<0.05),and significant differences in specificity and positive predictive value between PCR and serum antibody detection(X2=5.28 and 4.08,P<0.05).Conclusion ureA gene detection in feces by fluorescence PCR is of value for the diagnosis of HP infection.
7.The treatment of acute arterial ischemia in the extremities:report of 148 cases

ObjectiveTo sum up the experience in the treatment of acute arterial ischemia in the extremities. Methods From 1980 to 2001,148 patients with acute arterial ischemia in the extremities were treated by multiple-means such as: embolectomy, interventional treatment, thrombolytic and antiagglutinatives. Results The cure rate in patients treated within 12 hours was 95.5%,mortality was 4.5%,while the cure rate, alleviative rate, amputation rate and mortality in patients treated 12~24 hours after onset were 64.8%,17.6%,9.9%,7.7%,respectively and that were 20%,34.3%,25.7%,20% respectively when treatment started 24 hours after the onset. The cure rate in 19 patients treated by nonoperative means was 10.5%, alleviative rate was 73.3%, amputation rate was 15.8%. Conclusion Patients with acute arterial ischemia suffer a high mortality. Mortality and disability rate can be reduced by early diagnosis, appropriate treatment and effective management for the systemic diseases.
8.Expression of SOX2 and TGF-β1,Smad3 in gastric carcinoma and their relation

Objective To investigate the expression of SOX2,TGF-β1 and Smad3 in gastric cancer and their relationships with clinical pathological characteristics of gastric cancer. Methods Immunohistochemical staining was used to detect the expression of SOX2, TGF-β1 and Smad3 expression and the clinical pathological characteristics of gastric cancer were analyzed. Results Integrated optical density (IOD) values from the positive expression of SOX2, Smad3 in gastric cancer group were all significantly lower than normal gastric tissues (P < 0.05). IOD values from the positive expression of TGF-β1 in gastric cancer group were all significantly higher than normal gastric tissues (P < 0.05). IOD values from the positive expression of TGF-β1 in lower poorly differentiated gastric cancer tissues, with lymph node metastasis and Ⅲ+Ⅳ were all significantly higher (P < 0.05). Higher IOD values from expression of SOX2 and Smad3 were found in high-moderated differentiated gastric cancer tissues, without lymph node metastasis and Ⅰ+Ⅱgastric cancer (P < 0.05). SOX2, TGF-β1 and Smad3 in gastric cancer has nothing to do with age illness and sex (P >0.05). There was positive correlation between SOX2 and Smad3 expression in gastric cancer tissues, and negative correlation between TGF-β1 and Smad3 in gastric cancer. Conclusions Abnormal expression of SOX2, TGF-β1 and Smad3 protein is related to the onset, development and clinicopathologic features of gastric cancer.
9.Role of phosphatidylinositol 3-kinase/protein-serine-threonine kinases signal transduction pathway in EphB receptor-mediated neuropathic pain in rats

Objective To investigate the role of phosphatidylinositol 3-kinase (PI3K)/protein-serine-threonine kinases(Akt) signal transduction pathway in EphB receptor-mediated neuropathic pain in rats.Methods Forty-eight pathogen-free male SD rats aged 2-3 months weighing 150-180 g were randomly divided into 6 groups (n =8 each):groups sham operation (groups S1 and S2 ); groups chronic constrictive injury (CCI) (groups C1and C2 ) and groups EphBI-Fc (EphB receptor antagonist) + CCI (groups E1 and E2 ).Neuropathic pain was induced by placing 4 ligatures on left sciatic nerve at 1 mm intervals with 5-0 silk thread in groups C1,C2,E1 and E2.EphBI-Fc 0.5 μg in 5 μl normal saline was injected intrathecally 1 h before operation and at 1 and 2 d after operation (group E1 ) or on 5th day after operation (group E2).Normal saline 5 μl was injected intrathecally instead of EphBI-Fc 1 h before operation and at 1 and 2 days after operation (groups S1 and C1 ) or on 5th day after operation (groups S2 and C2 ).Pain withdrawal latency to noxious thermal stimulation (PWL) and pain withdrawal threshold to noxious mechanical stimulation (PWT) were measured before operation and at 1,3 and 5 d after operation.The animals were sacrificed at 5 d after operation after measurement of pain threshold.The lumbar segment of spinal cord (L4-6) was removed for determination of c-Fos,PI3K and phosphorylated Akt(p-Akt) expression.Results CCI significantly reduced PWL and PWT and up-regulated spinal c-Fos,PI3K and p-Akt expression in groups C1 and C2 as compared with groups S1 and S2.EphB1-Fc significantly decreased hyperalgesia and the upregulated spinal Fos,PI3K and p-Akt protein expression induced by CCI in groups E1 and E2 as compared with groups C1 and C2.Conclusion Spinal EphB receptor is involved in the development and maintenance of neuropathic pain through PI3K/Akt signal transduction pathway.
10.Association of three cytokines with graft versus host disease after allogeneic hematopoietic stem cell transplantation for leukemia

BACKGROUND: Recently, a few studies have reported the correlation between transforming growth factor-α (TGF-α) and graft-versus-host disease (GVHD); however, the combination of TGF-α with other cytokines in patients with chronic or acute GVHD requires further study.OBJECTIVE: To analyze the changes of serum tumor necrosis factor-α (TNF-α), interleukin-4 (IL-4), and transforming growth factor-α (TGF-α) in leukemic patients after allogeneic hematopoietic stem cell transplantation (Allo-HSCT) and investigate the effects of these cytokines on different grades of GVHD.DESIGN: Case control study.SETTING: Department of Hematology, Organ Transplantation Center, the Second Affiliated Hospital, General Hospital of Chinese PLA; Department of Nuclear Medicine, the Second Affiliated Hospital, General Hospital of Chinese PLA.PARTICIPANTS: Forty-two leukemic patients (23 males and 19 females, 16-68 years old, mean age of 35 years) who underwent Allo-HSCT for the first time were selected from the Department of Hematology, Organ Transplantation Center, the Second Affiliated Hospital, General Hospital of Chinese PLA and Department of Transplantation, the 307 Hospital of Chinese PLA from June 2005 to June 2007. Twelve patients had acute granulocytic leukemia (AGL), fifteen patients had acute lymphocytic leukemia (ALL), and fifteen patients had chronic granulocytic leukemia (CGL). Among the 42 patients, 37 underwent peripheral blood transplantation and five received bone marrow transplantation. Twenty-one patients had acute GVHD (18 cases in grades Ⅰ-Ⅱ and three cases in grades Ⅲ-Ⅳ) after Allo-HSCT, but the other 21 patients did not. Fourteen patients had chronic GVHD (five cases of limited type and nine cases of extensive type), but the other 28 patients did not. An additional 30 healthy subjects (18 males and 12 females, 20-70 years old, mean age of 44 years) were collected as a normal control group. All patients provided confirmed consent, and the study was approved by the local ethics committee.METHODS: Levels of serum TNF-α, IL-4, and TGF-α in leukemic patients with Allo-HSCT and normal subjects were measured by radio-immuno-assay, the cytokines levels of the patients with/without acute GVHD, of those with/without chronic GVHD and of different grades of GVHD were compared.MAIN OUTCOME MEASURES: Comparisons of serum TNF-α, IL-4, and TGF-α among the groups.RESULTS: All 42 leukemic patients and 30 healthy subjects were included in the final analysis. Levels of TNF-α, IL-4, and TGF-α in patients with acute or chronic GVHD were significantly higher than those in the normal subjects (P<0.05-0.01). Levels of TNF-α and IL-4 in patients without acute GVHD were significantly higher than those in the normal subjects (P<0.01,0.05). Levels of TNF-α, IL-4, and TGF-α in patients with acute GVHD were significantly higher than those in patients without acute GVHD (P<0.05). Levels of TNF-α, IL-4, and TGF-α in patients with chronic GVHD were significantly higher than those in patients without chronic GVHD (P<0.05). Levels of serum TNF-α and TGF-α in patients with acute GVHD of grades Ⅲ-Ⅳ or chronic GVHD of extensive type were significantly higher than those in patients with acute GVHD of grades Ⅰ-Ⅱ or chronic GVHD of limited type (P<0.05-0.01).CONCLUSION: After Allo-HSCT, dynamically monitoring changes of levels of TNF-α, IL-4, and TGF-α may serve as a possible means of predicting the onset of acute or chronic GVHD and may contribute considerably to deciding clinical severity of GVHD.
