1.Influence of COX-2 inhibitor on angiopoietin expression of human lung cancer xenografts in nude mice

AIM: To investigate the influence and significance of nimesulide (NIM), a selective cyclooxygenase 2 (COX-2) inhibitor, on angiopoietin expression of human lung cancer xenografts in nude mice. METHODS: Human A549 lung cancer cells were inoculated subcutaneously in athymic nude mice to establish xenograft models. The mice were divided into NIM group and control group randomly. Mice in NIM group and control group were administered with NIM (6 mg/kg) and saline into stomachs respectively once a day for 35 days. The inhibitory rate was calculated and expressions of angiopoietin-1, -2 (Ang-1, Ang-2) mRNA in xenografts were measured by semiquantitative reverse transcription-polymerase chain reaction (RT-PCR) analysis. Microvessel density (MVD) in xenografts was assessed immunohistochemically using antibody against FⅧ.RESULTS: NIM inhibited the xenografts growth significantly and the inhibitory rate was 43.02%. 2.Variation of blood platelet and granule membrane protein-140 in patients with type 2 diabetes mellitus

BACKGROUND: Parameters of platelet can reflect functions of coagulation. Granule membrane protein-140 (GMP-140), which is also called P-Selectin, in α-granules of platelets can reflect activities of platelet. Dysfunction of blood sugar and blood lipid in diabetic patients can result in complications such as thrombotic diseases.OBJECTIVE: To analyze the variation regulation of the platelet parameters and GMP-140 in patients with type 2 diabetes mellitus.DESIGN: A case-control study.SETTING:Department of Clinical Laboratory and Department of Endocrinology, Union Hospital Affiliated to Tongji Medical College, Huazhong University of Science and Technology.PARTICIPANTS: From March to April 2005, forty-five patients with type 2 diabetes mellitus, who were treated at the Department of Endocrinology,Union Hospital Affiliated to Tongji Medical College, Huazhong University of Science and Technology, were enrolled as diabetic group, and 45 healthy people, who were checked with health examination at the same period,were selected as control group. They all joined the experiment voluntarily. METHODS: Changes of quantity of blood platelet, mean platelet volume (MPV), platelet distribution width (PDW) and platelett-large cell ratio (PLCR) were analyzed with blood cell counter. GMP-140 was measured by enzyme-linked immunosorbent assay (ELISA), meanwhile, compared with those of the healthy people.MAIN OUTCOME MEASURES: Contents of the parameters of platelets and GMP-140 of subjects in the two group s.RESULTS: A total of 90 cases without drop out, 45 cases in the diabetic group and control group, respectively, were involved in the result analysis.①Comparison of the content of GMP-140 in the two groups: It was significantly higher in the diabetic group than that in the control group [(28.8±10.3) ,(9.1±5.6)μg/L(t=2.37,P < 0.01 )]. ②Comparison of parameters of blood platelet of subjects of the two groups: The MPV, PDW and P-LCR in patients of the diabetic group were markedly higher than those in the control group (t=2.02, 2.02, 2.02, P < 0.01 ), while the blood platelets (PLT)was dramatically lower than that in the control group (t=2.15, P < 0.01 ).CONCLUSION: The MPV, PDW, P-LCR and the content of GMP-140 of patients with type 2 diabetes mellitus significantly increase, while the PLT decreases, which indicate that platelet activity enhances and the consumption of platelet increases in patients with type 2 diabetes mellitus.
3.Antisense oligonucleotide of hypoxia-inducible factor-1alpha suppresses growth and tumorigenicity of lung cancer cells A549.

To study the role and mechanisms of hypoxia-inducible factor-1alpha (HIF-1alpha) on the growth and tumorigenicity of lung cancer cells A549, the antisense oligonucleotide of HIF-1alpha was transfected to A549 cells. The effect of the antisense oligonucleotide on tumor growth in vitro and in vivo was evaluated by the growth rate suppression of A549 cells and subcutaneous implanted tumor in nude mice, and the effect on tumorigenicity was evaluated by the expression inhibition of angiogenic factors, the microvessel density (MVD) and vascular endothelial growth factor (VEGF) protein expression which were detected by immohistochemistry and western blot respectively. This study revealed that in vitro the growth rate of antisense oligonucleotide group was significantly decreased as compared with that of control group, sense oligonucleotide group and false-sense oligonucleotide group; in vivo the weight of implanted tumors in nude mice of antisense oligonucleotide group was 1.51 +/- 0.40 g, which was significantly lower than that of control group (2.79 +/- 0.33 g), sense oligonucleotide group (2.81 +/- 0.45 g) and false-sense oligonucleotide group (2.89 +/- 0.39 g) and the inhibitory rate was 47%. Both MVD and VEGF protein expression were significantly inhibited in antisense oligonucleotide group compared with those in other groups. These results indicated that antisense oligonucleotide of HIF-1alpha could inhibit lung cancer cells A549 growth in vitro and in vivo, and the mechanism may be due to the inhibition of vascular growth and VEGF protein expression.
4.Feasibility of umbilical cord blood stem cell transplantation for the treatment of diabetic lower limb ischemia

BACKGROUND:Diabetic lower limb ischemia is prone to involve distal lower limb arteries, and a conventional treatment is often unable to obtain the ideal effect. OBJECTIVE:To investigate the effect and safety of umbilical cord blood stem cel transplantation in the treatment of diabetic lower limb ischemia. METHODS: A diabetic rat model of lower limb ischemia was established, and along the femoral artery, five points
were selected for injection of human umbilical cord mesenchymal stem cel suspension, 20 μL per point. At 1, 2, 4 weeks after transplantation, transcutaneous oxygen pressure, vascular density and vascular endothelial growth factor level in the ischemic region, and incidence of adverse reactions were recorded. RESULTS AND CONCLUSION:At 1, 2 and 4 weeks after transplantation, the transcutaneous oxygen pressure, vascular density and vascular endothelial growth factor level in the ischemic region were found increasing, which were significantly different from those before transplantation (P < 0.05). At different time after transplantation, al animals had no inflammatory reactions such as skin bleeding and dermatitis, and local red, sweling, hot, pain, and had no tumor-like growth in organs. These findings indicate that umbilical cord blood stem cel transplantation can safely and significantly improve symptoms of diabetic lower limb ischemia, which has certain application feasibility. 6.Application of E-flow in ultrasound examination of fetal pulmonary vein

Objective To explore the value of E-flow technique in ultrasound examination of fetal pulmonary vein.Methods The pulmonary veins of 332 fetus were examined using lungs as acoustic window by two-dimensional echocardiography(2DE), color Doppler flow imaging (CDFI) and E-flow.Visualization ratios of the three methods were compared.Results The fetal pulmonary veins were displayed as following:7 cases were showed by 2DE,49 cases by CDFI and 73 cases by E-flow among 118 cases during 12 - 22 gestational weeks.The visualization ratio of E-flow was higher than that of the other two methods (P <0.001).Fourty-one cases were showed by 2DE, 171 cases by CDFI and 177 cases by E-flow among 214 cases during 23 - 40 gestational weeks.The visualization ratios of E-flow and CDFI were higher than that of 2DE (P < 0.001).The visualization ratio of E-flow and CDFI has no distinct differrence (P > 0.0167).Conclusions E-flow was sensitive to pulmonary vein blood flow and better than 2DE and CDFI in early stage of the second trimester.
7.Expression and significance of cyclooxygenase 2 gene in lung cancer.

To study the expression of cyclooxygenase 2 (COX-2) gene and its relationship with clinicopathological characteristics of lung cancer, expression of the COX-2 mRNA was evaluated by reverse transcription polymerase chain reaction (RT-PCR) in cancerous tissues and paired adjacent non-cancerous tissues from 56 patients and benign lesions from 12 patients. Our results showed that expression of COX-2 gene was detected in a significantly greater proportion of cancerous tissues (60.7%) than adjacent noncancerous tissues (10.7%, P<0.01) and benign lesions (3/12, P<0.05). Expression of COX-2 gene was higher in adenocarcinoma than in squamous carcinoma (P<0.01). There was no significant relationship between COX-2 gene expression and patients' age, sex, histological type of tumors, differentiation degree and TNM stages (P>0.05). The up-regulation of COX-2 gene in lung cancer tissues especially in adenocarcinoma suggested that COX-2 may play a role in the lung carcinogenesis and COX-2 gene may serve as a potential therapeutic target in lung cancer.

8.Effects of glucocorticoid on IL-13-induced Muc5ac expression in airways of mice.

To study the effects of glucocorticoid on the IL-13-induced Muc5ac expression in airways of mice, and investigate its role in mucus secretion of airways, 24 pathogen-free BALB/c mice were randomly divided into 3 groups. IL-13 group received an nasal instillation of 100 microg of recombinant murine IL-13 solution on days 1, 3 and 5. In dexamethasone group, dexamethasone (0.5 mg/kg) was administered intraperitoneally 24 h before and 1 h before the first instillation of IL-13 and on 4 consecutive days (day 0 to day 5, 6 consecutive days in total), while control group was not treated with IL-13 or dexamethasone. Bronchoalveolar lavage fluid (BALF) was collected and eosinophils were counted, and expression of Muc5ac mRNA and protein in lungs were detected by reverse transcription-polymerase chain reaction (RT-PCR) technology and immunohistochemical assay respectively. Our results showed that the number of mice, with positve Muc5ac protein expression, expression of Muc5ac mRNA and eosinophils in BALF after IL-13 treatment were all significantly higher than that of control group (all P<0.01). Despite eosinophils reduced (P<0.01), the number of mice with positive Muc5ac protein expression, expression of Muc5ac mRNA afterdexamethasone treatment didn't decreas significantly as compared with that of IL-13 group. It is concluded that IL-13 can up-regulate the expression of Muc5ac mRNA and protein, which may play a pivotal role in the mucus overproduction of airways. Dexamethasone can suppress IL-13-induced eosinophilic infiltration in lung but can't inhibit the mucus overproduction.

9.Diagnosis of infectious mononucleosis by combined detection of atypical lymphocytes and transaminase.

In order to explore the value of combined detection of atypical lymphocytes (ATL) and transaminase (alanine aminotransferase, ALT; asparate aminotransferase, AST) in the diagnosis of infectious mononucleosis (IM), The data of blood routine and liver function were collected from 54 IM patients, 34 acute hepatitis (AH) patients, 44 upper respiratory infection (URI) patients in Union Hospital during March 2002 to March 2005. Same data were also collected from 40 healthy children as normal control. These data were analyzed retrospectively. Both proportion of atypical lymphocytes and enzyme activity of transaminase were elevated simultaneously (ALT > 40 IU/L, AST > 45 IU/L) in 57.4% (31/54) IM patients. There was significant difference (P < 0.01) between IM group and the other groups. Combined detection of atypical lymphocytes and transaminase can be regarded as a diagnostic marker of infectious mononucleosis.
