1.CARM1 is required to maintain stemness of amniotic fluid-derived stem cells

BACKGROUND:Studies have shown that methylation modification using CARM1-catalyzed histone H3R17/R26 can maintain the stemness of embryonic stem cel s. However, mechanism underlying CARM1 effect on the stemness of amniotic fluid-derived stem cel s is stil unclear.
OBJECTIVE:To investigate the function and underlying molecular mechanism of CARM1 to maintain stemness in the amniotic fluid-derived stem cel s.
METHODS:Amniotic fluid-derived stem cel s from term pregnancy were isolated and cultured. RT-PCR was used to identify the stem cel mark and CARM1 gene expression. CARM1 expression in amniotic fluid-derived stem cel s was knocked down by using two shRNA. RT-qPCR was used to detect the silencing efficiency, and western blot employed to examine the methylation level of Arginines 17 at N terminus of histone 3 (H3mR17). Moreover, the expression of embryonic stem cel markers, including OCT4, SOX2 and NANOG, were detected.
RESULTS AND CONCLUSION:Amniotic fluid-derived stem cel s from term pregnancy could express CARM1 and stem cel markers, including OCT4, SOX2, Nanog and KLF4. Both of the shRNAs could knock down the expression of CARM1 efficiently. When CARM1 was knocked down, the H3mR17 level was decreased and OCT4, SOX2 expression was also reduced, but NANOG expression had no change. Al these indicate that CARM1 is required for amniotic fluid-derived stem cel s to maintain stemness through regulating OCT4 and SOX2 expression.
2.Effect of lifestyle intervention on high-risk adults for hypertension

ObjectiveTo evaluate the effect of lifestyle intervention on high-risk individuals for hypertension. MethodsOne hundred and four prehypertensive adults who received physical examination during October 2005 and October 2007 and showed at least 2 hypertension risk factors were randomly assigned to the experimental or control group.A muhicomponent behavioral intervention,including health education(e.g.awareness of disease),metary gmdance,physical.exercise,and psychologtcat improvement,Was conducted for 2 years.The awareness of disease,blood pressure,bedy weight,and the change of risk factors were compared.ResultsAt 2 years,the awareness of disease in the experimental group wag significantly improved,and the blood pressure,body weight,and risk factors were obviously reduced.Although the awareness of disease and the risk factors in the control group were improved,the blood pressure and body weight were increased.The difference in average systolic blood pressure,average diastolic blood pressure,body weight,and the awareness of disease between the two groups were 13 mm Hg(1 mm Hg=0.133 kPa),7.9 nlnl Hg,3.7 kg,and 37.6%,respectively(all P<0.05).ConclusionThe lifestyle intervention among high-risk individuals for hypertension could effectively reduce blood pressure and body weight. Improved lifestyle would result in less risk factors.
3.Analysis of infected characters and its related factors of nosocomial infection in children leukemia

Objective To explore the related factors of nosocomial infection in children with leukemia and its character.Methods Retrospective analyzed 96 leukemia children with nosocomial infection who were come from 220 patients with acute leukemia.The information of the infection rate at different chemotherapy stages,different hospitalization time,site of infection and strain condition were recorded.Results (1) The infection rate of leukemia children at induction and consohdation stage were 85.7% (24/28) and 76.7% (23/30) respectively.(2) The infection rate increased with the duration of hospitalization periods.The infection rate was 8.6% (3/35) at 0-7 d hospitalization periods and 62.7% (42/67) at 15-30 d hospitalization periods.(3) The infection was prone to occurring at respiratory tract (58.3%,56/96),followed by gastrointestinal tract (17.7% (17/96)),blood (13.5% (13/96)),skin (8.3% (8/96)) and other parts (2.1% (2/96)).(4) The most frequency stain listed as Escherichia coli(21.8% (26/96)),and Citrobacter (16.8% (20/96)).The lower detection rates of the bacteria listed as Candida albicans (2.5% (3/96)),Aspergillus(1.7% (2/96)) and yeast (0.8% (1/96)).Condusion Even though the survival rates of leukemia children is increasing due to technology development,nosocomial infection frequently occur.The high infection rates occur at chemotherapy induction and consolidation phase.Therefore intervention should be taken based on nosocomial infection in children leukemia such improving the environment of ward,enhancing the immune ability of children in order to decrease the incidence of hospital infection.

5.Preparation and Quality Control of Qingyan Tea Bag

OBJECTIVE:To prepare Qingyang tea bag and establish the standard of quality control.METHODS:The Flos Lonicera and Radix Glycyrrhize were identified by TLC.The content of chlorogenic acid,the main active component in the tea derived from Flos Lonicera,was determined by TLCs.RESULTS:Linearity was found in the range from1.2?g~4.8?g of chlorogenic acid,r=0.9993;the average recovery was99.50%and RSD=2.39%.CONCLUSION:The method was found to be simple,precise and may be used for the quality control of the preparation.
6.The relationship of serum C-peptide levels and microvascular complications in type 2 diabetes mellitus

Objective To evaluate the relationship between serum C‐P levels and microvascular complications in patients with T 2DM. Methods The clinical data of 434 patients with T2DM were retrospectively analyzed. All subjects were divided into three groups based on ΔC‐P (2 hC‐P - FC‐P) tertiles :ΔC‐P1(≤2.2 ng/ml) ,ΔC‐P2(2.3~4.0 ng/ml) and ΔC‐P3(≥4.1 ng/ml). Results The levels of BMI ,TG ,FC‐P were lower ,and the course and HbAl c were higher in ΔC‐P1 group. ΔC‐P level was positively associated with BMI and TG (P< 0.05) ,and negatively associated with prevalence of DR and chronic kidney disease (CKD)in diabetes ,course and HbA1 c.ΔC‐P levels decreased gradually with the progression of DR and CKD. Logistic regression analysis showed that lower ΔC‐P level was the independent risk factor for microvascular complications after adjustment for related risk factors. Conclusion Serum ΔC‐P level is an independent factor for the development of diabetic microvasculr complications in T 2DM.
7.Clinical analysis of 179 patients of community-acquired pneumonia in out-patients

Objective To analyse the clinical features of community-acquired pneumonia(CAP)in out-patients.Methods A retrospective study was performed on 179 consecutive patients with CAP in fever clinic of Xuanwu Hospital during two months.Results Most of CAP patients were young(aged 14 to 30 years,accounting for 58.1%)and had no-existing diseases(72.6%).Within 2 to 4 days after onset of fever,50%~80% of patients were diagnosed as CAP.About 1/3 of patients had no respiratory symptoms except for fever.Cough and sputum production was seen in 52.5% of patients.WBC was normal in 63.1%.Unilateral pneumonia often occurred(accounting for 82.1%).After treatment with antibiotics,fever in 80%(42/52)of cases disappeared within 3 days in follow-up patients.Conclusion In fever clinic,most patients are young and have no-existing diseases.They can be identified as CAP quickly and have good outcomes.

Long non-coding RNA (lncRNA) widely exists in living organisms.It,whose length is more than 200 bp,can regulate the genic expression in the epigenetic,transcriptional and post-transcriptional level.Therefore,it can influence cell proliferation,apoptosis,vitality,immune response and oxidative stress.In recent years,the study of lncRNA is developing rapidly in the ophthalmic research,and this article reviews the latest research progress on mechanism of lncRNA in the ophthalmology disease.
9.Antioxidant and immunoregulatory effects of Centella asiatica extracts

AIM: To determine the antioxidant and the immunoregulatory effects of Centella asiatica extracts. METHODS: Centella asiatica was extracted with alcohol and different organic solvent. The content of polyphenol was determined by Folin-ciocalteau method. The efficacy of the extracts to scavenge the hydroxy radical (OH·), 1,1-diphenyl-2-picryl-hydrazyl radical (DPPH·) and intra-cellular reactive oxygen species (ROS) was measured. Lymphocyte proliferation was assessed to observe the influence of Centella asiatica extracts. RESULTS: (1) Centella asiatica was found to have abundant polyphenol extrated from different phases and in a descending order as follows: ethyl acetate extracts>n-butanol extracts>water extracts>ligarine extracts. (2) Extracts of Centella asiatica exhibited the scavenging efficacy of OH· and DPPH· free radicals, in which the acetic ether extracts showed the significant effect. (3) The acetic ether extracts had significant ability to inhibit the generation of ROS in stimulated lymphocytes. (4) The acetic ether extracts suppressed the lymphocyte proliferation. (5) The active ingredient was identified as flavone. CONCLUSION: Flavones in the Centella asiatica posseses antioxidant activity and effectively inhibits lymphocyte proliferation, showing ability of immunosuppre-ssion.
10.Expression of VEGF and its correlation with clinicopathological factors of cervical carcinoma ABSTRACT

Objective This study was designed to examine the expression of VEGF and its correlation with clinicopathological factors of cervical carcinoma. Methods mmunohistochemistry S-P method was used to detect the expression of VEGF in 10 cases of nomal cevical epithelium( NCE), 18 cases of cervical intraepithelial neoplasm (CIN) and 77 cases of early invasive carcinoma of cervix (ICC). Results The positive rate of VEGF protein in NCE,CIN,ICC was 10% (1/10) ,44.4% (8/18) ,74. 0% (57/77), respectively. The defference was significant (P <0. 05). Expresson of VEGF was not related with tumor stage, histologic types and stroma infiltration, but related with cell differentiation and intravascular involvement、lymph node metastasis. Conclusion VEGF protein had a positive correlation with cell differentiation and intravascular involvement 、 lymph node metastasis. the expression of VEGF gene result in the occurrence and development of cervical carcinoma by inducing the angiogenesis.
