1.Current study of hypercoagnlability in multiple myeloma
Journal of Leukemia & Lymphoma 2011;20(11):691-694
Mutiple myeloma(MM)has been associated with the hypercoagulable states and the development of venous thromboembolic events(VTE). Described pathogenetic mechanisms that are not exclusive to MM include the abnormality of blood clotting components, the dysfunction of fibrinolysis, the disorder of platelets, the abnormity of blood stream and blood viscosity,etc..It has been proved that high levels of vascular endothelial growth factor (VEGF),von Willebrand factor (vWF),increased production of inflammatory cytokines and higher expression of COX-2 in MM seem to be associated with the hypercoagulability. In addition, the treatment of MM may also result in the treatment-associated hypercoagulable states.

Objective: To study the influence of emodin on fluorouracil(5-FU) induced apoptosis by elevating reactive oxygen species(ROS)of human hepatocellular carcinoma cell lines HepG2. Methods: The intracellular ROS levels were detected when exposed to various concentrations of emodin for the indicated periods by flow cytometry(FCM).MTT assay and FCM were used to assess cell viability, apoptosis and cell cycle distribution after treatment with 5-FU alone or combined with various concentrations of emodin.Transmission electron microscope was applied to observe the ultrastructural change of cells. Results: Emodin elevated intracellular ROS level in a dose-and time-dependent manner, and intracellular ROS level of HepG2 cell incubated in 4 ?g/ml emodin reached peak in 2-4 h.Emodin combined with 5-FU sensitized HepG2 cell to 5-FU induced apoptosis via generation of ROS in a time-and dose-dependent manner. Conclusion: Enhancement of ROS generation by emodin may be a novel strategy to improve the curative effect of chemotherapeutic drugs.

Objective To evaluate the quality of clinical trials about acupuncture and moxibustion treatment of vertebrobasilar insufficiency in China. Methods Literature of clinical trials of acupuncture was collected and analyzed, evaluated by the randomized controlled test criteria and the clinical assessing method in Cochrane handbook of international Cochrane cooperation net. Results Sixteen literatures were brought in.87.5% of the included literature had no clear description of random methods, and no allocation concealment, use of blind methods, sample size calculation,intention-to-treat analysis.Among them,62.5% had explicit diagnosis criteria,12.5% with correct randomizing method,62.5% with explicit evaluation criteria,however,31.22% with no words referring to the comparability among the groups. Conclusions Though acupuncture and moxibustion have been widely applied in prevention and treatment of vertebrobasilar insufficiency,it can not provide evidences of higher reliability for clinical treatment due to less clinical randomized controlled tests and lower quality, which severely hinder testing and verifying of clinical therapeutic effects of acupuncture and moxibustion. It is proposed that multiple central and randomized controlled test should be made,so as to search for feasible acupuncture and moxibustion methods with definite therapeutic effect for vertebrobasilar insufficiency, and provide basis for further systematical evaluation.

Objectives To explore the changes of serum interleukin-33 (IL-33), IgA anti-endothelial cell antibodies (AECA) and IgA anticardiolipin antibodies (ACA) in acute stage of Henoch-Sch?nlein purpura (HSP) in children and its clinical signiif-cance. Methods Thirty-seven children with acute HSP admitted to hospital were selected from Nov.2012 to Jan.2013. Twenty healthy children were selected as healthy controls. The serum levels of IL-33 were measured by double anti-body sandwich en-zyme-linked immunosorbent assay (ELISA) and the serum levels of AECA-IgA and ACA-IgA were detected by double antigen sandwich ELISA. Results Serum levels of IL-33, AECA-IgA and ACA-IgA in patients with acute HSP were signiifcantly higher than those in healthy controls (P<0.001). According to different clinical types, the serum levels of IL-33, AECA-IgA and ACA-IgA were signiifcantly higher in mixed-type HSP (with joint involvement and gastrointestinal symptoms and/or kidney damage) than those in HSP patients with only joint, abdomen or renal involvement (P<0.05). Serum levels of IL-33 in acute stage of HSP had a positive correlation with AECA-IgA (r=0.752, P<0.001) and ACA-IgA (r=0.788, P<0.001). Conclusions The immune pathological injury in vessels caused by the interaction of IL-33, AECA-IgA and ACA-IgA may be the important pathogenesis of HSP, which may provide a clue for the early diagnosis and treatment for HSP.

Objective: To establish the permanent tooth germ missing animal model for future research on the root resorption of deciduous tooth in the absence of permanent tooth germ. Methods: The permanent tooth germ missing animal model was established by surgical removal of the permanent tooth buds in a male 11-week-old Beagle dog. Root resorption of the deciduous teeth without permanent successors was observed by taking periapical films periodically, and compared with physiological root resorption. Once the sign of root resorption of the deciduous teeth without permanent successors was detected on radiographic films, the animal was sacrificed and the mandibular bone was collected for histological study. Results: Root resorption of the deciduous teeth with the presence of permanent tooth germ started at 20 weeks after birth, while root resorption of deciduous teeth without permanent tooth germ started 26-27 weeks which was significantly delayed. Histological studies showed that a large number of multinucleated giant cells were present on the pulpal surface of the root, while only few of them were seen on the outer surface. Conclusion: The permanent tooth germ missing animal model was successfully established in this study which simulated the case of congenital absence of permanent tooth germ in human. Root resorption of deciduous tooth without permanent tooth germ was significantly delayed than the deciduous tooth with permanent tooth germ.
