1.Construction of expression vector of follicular inhibin and HBsAg fusion gene and expression in Hela cell

Objective:To construct inhibin expression vector with high immunogenicity.Methods:S gene with enzyme site sequence of EcoR I and Nde I was amplified by PCR from pCMV-S;Inhibin(1-32)gene with enzyme site sequence of Nde I and Hind III was cut from pUI;S and inhibin(1-32)gene were directly inserted into pcDNA3.1(-)and the recombinant pCIS was identified by restriction endonculease digestion and sequencing;then pCIS was transfected into HeLa cells.SDS-PAGE and ELISA was employed to detect the cell product.Rats were immunized with inhibin fused expression plasmid pCIS and ELISA was used to detect the blood antibody against inhibin.Results:The recombinant eukaryocyte expression vector of inhibin was constructed.The sequence of the insert was identical to designed gene.Fusion protein weight was about 29 kD and it had inhibin immunologic activity.Rats immunized with pCIS induced antibody against inhibin.Conclusion:The fusion gene expression vector was successfully constructed,and it set up the basis of inhibin gene immunization to induce multiple bear for single birth animals.
2.Various infected markers in early diagnosis of blood bacterial infections

Objective To assess the early diagnostic value of white blood cell count (WBC), neutrophil percentage (NEU%), C-reactive protein (CRP) and procalcitonin (PCT) in blood bacterial infection.Methods Clinical data of 114 patients with bacterial blood stream infections(BSI group) and 247 patients without bloodstream infections (control group) admitted to Taiyuan Third People''s Hospitalin 2015 were retrospectively analyzed.The WBC, NEU%, CRP and PCT were measured in all patients.Receiver opearating characteristic curve (Roc) was used to evalute the accuracy of WBC, NEU%, CRP and PCT in the diagnosis of blood stream bacterial infectious.Results There were no significant differences in gender and age between two groups (χ2=0.7731, t=0.9900, both P>0.05).The WBC, NEU%, CRP and PCT levels in BSI group were significantly higher than those in control group (all P<0.05).There were no significant differences in WBC, NEU% and CRP levels between G+ bacterial and G-bacterial infections in bacteremia group (P>0.05), while there was significant difference in PCT level (P<0.05).In diagnosis of bacteremia, the area under the ROC curve of PCT(0.827) was the largest, followed by CRP (0.721), NEU% (0.677) and WBC (0.593), the differences were statistically significant(Z=2.332, 3.355 and 3.786, P<0.05 or <0.01).Conclusion WBC, NEU%, CRP and PCT levels are of certain diagnostic value for bacterial bloodstream infections, particularly the PCT.In addition, PCT can predict G+ bacterial and G-bacterial infections.
3.Application of non-contrast-enhanced magnetic resonance angiography in renal artery imaging

Objective: To discuss the effect of non-contrast-enhanced magnetic resonance angiography in renal artery imaging. Methods:Twenty patients were selected with suspected as the research object, using non-contrast-enhanced MR examination, simultaneously enhanced MR examination, and relatively non-enhanced imaging examination effect on the results for statistical analysis and processing. Results: Non-contrast-enhanced ratio comparing contrast-enhanced, not statistically significant (x2=1.058, P>0.05). Conclusion:Non-contrast MR angiography is an effective, safe, non-invasive, high accuracy of the evaluation of renal artery screening tool, worthy of clinical application.
4.Effect of intracavitary therapy on acute pulmonary embolism

Objective To explore the effect of intracavitary therapy on acute pulmonary embohsm.Methods Fifteen patients were selected as our subjects,who suffered acute pulmonary embolism and received percutaneous catheter thrombus crashing and catheter directed thrombolysis in Beijing Military.Region General Hospital from January 2009 to June 2011.Local injection of Urokinase was performed with a total amount of 500 000 U in catheter directed thrombolysis.After thrombolysis,low molecular Heparin was administered to patients for 7-10 days and oral administration of Warfarin was performed for 3-6 months.Clinical symptoms,improvement of physical signs,complications,changes of mean pulmonary arterial pressure(mPAP) and arterial partial pressure of oxygen(PO2),and the opening of pulmonary artery were recorded.Results The pulmonary arteries of the 12 patients were completely opened,and partially opened in 3 patients.The effective rate was 100% (15/15).mPAP was reduced from (40.07 ±5.97) mmHg to (20.00 ±4.66) mmHg (t =-1.128,P < 0.05),PO2 was increased from (50.26 ± 9.30) mmHg to (80.49 ± 9.04) mmHg (t =1.246,P < 0.05).Patients were followed-up for 3-6 months and no recurrence case was seen.Conclusion The interventional therapy is effective,safe and practicable in the treatment of acute pulmonary embolism.
5.Study on the Application of Chelating Resins for Removal of Heavy Mental from Extracts of Folium Ginkgo

Objective To study the feasibility of using chelating resins D751 and D403 to remove the heavy metals from the extracts of Folium Ginkgo (FG). Methods Heavy metal removal experiments were performed on automatic control platform in industry mode. With dry ointment yield,total content of flavones,comparability of HPLC and removal rate of heavy metal as the indexes,the effects of two kinds of chelating resins on removing heavy metal were compared. Result After removing heavy metal by resins D751 and D403,the loss rate of the dry extract were less than 6%,and the heavy metal contents were lower than national limits;HPLC similaritites were higher than 0.98,and the loss rates of total flavones were less than 5%. Conclusion Chelating resins D751 and D403 can be used to remove heavy metal from extracts of Folium Ginkgo.
6.CD147 affects autophagy of prostate cancer PC-3 cells

[ ABSTRACT] AIM: To study the autophagy of prostate cancer PC-3 cells induced by CD147 in vitro.ME-THODS:Themethod of amino acid starvation to induce autophagy was used.The expression of CD147 was detected by Western blotting.To study the functional effects of CD147 on autophagy in prostate cancer PC-3 cells, the down-regulation of CD147 expression was induced by the technique of RNAi.The conversion of autophagic marker protein LC3-I to LC3-II was determined by Western blotting.The cell death after starvation-induced autophagy was analyzed by trypan blue exclu-sion assay.RESULTS:The CD147 expression gradually increased in starvation-induced autophagy.The down-regulation of CD147 significantly increased the expression of autophagy-related protein LC3-II compared with control group.Mean-while, the cell death rates increased from (19.3 ±3.1)%and (22.3 ±3.5)%in control groups to (38.4 ±3.1)%in si-lencing the expression of CD147 in the PC-3 cells (P<0.05).CONCLUSION:CD147 inhibits starvation-induced auto-phgy and autophagy death in the prostate cancer PC-3 cells.
7.Review of the role of bone morphogenetic protein family and its receptors in the reproductive modulation

OBJECTIVE:Bone morphogenetic protein (BMP) plays a vital role in the prevention and treatment of skeleton diseases, recently researches on the molecular mechanism of sheep prolific FecB gene indicated that BMP and its receptors have important influence on animal follicular development. In this study the influence of different type BMPs andits receptors on the follicular development was reviewed in order to explore effective modulation on animal reproduction.DATA SOURCES: Computer was applied to retrieve Medline database on the related literatures from January 1998 to June 2005. The retrieval words were "BMP" and "BMPR" that combined respectively. Language in the articles was limited to English. Simultaneously related articles were also computer searched in China periodical full text database and Wanfang databases from January 1996 to December 2005 with the retrieval words of "BMP, BMPR", that limiting the article language to Chinese.STUDY SELECTION: At first, the document was retrieved, altogether 200 studies on BMP and its receptors were enrolled including 140 Chinese literatures and 60 English literatures.DATA EXTRACTION: these literatures were screened and 30 were included for relating to the BMP characteristic, as well as the influence of BMP and its receptors on follicular development and reproductive endocrine.DATA SYNTHESIS: Of the 30 literatures, 18 experiments discussed the function of BMP and its receptors and its signal transduction mechanism,12 were about the influence of different BMP on reproductive cell secretion, as well as receptor mutation on ovulation.CONCLUSION: BMPs family plays vital role in animal reproductive modulation, current experiments prove that the changes of signal transduction due to BMP receptor gene mutation has made breakthrough for the exploration of the prolific mechanism in sheep. Moreover studies on follicular development modulation and ovulation mechanism are liable to provide theoretical reference for the prolific modulation in sheep or other species.
8.Association between postprandial triglyceride and the severity of coronary artery disease in patients with metabolic syndrome

Objective To investigate the association between postprandial triglyceride and the severity of coronary artery disease in patients with metabolic syndrome (MS). Methods AlI of 91 patients with MS were recruited for this study. Thirty-one patients with normal fasting and postprandial triglyceride was in MS1 group, 29 patients with normal fasting triglyceride and postprandial hypertrigtyceridemia was in MS2 group, and 31 patients with fasting hypertriglyeeridemia was in MS3 group. Blood triglyceride at the time of postprandial 4 hours was measured and the quantity of coronary artery branch disease was determined by coronary angiography. The relationship between them was analyzed. Result There was significant positive correlation between the quantity of coronary artery branch disease and the level of blood triglyceride at the time of postprandial 4 hours (r = 0.42, P < 0.01 ). Conclusions It is important to detect the level of blood triglyceride at the time of postprandial 4 hours. Prompt intervention maybe decrease the incidence and mortality rate of cardiovascular disease in patients with MS.
9.Advances in research on altered cardiomyocyte autophagy and its regulatory mechanisms in the elderly

Cardiomyocyte autophagy plays an important role in maintaining normal cardiac structure and function.Recent studies have shown that cardiomyocyte autophagy is decreased in the aging heart.The expression of autophagy-related genes Atg5, Atg7 and Beclin1 decreases in the aging myocardium.Decreased cardiomyocyte autophagy in the aging heart is associated with dysregulation of phosphatidylinositol-3-kinase(PI3K)/serine-threonine kinase(Akt)/mammalian target of rapamycin(mTOR), adenosine monophosphate-activated protein kinase(AMPK)and/or the SIRT1 signaling pathways.In addition, reactive oxygen species and some neural hormonal factors such as endothelin-1 can also mediate the decrease of cardiomyocyte autophagy in cardiac aging.The regulation of cardiomyocyte autophagy may provide new strategies for the prevention and treatment of cardiomyopathy in the elderly.
10.Clinical effect of Tongxinluo capsule in the treatment of women with microvascular angina

Objective To analyze the clinical effect of Tongxinluo capsule in the treatment of female patients with microvascular angina .Methods 122 female patients with microvascular angina pectoris were selected ,and they were randomly divided into conventional group and observation group ,61 cases in each group .On the basis of the two groups in the same treatment ,including antiplatelet ,lipid,the observation group was given Tongxinluo capsule orally , treatment for 3 months.The therapeutic effect was compared between the two groups .Results The total effective rate of the observation group was 93.44%,which of the conventional group was 80.33%,the difference between the two groups was statistically significant (χ2 =4.603,P=4.603).The total effective rate of exercise electrocardiogram ( ECG ) improvement in the observation group was 91.80%, which of the conventional group was 75.41%, the difference between the two groups was statistically significant (χ2 =4.552,P=0.048).Conclusion Tongxinluo capsule in the treatment of female patients with microvascular angina pectoris has good clinical curative effect ,it can obviously improve the clinical symptoms ,promote disease recover ,which is worthy of popularizing .
