1.Advances in biological predictors of radioactive esophagitis

Radioactive esophagitis is one of the most common complications in thoracic tumor radiotherapy,biological factors such as single nucleotide polymorphisms (SNPs),miRNAs,and HIV infection may play key roles in the occurrence and development of radioactive esophagitis,and they have become an active field in protection areas of radiotherapy.We can identify the patients who may cause radioactive esophageal in high dose radiotherapy as early as possible,and modify the treatment plan to protect the esophagus.Therefore the biological factors of radioactive esophagitis are of important clinical significance.
2.Clinicopathological and immunohistochemical analysis of 71 cases of GIST

Objective To study the clinicopathological and immunophenotypical features of gastrointestinal stromal tumor (GIST). Methods A SP Immunohistochemical method was selected to detect the 71 cases of GIST for a panel of antibodies CD117, CD34, Vim, Act, S-100. Results The patient's ages ranged from 21 years to 86 years (mean 55 years) including 34 male and 37 female. Most cases were diagnosed by bellyache and alimentary tract bleeding. Tumor size varied from 0.3 cm to 23 cm (mean 5.8 cm). GIST were composed of spindle cells and (or) epithelioid cells. Various sized eosinophilic globules were observed between the tumor cells and were designated as skeinoid fibers. The positive rate by immunohistochemical methods for CD117 and CD34 were 94.4 % and 73.2 % respectively. Conclusion GIST predominantly occurred in middle aged or older patients over 40 years. The histological characters are similar to smooth muscle tumor and schwannoma. CD117 and CD34 are more specific and sensitive markers. GIST may have been derived from interstitial cell of cajal or show differentiation toward interstitial cell of Cajal.
3.Preparation and Quality Evaluation of Shenhuang Vaginal Thermosensitive in situ Gels

Objective:To prepare and evaluate Shenhuang vaginal thermosensitive in situ gels. Methods:Shenhuang vaginal ther-mosensitive in situ gels( SHV-TISG)were prepared with poloxamer 407 and poloxamer 188 as the materials. The viscosity change at different temperatures and the gelation temperature of SHV-TISG and SHV-TISG diluted by stimulant vaginal fluid were measured,the in vitro drug release of the preparation was evaluated by a membraneless diffusion method,and the safety of SHV-TISG was detected by vaginal irritation test. Results:The preparation showed the vaginal injectability and gelationability in vivo without vaginal irritation. Gel erosion was the principal mechanism of the drug release from the gels. Conclusion:The preparation of SHV-TISG is feasible in tech-nique,and the properties meet the requirements of vaginal topical use.
4.Optimization of Extraction Technology for Baiyi Rectal Solution

OBJECTIVE:To optimize the extraction technology of Baiyi rectal solution. METHODS:The effects of water amount and extraction time on extraction technology were investigated by central composite design-response surface method using OD value of berberine hydrochloride and ferulic acid and the extract yield as index. Validation test was also conducted. RESULTS:The optimal extraction technology was as 11-fold water,decocting 70 min for the first time;5-fold water,decocting 30 min for the second time. In validation test,OD value of first extraction was 0.940 0,and the bias between observed and predicted values was -2.07%;that of second extraction was 0.851 8,and the bias was -2.41%. CONCLUSIONS:The extraction technology of Baiyi rectal solution is reasonable,feasible and suitable for industrial production.
5.Clinical Effect of Four Oral Vitamins Combined with FE Complex Enzyme Rash on Recurrent Oral Ulcer and Serum Inflammatory Factors Levels

Objective:To explore the clinical effect of four oral vitamins [vitamin E (Vit E) +folic acid (FA)+ vitamin B2 (Vit B2) + vitamin B12 (Vit B12)] combined with FE complex enzyme rash on the recurrent oral ulcer (ROU) and on the serum inflammatory factors levels.Methods:126 cases with ROU in our hospital from January 2014 to February 2016 were selected as research objectives and randomly divided into two groups.FE complex enzyme was provided to the control group,while FE complex enzyme and four Vietnam (Vit E+FA+Vit B2+Vit B12) were given to the observation group.The local efficacy,long-term efficacy,serum inflammatory factor levels before and after treatment as well as the incidence of adverse drug reactions were recorded and compared between two groups.Results:Compared with the control group,the pain index was significantly decreased on 30th treating day in the observation group(P<0.01),and the average ulcer period was shorten (P<0.01).After being treated for 6 months,the overall effective rate was 95.2% in the observation group,which was significantly higher than that of the control group (81.0%,P<0.05).The serum TNF-α and IL-17 levels on the 30th treating day was significantly lower in both groups after treatment than those before treatment (P<0.01),but the serum 1L-2 level was significantly increased (P<0.01),and the improvement of each above inflammatory factors in the observation group were more significant than those of the control group (P<0.01).No significant difference was found in the incidence of adverse reactions between two groups (P>0.05).Conclusions:Four oral Vietnam combined with FE complex enzyme could promote the ulcer wound healing,reduce the pain,regulate the body to promote/anti-inflammatory factor balance and improve the long-term efficacy in the treatment of patients with ROU with high safety.

7.Composition and associated factors of radiological examination in major trauma patients

Objective To explore the determinant factors influencing the constituent parts of radiological examination in severe trauma patients so as to provide scientific basis for optimized strategy of radiological examination. Methods A prospective study was carried out from April to July 2010 in a tertiary hospital. Clinical data of 60 severe trauma patients admitted to emergency department and ICU were recorded. The type, number and site of trauma under radiological examination were described and compared among different stages of treatment. The correlation between number of radiological examinations and age, number of injured site, injury severity score (ISS), Glasgow Coma Scale (GCS), ICU stay and overall length of hospital stay were analyzed. Results (1) The majority of radiological examinations in 60 patients were radiography and CT, with a corresponding median number of 6.0(3.0~ 11.0, IQR)and 10.0(8.0 ~ 13.8, Qr) times per patient. (2) The numbers of radiography examinations requested in emergency room, emergency ICU and general ward were quite approximately equal (x2 =4.043 ,P =0. 132), while CT examinations were mainly requested in emergency room and emergency ICU (x2 = 20. 274 , P < 0. 001). (3) The numbers of radiological examinations requested for different sites of injury were quite significantly different between radiography and CT during different stages of treatment (x2 = 114.609, 75.932, P < 0.01). (4 ) The number of CT scan requested was positively correlated with number of injured site, ISS, ICU and overall length of hospital stay (r =0.273,0.369,0.523,0.417,all P <0.05). Conclusions The sum of radiological examinations in severe trauma patients was great mainly in radiography and CT, and CT was more predominantly requested. The number of CT scans examinations was positively correlated with severity of injury and length of ICU stay. Further study is warranted to optimize radiological examination in severe trauma patients.
8.Cloning, Sequence Analysis and Prokaryotic Expression ofGGPS Gene fromLepidium apetalum

This study was aimed to clone the GGPS (geranylgeranyl pyrophosphate synthase) gene from Lepidium apetalum, to analyze its sequence, and to express the protein in E.coli expression system. Specific PCR cloning primers were designed for GGPS gene from Lepidium apetalum according to the full-length sequence from a previous transcriptome sequencing project. PCR amplification was performed with this primer pair on a leaf cDNA template. TA cloning, sequencing and sequence analysis were performed.GGPS gene from Lepidium apetalum was expressed in the E.coli expression system. The results showed that the full-lengthGGPS cDNA from Lepidium apetalum was 1 146 bp coding a protein of 381 amino acids. The LaGGPS protein had an isoprenoid synthase domain. According to a phylogenetic tree constructed with multiple alignment of GGPS protein sequences from various plant species, GGPS protein from Lepidium apetalum was the closest to Arabidopsis thaliana and Sinapis alba. The prokaryotic expression vectorpET-32a-LaGGPS was also constructed successfully. The protein was expressed in E.coli BL21 strain. It was concluded that the cloning and prokaryotic expression of LaGGPS gene provided a foundation for a follow-up research of its function with protein purification and activity analysis.
9.Investigation of “practice teaching methods ”for musculoskeletal ultrasound imaging

Objective To discuss the effect of “practice teaching methods ” in musculoskeletal ultrasound diagnosis training process .Methods Thirty refresher doctors for a month with the method training .The effect training is evaluated by objective exam and questionnaire survey .Results Thirty refresher doctors received tests before and after the training .Test results showed that the accuracy after the training was significantly higher than that of before ( P<0.05 ) .Refresher doctors survey about satisfaction of the training method showed that whole satisfaction is 100%, in which 70%were very satisfied , 30%were satisfied .The self-assessment results for the improvement of the level of diagnosis musculoskeletal ultrasound showed that 45%doctors are very satisfied ,40%doctors are satis-fied, and 15%is not satisfied .Conclusions With the implementation of “teaching by practice” method, refresher doctors could master the knowledge of musculoskeletal anatomy as soon as possible , and have the ability to diagnose common musculoskeletal diseases independently .
10.Effect of Astilbin on expressions of perforin and granzyme B in activated T cells of mouse heart transplantation model with acute rejection

Objective To investigate the effect of astilbin on expressions of perforin and granzyme B in activated T cells of mouse heart transplantation model with acute rejection.Methods Cardiomyocytes of BALB/C mouse and spleen cells of C57BL/6 mouse were harvested and made into single cell suspensions.The cardiomyocytes(2?10~5 ml~(-1))as stimulators and spleen cells(1?10~6 ml~(-1)) as responsers were mixed and cultured.The model of mouse heart transplantation with acute rejection in vitro was therefore established.There were two groups in the experiment.Control group is the mixed culture of the cardiomyocytes and spleen cells;Astilbin group is the mixed culture of the cardiomyocytes and spleen cells with astilbin(15?g/ml).Apoptosis of T cells were analyzed by TUNEL assay.The expressions of perforin and granzyme B were measured by RT-PCR.Results Apoptosis of activated T cells in Astilbin group was significantly increased than that of the control group(P
