Objective:To observe the therapeutic effect and safety of warfarin on patients with atrial fibrillation. Methods:A total of 126 patients with atrial fibrillation from our hospital during Jun 2012-Jun 2013 were selected. According to hiding number random method,they were divided into aspirin group (n=63)and warfarin group (n=63).Coagulation function,blood lipid levels and end-point events were compared between two groups.Results:Compared with aspirin group,after treatment,there were significant reductions in levels of total cholesterol [(5.8 ±0.5)mmol/L vs.(5.2±0.7)mmol/L],triglyceride [(2.6±0.4)mmol/L vs.(2.4±0.3)mmol/L]and low density lipoprotein cholesterol [(2.7±0.5)mmol/L vs.(2.4±0.3)mmol/L],significant rise in level of high den-sity lipoprotein cholesterol [(1.1±0.2)mmol/L vs.(1.3±0.2)mmol/L],prothrombin time [(28.3±11.7)s vs. (36.9±10.4)s]and it′s international normalized ratio [(1.9±0.4)vs.(2.4±0.5)]in warfarin group,P <0.05 all.Incidence rate of endpoint events such as cerebral infarction and peripheral artery embolism etc.in warfarin group was significantly lower than that of aspirin group (3.17% vs.23.81%,P <0.01).The incidence rates of complications were 23.81% and 26.98% in warfarin group and aspirin group respectively,and they had no signifi-cant difference,P >0.05. Conclusion:For atrial fibrillation,the therapeutic effect and safety of warfarin is better than that of aspirin,is worth extending.

Objective To study the hemodynamic changes of rabbit VX2 liver tumors before and after ethanol injection by perfusion imaging of MSCT and investigate the correlation between perfusion parameters and microvessel density (MVD). Methods All of 15 Japanese long-ear white rabbits were divided into control group and experiment group. All rabbits were inoculated with VX2 liver tumor. Perfusion imaging of MSCT scans were conducted in 5 rabbits in the control group on the 14 th day after VX2 tumor inoculation, and the borders of the tumors were stained with immunohistochemical stains, and MVD was measured by anti-CD34. Perfusion imaging of MSCT scans were conducted in all 10 rabbits in the experiment group on the 14 th day after VX2 tumor inoculation. Then absolute alcohol was injected into the tumors by laparotomy. CT scans were conducted 3 and 30 days after absolute alcohol injection, and the borders of the tumors were stained immunohistochemically, and MVD was measured by anti-CD34. The differences of perfusion parameters such as hepatic blood ( BF), hepatic blood volume (BV), mean transit time ( MTT),permeability of capillary vessel surface (PS), and hepatic arterial fraction (HAF), hepatic arterial perfusion (HAP) were compared to evaluate the liver hemodynamic changes. Statistical repeated measurement t test, correlation analysis were performed. Results BF of border of the tumor between pretreatment, 3 days after ethanol injection and 30 days after ethanol injection were ( 280. 62 ± 59. 87 ),(322.03 ± 86. 94 ), ( 177.05 ± 75.96) ml · min -1; HAF were 0. 59 ± 0. 08, 0. 89 ± 0. 12, 0. 23 ± 0. 07;HAP were ( 189. 26 ± 25.46), ( 251.57 ± 31.78 ), (40. 90 ± 5.17 ) ml · min - 1 · ml - 1. HAP increased significantly after ethanol injection 3 days ( P ＜ 0. 05 ); BF, HAF, HAP decreased significantly 30 days after ethanol injection compared with 3 days after ethanol injection ( P ＜ 0. 05 ); BF, HAF, HAP decreased significantly 30 days after ethanol injection compared with pre-treatment ( P ＜ 0. 05 ). It showed positive correlation between BF, PS, HAF, HAP and MVD (r=0. 916, 0. 726, 0. 870, 0. 889; P ＜0. 05). MVD decreased significantly compared with the control group (43.9 ± 4. 0)/HP 30 days after ethanol injection (21.8±3.5)/HP (t = 12.271, P ＜0.05). Conclusion Perfusion imaging of MSCT can detect the hemodynamic changes in rabbit VX2 liver tumors, and also in tumors before and after ethanol injection. CT perfusion can take the place of MVD to evaluate the tumor angiogenesis.

The course of pharmaceutical analysis for pharmacy undergraduate students in National University of Singapore and China Pharmaceutical University was compared, in terms of curriculum, lecturing, assessment and practical teaching.This study provides in-depth analysis by the lecturers from both universities,and can provide reference for teaching reform and course construction of pharmaceutical analysis.

Objective To study a histopathological classification system for hippocampal cell loss in patients suffering from mesial temporal lobe epilepsy.Methods Fifty-one surgically resected specimens were microscopically examined with respect to neuronal cell loss in hippocampal subfields CA1—CA4.Clinical data including age at first seizure,with or without initial precipitating injuries (IPIs),latecy,epilepsy duration,Engel score of half and one year after surgery were collected to analyse the clinical characteristics between different pathological types.Eight additional hippocampal specimens obtained from neurologically healthy autopsies served as controls.Results (1)Five distinct patterns were recognized within a consecutive cohort of anatomically well-preserved surgical specimens.Type 1 a (21/51,41.18 ％):severe cell loss in CA1 and moderate neuronal loss in all other subfields excluding CA2; Type 1b(18/51,35.2％):severe cell loss in all sectors; Type 2(9/51,17.65％):severe neuronal loss restricted to sector CA1 ;Type 3 (1/51,1.96％):severe neuronal loss restricted to the hilar region; no mesial temporal sclerosis (2/51,3.92％):a group comprised hippocampi with neuronal cell densities not significantly different from age matched autopsy controls.(2)The patients of type 1 a and 1 b had younger age of first onset,longer latency and duration,more frequency of IPIs,the proportion of ourrence of febrile seizures in type 1a was 10/19,type 1b was 7/16,type 2 was 4/7,type 3 was 0 (x2 =11.790,P =0.019).(3) The patients of type 1a and 1 b had better postsurgical outcome.Conclusion Type 1 is the most common type of mesial temporal sclerosis,which have better postsurgical outcome than the other 3 types.

Objective To observe the effect of JWSNS serum on proliferation and apoptosis hepatic stellate cells.Methods After being added different concentrations of JWSNS (the low concentrations of JWSNS:0.78 g/ml of crude drug; the medium concentration group of JWSNS:1.56 g/ml of crude drug; the high concentration group of JWSNS:3.12 g/ml of crude drug) drug-containing serum in vitro HSC-T6 cells for 12h,24 h and 48 h respectively,detected serum HSC-T6 proliferation with MTT colorimetry method and measured HSC-T6 apoptosis with flow cytometry and TUNEL method.Results (①) After applied JWSNS on rats HSC-T6,the Cell proliferation was inhibited which showed a time-concentration dependence.The differences were significant when comparing each JWSNS group with the control group (P＜0.01).High concentration of JWSNS group showed significant difference when compared with Biejiaruangan tablets group (P＜0.05) with high concentration of JWSNS (0.399± 0.041) ％ after 48h,and Biejia-Ruangan tablets (0.429± 0.037) ％ after 48 h.② Flow cytometry analysis showed each JWSNS group and Biejiaruangan tablets group had significant increased cell apoptosis when compared with the control group (P＜0.05) after 12 h,24 h,and 48 h.JWSNS medium concentration group [12 h was (17.83±0.25)％,24 h was (26.06±0.26)％,48 h was (39.30±2.25) ％] and JWSNS high concentration group [12 h was (27.15±0.29)％,24 h was (38.96±0.51)％,48 h was (49.34± 0.77) ％] had a significant increased cell apoptosis compared to the Biejia-Ruangan tablets group [ 12 h was (8.31 ± 0.30) ％,24 h was (16.25 ± 0.25) ％,48 h was (27.12± 0.39) ％].③ TUNEL detection showed that each concentration of JWSNS group [the low concentration of JWSNS:was (25.1 ± 1.48)％,medium concentration group of JWSNS was(39.30±2.25)％,high concentration group of JWSNS was(39.30±2.25)％] had a significant increased cell apoptosis rate than Biejiaruangan tablets group (30.0± 3.92) after 48 h (P＜0.01).Conclusion JWSNS containing serum can inhibit the proliferation of HSC-T6 in vitro,promote the apoptosis

Objective To investigate the hallux nail flap with the tibia lateral diamond shaped flap of the second phalange,joints,tendon combined tissue combination reengineering the surgical methods and clinical effects of thumb.Methods From May,2010 to February,2015,thcrc had fifteen cases of the thumb defect on Ⅱ-Ⅳ degree of the patients in our hospital,and we took anastomosis vascular pedicle of the hallux nail flap and thumb reconstruction with a second phalange with the tibia lateral diamond shaped flap,joint,and tendon composite tissue combination to reconstruct the thumb.The provided district of the second metatarsal was amputated to repair the ligaments between metatarsal bones.And we took the second toe bone nail flap covering the hallux toe area wound in anterograde shift.Results 15 cases of thumb reconstruction survived.The patients were followed-up for 6 to 30 months,the shape of the reconstructed thumb was closed to the contralateral thumb.The thumb to the finger and the palm function was good,two-point discrimination of the finger pulp were 0.6-1.0 cm.According to the upper limb function of the Chinese Medical Association Hand Surgery Society,the upper limb partial function of the evaluation trial standard:excellent in 11 cases,good in 4 cases.The district was recovered well,normal walking and running function were not restricted.Long term follow-up showed no blisters formation and skin ulceration.No nail deformity,good growth.Conclusion The hallux nail flap with the tibia lateral diamond shaped flap of the second phalange,joints,tendon combined tissue combination to reconstruct the thumb of the defect on Ⅱ-Ⅳ degree can restore good function and feeling,but also to have beautiful appearance.It is an effective method for reconstruction of thumb defect on Ⅱ-Ⅳ degree.

Objective:To study the expression of ULBP2 protein in the brain tissues of patients with drug-refractory epilepsy and its clinical significance.Methods:Gene-chip,immunofluorescence and Western blot were used to test expression of ULBP2 in the surgically removed brain tissue of patients with drug-refractory epilepsy from the brain bank of our department(n=42),and the results were compared with that of normal controls (n=12).Results:The relative increasing expression of ULBP2-gene in the brain of patients with drug-refractory epilepsy,and ULBP2 protein expression was significantly increased in temporal lobe cortex of patients with drug-refractory epilepsy as compared with the same regions of the controls specimens.Conclusion:The results indicate that the overexpression of ULBP2 may be involved in the pathophysiology of drug-refractory epilepsy.

Objective To evaluate the clinical efficacy and side effects of sunitinib in the treatment of advanced renal cell carcinoma. Methods Forty-five patients with advanced renal cell carcinoma and an average age of 48.6 yrs were treated with sunitinib. Among the study group, 25 were male and 20 were female. In group one, patients received sunitinib treatment in repeated six week cycles consisting of four weeks of sunitinib 50 mg daily followed by two weeks off treatment (schedule 4/2). In group two, a single daily dose of sunitinib 37.5 mg was administrated to 20 patients without off treatment. A CT scan was used to evaluate the treatment efficacy after each cycle and the side effects were recorded accordingly. Results Clinical efficacy could be evaluated in 40 patients. Of these, two achieved complete response, eight achieved partial response, 27 were stable and the remaining eight experienced disease progression with four patients dying during the study period. The side effects of sunitinib in group one and in group two included hypertension 32% (8/25) and 10% (2/20), P=0.02; liver function impairment 32% (8/25) and 20% (4/20), P=0.011; hand-foot skin reaction 68% (17/25) and 60% (12/20), respectively. The incidence of major side effects of sunitinib were different in Chinese patients than from what had been previously reported in studies conducted in US and Europe. Generally, most of the sunitinib side effects were easy to manage. Conclusions There weredifferences between the two groups of Chinese patients treated with different sunitinib protocols. The protocol of sunitinib 37.5mg daily without off-treatment was better than the protocol of sunitinib 50mg daily (schedule 4/2) in regard to liver function impairment and hypertension.

Objective To investigate the efficacy of Sunitinib in treating metastatic non-clear cell renal cell carcinoma (RCC).Methods Twenty-two metastatic non-clear cell subtype renal cell carcinoma patients with a median age of 46 years (29 -76 years) were treated with Sunitinib.Fourteen cases were found have metastasis post radical nephrectomy,and the remaining eight cases with metastasis received cytoreductive surgery.Pathological diagnosis showed 12 papillary RCCs,one chromophobe RCC,three collecting duct RCCs,and six unclassified RCCs.The metastatic lesions were located in the lung,lymph nodes,adrenal gland,bone,liver,and thyroid gland.The patients were given the treatment of sunitinib 50 mg qd four weeks on and two weeks off.The median time of treatment was 11 months (4.5 - 24 months).Results The objective control rate was 73％.Three papillary RCC and one chromophobe RCC reached partial remission (PR) and 12 cases maintained stable disease (SD) for more than 12 weeks.And the remaining six cases progressed (PD).Conclusions Sunitinib has definitive efficacy in metastatic papillary RCC,chromophobe RCC,collecting duct RCC and unclassified RCC.Metastatic lesions in lungs and lymph nodes might be more sensitive to Sunitinib.

OBJECTIVETo construct a recombinant lentiviral vector that stably express hepatocyte nuclear factor 6 (HNF6) in colorectal cancer cell line and examine its effects on the invasive ability of SW620 cells.

METHODSThe lentiviral vector pLeno-DCE-HA-HNF6 was constructed and transfected into 293T cells. The supernatant containing the lentivirus particles was harvested to determine the virus titer. A stable cell line was established by infecting SW620 cells with the lentivirus particles, and the transfection efficiency was examined by fluorescence microscopy and flow cytometry. The invasion ability of the transfected SW620-HNF6 cells was assessed by wound healing and transwell assays.

RESULTSThe recombinant lentiviral vector was correctly constructed and verified by sequencing. SW620-HNF6 cell line with stable HNF6 expression was established successfully, and the transfection efficiency reached 82.3%. Western blotting and quantitative PCR demonstrated significantly upregulated HNF6 expression in SW620-HNF6 cells, which showed obviously suppressed invasive ability in wound healing and transwell assays.

CONCLUSIONWe have successfully established a colorectal cancer cell line SW620-HNF6 stably expressing HNF6, which shows a lowered migration activity in vitro.
