1.Role of the Nrf2 pathway in liver diseases
International Journal of Pediatrics 2014;(5):462-464,465
Nrf2 signaling pathway is a major mechanism in the cellular defense against oxidative or electrophilic stress, which regulates series of detoxifying and antioxidant defense genes expression in the liver. The Nrf2 signaling pathway collectively exhibits diverse biological functions against viral hepatitis, alcoholic and nonalcoholic liver disease, fibrosis, and cancer via target gene expression. This review discusses the role of the Nrf2 pathway in liver pathophysiology and the potential application of Nrf2 as a therapeutic target of prevention and treatment of liver diseases.

Objective To study the relationship between morphological and functional changes of melanocytes and their effect on the graft color. Methods Eighteen black whited guinea pigs were divided into nine groups randomly and biopsied at 1 d, 2 d, 3 d, 7 d, 14 d, 21 d, 28 d, and 90d after skin grafting, respectively. Then skin spliting and Dopa reaction techniques were used to demonstrate melanocytes. And computer assitant imagine analysis system was applied to observe and quantitate the morphological changes of melanocytes at various time points in three postoperative months. Results During three days after grafting, resulting from the surgery wound and shortage of blood and oxygen supply, the processes of the melanocytes broke off, accompanied by reduction of whole dimension of the cells, size as well as number of the processes. The indices above returned to the normal level between second week and third week after grafting. It was an evident characteristic that number of melanocytic processes became shorter and stouter in the end of the experiment. conclusion Functional changes of melanocytes in the skin grafts are in accord with their morphological changes. Compared to those in the sizes of melanocytes, changes in the processes of the cell are more sensitive in reflection of function of melanocytes. The shorter and stouter processes of melanocytes in the end of the experiment are suitable to the smaller epidermal melanin unit and more efficient in transformation of melanin into the keratinocytes.

Objective To study the correlation of replication efficiency and antigen expression of hepatitis B virus (HBV) strains in patient sera and transfected cells. Methods Quantification of five maternal serum HBV DNA was carried out using dot hybridization and polymerase chain reaction-enzyme linked immunosorbent assay (PCR-ELISA), hepatitis B surface antigen (HBsAg) and hepatitis B e antigen (HBeAg) levels in the sera were determined by ELISA. Full-length genomes of HBV strains cloned from five pregnant women were separately used to transfect HepG2 cells. The levels of HBsAg and HBeAg in the supernatant of transfected cells were determined with Abbott EIA kits, the replication efficiency of intracelluar replicative intermediates and extracellular HBV DNA of viral particles was analyzed by Southern blot and hybridization. Results Intracellular and extracellular HBV replication and antigen expression of cloned HBV strains showed positive correlation tendency with the HBV DNA and antigen levels in serum samples. Conclusions Virus replication efficiency and expression of HBsAg and HBeAg by full-length HBV clones are in relative coincidence with the biological characteristics of HBV strains in patients. Cell transfection can be used to study the biological characteristics of individual isolates.

Objective To investigate the correlative factors that affect the outcome of pregnancy complicated with burn injuries and to provide the treatment proposal for the pregnant women with vary gestational age. Methods Retrospective analysis of 13 cases of burn injuries during pregnancy was made. Results All patients required standard treatment to heal the burn injuries. During the first trimester, abortions occurred in two of the four patients; Abortions occurred in two and premature labour occurred in one of the nine patients who were injured during their second or third trimester. Conclusion Abortions or premature labour appeared during pregnancy as complications of the treatment of burn injuries. Enough fluid therapy in time in early phase of burn is necessary for mother survival and placental perfusion.

Objective To investigate the correlation between hope and medical coping styles among patients with diabetic foot ulcer.Method Fifty-four inpatients and outpatients with diabetic foot ulcer were investigated by the demographic questionnaire, the Herth hope index(HHI)and medical coping modes questionnaire(MCMQ)to investigate the status of their hope and coping styles and the correlation between them. Results The average score of hope was(36.50±5.06),which was at the high level.Confrontation was lower than the norm in coping styles(all P<0.05)and the differences between avoidance,acceptance-resignation and norm were not statistically significant(P>0.05).The average score on hope was positively correlated with confrontation but negatively correlated with avoidance(all P<0.05),and there was no significant correlation with acceptance-resignation(all P>0.05).Conclusions The hope of patients with diabetic foot is high.It is positively correlated with confrontation and negatively correlated with acceptance-resignation in coping styles.Nurses should instruct the patients to adopt positive coping styles, avoid negative coping styles and keep them at a higher level of hope.

Objective To explore the nursing intervention of adverse reactions of Iohexol Injection using CT scanning. Methods According to the different nursing intervention mode of patients in our hospital in January January 2015 to January 2017 performed CT scanning using the Iohexol Injection group in 50 cases as control group with routine nursing intervention, the observation group used routine nursing quality nursing intervention;adverse reaction a detailed record of the two groups of patients, and the incidence of adverse reactions compared to investigate the effect of nursing intervention of adverse reactions using the Iohexol Injection CT scan. Results Routine nursing quality nursing intervention (observation group) is better than the clinical effect of CT scanning using Iohexol Injection in patients with routine nursing intervention (control group) clinical the effect and adverse reaction rates in patients with lower than the control group, the difference was statistically significant (P<0.05). Conclusion CT scan. With Iohexol Injection patients regular nursing care, high quality nursing mode intervention effect is remarkable, can effectively reduce the incidence of adverse reactions in patients, it is worthy of clinical widely used.

Objective To evaluate the effect of morphine on the growth of subcutaneous tumor of human gastric cancer cells in nude mice. Methods Thirty SPF male BALB/C nude mice, aged 4-5 weeks, weighing 15-20 g,in which the model of subcutaneous tumor of human gastric cancer cell line MGC-803 was established, were randomly divided into 3 groups (n=10 each) using a random number table:control group (group C),normal saline group (group N),and morphine group (group M).The mice in group C received no treatment.Morphine 20 mg/kg was intraperitoneally injected once a day for 14 consecutive days in group M,while normal saline 1.5 ml/kg was given instead of morphine in group N.The caliper was used to measure the tumor size every 2 days starting from 3 days after beginning of administration, and the relative tumor volume was calculated. The nude mice were sacrificed on 15th day,and the tumor tissues were obtained for determination of nuclear factor-kappa B activity and Bcl-2 and Bax protein and mRNA expression by real-time reverse transcriptase polymerase chain reaction and Western blot. Results Compared with group C,the relative tumor volume was significantly decreased, the activity of nuclear factor-kappa B in tumor tissues was significantly decreased, the expression of Bcl-2 protein and mRNA was significantly down-regulated,and the expression of Bax protein and mRNA was significantly up-regulated at each time point in group M (P<0.05),and no significant change was found in each parameter mentioned above in group N (P>0.05).Conclusion Morphine can inhibit the growth of subcutaneous tumor of human gastric cancer cells in nude mice, and the mechanism is associated with promotion of apoptosis in tumor cells.
