1.Characteristics of Ambulatory Electrocardiogram in Patients with Chronic Atrial Fibrillation and Optimal Ventricular Rate Control

Objective:To observe the characteristics of 24 hour ambulatory electrocardiogram in patients with chronic atrial fibrillation (AF) and optimal ventricular rate control. Methods:Holter monitoring was performed in 122 hospitalized patients,aged 17—85 (mean 51.57±11.94) years,with organic heart disease and chronic AF (New York Heart Association functional class Ⅱ-Ⅲ) in whom the ventricular rate were well-controlled by digoxin.Maximum,minimum and mean ventricular rates,the number of ventricular pauses of >1.5 second,and the longest ventricular pauses during the monitoring were recorded.Follow-up data were collected in 72 patients. Results:The mean,fastest and lowest ventricular rates over 24 hours were 78.53±8.12(65—90),143.79±28.85(90-183) and 51.04±7.52(34—71) bpm,respectively.Bradycardia of less than 50 and 45 bpm occurred in 62 and 32 of the 122 patients,respectively.Ventricular pauses of more than 1.5 and 3.0 seconds were noted in 95 and 14 of the 122 patients,respectively.The longest ventricular pause was 2.38±0.55(4.64—1.60) seconds.During follow-up,there were no worsening in these patients. Conclusion:Ventricular pauses at rest and nocturnal bradycardia were common in patients with chronic atrial fibrillation who had ventricular rates controlled.The dosage of digoxin should not be changed,nor pacemaker insertion is needed.
2.Expression of CD_(44) Molecules in Psoriatic Epidermis

The expression of CD 44 in psoriatic epidermis was studied with immunohistochemical staining. Overexpression of CD 44 was observed in involved proliferative epidermis as compared with uninvolved and cured epidermis in psoriatic patient. Several CD 44 isoforms based on analysis by reverse transcriptase polymerase chain reaction (RT PCR) were mainly CD 44 S and CD 44 E. These results suggest that CD 44 may be an important substance in the proliferation of keratinocytes, such as psoriasis.
3.Malnutrition related factors in patients with gastric cancer

Malnutrition is one of the most important factors for the prognosis of patients with gastric cancer,but the mechanism of malnutrition is unclear at present. These patients are at risk of nutritional deple-tion from anorexia,dysphagia,and may be associated with the presence of two or more of the following cyto-kines with metabolic activity such as TNF-α,IL-1,IL-6,interferon,leptin,nuclear factor-κB and so on, which are secreted by the host-tumor interaction. The cytokines constitute a sophisticated regulatory network. Therefore,the study of the relationship between malnutrition and related factors in patients with gastric cancer may improve the nutritional status and prognosis of these patients.
4.Effects of FTY720 pretreatment on ventilator-induced lung injury in rats

Objective To study the effects of FTY720 pretreatment on ventilator-induced lung injury (VILI) in rats in order to explore the role of sphingosine-l-phosphate receptor 1 (S1PR1).Methods Forty healthy adult male SD rats weighing 300-350 g were randomly (randlom number) divided into 4 groups (n=10 each):group CV (conventional tidal volume VT =8 mL/kg),group HV (high tidal volume VT =40 mL/kg),group HF and group HFW.The rats in group HF received intra-gastric administration of FTY720 10 mg · kg-1 · d-1 for 7 days,while additional dose of W146 (S1PR1 antagonist) 1 mg · kg-1 · d-1 was administrated in group HFW before high tidal volume ventilation.After 4-hour mechanical ventilation,arterial blood samples were obtained for blood gas analysis before the animals were sacrificed.The lungs were harvested for histopathologic observation.Apoptosis rate in lung tissue was determined with flow cytometry.W/D lung weight ratio,pulmonary permeability index (PPI),total protein,and TNF-α,IL-1β in bronchoaveolar lavage fluid (BALF) were measured.All data were analyzed by oneway analysis of variance (ANOVA),The intergroup comparisons were analyzed by the least-significantdifference (LSD) test by using SPSS version 20.0 software.Differences were considered statistically significant if P < 0.05.Results Compared with group CV,the level of PaO2 [(73.6 ± 8.9) vs.(50.5 ± 6.0)] was decreased,the levels of W/D [(3.12 ± 0.27) vs.(5.12 ± 0.56)],PPI [(0.08 ±0.03) vs.(0.30 ± 0.06)],apoptosis rate [(10.6 ±2.9) vs.(48.5±6.7)],total protein [(5.8 ±2.1) vs.(15.4±5.6)] and TNF-α [(24.3±5.7) vs.(108.4±16.0)] andIL-1β [(90.6±14.1) vs.(338.5 ± 44.3)] were increased in group HV (P < 0.05).Compare with group HV,PaO2 [(50.5 ± 6.0) vs.(65.9±10.3)] was increased,W/D[(5.12±0.56)vs.(3.85±0.37)],PPI[(0.30± 0.06)vs.(0.14±0.03)],apoptosis rate [(48.5 ±6.7)vs.(25.6 ±5.3)],total protein [(15.4±5.6) vs.(8.9±2.5)],TNF-cα [(108.4±16.0) vs.(75.6±10.3)] andIL-1β [(338.5 ±44.3) vs.(188.9 ±33.8)] in BALF were decreased in group HF (P <0.05).PaO2 [(50.5±6.0) vs.(59.7±7.8)] was higher,W/D[(5.12±0.56) vs.(4.44±0.30)],PPI [(0.30±0.06)vs.(0.19±0.09)] andIL-1β[(338.5-±44.3) vs.(246.8±24.6)] levels were lower in group HFW than those in group HF There were no significant differences in apoptosis rate[(48.5±6.7)vs.(41.3±6.8)],totalprotein[(15.4±5.6)vs.(10.4±2.7)] and TNF-α level [(108.4 ± 16.0) vs.(97.5 ± 10.3)] between HFW group and HF group (P > 0.05).Compare with group CV,The histopathologic damage of lung tissue was obvious in group HV and group HFW,it was attenuated by pretreatment with FTY720 in group HF.Conclusions FTTY720 pretreatment provides protective effects against ventilation-induced lung injury in rats,and S1PR1 may mediate the protection through reducing apoptosis rate and inflammatory reaction.
5.Effect of Case Method on the Clinical Teaching of Medical Oncology

Objective Explore the effect of case method on clinical teaching of Medical Oncology. Methods The case method was taken in the teaching of Clinical Oncology for trial group,and the traditional method teaching was taken for contrast group of our university. At the end of term, we made a questionnaire and statistics examination records. Results (1) It was conducive to good learning ability. Case method teaching in Clinical Oncology could cultivate student's ability to study independently. (2) Comparing examination records of the two groups, it also showed a significant difference ( <0.05) . Conclusion (1) Case method teaching has good teaching effect, and it can cultivate student's ability to study independently. (2) Application of case teaching method in Clinical Oncology received a very good teaching result, but the assessment system should be established and improved.
6.The Clinical Diagnosis and Treatment of 143 Cases of Hodgkin's Lymphoma

Objective To investigate the clinical characteristics of hodgkin's lymphoma (HL) patients . Method The clinical data of HL patients in our hospital from January 1, 2003 to December 31, 2012 were analyzed retrospectively. The contents of investigation included gender, age, pathological type, first symptom, the incidence locations, clinical stages, extranodal involvement locations, ECOG and treatments.Results A total of 143 cases of HL were included, the M/F ratio was 2.5:1 with a median age of 37 years (11~73years) . The most frequent subtypes of HL were mixed cellular (34.3%) and nodular sclerosis (28.0%) . The nodal lymphomas accounted for 97.2%, while the extranodal lymphomas occupied 2.1%. The nodal lymphomas with extranodal invasion accounted for 29.5%, and the common locations were spleen (14.4%), lung (5.8%) and bone (5.0%) . The most clinical stage of all patients was stage Ⅰ/Ⅱ (60.8%), The differences were no significant in gender, pathological type, first symptom, the incidence locations and the effective rate of the first-line treatment (P>0.05), while were statistically significant in B symptoms, ECOG and treatments betweenⅠ/Ⅱgroups andⅢ/Ⅳgroups (P<0.05) . Conclusions In this group,the incidence of HL patients were mainly found in middle-aged people, males were lager than females.The pathological type, first symptom and the incidence locations had no obvious effect on clinical stage. The clinical stage had no obvious influence on the effective rate of the first-line treatment.
7.Characteristics of bone marrow-derived mesenchymal stem cells of different species: An in vitro comparison study

BACKGROUND: Bone marrow-derived mesenchymal stem cells (BMSCs) can not only differentiate into multiple nonhematopoietic cell lineages, but seek out damaged tissues and repair them as well. Hence, they were largely studied for their potential clinical use. However, their biological characteristics have not been fully discovered. OBJECTIVE: To compare the biological characteristics of BMSCs of different species cultured in vitro, in order to provide basis for the clinical research of stem cell therapy.DESIGN: Randomized controlled observation was designed.SETTING: Medical Research Department of General Hospital of Guangzhou Military Area.MATERIALS: The experiment was performed in Medical Research Department of General Hospital of Guangzhou Military Area from June 2004 to July 2005. Thirty SD rats weighing (160±20) g, aged 35 to 40 days, 30 Kunming mice weighing (16.0±2.0) g, aged about 40 days, 8 New Zealand white rabbits weighing (2.0±0.2) kg, aged 80 to 90 days and 10 healthy volunteers (25-32 years old) were selected. All the animals were of clean grade, which were purchased from the Animal Center of Southern Medical University.METHODS: The BMSCs of mice and rats were prepared according to the protocol developed in the Caplan laboratory, while those of rabbits and human were isolated from bone marrow suspension obtained by iliac puncture.The morphology of BMSCs was observed by light microscope and transmission electron microscope. Cell growth curve was tested by MTT. Expression of Stro-1 was analyzed by immunofluorescence cytochemistry and flow cytometry. To evaluate the specific response of BMSCs to osteogenic supplements(10 nmol/L dexamethasone, 10 mmol/L β-glycerophosphate,and 50 mg/L ascorbic acid), the activity of alkaline phosphatase (AKP) activity was tested by a commercial kit. Expression of osteocalcin was examined by immunocytochemistry and hydroxyapatite crystals were shown by von Kossa staining. Adipogenic differentiation was evaluated by estimating percent of cells containing Oil Red-O- stained oil droplets.MAIN OUTCOME MEASURES: ① Morphological observation and growth situation of BMSCs. ② Expression of Stro-1: BMSC marker. ③ Differentiation in osteogenic medium.RESULTS: ①The morphology of adherent BMSCs ofthose four species observed by optic microscopy was obviously different. When they became mature or aged, the mouse cells turned into flat shape, irregularly polygonal, fell to pieces and deposited on the flask-bottom, while the rat, rabbit and human cells would enlarge and become polygonal, vacuoles would appear in their cytoplasm, finally, the cells were detached from the flask-bottom, floating off like cotton wool. The cultures of different species also had some commonness, such as poly-layer growing manner, without contact inhibition and consisting of two groups. Cells of one group grew into colonies from single cells and expanded quickly, while cells of the other group were sporadic and did not proliferate. Electron microscopy revealed that all of the primary cells had microvilli and that they could be divided into two subpopulations according to their ultrastructures. Some cells were rich in organelles and most chromatin was euchromatin, while the other subpopulation cells had much fewer organelles and more heterochromatin. Growth curves of BMSCs of different species were almost the same. ② The positive rate of human adherent bone marrow-derived cells for Stro-1: BMSC marker was (91.4±8.3) %, and that of mouse adherent cells was (83.5±6.2) % .③Treated with osteogenic supplements, mouse BMSCs differentiated into adipose tissue, rabbit ones died, while rat and human ones differentiated into osteocytes. BMSCs also demonstrated spontaneous differentiation in vitro.CONCLUSION: Mouse, rat, rabbit and human BMSCs can be easily expanded in vitro, although the harvest of the current method is a mixture of mesenchymal cells with various maturities, most of which are poor-differen-tiated cells. BMSCs of those species are different in morphology and response to the same inductive supplements. Therefore, in order to establish a kind of stem cell therapy, it is necessary not only for evidence from animal experiments but for that from human experiments as well.
8.The effect of hyperbaric oxygen combined with drug treatment on the cell cycle of colon carcinoma Lovo cells

Objective To investigate the effect of hyperbaric oxygen(HBO)combined with 5-fluorouracil (5-FU)chemotherapy on colon carcinoma Lovo cells.Methods Lovo ceils were exposed to 0.20 MPa HBO combined with 5-FU at different concentrations.The cell cycle was monitored with flow cytometry,and cell proliferation was detected using a methylthiazol tetrazolium test(MTT).Results ①After exposure to HBO,phase accumulation of Lovo cells was significantly higher than in the control group,and the accumulation of Lovo cells exposed to HBO after 24 h was significantly higher than at 12 and 48 hours.②There was difference between HBO(0.20 MPa,post 24 h)+ 5-FU(≤8 pM)group and 5-FU(≤8 μM,12 h)group on the inhibition of cells proliferation(P <0.05),and significan difference could be seen between the two groups after treatment for 48 h(P < 0.01); ③There was no significant difference between HBO + 5-FU group and 5-FU group treatment with the concentration(16,32,64 μM)of 5-FU(P > 0.05)for 12 h ;however,the difference could be seen after treatment for 48 h between the two groups(P < 0.05).Conclusion The lethal effect on Lovo cells of 5-FU can be enhanced by exposure to 0.20 MPa HBO,especially with low concentrations of 5-FU.
9.Effects of hyperbaric oxygen on the expression of hypoxia inducible factor-1α mRNA and vascular endothelial growth factor mRNA in colon adenocarcinoma cells

Objective To investigate the effects of hyperbaric oxygen (HBO) on the expression of hypoxia inducible factor-1α (HIF-1α) mRNA and vascular endothelial growth factor (VEGF) mRNA in colon adenocarcinoma cells. Methods SW480 colon cancer cells were divided into a control group and an HBO group, which was in turn divided into various subgroups according to the various HBO pressures (0.15 MPa, O. 20 MPa, 0.25 MPa) and exposure times (12, 24 and 36 h) tested. The expression of HIF-1α and VEGF mRNA was evaluated with RT-PCR after the various treatments. Results The expression of HIF-1α mRNA was lower in the SW480 cells than in the controls after exposure to 0.25 MPa HBO for 12 h. No expression of HIF-1α mRNA was detected after exposure to 0.25 MPa HBO for 24 h. The expression of VEGF mRNA was lower after exposure to 0.25 MPa HBO for 36 h than in the controls. Conclusion Absence of HIF-1α mRNA expression and decreased expression of VEGF mRNA in SW480 can be observed after exposure to 0.25 MPa HBO for 24 h, suggesting that HBO may inhibit the formation of tumor blood vessels through down-regulating the expression of these mRNAs.
10.A study of line bisection and number bisection in patients with schizophrenia

Objective To investigate the existence of neglect and explore the relationship between spacial attentional deficits and hemisphere in patients with schizophrenia. Methods 51 schizophrenic patients and 55 healthy controls were measured with line bisection test and number bisection test、 Results In the Line Bisection Test,shcizophrenic patients group showed a negative deviation, in which the 2cm line( -0.03 ± 0. 74 )% and mean ( - 0. 02 ± 0. 03 ) % deviation rates were more significantly leftward than that of the normals ( ( 0. 00 ±0.02 ) %, ( - 0.00 ± - 0.01 ) %, P < 0. 05 ) respectively. The mean deviation rates of the patients (P < 0. 01 ) and control groups (P < 0. 05 ) were also significantly leftward compared with zero value. In the number bisection test,schizophrenic patients group showed a negative deviation and a significant leftward bias on the interval 5 ( - 0.07 ±0.18)% ,7( -0.08 ±0.22)% and 9( -0.09 ±0.28)% ) than that of the normals(( -0.01 ±0.05)% ,(0.01± 0. 08 ) % and (0.00 ± 0.14 ) %, P < 0.05 respectively). Conclusion Schizophrenic patients have a right-neglect, suggesting that spacial attentional deficits exist, and it may be related to the left cerebral hemisphere dysfunction.
