1.Clinical significance of peripheral blood leukocyte and lymphocyte function-associated antigen-1 expression in renal allograft recipients

Objectives To inspect the levels of peripheral blood leukocyte and lymphocyte functionassociated antigen-1(LFA-1)expression in renaI allograft recipients before and after renal transplantation.Methods Using monoclonal antibody-flow cytometer fluorescence technique,the expression of LFA-1 in lymphocytes of 33 patients with renal transplantation was detected at 1 day before operation and 3,6,9,15 days after operation by flow cytometric immunological technique,Results The level of LFA-1 expression after transplantation was significantly decreased and maintained at a low level.and ascended suddenly during the course of graft rejection.The level of LFA-1 expression in rejection was obviously higher than in non-rejection and after the eontrollring of rejection.Conclusions LFA-1 might Dlay an important role in graft rejection.Monitoring of the expression of LFA-1 is helpful for the diagnosis of the graft rejection.
2.Influences of Chuan Shen Tong Injection on Epidermal Growth Factor and Its Receptor in Human Benign Prostatic Hyperplasia

Objective:To investigate the mechanism of Chuan Shen Tong Injection (CSTI) for benign prostatic hyperplasia (BPH).Methods:The distribution of epidermal growth factor (EGF) and epidermal growth factor receptor(EGFR) expression were detected with immunohistochemical technique in 49 samples of the excised prostate. Among them, 18 samples were from patients without intraglandular injection of CSTI (group A) as controls, 21 from those receiving injection but without effect (group B) and 10 from those receiving injection with effect but recur (group C).Results: The positive expression rates of interstitial EGF and EGFR in group B (81% and 71.4% respectively) increased as compared with group A (33.3% and 33.3% respectively.,P

To investigate the correlation between activation of B lymphocytes and CD11a, CD54 expression in renal allograft recipients, the dynamic changes in CD19, CD11a and CD54 levels in lymphocytes at different time points were examined by using mAb-FACS-immunofluores-cence technique in 10 renal allograft recipients in stable course(stable group) and 10 with acute rejection(rejection group) after operation. The expression levels of CD19 and CD54 in lymphocytes were significant higher than those of the pre-operation levels in both stable group and rejection group on the 3rd day after kidney transplantation, and the CD19 expression level was significant higher before rejection in the rejection group than that in stable group at the same time points; The expression of CDlla increased from (31. 9 ?12. 4)% to (49. 5 ? 20. 2)% during rejection and decreased significantly after anti-rejection treatment. ObjectiveTo explore the expression and clinical significance ofaquaporin (AQP) 1 and AQP 4 in human pulmonary adenocarcinoma H1299 cell line.MethodsH1299 cell line in human pulmonary adenocarcinoma(pulmonary adenocarcinoma group) were obtained,the expressions of AQP1 and AQP4 in mRNA level and their locations were determined in H1299 cell line respectively by semiquantitative reverse transcription polymerase chain reaction (RT-PCR) and Western blot analysis.The migration of tumor cells were observed by Matrigel invasion assay.Then normal tissues adjacent of pulmonary adenocarcinoma (above cancer line 3 cm,no tumor cell with pathological proven) were as control group.ResultsThe results of RT-PCR showed that AQP1,AQP4 mRNA was 1.030 ± 0.070 and 1.140 ± 0.190 in conlrol group,which were lower than those in pulmonary adenocarcinoma group (2.021 ± 0.250 and 2.180 ±0.180)(P<0.05 ).The results of Western blot showed AQP1,AQP4 located on the membrane of H1299 cell.Both AQPI and AQP4 mRNA expressed very high in pulmonary adenocarcinoma group,while expressed very low in control group (P<0.05).Matrigel invasion assay showed that the invasion was positively related to AQP1,AQP4(r =0.351,P < 0.05 ).ConclusionAQP1,AQP4 significantly over express in H1299 cell line,both of them phy important roles in the growth of tumor tissue and cell migration.
6.Treatment and effective analysis on maxillary complex injury.

OBJECTIVETo analyze the characteristics of cases of maxillary complex injury, to sum up experiences and lessons in the clinical first aid process, to evaluate different treatment effects of maxillary complex fracture and investigate the reasons for the failure of the treatment process.
METHODS152 cases with maxillary complex injury were analyzed retrospectively.
RESULTS2 cases (1.32%) died during emergency treatment. The cure rate was 62.67% (94/ 150) in the 150 survival cases. The cure rate of closed fractures were better than open ones and surgical treatments than non-surgical ways (P < 0.05); the cure rate of surgical treatment in stage I was higher than II (P < 0.01).
CONCLUSIONFor patients with complex injuries of maxillary, first aid failure is mainly due to bleeding or asphyxiation. Therapeutic efficacy is influenced by treatment time, surgical approach, type of treatment and so on. The key to success of treatment is dependent on the early firm internal fixation and the functional protection and restoration of injured organs or tissues.

7.Mediating effect of nursing staff′s character advantage between job satisfaction and subjective well-being

Objective:To explore the status quo of job satisfaction, character advantage and subjective well-being of nursing staff, and to explore the relationship among the three, so as to provide theoretical basis for improving the subjective well-being of medical staff.Methods:Using cluster random sampling method, 582 nursing staff in Jinzhou City of Liaoning Province were selected as the survey objects. The General Information Questionnaire, Minnesota Satisfaction Questionnaire, Character Strengths Scale and Subjective Well-being Scale were used to investigate the respondents. The mediating effect of character strengths on job satisfaction and subjective well-being was analyzed by structural equation model. The mediating effect was analyzed and verified by bootstrap method.Results:The score of Minnesota satisfaction questionnaire, Character Strengths Scale and Subjective Well-being Scale of nursing staff were 70.5 ± 10.8, 45.0 ± 6.8 and 24.2 ± 6.3 respectively. The results of correlation analysis showed that there was a positive correlation between the total score of job satisfaction and the total score of character strengths of nursing staff ( r=0.214, P<0.01), and the total score of subjective well-being ( r=0.373, P<0.01). The total score of subjective well-being was positively correlated with the total score of character strengths ( r=0.123, P<0.01). Conclusions:The character strengths of nursing staff plays a mediating role between job satisfaction and subjective well-being.
8.Breeding of Actinobacillus succiniogenes mutants with improved succinate production based on metabolic flux analysis.

It is very important to obtain high yield mutant strains on the base of metabolic flux analysis of Actinobacillus succinogenes S.JST for the industrial bioconversion of succinic acid. The metabolic pathway was analized at first and the flux of the metabolic networks was calculated by matrix. In order to decrease acetic acid flux, the strains mutated by soft X-ray of synchronous radiation were screened on the plates with high concentration of fluoroacetic acid. For decreasing the metabolic flux of ethanol the site-directed mutagenesis was carried out for the reduction of alcohol dehydrogenase(Adh) specific activity. Then the enzyme activity determination and the gene sequence analysis of the mutant strain was compared with those of the parent strain. Metabolic flux analysis of the parent strain indicated that the flux of succinic acid was 1.78(mmol/g/h) and that the flux of acetic acid and ethanol were 0.60 (mmol/g/h) and 1.04( mmol/g/h), respectively. Meanwhile the metabolic pathway analysis showed that the ethanol metabolism enhanced the lacking of H electron donor during the synthesis of succinic acid and that the succinic acid flux was weakened by the metabolism of byproducts ethanol and acetic acid. Compared with the parent strain, the acetic acid flux of anti-fluoroacetic mutant strain S.JST1 was 0.024 (mmol/g/h), decreasing by 96%. Then the enzyme determination showed that the specific activity unit of phosphotransacetylase(Pta) decreased from 602 to 74 and a mutated site was founded in the pta gene of the mutant strain S.JST1. Compared with that of the parent strain S.JST1 the ethanol flux of adh-site-directed mutant strain S.JST2 was 0.020 (mmol/g/h), decreasing by 98%. Then the enzyme determination showed that the specific activity unit of Adh decreased from 585 to 62 and the yield of end product succinic acid was 65.7 (g/L). The interdiction of Adh and Pta decreased the metabolism of byproducts and the H electron donor was well balanced, thus the succinic acid flux was strengthened by the redundant carbon flux from these byproducts. The mutant strain S.JST2 obtained in this paper deserves being extended to application of industrial fermentation.

9.Effects of Allogeneic Transplantation of T Helper-2 Cells on Graft-versus-Host Disease and Graft-versus-Leukemia Effect

This study was undertaken to explore whether the graft-versus-host-disease could be decreased and graft-versus-leukemia effect be retained by transplantation of allogeneic T helper-2 (Th2) cells. T cells from C57BL/6(H-2b) mice were incubated and polarized with rmIL-4, Con A and ionomycin in vitro, and then, the T cells were mixed with marrow cells and transplanted into recipient BALB/c(H-2d) mice bearing erythroleukemia cells. The occurence of GVHD and GVL effect was observed. The results showed that the mean survival time in the groups of untreated control, cyclophosphamide treatment, marrow and spleen T cell transplantation and marrow and Th2 cell transplantation was 10.6 +/- 1.3, 18.7 +/- 4.2, 22.7 +/- 7.4 and 36.9 +/- 10.8 days, respectively. In untreated control and cycophosphamide treatment groups, all of ten mice died from leukemia. Nine of ten mice died from GVHD in marrow and spleen T cells transplantation group. In marrow and spleen Th2 cell transplantation group, three of ten mice died from GVHD, and GVHD was not occurred in the other seven mice, and there was no any evidence of leukemia in two mice on 50 days after transplantation. It was concluded that tranplantation with polarized Th2 cells could relieve GVHD, and at the same time retain the GVL effect.
10.MRI Average Diffusion Kurtosis on Differential Diagnosis of Breast Lesions

Purpose To synthetically evaluate MR diffusion kurtosis imaging (DKI) using Meta-analysis to differentiate diagnostic value of benign and malignant breast lesions and quantitatively analyze mean kurtosis (MK) values of benign and malignant breast lesions in an attempt to provide evidence-based facts for clinical decision making. Materials and Methods Computer-based retrieval of Chinese and English literatures concerning differential diagnosis of benign and malignant breast lesions with application of DKI publicly published at home and abroad in PubMed, Ebsco, Embase, CNKI, Cochrane Library, CBM, VIP Data database from January. 1, 2006 to September. 30, 2017. Two valuators independently screened and extracted data according to inclusion and exclusion criteria, and drew on QUADAS for quality assessment, Stata 12.0 and Meta-Disc 1.4 software for Meta analysis and extraction of relevant parameters. Results Totally 8 up-to-standard literatures were included, including 3 in English and 5 in Chinese. The total number of focus was 689. Corresponding effect model analysis revealed that MK value in malignant breast lesion group was higher than that in benign lesion group, the difference of which was statistically significant. The optimal diagnostic threshold was determined by the maximum Youden index, which was approximately 0.71 in size,with combined sensitivity of 0.907(95% CI 0.860-0.939),specificity of 0.910(95% CI 0.856-0.945),positive likelihood ratio of 10.053(95% CI 6.293-16.059),negative likelihood ratio of 0.102(95% CI 0.068-0.154),and diagnostic test odds ratio of 98.203(95% CI 55.014-175.296).The area under receiver operating characteristic curve was 0.96.MK value suggested 94% posterior malignant rate of malignant breast lesions, and 13% malignant rate of benign breast lesions. Conclusion MK value has high diagnostic value for benign and malignant breast lesions with good sensitivity and specificity.
