1.Study on clinical outcome of frozen-thawed transfer cycles with different cryopreservation duration of pro-grammed freezing and vitrification embryos

Objective To compare the influence of cryopreservation duration on quality of rozen-thawed embryo and clinical outcome in frozen-thawed transfer cycles. Methods A retrospective study was performed to evaluate the clinical outcomes and quality of rozen-thawed embryo of frozen embryo transfer(FET)cycles includ-ing freezing cycles and vitrification cycles. FET cycles were divided into 4 group acorrding to the storage duration of vitrified embryos:1~6 months group ,7~12 months group ,13~24 months group and ≥ 25 months group. Result There was no significant difference in recovery rate ,clinical pregnancy rate and abortion rate (98.1%, 99.0%,96.8%&95.7%;54.4%,55.4%,49.6%&51.5%;0.7%,8.9%,9.4%&11.1%;P>0.05). The embryos recovery rate and clinical pregnant rate after vitrification were significantly higher than programmed freezing (P<0.05). The blastocyst recovery rate in group A was significantly higher than the cleavage stage embryo recov ery rate in group B(P<0.05). Conclusions Vitrification has better effects on cryopreservation and clinical outcomes. Blastocyst may be the best stage for vitrifying and freezing of embryo.
2.Effects of Electroacupuncture in Different Time on Variations of Fractional Anisotropy Mean Value of Diffusion Tensor Tractography in Spinal Cord Injured Rats

Objective To explore the effects of electroacupuncture in different time on the variations of fractional anisotropy (FA) mean value in diffusion tensor tractography (DTT) in spinal cord injured (SCI) rats. Methods 48 Sprague-Dawley rats were performed with Allen's method by NYU blow device, resulting in T10-11 SCI. They were divided into groups according to random table. Control group (n=16) received no treatment. Experimental group (Group A, n=32) received electric stimulation in the scalp surface projection area of motor area and local electric stimulation at damaged site. Group A was divided into Group A1 (n=16) and Group A2 (n=16) again. Group A1 received electric stimulation 3 days after SCI, while Group A2 received electric stimulation 2 weeks after SCI. They were assessed with Basso-Beattie-Bresnahan (BBB) scores and routine MRI and diffusion tensor imaging (DTI). Results There was no significant difference in BBB score among all the groups at the first week (P>0.05). BBB scores of the Groups A1 and A2 were significantly different from the control group at the second and fourth week (P<0.001), and the Groups A1 from A2 (P<0.001). 2~4 hours and 3 days after modeling, there was no significant difference in the FA value among the groups (P>0.05). There was significant difference among the Groups A1, A2 and the control group at the fourth week (P<0.001). Group A1 was better than Group A2 (P<0.001). DTT images of all the groups were successful reconstructed, the Groups A1 and A2 repaired better than the control group. Conclusion The earlier electroacupuncture is taken after spinal cord injury, the better the outcome is.
3.Relationship between the expression of E-cadherin and ki-67 in hepatocellular carcinoma tissues and prognosis of the patients after hepatectomy

Objective: To study the relationships between the expressions of E-cadherin and ki-67 in the tissues of heatocellular carcinoma(HCC)and the prognosis of HCC patients after hepatectomy as well as their clinical pathology. Methods: We examine the expressions of E-cadherin and ki-67 in 255 HCC tissues by tissue microarray and PV-6000 two-step method of immunohistochemistry and analyze the correlations between their expressions and clinical pathological data, 1-year recurrent rate and overall survival time after hepatectomy. Results: The expression of E-cadherin correlated with the tumor size and the 1-year recurrent rate of positive group was higher than that of the negative group. The expression of ki-67 correlated with vascular invasion and differentiation of the tumor, the positive group showed a higher 1-year recurrent rate and a shorter overall survival time. Multivariate Cox regression analysis indicated that the expression of ki-67 was an independent risky factor. Conclusions: The negative expression of E-cadherin and the positive expression of ki-67 predict a higher recurrent rate of early stage. The expression of ki-67 is an independent risky factor which can be used to evaluate the prognosis of patients with HCC after hepatectomy.

5.Effects of hydroxyl fasudil on vascular relaxation and contraction activity

6.Hepatic T2 value in evaluation of HBV based acute-on-chronic liver failure

Objective To investigate the value of hepatic T2 value in evaluation of chronic HBV-related acute-on-chronic liver failure (HBV-ACLF).Methods The HBV-ACLF group,chronic hepatitis B group and control group who underwent liver MRI (M-GRASE sequence) were enrolled.The T2 map was produced from the post-processing software,and the mean T2 and R2 value of liver was calculated.The blood biochemical indexes from HBV-ACLF and chronic hepatitis B group were collected in 2 days pre-MR scaning.The differences of T2 and R2 values among 3 groups and the correlation between biochemical indexes and T2 value were analyzed.ROC curve was conducted to evaluate diagnostic efficiency of T2 value for HBV-ACLF.Results There were significant differences of T2value (x2 =19.074,P<0.001) or R2 value (F=10.411,P<0.001) among the 3 groups.The AUC of T2 value for diagnosing HBV-ACLF was 0.86 (P<0.001),with the cut-off value 57.73 ms (R2=0.017).Moderate positive correlation was shown between T2 values and international normalized ratio (INR),prothrombin time (PT),haluronicacid (HA) values (rs =0.65,0.67,0.39,all P<0.05),and moderate negative correlation was shown between T2 values and prothrombin activity (PTA),albumin (ALB),prealbumin (PA) values (rs =-0.67,-0.48,-0.37,all P<0.05).Conclusion T2 or R2 value could reflect the liver function,and were correlated with some biochemical indexes,which illustrated a good diagnostic efficiency for diagnostic of HBV-ACLF.
7.Effects of frozen thawed embryos versus frozen thawed blastocysts on transfer period and delivery outcomes

BACKGROUND:Since the first frozen embryo transplantation succeeded in 1983, embryo cryotechnique has been an important component in human assisted reproductive techniques. It is controversial which embryos and blastocysts after cryopreservation is selected.
OBJECTIVE:To compare the birth outcomes and neonatal status of frozen thawed embryos and blastocysts.
METHODS:In frozen thawed embryo group (n=1 273) and frozen thawed blastocyst group (n=471), we compared pregnancy rate, abortion rate, ectopic pregnancy rate, premature delivery rate, average premature gestational weeks, term yield, average ful-term gestational age, newborn sex, birth weight, birth defects and so on.
RESULTS AND CONCLUSION:There were frozen thawed blastocyst thaw cycles in 478 cases, 471 cases of transplantation period (including 7 cases without blastocyst transfer were canceled), 236 cases of pregnancy, 201 cases of delivery. Delivery number was 251, including 140 boys and 111 girls. The third day embryo thawing cycle of freezing and thawing appeared in 1 280 cases, 1 273 cases of transplantation period (including 7 cases of no embryo transplantation were canceled), 415 cases of pregnancy, 343 cases of delivery. The delivery number was 431, including 225 boys and 206 girls. Rate of pregnancy was significantly higher in frozen thawed blastocysts compared with frozen thawed embryos. No significant differences were detected in rate of miscarriage, ectopic pregnancy rate, premature delivery rate, average premature gestational weeks, term yield, average ful-termgestational age, newborn sex, and birth weight between frozen thawed embryos and blastocysts. There was no increase in birth defects between frozen thawed embryos and blastocysts. Results suggested that no significant difference was detected in birth outcomes and neonatal status of frozen thawed embryos and blastocysts. Nevertheless, pregnancy outcome is better in frozen thawed blastocysts than frozen thawed embryos.
8.Effect of three different fertilization modes of frozen-thawed embryos and blastocysts on pregnancy outcome

Objective To compare the effect of three different fertilization modes of frozen-thawed embryos and blastocysts on the pregnancy outcome after thawed transplantation .Methods 3 586 cycles of frozen embryos transplantation (FET ) treatment in our center from January 2012 to December 2013 were retrospectively analyzed and divided into 3 groups according to the fertilization modes .The group A adopted the conventional in vitro fertilization (IVF) of frozen-thawed embryo transfer in 2 008 cases and blas-tocysts transfer in 954 cases .The group B adopted intracytoplasmic sperm injection (ICSI) of frozen-thawed embryo transfer in 361 cases and blastocysts transfer in 129 cases ;the group C was given early remedy intracytoplasmic sperm injection (r-ICSI) of frozen-thawed embryo transfer in 87 cases and blastocysts transfer in 47 cases .The recovery rate of embryo and blastocyst ,pregnancy rate ,abortion rate ,ectopic pregnancy rate were compared for determining whether the differences among the indexes existed . Results The thawing recovery rate ,pregnancy rate ,abortion rate and ectopic pregnancy rate in the three different fertilization modes of embryo and blastocyst had no statistically significant differences (P>0 .05) .Conclusion Compared with conventional in vitro fertilization and intracytoplasmic sperm injection ,early r-ICSI is one of effective treatment means .

Objective The purpose of this study was to evaluate the practicability and efficiency of iFlow assistant function in lower extremity vascular disease angioplasty.Methods Retrospective analysis to 22 patients with lower extremity vascular disease in our single center from Septmber 2013 to November 2013, 19 males and 3 females with mean age(66.31 ± 7.46)years, iFlow technology was utilized to analyze the Region Of Interest(ROI) Peak Time(PeakTime) in pre-and post-angioplasty of lower extremity artery and assessed the state of vessel stenosis and blood flow perfusion, as well Doppler ultrasound in postoperative was used to double identify iFlow results.The results of vessel lesion, such as state of stenosis and occlusion were diagnosed by two senior doctors individually, as well as ROI Peak Time was recorded by technician.Results The iFlow technique performed that the ROI PeakTime in the region of interest was significantly decreased in postoperative in comparison to pre-angioplasty [(0.48 ± 0.39) s vs.(1.32 ± 0.60) s, t =8.836, P < 0.001].The Doppler ultrasound in postoperative showed that the ankle-brachial index was significantly increased compared within preoperative (0.97 ± 0.16 vs.0.49 ± 0.11, t =-10.205, P < 0.001).The results of iFlow technique in intraoperative were consistent with those of Doppler ultrasound in postoperative.Conclusion iFlow technique is effective to evaluate hemodynamic change of blood flow in vessel lesion in pre-and postangioplasty, which is assistive technique to assess the efficiency and prognosis in angioplasty.
