1.Effect of Ebosin on the inflammatory cytokines in THP-1 cells.

Ebosin is a novel exopolysaccharide produced by Streptomyces sp.139 with remarkable activity against rheumatic arthritis in vivo. In this paper, we reported effects of Ebosin on the inflammatory cytokines including interleukin-1beta (IL-1beta), interleukin-6 (IL-6) and tumor necrosis factor alpha (TNFalpha) in THP-1 cells. With the special fluorogenic peptide as substrates, the enzymatic activities of interleukin-1beta converting enzyme (ICE) and TNFalpha-converting enzyme (TACE) were inhibited by Ebosin separately. Using the real-time reverse transcription polymerase chain reaction (real-time PCR), the mRNA synthesis of the three cytokines were identified decline separately by Ebosin. The secretion quantum of three cytokines in THP-1 cells with Ebosin was lower than that of normal THP-1 cells determined by ELISA assay and Western blotting. All of these results showed that Ebosin has remarkably suppressed synthesis of the three cytokines in THP-1 cells through different pathways. The primary study of Ebosin on anti-inflammation mechanism was promoted developing the new drugs treating rheumatic arthritis.
2.ISOLATION AND CULTURE OF FETAL MURINE EPIDERMAL STEM CELLS IN VITRO AND rAAV_2/eGFP GENETIC TRANSFECTION

Objective To isolate,culture and identify epidermal stem cells(ESCs).The gene of enhanced green fluorescent protein(eGFP) was introduced via recombinant adeno-associated virus(rAAV) infection into the epidermal stem cells in vitro and the transfection efficiency under various tite of culture was determined. Methods 1.Getting fetal Wistar rats'dissociated single epidermal cells.2.Making laminin and CollagenⅣ the substitution of basal memrane,ESCs were isolated by adhering to murine laminin and CollagenⅣ.3.Epidermal stem cells were cultured in twenty-four-well plate,and cells number was controled 5?10~4 or so.After epidermal stem cells adhered to plate,diluted rAAV_2/eGFP virus fluid with serum-free medium.The diluted rAAV_2/eGFP virus fluid was added to the twenty-four-well plate according to the different MOI(viru gene/cells).The number of green fluorescence cells were counted under fluorescence microscope.The transfection efficiency was determined. Results 1.ESCs had better adhesive ability to laminin and higher colony formation efficiency(CFE) than that of keratinocytes.2.ESCs wer strongly positive with immunocytochemical staining of integrin ?1 and keration 19(K19).3.After rAAV_2/eGFP genetic transfection of ESCs,the positive cloning expressed highly eGFP gene along with time.Conclusion 1.These results suggest that rats'epidermal stem cells could been successfully isolated and enriched in vitro by means of rapid adherence to laminin and CollagenⅣ.After rAAV_2/eGFP genetic transfection of epidermal stem cells,the positive cloning expressed highly eGFP gene stably along with time.
3.AN ULTRASTRUCTURAL STUDY OF SMOOTH MUSCLE CELLS IN THE THECA EXTERNA OF OVARIAN FOLLIELES IN THE RAT

This investigation was made on 30 adult female rats weighing approximately 150~200g. during estrus as judged from vaginal smear examination. Ovaries which investigated with electron microscope, were fixed in 3% glutaraldehyde. Other ovaries were embeded in paraffin and examined under light microscope.Theca folliculi of large antral follicles were examined with electron microscope. Three kinds of theca cells were distinguished. They were secretory cell, fibroblast and smooth muscle cell. The secretory cell showed the characterastics of the steroidproducing cell, having smooth endoplasmic reticulum, mitochondria and lipid droplets. The fibroblast contained rough endoplasmic reticulum, free ribosomes and mitochondria in the cytoplasm. The smooth muscle cell was located in the outer layer of theca folliculi, and its structural features were typical according to the criteria of Somlyo and Somlyo: (1) pinocytotic vesicles of the cytoplasmic membranes; (2) myofilaments in the cytoplasm; (3) dense bodies intimately related to the myofilaments; and (4) location of mitochondria and endoplasmic reticulum at the nuclear poles.The ultrastructure of smooth muscle cell, its differentiation and other problems were discussed.
4.Study of a novel compound 2460A with activities produced by fungus.

With IL-6R as target, a new compound 2460A was identified from fungus using HTS screening model. The taxonomics of the produced strain was confirmed to be Trichoderma hazianum rifai after sequencing analysis of rDNA-ITS (internal transcribed spacer). Results showed that this compound has a binding activity on IL-6R competed with IL-6, thus it is a new ligand of IL-6R originating from microbe. With MTT assay, the anti-tumor activities of 2460A were demonstrated on CM126 and HT-29 cell lines separately, the IC50 are 2.17 x 10(-5) mol x L(-1) and 1.8 x 10(-5) mol x L(-1) respectively. The compound affected lightly the HT-29 cell cycle at S phase. Studies for the anti-tumor activity of 2460A in vivo are in progress in our lab.
5.Meta-analysis of effects of dietary intervention on health outcome in patients with polycystic ovary syndrome

Objective:To analyze effects of dietary intervention on health outcome in patients with polycystic ovary syndrome.Methods:PubMed, Cochrane Library, Web of Science, Embase, China Biology Medicine disc, CNKI and WanFang Data were searched by the computer, and clinical RCTs about dietary intervention in patients with polycystic ovary syndrome were included. The retrieval time was up to August 2020. Two trained researchers independently screened the literature, extracted data and evaluated the quality. Meta-analysis was performed using RevMan 5.3 software.Results:A total of 9 clinical RCTs were included, including 697 patients with polycystic ovary syndrome. The results of Meta-analysis showed that compared with the control group, dietary intervention could reduce the body weight ( MD=-2.22, 95% CI -2.75--1.68, P<0.001) , body mass index ( MD=-1.05, 95% CI -1.24--0.87, P<0.001) , waist circumference ( MD=-2.35, 95% CI -4.17--0.53, P=0.01) , fasting blood glucose ( MD=-0.12, 95% CI -0.18--0.05, P<0.001) , fasting insulin ( MD=-4.77, 95% CI -5.59--3.96, P<0.001) , insulin resistance index assessed by steady-state model ( MD=-0.93, 95% CI -1.29--0.57, P<0.001) , luteinizing hormone ( MD=-0.46, 95% CI -0.90--0.03, P=0.040) , follicle-stimulating hormone ( MD=-0.16, 95% CI -0.31--0.01, P=0.030) , total testosterone ( MD=-0.33, 95% CI -0.53--0.13, P<0.001) , increase sex hormone-binding globulin ( MD=8.02, 95% CI 5.55-10.49, P<0.001) and pregnancy rate ( RR=18.38, 95% CI 4.47-75.55, P<0.001) of patients with polycystic ovary syndrome. Conclusions:Dietary intervention can improve the pregnancy rate of patients with polycystic ovary syndrome, improve the endocrine and metabolic status and reduce the level of insulin resistance and human measurable indicators.
