2.Comparison of single factor design and response surface design to optimize the reaction system for randomly amplified polymorphic DNA

Objective To obtain optimum scheme on reaction system for randomly amplified polymorphic DNA(RAPD) of Pseudomonas aeruginosa. Methods The optimum reaction systems of RAPD were obtained by single factor design and response surface design(RSD). Results The multiple optimum reaction systems caused by effect of interaction on multifactor were observed in single factor design, and the concentrations of key components of the optimum reaction system with good stability obtained by response surface design were 2.0 mmol/L of Mg 2+ , 0.5 ?mol/L of primer and 0.5 ng/?l of template. Conclusion To optimize the reaction system of RAPD, RSD is simpler, more scientific, and more reasonable than single factor design.
3.Relationship between body mass index and blood pressure, blood glucose, lipid profiles and blood uric acid in health check-up populations

Objective To evaluate the relationship between body mass index (BMI) and blood pressure,blood glucose,lipid profiles and blood uric acid in healthy populations in Hefei of Anhui province.Methods A total of 3 640 healthy adults were enrolled to analyze the relationship between BMI and blood pressure and serum levels of lipid profiles,uric acid and glucose.Results The mean BMI of the subjects was (25.5±2.8) kg/m2 (men vs women:(26.3±2.5) vs (23.9±2.8) kg/m2).BMI showed a significant difference in different age groups (P<0.05).Different BMI subgroup had statistically significant difference in blood pressure,glucose,lipid profiles and uric acid.In different gender and age groups,BMI,blood pressure,glucose,blood lipid and uric acid were found to be significantly different.Conclusion The prevalence of obesity and overweight is high in our health check-up populations.It should be important to prevent and control hypertension,high blood glucose,high blood uric acid and other chronic diseases.
4.Evolution of prevalence and risk factors of non-alcoholic fatty liver disease in the elderly:an epide-miologic survey

Objective This epidemiologic survey aims to investigate the changes in prevalence and risk factors for non-alcoholic fatty liver disease (NAFLD)in the elderly people who are living within Pudong San-lin region.Methods We retrospectively analyzed annual physical check-up data in 2012 and 2014 for elderly people living in this region (aged≥70years).Demographic variables including age,sex,blood pressure,body mass index (BMI)were recorded.Serum fasting glucose level was determined and liver and kidney function were assessed.Abdominal echo-Doppler examination was performed.NAFLD was diagnosed according to Chinese guideline on management of NAFLD (revised version).Results Among 2127 sub-jects with physical check-up in 2012,the occurrence rate of NAFLD was 27.46% (23.02% and 31.02%for men and women,respectively).Type 2 diabetes was detected in 224 subjects and 33.93% of them de-veloped NAFLD (26.73% and 39.84% for males and females,respectively).Among 1771 subjects who received physical check-up in 2014,the occurrence rate of NAFLD increased up to 32.29%,with NAFLD rate being 29.97% and 34.19% for men and women,respectively.In 185 subjects with type 2 diabetes, 49.19% developed NAFLD (47.56% and 50.49% for men and women,respectively).Thus,compared with the findings during physical check-up in 2012,the occurrence rate of NAFLD was significantly in-creased during physical check-up in 2014 for both male and female subjects of overall and diabetic popula-tions (P <0.05 for all comparisons).Further analysis showed that for 185 diabetic patients who had phys-ical check-up in 2014,systolic and diastolic blood pressures,BMI,and serum levels of fasting glucose lev-el,alanine aminotransferase,aspartate aminotransferase,total cholesterol,and triglyceride were significant-ly higher in patients with NAFLD than in those without NAFLD (all P <0.05).Multivariate regression a-nalysis revealed that BMI,total cholesterol and triglyceride levels were independent risk factors for occur-rence of NAFLD in diabetic patients.Conclusions This epidemiologic survey demonstrates a significant increase in the occurrence rate of NAFLD for the elderly people (especially with type 2 diabetes).Hyper-lipidemia,obesity,and hypertension are major risk factors for NAFLD in diabetic patients.
5.Ultrasound-targeted microbubble destruction combined with polyethylenimine mediated Bax gene silence in H9c2 cells

Objective To silence Bax gene in H9c2 cells by ultrasound-targeted microbubble destruction(UTMD) combined with polyethylenimine.Methods The shRNA expression plasmid targeting Bax gene was co-treated with microbubbles and polyethylenimine.Then the mixture added to H9c2 cells were irradiated by ultrasound.Grouping:① plasmid group;② Lipofectamine2000 + plasmid group; ③ UTMD + plasmid group;④ PEI + UTMD + plasmid group;⑤ PEI + SonoVue + plasmid group (without ultrasound irradiation).The transfection efficiency was assessed by fluorescence microscope and FCM.The expression of Bax mRNA and protein were detected by RT-PCR and Western Blot.Results Bax shRNA wasn't destroyed by ultrasound exposure.The transfection efficiency of PEI + UTMD + plasmid group was higher than any other groups obviously (P <0.05).The result of RT-PCR and Western Blot showed inhibition efficiency of Bax mRNA and protein expression in PEI + UTMD + plasmid group were (69.41 ± 4.91) % and (64.09 ± 2.38)% separately,which were higher than any other groups significantly (P <0.05).Conclusions UTMD combined with polyethylenimine can inhibit the expression of Bax gene in H9c2 cells effectively.
6.Changes in serum levels of Th22 cell-related cytokines and complements in patients with drug eruption before and after treatment

Objective To investigate changes in serum levels of Th22 cell ? related cytokines and complements in patients with drug eruption before and after treatment, and to explore their possible roles in the occurrence and development of drug eruption. Methods This study included 35 patients with drug eruption, and 35 sex?and age?matched healthy controls. Five milliliters of peripheral blood samples were collected from the controls and patients before and after treatment. Enzyme?linked immunosorbent assay(ELISA)was performed to measure serum levels of interleukin 22(IL?22)and IL?13, and the cytometric bead array(CBA)system was used to determine serum levels of tumor necrosis factor?α(TNF?α) and complement components C3a, C4a and C5a. Results Before treatment, the patients with drug eruption showed significantly higher serum levels of IL?22(40.85 ± 14.56 vs. 29.09 ± 8.66 ng/L, t=5.549, P<0.05), IL?13(869.94 ± 463.39 vs. 372.92 ± 151.75 ng/L, t=6.071, P<0.05), TNF?α(1.03 ± 0.64 vs. 0.44 ± 0.31 ng/L, t=4.321, P<0.05), complement C3a(55.21 ± 32.98 vs. 42.44 ± 14.26 ng/L, t=2.832, P<0.05), C4a(285.11 ± 123.91 vs. 237.00 ± 63.57 ng/L, t=2.257, P<0.05), and C5a(279.68 ± 127.72 vs. 215.98 ± 65.38 ng/L, t=2.495, P<0.05)compared with the controls. After treatment, the serum levels of IL?22, IL?13, TNF?α, complement C3a, C4a and C5a in patients decreased to(32.72 ± 11.77)ng/L,(456.21 ± 123.22)ng/L,(0.64 ± 0.39)ng/L,(45.47 ± 21.11)ng/L,(241.86 ± 84.12)ng/L and(239.61 ± 103.51)ng/L respectively, with a significant difference between the pretreatment and posttreatment values of these proteins(t = 4.443, 5.197, 3.572, 3.213, 2.728 and 4.772, respectively, all P ≤ 0.01). Additionally, the serum levels of IL?22 and IL?13 were still significantly higher in the patients than in the controls(both P < 0.05), while there were no significant differences in the serum levels of TNF?α, complement C3a, C4a or C5a between the patients and controls after treatment(all P > 0.05). Correlation analysis showed positive correlations between complement C3a and C4a serum levels(r = 0.660, P < 0.05), between C3a and C5a serum levels(r = 0.404, P < 0.05), between C4a and C5a serum levels(r = 0.501, P < 0.05), and between IL ? 22 and TNF ? α serum levels(r = 0.573, P = 0.005), but negative correlations between IL ? 22 and complement C3a serum levels(r = -0.490, P = 0.005), in patients before treatment. Conclusion The activation of Th22 cell?related cytokines and complements may play important roles in the occurrence and development of drug eruption, and IL?22 may participate in the regulation of complements.
